First report of association of potato virus X and potato virus Y and 'Candidatus Phytoplasma trifolii' in brinjal in India.

ABSTRACT: Symptoms of little leaf, leaf chlorosis and leaf malformations with mosaic mottling symptoms were observed in two brinjal varieties (Pusa Shyamla and Pusa Purple Cluster) in fields of IARI, New Delhi, India during 2014-2015. Electron microscopy, PCR and sequence analysis first time provided evidence of association of Candidatus Phytoplasma trifolii with potato virus X and potato virus Y in brinjal in India.

First report of '<i>Candidatus</i> Phytoplasma asteris' associated with yellowing, scorching and decline of almond trees in India.

Project description:The almond, a commercially important tree nut crop worldwide, is native to the Mediterranean region. Stone fruit trees are affected by at least 14 'Candidatus Phytoplasma' species globally, among which 'Candidatus Phytoplasma asteris' is one of the most widespread phytoplasma infecting Prunus dulcis, causing aster yellows disease. Recently, almond plantations of Nauni region were consistently affected by phytoplasma, as evidenced by visible symptoms, fluorescent microscopic studies and molecular characterization. During several surveys from May to September 2020-2022, almond aster yellows phytoplasma disease showing symptoms such as chlorosis, inward rolling, reddening, scorching and decline with an incidence as high as 40%. Leaf samples were collected from symptomatic almond trees and the presence of phytoplasma was confirmed through fluorescent microscopic studies by employing DAPI (4, 6-diamino-2-phenylindole) that showed distinctive light blue flourescent phytoplasma bodies in phloem sieve tube elements. The presence of phytoplasma in symptomatic almond trees was further confirmed using nested PCR with specific primer pairs followed by amplification of 16S rDNA and 16S-23S rDNA intergenic spacer (IS) fragments. Sequencing and BLAST analysis of expected amplicon of the 16S rDNA gene confirmed that the almond phytoplasma in Himachal Pradesh was identical to the aster yellows group phytoplasma. Phylogenetic analysis of 16S rDNA almond phytoplasma also grouped 'Prunus dulcis' aster yellows phytoplasma within 16SrI-B subgroup showed 94% nucleotide identity with 'Prunus dulcis' phytoplasma PAEs3 and 'Prunus dulcis' phytoplasma PAE28 from Iran. This research presents the first host report of 'Candidatus Phytoplasma asteris' infecting almonds in India, expanding the knowledge of the diversity and distribution of phytoplasma strains affecting almond trees globally.

Multilocus gene-specific characterization of 'Candidatus Phytoplasma australasia' associated with shoot proliferation disease of small cardamom in India.

Project description:Symptoms of excessive shoot proliferation were observed in the Njallani cultivar of small cardamom accompanied by stunting of stalks with fewer degenerated capsules at Nedumkandam Panchayat of Idukki district of Kerala in 2017. Five symptomatic Elettaria cardamomum shoot proliferation (ECSP) plant samples were collected and processed for DNA extraction and PCR assays utilizing universal phytoplasma 16S ribosomal-specific primers pair, P1/P7 followed by R16F2n/R16R2. Sequence comparison analysis of the R16F2n/R16R2 region of 16SrRNA gene showed 100% sequence identity with the 'Candidatus Phytoplasma australasia'- related strain. Phylogeny and virtual RFLP analyses of 16S rRNA gene sequences confirmed the association of 'Ca. P. australasia' strain subgroup D with ECSP disease. The association of 16SrII group was further established and validated by amplifying phytoplasma-specific multilocus candidate genes by utilizing specific primers of secA, secY, SAP11, and tuf genes. The multilocus gene sequence comparison analysis again confirmed the association of 'Ca. P. australasia' with the ECSP phytoplasma isolate. This is the first report of phytoplasma association with small cardamom.

First Report of Tobacco streak virus Infecting Kenaf (Hibiscus cannabinus) in India.

