Project description:Background: Cognitive impairment (CI) is an early and frequent symptom of multiple sclerosis (MS). Likewise, affective symptoms (e.g., depression and anxiety) and alterations in the processing of emotional stimuli have been frequently reported. Thus, abilities that integrate affective and cognitive processes such as decision making (DM) based on affective feedback are potentially valuable early diagnostic markers for MS. The available research on this topic, however, is still inconclusive and suffers from methodological issues. Methods: We compared DM ability in a clinically homogeneous cohort of 24 patients with early relapsing-remitting MS (RRMS) and 59 age-matched healthy controls (HCs). A modified version of the Iowa gambling task (IGT) allowed us to control for individual differences in search strategies during the risk exploration phase. Besides standard IGT measures (netscore, obtained play money, and learning index), we also examined reaction times and post-error slowing (PES) patterns as a proxy for abnormalities in the processing of affective feedback. Results: The performance of patients did not significantly deviate from HCs in any standard parameter of the modified IGT. Furthermore, although RRMS patients reacted significantly slower than HCs overall, we found similar patterns of PES in both groups, suggesting similarly efficient processing of affective feedback. Conclusion: We conclude that there is no specific deficit in affective feedback processing in early RRMS. Previous findings of IGT impairments in this patient group may thus not represent a genuine deficit in affective DM but rather be related to sample characteristics, general CI, and/or differences in individual search strategies. Future research should explore the potential influence of lesion volumes and locations on DM ability by employing brain imaging techniques.

Project description:In modeling the human affective system and applying lessons learned to human-robot interaction, the challenge is to handle ambiguous emotional states of an agency (whether human or artificial), probabilistic decisions, and freedom of choice in affective and behavioral patterns. Moreover, many cognitive processes seem to run in parallel whereas seriality is the standard in conventional computation. Representation of contextual aspects of behavior and processes and of self-directed neuroplasticity are still wanted and so we attempt a quantum-computational construction of robot affect, which theoretically should be able to account for indefinite and ambiguous states as well as parallelism. Our Quantum Coppélia (Q-Coppélia) is a translation into quantum logics of the fuzzy-based Silicon Coppélia system, which simulates the progression of a robot's attitude towards its user. We show the entire circuitry of the Q-Coppélia framework, aiming at contemporary descriptions of (neuro)psychological processes. Arguably, our work provides a system for simulating and handling affective interactions among various agencies from an understanding of the relations between quantum algorithms and the fundamental nature of psychology.

Project description:Emotional experience has a pervasive impact on choice behavior, yet the underlying mechanism remains unclear. Introducing facial-expression primes into a probabilistic learning task, we investigated how affective arousal regulates reward-related choice based on behavioral, model fitting, and feedback-related negativity (FRN) data. Sixty-six paid subjects were randomly assigned to the Neutral-Neutral (NN), Angry-Neutral (AN), and Happy-Neutral (HN) groups. A total of 960 trials were conducted. Subjects in each group were randomly exposed to half trials of the pre-determined emotional faces and another half of the neutral faces before choosing between two cards drawn from two decks with different assigned reward probabilities. Trial-by-trial data were fit with a standard reinforcement learning model using the Bayesian estimation approach. The temporal dynamics of brain activity were simultaneously recorded and analyzed using event-related potentials. Our analyses revealed that subjects in the NN group gained more reward values than those in the other two groups; they also exhibited comparatively differential estimated model-parameter values for reward prediction errors. Computing the difference wave of FRNs in reward vs. non-reward trials, we found that, compared to the NN group, subjects in the AN and HN groups had larger "General" FRNs (i.e., FRNs in no-reward trials minus FRNs in reward trials) and "Expected" FRNs (i.e., FRNs in expected reward-omission trials minus FRNs in expected reward-delivery trials), indicating an interruption in predicting reward. Further, both AN and HN groups appeared to be more sensitive to negative outcomes than the NN group. Collectively, our study suggests that affective arousal negatively regulates reward-related choice, probably through overweighting with negative feedback.

