Formation of a series of stable pillar[5]arene-based pseudo[1]-rotaxanes and their [1]rotaxanes in the crystal state.

ABSTRACT: A series of mono-amide-functionalized pillar[5]arenes with different lengths of N-ω-aminoalkyl groups as the side chain on the rim were designed and synthesized, which all formed pseudo[1]rotaxanes in the crystal state. And these pseudo[1]rotaxanes could be transformed into [1]rotaxanes or open forms in the crystal state. In addition, they were also studied in solution by (1)H NMR spectroscopy.

Project description:Some examples of atropoisomeric pseudorotaxanes in which the isomerism arises by the different conformations adopted by the wheel are reported here. Upon threading hexahexyloxycalix[6]arene 1 with ammonium axles 2+ or 3+ , bearing biphenyl or trifluoromethylbenzyl moieties, respectively, two atropoisomeric pseudorotaxanes were formed in which the calix[6]-wheel 1 adopts the 1,2,3-alternate and cone conformations. The interconversion between them cannot be obtained by simple rotation around the ArCH2Ar bonds of the calixarene wheel, which is blocked by the presence of the axle inside its cavity. Therefore, it can only be obtained through a mechanism of de-threading/re-threading of the axle. In all the examined cases, the 1,2,3-alternate and cone atropoisomers are, respectively, the kinetic and the thermodynamic ones.

Construction and Property Investigation of Serial Pillar[5]arene-Based [1]Rotaxanes.

Project description:Although the construction and application of pillar[5]arene-based [1]rotaxanes have been extensively studied, the types of stoppers for them are limited. In this work, we designed and prepared three series of pillar[5]arene-based [1]rotaxanes (P5[1]Rs) with pentanedione derivatives, azobenzene derivatives, and salicylaldehyde derivatives as the stoppers, respectively. The obtained P5[1]Rs were fully characterized by NMR (1H, 13C, and 2D), mass spectra, and single-crystal X-ray analysis. We found that the synergic C-H···π, C-H···O interactions and N-H···O, O-H···N hydrogen bonding are the key to the stability of [1]rotaxanes. This work not only enriched the diversity of pillar[n]arene family but also gave a big boost to the pillar[n]arene-based mechanically interlocked molecules.

Selective photoinduced charge separation in perylenediimide-pillar[5]arene rotaxanes.

Project description:The ability to control photoinduced charge transfer within molecules represents a major challenge requiring precise control of the relative positioning and orientation of donor and acceptor groups. Here we show that such photoinduced charge transfer processes within homo- and hetero-rotaxanes can be controlled through organisation of the components of the mechanically interlocked molecules, introducing alternative pathways for electron donation. Specifically, studies of two rotaxanes are described: a homo[3]rotaxane, built from a perylenediimide diimidazolium rod that threads two pillar[5]arene macrocycles, and a hetero[4]rotaxane in which an additional bis(1,5-naphtho)-38-crown-10 (BN38C10) macrocycle encircles the central perylenediimide. The two rotaxanes are characterised by a combination of techniques including electron diffraction crystallography in the case of the hetero[4]rotaxane. Cyclic voltammetry, spectroelectrochemistry, and EPR spectroscopy are employed to establish the behaviour of the redox states of both rotaxanes and these data are used to inform photophysical studies using time-resolved infra-red (TRIR) and transient absorption (TA) spectroscopies. The latter studies illustrate the formation of a symmetry-breaking charge-separated state in the case of the homo[3]rotaxane in which charge transfer between the pillar[5]arene and perylenediimide is observed involving only one of the two macrocyclic components. In the case of the hetero[4]rotaxane charge separation is observed involving only the BN38C10 macrocycle and the perylenediimide leaving the pillar[5]arene components unperturbed.

Spacer Length-Independent Shuttling of the Pillar[5]arene Ring in Neutral [2]Rotaxanes.

Project description:For a series of neutral [2]rotaxanes consisting of a pillar[5]arene ring and axles possessing two stations separated by flexible spacers of different lengths, the free energies of activation for the ring shuttling between the stations were found to be independent of the spacer length. The constitution of the spacer affects the activation energies: replacement of CH2 groups by repulsive oxygen atoms in the axle increases the barrier. The explanation for the observed length-independence lies in the presence of a barrier for re-forming the stable co-conformation, which makes the ring travel back and forth along the thread in an intermediate state.

Diversity Oriented Preparation of Pillar[5]arene-Containing [2]Rotaxanes by a Stopper Exchange Strategy.

Project description:Rotaxane building blocks bearing 3,5-bis(trifluoromethyl) benzenesulfonate (BTBS) stoppers have been efficiently prepared from a pillar[5]arene derivative, 3,5-bis(trifluoromethyl) benzenesulfonyl chloride (BTBSCl) and different diols, namely 1,10-decanediol and 1,12-dodecanediol. The BTBS moieties of these compounds are good leaving groups and stopper exchange reactions could be achieved by treatment with different nucleophiles thus affording rotaxanes with ester, thioether or ether stoppers.

Recognition Selectivities of Lasso-Type Pseudo[1]rotaxane Based on a Mono-Ester-Functionalized Pillar[5]arene.

