Body fat mobilization in early lactation influences methane production of dairy cows.

ABSTRACT: Long-chain fatty acids mobilized during early lactation of dairy cows are increasingly used as energy substrate at the expense of acetate. As the synthesis of acetate in the rumen is closely linked to methane (CH4) production, we hypothesized that decreased acetate utilization would result in lower ruminal acetate levels and thus CH4 production. Twenty heifers were sampled for blood, rumen fluid and milk, and CH4 production was measured in respiration chambers in week -4, +5, +13 and +42 relative to first parturition. Based on plasma non-esterified fatty acid (NEFA) concentration determined in week +5, animals were grouped to the ten highest (HM; NEFA > 580 μmol) and ten lowest (LM; NEFA < 580 μmol) mobilizing cows. Dry matter intake (DMI), milk yield and ruminal short-chain fatty acids did not differ between groups, but CH4/DMI was lower in HM cows in week +5. There was a negative regression between plasma NEFA and plasma acetate, between plasma NEFA and CH4/DMI and between plasma cholecystokinin and CH4/DMI in week +5. Our data show for the first time that fat mobilization of the host in early lactation is inversely related with ruminal CH4 production and that this effect is not attributed to different DMI.

Project description:Rumen-protected glucose (RPG) plays an important role in alleviating the negative energy balance of dairy cows. This study used a combination of rumen microbes 16S and metabolomics to elucidate the changes of rumen microbial composition and rumen metabolites of different doses of RPG's rumen degradation part in early-lactation dairy cows. Twenty-four multiparous Holstein cows in early lactation were randomly allocated to control (CON), low-RPG (LRPG), medium-RPG (MRPG), or high-RPG (HRPG) groups in a randomized block design. The cows were fed a basal total mixed ration diet with 0, 200, 350, and 500 g of RPG per cow per day, respectively. Rumen fluid samples were analyzed using Illumina MiSeq sequencing and ultrahigh-performance liquid chromatography coupled to quadrupole time-of-flight mass spectrometry. MRPG supplementation increased bacterial richness and diversity, including increasing the relative abundance of cellulolytic bacteria, such as Ruminococcus, Lachnospiraceae_NK3A20_group, Ruminiclostridium, and Lachnospiraceae_UCG-008 MRPG significantly increased the concentrations of acetate, propionate, butyrate, and total volatile fatty acid in the rumen. Ruminal fluid metabolomics analysis showed that RPG supplementation could significantly regulate the synthesis of amino acids digested by protozoa in the rumen. Correlation analysis of the ruminal microbiome and metabolome revealed some potential relationships between major bacterial abundance and metabolite concentrations. Our analysis found that RPG supplementation of different doses can change the diversity of microorganisms in the rumen and affect the rumen fermentation pattern and microbial metabolism and that a daily supplement of 350 g of RPG might be the ideal dose.IMPORTANCE Dairy cows in early lactation are prone to a negative energy balance because their dry matter intake cannot meet the energy requirements of lactation. Rumen-protected glucose is used as an effective feed additive to alleviate the negative energy balance of dairy cows in early lactation. However, one thing that is overlooked is that people often think that rumen-protected glucose is not degraded in the rumen, thus ignoring its impact on the microorganisms in the rumen environment. Our investigation and previous experiments have found that rumen-protected glucose is partially degraded in the rumen. However, there are few reports on this subject. Therefore, we conducted research on this problem and found that rumen-protected glucose supplementation at 350 g/day can promote the development and metabolism of rumen flora. This provides a theoretical basis for the extensive application of rumen bypass glucose at a later stage.