Project description:Executive function is thought to be the coordinated operation of multiple neural processes and allows to accomplish a current goal flexibly. The most important function of the prefrontal cortex is the executive function. Among a variety of executive functions in which the prefrontal cortex participates, decision-making is one of the most important. Although the prefrontal contribution to decision-making has been examined using a variety of behavioral tasks, recent studies using fMRI have shown that the prefrontal cortex participates in decision-making under free-choice conditions. Since decision-making under free-choice conditions represents the very first stage for any kind of decision-making process, it is important that we understand its neural mechanism. Although few studies have examined this issue while a monkey performed a free-choice task, those studies showed that, when the monkey made a decision to subsequently choose one particular option, prefrontal neurons showing selectivity to that option exhibited transient activation just before presentation of the imperative cue. Further studies have suggested that this transient increase is caused by the irregular fluctuation of spontaneous firing just before cue presentation, which enhances the response to the cue and biases the strength of the neuron's selectivity to the option. In addition, this biasing effect was observed only in neurons that exhibited sustained delay-period activity, indicating that this biasing effect not only influences the animal's decision for an upcoming choice, but also is linked to working memory mechanisms in the prefrontal cortex.

Project description:During the last decades, research on binary decision making elucidated some of the basic neural mechanisms underlying the decision-making process. Recently, the focus of experimental as well as modeling studies began to shift from simple binary choices to decision making with multiple alternatives. In this article, we address the question how different numbers of choice alternatives might be handled and encoded in the brain. We present a minimal, biophysically realistic spiking neuron model for decision making with multiple alternatives. Our model accounts for the relevant aspects of recent experimental data of a random-dot motion-discrimination task on both the cellular and behavioral level. Notably, all network parameters and inputs in our network are independent of the number of possible alternatives used in the tested experimental paradigms (2 and 4 alternatives and 2 alternatives with an angular separation of 90 degrees ). This avoids the use of extra top-down regulation mechanisms to adapt the network to the number of choices. Furthermore, we show that increasing the number of neurons encoding each choice alternative is positively related to the network's capacity of choice-number-independent decision making. Consequently, our results suggest a physiological advantage of a pooled, multineuron representation of choice alternatives.

Project description:The response behaviors in many two-alternative choice tasks are well described by so-called sequential sampling models. In these models, the evidence for each one of the two alternatives accumulates over time until it reaches a threshold, at which point a response is made. At the neurophysiological level, single neuron data recorded while monkeys are engaged in two-alternative choice tasks are well described by winner-take-all network models in which the two choices are represented in the firing rates of separate populations of neurons. Here, we show that such nonlinear network models can generally be reduced to a one-dimensional nonlinear diffusion equation, which bears functional resemblance to standard sequential sampling models of behavior. This reduction gives the functional dependence of performance and reaction-times on external inputs in the original system, irrespective of the system details. What is more, the nonlinear diffusion equation can provide excellent fits to behavioral data from two-choice decision making tasks by varying these external inputs. This suggests that changes in behavior under various experimental conditions, e.g. changes in stimulus coherence or response deadline, are driven by internal modulation of afferent inputs to putative decision making circuits in the brain. For certain model systems one can analytically derive the nonlinear diffusion equation, thereby mapping the original system parameters onto the diffusion equation coefficients. Here, we illustrate this with three model systems including coupled rate equations and a network of spiking neurons.

Project description:Impulse Control Disorder (ICD) in Parkinson's disease is a behavioral addiction induced by dopaminergic therapies, but otherwise unclear etiology. The current study investigates the interaction of reward processing variables, dopaminergic therapy, and risky decision-making and subjective feelings in patients with versus without ICD. Patients with (n = 18) and without (n = 12) ICD performed a risky decision-making task both 'on' and 'off' standard-of-care dopaminergic therapies (the task was performed on 2 different days with the order of on and off visits randomized for each patient). During each trial of the task, participants choose between two options, a gamble or a certain reward, and reported how they felt about decision outcomes. Subjective feelings of 'pleasure' are differentially driven by expectations of possible outcomes in patients with, versus without ICD. While off medication, the influence of expectations about risky-decisions on subjective feelings is reduced in patients with ICD versus without ICD. While on medication, the influence of expected outcomes in patients with ICD versus without ICD becomes similar. Computational modeling of behavior supports the idea that latent decision-making factors drive subjective feelings in patients with Parkinson's disease and that ICD status is associated with a change in the relationship between factors associated with risky behavior and subjective feelings about the experienced outcomes. Our results also suggest that dopaminergic medications modulate the impact expectations have on the participants' subjective reports. Altogether our results suggest that expectations about risky decisions may be decoupled from subjective feelings in patients with ICD, and that dopaminergic medications may reengage these circuits and increase emotional reactivity in patients with ICD.