Project description:Two types of mono-ester-functionalized pillar[5]arenes, P1 and P2, bearing different side-chain groups, were synthesized. Their host-guest complexation and self-inclusion properties were studied by 1H NMR and 2D nuclear overhauser effect spectroscopy (NOESY) NMR measurements. The results showed that the substituents on their phenolic units have a great influence on the self-assembly of both pillar[5]arenes, although they both could form stable pseudo[1]rotaxanes at room temperature. When eight bulky 4-brombutyloxy groups were capped on the cavity, instead of methoxy groups, pseudo[1]rotaxane P1 became less stable and its locked ester group in the inner space of cavity was not as deep as P2, leading to distinctly different host-guest properties between P1 and P2 with 1,6-dibromohexane. Moreover, pillar[5]arene P1 displayed effective molecular recognition toward 1,6-dichlorohexane and 1,2-bromoethane among the guest dihalides. In addition, the self-complex models and stabilities between P1 and P2 were also studied by computational modeling and experimental calculations.

Visualization and quantitation of electronic communication pathways in a series of redox-active pillar[6]arene-based macrocycles

Project description:While oxidized pillar[5]arenes with 1-5 benzoquinone units are known, very few examples of oxidized pillar[6]arenes have been reported. We describe here the synthesis, characterization and electrochemical behavior of a series of macrocyclic hosts prepared by the stepwise oxidation of 1,4-diethoxypillar[6]arene, resulting in high-yield and high-purity isolation of two constitutional isomers for each macrocycle, in which two, three or four 1,4-diethoxybenzene units are replaced by benzoquinone residues. A careful structural comparison with their counterparts in the pillar[5]arene framework indicates that the geometries of the macrocycles are better described as non-Euclidean hyperbolic hexagons and elliptic pentagons, respectively. A comprehensive computational study to determine anisotropic induced current density (ACID) allows us to visualize and quantify through-space and through-bond communication pathways along the macrocyclic belt. Experimental and simulated voltammetric data, as well as UV-vis spectra, of the new macrocycles afford insights into the various electronic communication pathways in these compounds. Partially oxidised pillar[n]arenes exhibit interesting redox behaviour, but the pillar[6]arenes are poorly characterised in this context. Here stepwise oxidation of 1,4-diethoxypillar[6]arene allows detailed study of the structural and electrochemical properties of molecules with 2, 3, and 4 oxidised units.

A pillar[5]arene-based [2]rotaxane lights up mitochondria.

Project description:Subcellular organelle-specific reagents for simultaneous targeting, imaging and treatment are highly desirable for cancer therapy. However, it remains a challenge to fabricate a single molecular platform containing a targeting group, imaging and therapeutic agents through traditional synthesis. Due to their superior sensitivity and photostability, fluorescent probes with aggregation-induced emission (AIE) characteristics have attracted more and more attention in studying the process of translocation, drug release, and excretion of nanomedicines in vitro or in vivo. We construct a pillar[5]arene-based [2]rotaxane (R1) by employing tetraphenylethene (TPE) and triphenylphosphonium (TPP) moieties as stoppers; the TPE unit retains the aggregation-induced emission (AIE) attribute and the TPP group is used as a mitochondria-targeting agent. R1 exhibits enhanced AIE, high specificity to mitochondria, and superior photostability. By introducing doxorubicin (DOX) into R1, prodrug R2 is constructed as a dual-fluorescence-quenched Förster resonance energy transfer (FRET) system, in which the TPE-based axle acts as a donor fluorophore and the DOX unit acts as the acceptor. Upon hydrolysis of R2 in endo/lysosomes, the fluorescences of the carrier and the drug recover. R1 is further utilized as a drug delivery platform to conjugate other anticancer drugs containing amine groups through imine formation to prepare prodrugs. The anticancer drugs are released from these prodrugs in the cells upon hydrolysis of the pH-responsive imine bonds.

Porous pillar[6]arene-based polymers for reversible iodine capture.

Project description:Iodine in nuclear waste can cause serious environment pollution and health risks, and has thus driven more development of materials for iodine capture. Herein, a novel porous pillar[6]arene-based polymer (P-P6APs) was easily prepared as a supramolecular adsorbent for iodine via a one-step crosslinking reaction between per-hydroxylated pillar[6]arene and decafluorobiphenyl. Nitrogen adsorption tests demonstrated that this material possessed a satisfactory surface area (S BET = 366 m2 g-1) and pore diameter (3.8 nm), which was due to the macrocyclic scaffolds. Compared with commercially available activated carbon, P-P6APs exhibited superior adsorption efficiency toward volatile iodine not only in the solution phase (water and n-hexane) but also in the gas phase. This outcome was mainly ascribed to nonspecific adsorption by the pore structure of the crosslinked material coupled with multiple intermolecular binding sites of the macrocyclic scaffold. Moreover, this supramolecular absorbent was recyclable and could be reused 5 times with no obvious loss of performance.

Crystal structure and supra-molecular features of a bis-urea-functionalized pillar[5]arene.

Project description:The crystal structure of a bis-urea derivative based on A1/A2-functionalized pillar[5]arene (DUP) that encapsulates dimethyl formamide (DMF) inside the macrocyclic cavity is reported. The crystal structure of DUP·DMF, C63H70N4O12·C3H7NO, reveals that out of two urea functionalized spacers, one arm is oriented above the macrocyclic cavity with strong hydrogen-bonding inter-actions between the urea H atoms and DMF guest, whereas, the other arm is positioned away from the macrocycle, leading to inter-molecular hydrogen-bonding inter-actions between the urea H atoms of two adjacent pillar[5]arene macrocycles, resulting in the formation of a supra-molecular dimer.
