Behavioral performance is provided in a table. Fig. 2 of the article is based on this table. Brain regions showing time effect are provided in a table. Real-time observation of Cooper pair splitting showing strong non-local correlations.

Project description:Controlled generation and detection of quantum entanglement between spatially separated particles constitute an essential prerequisite both for testing the foundations of quantum mechanics and for realizing future quantum technologies. Splitting of Cooper pairs from a superconductor provides entangled electrons at separate locations. However, experimentally accessing the individual split Cooper pairs constitutes a major unresolved issue as they mix together with electrons from competing processes. Here, we overcome this challenge with the first real-time observation of the splitting of individual Cooper pairs, enabling direct access to the time-resolved statistics of Cooper pair splitting. We determine the correlation statistics arising from two-electron processes and find a pronounced peak that is two orders of magnitude larger than the background. Our experiment thereby allows to unambiguously pinpoint and select split Cooper pairs with 99% fidelity. These results open up an avenue for performing experiments that tap into the spin-entanglement of split Cooper pairs.

Data showing atherosclerosis-associated differentially methylated regions are often at enhancers.

Project description:Atherosclerosis involves phenotypic modulation and transdifferentiation of vascular smooth muscle cells (SMCs). Data are given in tabular or figure format that illustrate genome-wide DNA methylation alterations in atherosclerotic vs. control aorta (athero DMRs). Data based upon publicly available chromatin state profiles are also shown for normal aorta, monocyte, and skeletal muscle tissue-specific DMRs and for aorta-specific chromatin features (enhancer chromatin, promoter chromatin, repressed chromatin, actively transcribed chromatin). Athero hypomethylated and hypermethylated DMRs as well as epigenetic and transcription profiles are described for the following genes: ACTA2, MYH10, MYH11 (SMC-associated genes); SMAD3 (a signaling gene for SMCs and other cell types); CD79B and SH3BP2 (leukocyte-associated genes); and TBX20 and genes in the HOXA, HOXB, HOXC, and HOXD clusters (T-box and homeobox developmental genes). The data reveal strong correlations between athero hypermethylated DMRs and regions of enhancer chromatin in aorta, which are discussed in the linked research article "Atherosclerosis-associated differentially methylated regions can reflect the disease phenotype and are often at enhancers" (M. Lacey et al., 2019).

Data showing regional differences in rat brain monoaminergic function.

Project description:Chemical neurotransmitters (such as dopamine) modulate cognitive function via ascending projections to various cortical and sub-cortical brain regions. This report describes and links to a relatively large dataset (up to N = 112) compiled from control (untreated) brain samples taken during a series of experimental in vivo studies. The dataset is freely available, to explore the normal interrelationships between levels of neurotransmitter (e.g., dopamine, serotonin), across brain regions implicated in both normal reward and drug addiction, as well as in disorders such as schizophrenia (e.g., nucleus accumbens, prefrontal cortex). Most experimental studies run with a relatively small control group, so there is a lack of baseline data on the expected levels of neurotransmitters and their metabolites in different brain regions. Accordingly, the available dataset has been compiled from a number of studies run in the same laboratory, and using closely similar behavioural procedures, sampling selected brain regions of a priori interest. These collated data can be used to explore differences in the distribution of the monoamines and their metabolites, patterns of neurotransmitter intercorrelations, both between and within different brain structures and including some consideration of laterality effects.

Brain Regions Showing White Matter Loss in Huntington's Disease Are Enriched for Synaptic and Metabolic Genes.

