Project description:This paper launches an open access DNA barcoding project "AUSBS" under the Barcoding of Life Datasystems (BOLD). The aims of the project are to help scientists who lack the necessary morphological knowledge to identify known species using molecular markers, to aid native bee specialists with the recognition of species groups that morphologically are difficult to define, and, eventually, to assist with the recognition of new species among known species. Using integrative taxonomy, i.e. morphological comparison to type specimens in Australian museum collections combined with phylogenetic analysis of a fragment of the mitochondrial DNA cytochrome c oxidase subunit I (mtCOI) gene sequences led to the recognition of four new species of Euhesma Michener (Hymenoptera: Colletidae: Euryglossini) collected during intensive surveys in remote Australian conservation areas, which are described. The new species are Euhesma micans, Euhesma lyngouriae, and Euhesma aulaca in a species group associated with Eremophila flowers, and Euhesma albamala in the walkeriana species group.

Project description:The bee genus Chilicola Spinola (Xeromelissinae) is recorded from the State of Bahia, Brazil for the first time, based on a new species of the megalostigma group of the subgenus Hylaeosoma Ashmead. Chilicola (Hylaeosoma) kevanisp. n. is described and figured from males collected in Wesceslau Guimarães, Bahia. The species can be distinguished on the basis of coloration, size, integumental sculpturing, and structure of the hidden metasomal sterna and genitalia. A revised key to the species of the megalostigma group is provided.

Project description:High-altitude ecosystems are found in mountain chains and plateaus worldwide. These areas tend to be underrepresented in insect biodiversity assessments because of the challenges related to systematic survey at these elevations, such as extreme climatic and geographic conditions. Nonetheless, high-altitude ecosystems are of paramount importance because they have been seen to be species pumps for other geographic areas, such as adjacent locations, functioning as buffers for population declines. Moreover, these ecosystems and their biodiversity have been proposed to be fast-responding indicators of the impacts caused by global climate change. Bees have been highlighted among the insect groups that have been affected by these problems. This work used bees as a proxy to demonstrate and reinforce the importance of systematic surveys of high-altitude ecosystems. Here, field collections were undertaken and an updated review was conducted for the native bee biodiversity of the high-altitude ecosystem found at the Andes system of central Chile, including the phenological trends of these insects during the flowering season. Of the 58 species that have been described for this location, we were able to confirm the occurrence of 46 of these species as a result of our sampling. In addition, thanks to these recent collections, a new species of Xeromelissa Cockerell is described in the present work. These findings highlight the need for further high-altitude insect surveys of this biome, which include both temporal and spatial complexity in their design, to allow for accurate assessment of bee species diversity and compositional changes in these mountain regions.

Project description:The bee subgenus Chilicola (Heteroediscelis) Toro & Moldenke, 1979 (Hymenoptera, Colletidae, Xeromelissinae) is revised. The subgenus is considered endemic to Chile and occurs across a broad range of habitats. Eight new species are described: Chilicola (Heteroediscelis) charizard Monckton, sp. n., Chilicola (Heteroediscelis) curvapeligrosa Monckton, sp. n., Chilicola (Heteroediscelis) guanicoe Monckton, sp. n., Chilicola (Heteroediscelis) katherinae Monckton, sp. n., Chilicola (Heteroediscelis) lickana Monckton, sp. n., Chilicola (Heteroediscelis) mayu Monckton, sp. n., Chilicola (Heteroediscelis) packeri Monckton, sp. n., and Chilicola (Heteroediscelis) randolphi Monckton, sp. n. One of the existing species, Chilicola (Heteroediscelis) valparaiso Toro & Moldenke, 1979, syn. n., is treated as a junior synonym of Chilicola (Heteroediscelis) mantagua Toro & Moldenke, 1979, and the nine remaining valid species are redescribed. Thoroughly illustrated keys to species for males and females are provided, along with habitus images, images of male terminalia, distribution maps for each species, and a map of relevant Chilean biogeographic regions. Results of phylogenetic analyses are presented, based upon 74 morphological characters and on CO1 barcode sequences, analyzed both separately and as a combined dataset. Monophyly of the subgenus is supported, and groupings within the subgenus are discussed in light of a biogeographic analysis of their species distributions (spatial analysis of vicariance), whereby divergence between taxa is found to occur primarily via north-south disjunctions.