Project description:Simple Summary Within anthropogenic sources, agriculture contributes the most to greenhouse gas (GHG) emissions. The on-field assessment of methane emissions from livestock is crucial for testing and implementing mitigation strategies to reduce the deleterious effects of GHG on climate. Therefore, this article presents a longitudinal study in which measurements of enteric methane emissions from dairy cows (Jersey and Holstein-Freisian raised under a semi-extensive management system) were performed with a tool previously described as reliable, portable, and cost-effective: the laser methane detector (LMD). The results showed breed-related differences in methane emissions when milk yield was not considered, challenging us to rethink milk traits and breeding choices. Moreover, methane emissions were lower in cows in early lactation as well as in primiparous, likely reflecting concentrate supplementation and energy metabolism. The study pointed out the opportunity to design and test regional tailored mitigation strategies, including nutritional management, for higher methane emission periods within cows’ lactation cycle, while ensuring the enhancement of productivity. Abstract Reversing climate change requires broad, cohesive, and strategic plans for the mitigation of greenhouse gas emissions from animal farming. The implementation and evaluation of such plans demand accurate and accessible methods for monitoring on-field CH4 concentration in eructating breath. Therefore, this paper describes a longitudinal study over six months, aiming to test a protocol using a laser methane detector (LMD) to monitor CH4 emissions in semi-extensive dairy farm systems. Over 10 time points, CH4 measurements were performed in dry (late gestation) and lactating cows at an Azorean dairy farm. Methane traits including CH4 concentration related to eructation (E_CH4) and respiration (R_CH4), and eructation events, were automatically computed from CH4 measured values using algorithms created for peak detection and analysis. Daily CH4 emission was estimated from each profile’s mean CH4 concentration (MEAN_CH4). Data were analyzed using a linear mixed model, including breed, lactation stage, and parity as fixed effects, and cow (subject) and time point as random effects. The results showed that Holsteins had higher E_CH4 than Jersey cows (p < 0.001). Although a breed-related trend was found in daily CH4 emission (p = 0.060), it was not significant when normalized to daily milk yield (p > 0.05). Methane emissions were lower in dry than in lactation cows (p < 0.05) and increased with the advancement of the lactation, even when normalizing it to daily milk yield (p < 0.05). Primiparous cows had lower daily CH4 emissions related to R_ CH4 compared to multiparous (p < 0.001). This allowed the identification of periods of higher CH4 emissions within the milk production cycle of dairy cows, and thus, the opportunity to tailor mitigation strategies accordingly.

Project description:Methane (CH4) is produced as an end product from feed fermentation in the rumen. Yield of CH4 varies between individuals despite identical feeding conditions. To get a better understanding of factors behind the individual variation, 73 dairy cows given the same feed but differing in CH4 emissions were investigated with focus on fiber digestion, fermentation end products and bacterial and archaeal composition. In total 21 cows (12 Holstein, 9 Swedish Red) identified as persistent low, medium or high CH4 emitters over a 3 month period were furthermore chosen for analysis of microbial community structure in rumen fluid. This was assessed by sequencing the V4 region of 16S rRNA gene and by quantitative qPCR of targeted Methanobrevibacter groups. The results showed a positive correlation between low CH4 emitters and higher abundance of Methanobrevibacter ruminantium clade. Principal coordinate analysis (PCoA) on operational taxonomic unit (OTU) level of bacteria showed two distinct clusters (P < 0.01) that were related to CH4 production. One cluster was associated with low CH4 production (referred to as cluster L) whereas the other cluster was associated with high CH4 production (cluster H) and the medium emitters occurred in both clusters. The differences between clusters were primarily linked to differential abundances of certain OTUs belonging to Prevotella. Moreover, several OTUs belonging to the family Succinivibrionaceae were dominant in samples belonging to cluster L. Fermentation pattern of volatile fatty acids showed that proportion of propionate was higher in cluster L, while proportion of butyrate was higher in cluster H. No difference was found in milk production or organic matter digestibility between cows. Cows in cluster L had lower CH4/kg energy corrected milk (ECM) compared to cows in cluster H, 8.3 compared to 9.7 g CH4/kg ECM, showing that low CH4 cows utilized the feed more efficient for milk production which might indicate a more efficient microbial population or host genetic differences that is reflected in bacterial and archaeal (or methanogens) populations.