Project description:BACKGROUND:The earliest white matter changes in Huntington's disease are seen before disease onset in the premanifest stage around the striatum, within the corpus callosum, and in posterior white matter tracts. While experimental evidence suggests that these changes may be related to abnormal gene transcription, we lack an understanding of the biological processes driving this regional vulnerability. METHODS:Here, we investigate the relationship between regional transcription in the healthy brain, using the Allen Institute for Brain Science transcriptome atlas, and regional white matter connectivity loss at three time points over 24 months in subjects with premanifest Huntington's disease relative to control participants. The baseline cohort included 72 premanifest Huntington's disease participants and 85 healthy control participants. RESULTS:We show that loss of corticostriatal, interhemispheric, and intrahemispheric white matter connections at baseline and over 24 months in premanifest Huntington's disease is associated with gene expression profiles enriched for synaptic genes and metabolic genes. Corticostriatal gene expression profiles are predominately associated with motor, parietal, and occipital regions, while interhemispheric expression profiles are associated with frontotemporal regions. We also show that genes with known abnormal transcription in human Huntington's disease and animal models are overrepresented in synaptic gene expression profiles, but not in metabolic gene expression profiles. CONCLUSIONS:These findings suggest a dual mechanism of white matter vulnerability in Huntington's disease, in which abnormal transcription of synaptic genes and metabolic disturbance not related to transcription may drive white matter loss.

Behavioral impairments caused by injections of the protein synthesis inhibitor anisomycin after contextual retrieval reverse with time.

Project description:Recent accounts of memory suggest that retrieval of a learning experience transforms that memory into a labile state that requires a period of protein synthesis to be reconsolidated into a fixed state. In this article, we show that the impairments in behavior caused by the protein synthesis inhibitor anisomycin given after retrieval are temporary and are thus not likely to reflect disruptions in a protein synthesis-dependent reconsolidation process. Mice received injections of anisomycin after either initial acquisition or retrieval of contextual fear conditioning. When anisomycin injections followed acquisition, freezing was impaired during memory tests the next day and 21 days later. When anisomycin injections followed normal retrieval of contextual fear conditioning, freezing was impaired the next day but recovered to levels of control mice when testing occurred 21 days later. This recovery effect occurred after short or long durations of exposure during the retrieval period and was specific to the conditioning context. These results suggest that anisomycin injections after retrieval do not retroactively affect the memory from conditioning.

Ascending neurons convey behavioral state to integrative sensory and action selection brain regions.

Project description:Knowing one's own behavioral state has long been theorized as critical for contextualizing dynamic sensory cues and identifying appropriate future behaviors. Ascending neurons (ANs) in the motor system that project to the brain are well positioned to provide such behavioral state signals. However, what ANs encode and where they convey these signals remains largely unknown. Here, through large-scale functional imaging in behaving animals and morphological quantification, we report the behavioral encoding and brain targeting of hundreds of genetically identifiable ANs in the adult fly, Drosophila melanogaster. We reveal that ANs encode behavioral states, specifically conveying self-motion to the anterior ventrolateral protocerebrum, an integrative sensory hub, as well as discrete actions to the gnathal ganglia, a locus for action selection. Additionally, AN projection patterns within the motor system are predictive of their encoding. Thus, ascending populations are well poised to inform distinct brain hubs of self-motion and ongoing behaviors and may provide an important substrate for computations that are required for adaptive behavior.

The time course of word retrieval revealed by event-related brain potentials during overt speech.

Project description:Speech production is one of the most fundamental activities of humans. A core cognitive operation involved in this skill is the retrieval of words from long-term memory, that is, from the mental lexicon. In this article, we establish the time course of lexical access by recording the brain electrical activity of participants while they named pictures aloud. By manipulating the ordinal position of pictures belonging to the same semantic categories, the cumulative semantic interference effect, we were able to measure the exact time at which lexical access takes place. We found significant correlations between naming latencies, ordinal position of pictures, and event-related potential mean amplitudes starting 200 ms after picture presentation and lasting for 180 ms. The study reveals that the brain engages extremely fast in the retrieval of words one wishes to utter and offers a clear time frame of how long it takes for the competitive process of activating and selecting words in the course of speech to be resolved.

A BAYESIAN TIME-VARYING EFFECT MODEL FOR BEHAVIORAL MHEALTH DATA.