Project description:The natural occurrence of Tobacco streak virus (TSV) in Hibiscus cannabinus was detected by enzyme-linked immunosorbent assay using an antiserum raised against TSV and reverse transcription polymerase chain reaction (PCR) using primers specific for the coat protein gene of the virus. Sequence analysis of the PCR products showed 99.6 and 99.5% of maximum identity at nucleotide and amino acid levels, respectively with TSV onion isolate from Kurnool (HM131490).This is the first report of the natural occurrence of TSV on kenaf in India.

First report of apple stem grooving virus infection in loquat from India.

Project description:Loquat, commonly known as Eriobotrya japonica, is a major subtropical fruit from the Rosaceae family that is native to China but also found in most of Europe and Asia, including India. Apple stem grooving virus (ASGV) infecting loquat was detected using leaf samples collected from Himachal Pradesh (India) through DAC-ELISA followed by RT-PCR assays targeting coat protein (CP), movement protein (MP) and replicase (Rep) regions of ASGV genome. Sequencing of RT-PCR amplicons and sequence analyses revealed that CP, MP and Rep sequences of ASGV loquat Indian isolate of the current study shared a maximum of 98-100% nucleotide sequence identities with the corresponding sequences of available ASGV Indian isolates [LN559078, HE978837, MZ127820, MN912568]. Phylogenetic tree based on each sequenced gene confirmed the genetic diversity of ASGV. To the best of our knowledge, and based on review of the literature, this is the first report of ASGV infection in loquat from India.

First report of lily mottle virus on lily (Lilium sp.) in southern India.

Project description:Lily samples collected from commercial cut flower growing field at Nilgiris, Tamil Nadu, India were serologically tested through direct antigen coating-ELISA and dot immuno binding assay for the infection of potyvirus through potyvirus group specific polyclonal antibody. The RT-PCR using potyvirus universal degenerate primers (PNIbF1/PCPR1) showed the presence of a potyvirus and the sequencing results showed the association of lily mottle virus (LMoV). Further LMoV was confirmed by RT-PCR amplification using newly designed primer pair covering entire coat protein gene and sequence results showed 100% nucleotide identity with the LMoV reported from South Korean. To our knowledge, this is the first confirmed evidence for the occurrence of LMoV on lily in southern India.

First report of cucumber mosaic virus infecting antamul vine (Tylophora indica) in India.

Project description:Tylophora indica (Burm f.) Merrill; commonly known as antamul, is an important medicinal herb. Typical yellow rings or irregular yellow spot and in severe condition necrotic rings were observed on the leaves of the crop. The examination of symptomatic leaf samples under transmission electron microscopy revealed the presence of spherical virions confirmed the association of cucumo like virus group. A novel degenerate primers pair was designed by multiple sequence alignment of RdRP region and used in RT-PCR to amplify a ~?410 bp genomic fragment. The sequence of the amplified fragment shared 97-98% sequence identity with cucumber mosaic virus (CMV). This study is the first report of the association of CMV with the yellow ring symptom of antamul in India.

Case report: first symptomatic Candidatus Neoehrlichia mikurensis infection in Slovenia.

Project description:BackgroundCandidatus Neoehrlichia mikurensis (CNM) is an emerging tick-born pathogen and usually causes symptomatic infection only in immunocompromised patients. Apart from one described case found in the literature where cultivation was successful, all cases so far were diagnosed by using broad-range 16S rDNA PCR.Case presentationOur patient presented with a prolonged febrile state of unknown origin. Clinical presentation, extensive medical workup and classic microbiologic testing were non-conclusive. Several infectious agents and other causes for the febrile state were excluded. In the end, a broad-range 16S rDNA PCR was to be performed to confirm the diagnosis of CNM infection. Treatment was successful with doxycycline.ConclusionsDue to the obscurity of the pathogen, diagnostic workup in CNM is prolonged and challenging. More awareness is need about this emerging infectious disease in countries with high prevalence of tick-borne diseases as standard microbiological methods are not successful in confirming the diagnosis.