Project description:BackgroundVaccine hesitancy is a significant threat to global health. A key part of addressing hesitancy is to ensure that public health messaging prioritises information that is considered important to the public. This study aimed to examine how different vaccine characteristics affect public preferences for vaccines in New Zealand, what trade-offs they are willing to make between different vaccine characteristics, and how their preferences are affected by their vaccine-related conspiracy beliefs and COVID-19 vaccination status.MethodsAn online discrete choice experiment (DCE) was designed to elicit individual preferences about vaccines using the 1000minds platform. Members of the general population of New Zealand aged ≥ 18 years were invited to complete the DCE. Participants were asked to indicate their preference between two options showing different combinations of vaccine characteristics. Data on sociodemographic characteristics were collected. Beliefs were measured using the vaccine conspiracy beliefs scale (VCBS) with scores ≥ 19 indicating strong vaccine-related conspiracy beliefs. The DCE was analysed using the PAPRIKA method (Potentially All Pairwise RanKings of all possible Alternatives) and preferences compared between respondents with high versus low VCBS scores and vaccinated versus unvaccinated respondents for COVID-19.ResultsA total of 611 respondents from 15 regions completed the DCE. Mean (SD) age was 45.9 (14.7) years with most having had 2 or more doses of the coronavirus vaccine (86%). Mean (SD) VCBS score was 18.5 (12.4) indicating moderate vaccine-related conspiracy beliefs. Risk of severe adverse effects was the most highly valued vaccine characteristic, followed by vaccine effectiveness and duration of protection. Vaccine origin and route of administration were ranked least important. Respondents scoring high on the VCBS placed less value on the effectiveness of vaccines but greater value on development time and total number of doses (p < 0.001). COVID-19 unvaccinated respondents ranked development time and total number of doses more highly than those vaccinated respondents (p < 0.001).ConclusionsRisk of severe adverse effects, vaccine effectiveness and duration of protection were rated by the New Zealand public as the top three most important vaccine characteristics. This information is important for informing public health messaging to promote vaccine uptake and inform vaccine decision-making.

Project description:The contribution of sensory and decisional processes to perceptual decision making is still unclear, even in simple perceptual tasks. When decision makers need to select an action from a set of balanced alternatives, any tendency to choose one alternative more often-choice bias-is consistent with a bias in the sensory evidence, but also with a preference to select that alternative independently of the sensory evidence. To decouple sensory from decisional biases, here we asked humans to perform a simple perceptual discrimination task with two symmetric alternatives under two different task instructions. The instructions varied the response mapping between perception and the category of the alternatives. We found that from 32 participants, 30 exhibited sensory biases and 15 decisional biases. The decisional biases were consistent with a criterion change in a simple signal detection theory model. Perceptual decision making, thus, even in simple scenarios, is affected by sensory and decisional choice biases.

Project description:Decision making with several choice options is central to cognition. To elucidate the neural mechanisms of such decisions, we investigated a recurrent cortical circuit model in which fluctuating spiking neural dynamics underlie trial-by-trial stochastic decisions. The model encodes a continuous analog stimulus feature and is thus applicable to multiple-choice decisions. Importantly, the continuous network captures similarity between alternatives and possible overlaps in their neural representation. Model simulations accounted for behavioral as well as single-unit neurophysiological data from a recent monkey experiment and revealed testable predictions about the patterns of error rate as a function of the similarity between the correct and actual choices. We also found that the similarity and number of options affect speed and accuracy of responses. A mechanism is proposed for flexible control of speed-accuracy tradeoff, based on a simple top-down signal to the decision circuit that may vary nonmonotonically with the number of choice alternatives.