Project description:The integration of mobile health (mHealth) devices into behavioral health research has fundamentally changed the way researchers and interventionalists are able to collect data as well as deploy and evaluate intervention strategies. In these studies, researchers often collect intensive longitudinal data (ILD) using ecological momentary assessment methods, which aim to capture psychological, emotional, and environmental factors that may relate to a behavioral outcome in near real-time. In order to investigate ILD collected in a novel, smartphone-based smoking cessation study, we propose a Bayesian variable selection approach for time-varying effect models, designed to identify dynamic relations between potential risk factors and smoking behaviors in the critical moments around a quit attempt. We use parameter-expansion and data-augmentation techniques to efficiently explore how the underlying structure of these relations varies over time and across subjects. We achieve deeper insights into these relations by introducing nonparametric priors for regression coefficients that cluster similar effects for risk factors while simultaneously determining their inclusion. Results indicate that our approach is well-positioned to help researchers effectively evaluate, design, and deliver tailored intervention strategies in the critical moments surrounding a quit attempt.

Retrieval Practice Is Effective Regardless of Self-Reported <i>Need for Cognition</i> - Behavioral and Brain Imaging Evidence.

Project description:There is an emerging consensus that retrieval practice is a powerful way to enhance long-term retention and to reduce achievement gaps in school settings. Less is known whether retrieval practice benefits performance in individuals with low intrinsic motivation to spend time and effort on a given task, as measured by self-reported need for cognition (NFC). Here, we examined retrieval practice in relation to individual differences in NFC by combining behavioral and functional magnetic resonance imaging (fMRI) data. Using a within-subject design, upper-secondary school students (N = 274) learned a language-based material (Swahili-Swedish word-pairs), with half of the items by means of retrieval practice with feedback and half by study only. One week later, the students were tested on the word-pairs either in the classroom (n = 204), or in a fMRI scanner (n = 70). In both settings, a retrieval practice effect was observed across different levels of NFC (high or low). Relatedly, comparable fMRI effects were seen in both NFC subgroups. Taken together, our findings provide behavioral and brain-imaging evidence that retrieval practice is effective also for individuals with lower levels of NFC, which is of direct relevance for educational practice.

Distinct behavioral traits and associated brain regions in mouse models for obsessive-compulsive disorder.

Project description:BackgroundObsessive-compulsive disorder (OCD) is a mental disease with heterogeneous behavioral phenotypes, including repetitive behaviors, anxiety, and impairments in cognitive functions. The brain regions related to the behavioral heterogeneity, however, are unknown.MethodsWe systematically examined the behavioral phenotypes of three OCD mouse models induced by pharmacological reagents [RU24969, 8-hydroxy-DPAT hydrobromide (8-OH-DPAT), and 1-(3-chlorophenyl) piperazine hydrochloride-99% (MCPP)], and compared the activated brain regions in each model, respectively.ResultsWe found that the mouse models presented distinct OCD-like behavioral traits. RU24969-treated mice exhibited repetitive circling, anxiety, and impairments in recognition memory. 8-OH-DPAT-treated mice exhibited excessive spray-induced grooming as well as impairments in recognition memory. MCPP-treated mice showed only excessive self-grooming. To determine the brain regions related to these distinct behavioral traits, we examined c-fos expression to indicate the neuronal activation in the brain. Our results showed that RU24969-treated mice exhibited increased c-fos expression in the orbitofrontal cortex (OFC), anterior cingulate cortex (ACC), prelimbic cortex (PrL), infralimbic cortex (IL), nucleus accumbens (NAc), hypothalamus, bed nucleus of the stria terminalis, lateral division, intermediate part (BSTLD), and interstitial nucleus of the posterior limb of the anterior commissure, lateral part (IPACL), whereas in 8-OH-DPAT-treated mice showed increased c-fos expression in the ACC, PrL, IL, OFC, NAc shell, and hypothalamus. By contrast, MCPP did not induce higher c-fos expression in the cortex than control groups.ConclusionOur results indicate that different OCD mouse models exhibited distinct behavioral traits, which may be mediated by the activation of different brain regions.