Project description:A new genus and two new species of miniature clingfishes are described based on specimens collected from dense stands of macroalgae in intertidal and shallow subtidal areas along the coast of southern Australia. The new genus, Barryichthys, is distinguished from other genera of the Gobiesocidae by unique features of the adhesive disc, including elongate papillae in adhesive disc regions A and B, the reduction and/or loss of several elements of the cephalic lateral line canals, the lower gill arch skeleton, and the neurocranium, and by having two distinct types of pectoral-fin rays. Barryichthyshutchinsi is described based on 19 specimens (12.4-18.7 mm SL) from Western Australia and South Australia. Barryichthysalgicola is described based on 22 specimens (9.0-21.0 mm SL) from Victoria, New South Wales and Tasmania. The new species are distinguished from each other by characters of body and head shape, vertebral counts, and aspects of live colour pattern. The new genus shares several characters in common with Parvicrepis, another genus of miniature gobiesocids from southern Australia that also inhabits macroalgae habitats. The many reductions and novel characters of Barryichthys are discussed within the context of miniaturisation.

Project description:A new species of sponge-inhabiting barnacle, Membranobalanus porphyrophilus sp. nov., is described herein. This species can be distinguished from all other congeners by a combination of characters, in particular by the shapes of the tergum and scutum and the armament of the cirri. COI sequence data from the type specimens have been lodged with GenBank and a morphological key to the species of Membranobalanus is provided to aid future research. The host of the new species is the southern Australian endemic demosponge Spheciospongia purpurea. The new species of barnacle is thought to be host species specific.

Project description:BACKGROUND: The past several years have seen a flurry of papers seeking to clarify the utility and limits of DNA barcoding, particularly in areas such as species discovery and paralogy due to nuclear pseudogenes. Heteroplasmy, the coexistence of multiple mitochondrial haplotypes in a single organism, has been cited as a potentially serious problem for DNA barcoding but its effect on identification accuracy has not been tested. In addition, few studies of barcoding have tested a large group of closely-related species with a well-established morphological taxonomy. In this study we examine both of these issues, by densely sampling the Hawaiian Hylaeus bee radiation. RESULTS: Individuals from 21 of the 49 a priori morphologically-defined species exhibited coding sequence heteroplasmy at levels of 1-6% or more. All homoplasmic species were successfully identified by COI using standard methods of analysis, but only 71% of heteroplasmic species. The success rate in identifying heteroplasmic species was increased to 86% by treating polymorphisms as character states rather than ambiguities. Nuclear pseudogenes (numts) were also present in four species, and were distinguishable from heteroplasmic sequences by patterns of nucleotide and amino acid change. CONCLUSIONS: Heteroplasmy significantly decreased the reliability of species identification. In addition, the practical issue of dealing with large numbers of polymorphisms- and resulting increased time and labor required - makes the development of DNA barcode databases considerably more complex than has previously been suggested. The impact of heteroplasmy on the utility of DNA barcoding as a bulk specimen identification tool will depend upon its frequency across populations, which remains unknown. However, DNA barcoding is still likely to remain an important identification tool for those species that are difficult or impossible to identify through morphology, as is the case for the ecologically important solitary bee fauna.

Project description:Despite intense interest in bees, no genomes are available for the bee family Colletidae. Colletes gigas, one of the largest species of the genus Colletes in the world, is an ideal candidate to fill this gap. Endemic to China, C. gigas has been the focus of studies on its nesting biology and pollination of the economically important oil tree Camellia oleifera, which is chemically defended. To enable deeper study of its biology, we sequenced the whole genome of C. gigas using single-molecule real-time sequencing on the Pacific Bioscience Sequel platform. In total, 40.58 G (150×) of long reads were generated and the final assembly of 326 scaffolds was 273.06 Mb with a N50 length of 8.11 Mb, which captured 94.4% complete Benchmarking Universal Single-Copy Orthologs. We predicted 11,016 protein-coding genes, of which 98.50% and 84.75% were supported by protein- and transcriptome-based evidence, respectively. In addition, we identified 26.27% of repeats and 870 noncoding RNAs. The bee phylogeny with this newly sequenced colletid genome is consistent with available results, supporting Colletidae as sister to Halictidae when Stenotritidae is not included. Gene family evolution analyses identified 9,069 gene families, of which 70 experienced significant expansions (33 families) or contractions (37 families), and it appears that olfactory receptors and carboxylesterase may be involved in specializing on and detoxifying Ca. oleifera pollen. Our high-quality draft genome for C. gigas lays the foundation for insights on the biology and behavior of this species, including its evolutionary history, nesting biology, and interactions with the plant Ca. oleifera.
