A Comparative Analysis of Sonic Defences in Bombycoidea Caterpillars.

ABSTRACT: Caterpillars have long been used as models for studying animal defence. Their impressive armour, including flamboyant warning colours, poisonous spines, irritating sprays, and mimicry of plant parts, snakes and bird droppings, has been extensively documented. But research has mainly focused on visual and chemical displays. Here we show that some caterpillars also exhibit sonic displays. During simulated attacks, 45% of 38 genera and 33% of 61 species of silk and hawkmoth caterpillars (Bombycoidea) produced sounds. Sonic caterpillars are found in many distantly-related groups of Bombycoidea, and have evolved four distinct sound types- clicks, chirps, whistles and vocalizations. We propose that different sounds convey different messages, with some designed to warn of a chemical defence and others, to startle predators. This research underscores the importance of exploring acoustic communication in juvenile insects, and provides a model system to explore how different signals have evolved to frighten, warn or even trick predators.

Project description:BackgroundHerbivorous insects represent a major fraction of global biodiversity and the relationships they have established with their food plants range from strict specialists to broad generalists. Our knowledge of these relationships is of primary importance to basic (e.g. the study of insect ecology and evolution) and applied biology (e.g. monitoring of pest or invasive species) and yet remains very fragmentary and understudied. In Lepidoptera, caterpillars of families Saturniidae and Sphingidae are rather well known and considered to have adopted contrasting preferences in their use of food plants. The former are regarded as being rather generalist feeders, whereas the latter are more specialist.New informationTo assemble and synthesise the vast amount of existing data on food plants of Lepidoptera families Saturniidae and Sphingidae, we combined three major existing databases to produce a dataset collating more than 26,000 records for 1256 species (25% of all species) in 121 (67%) and 167 (81%) genera of Saturniidae and Sphingidae, respectively. This dataset is used here to document the level of polyphagy of each of these genera using summary statistics, as well as the calculation of a polyphagy score derived from the analysis of Phylogenetic Diversity of the food plants used by the species in each genus.

Comparative transcriptomics of albino and warningly-coloured caterpillars.

Project description:Coloration is perhaps one of the most prominent adaptations for survival and reproduction of many taxa. Coloration is of particular importance for aposematic species, which rely on their coloring and patterning acting as a warning signal to deter predators. Most research has focused on the evolution of warning coloration by natural selection. However, little information is available for color mutants of aposematic species, particularly at the genomic level. Here, I compare the transcriptomes of albino mutant caterpillars of the aposematic wood tiger moth (Arctia plantaginis) to those of their full sibs having their distinctive orange-black warning coloration. The results showed >290 differentially expressed genes genome-wide. Genes involved in the immune system, structural constituents of cuticular, and immunity were mostly downregulated in the albino caterpillars. Surprisingly, higher expression was observed in core melanin genes from albino caterpillars, suggesting that melanin synthesis may be disrupted in terminal ends of the pathway during its final conversion. Taken together, these results suggest that caterpillar albinism may not be due to a depletion of melanin precursor genes. In contrast, the albino condition may result from the combination of faulty melanin conversion late in its synthesis and structural deficiencies in the cuticular preventing its deposition. The results are discussed in the context of how albinism may impact individuals of aposematic species in the wild.

Analysis on the Complete Mitochondrial Genome of Andraca theae (Lepidoptera: Bombycoidea).

Project description:The bombycid moth, Andraca theae (Matsumura) (Lepidoptera: Bombycoidea) is an important pest of tea in southeastern China. In the present study, the mitochondrial genome (mitogenome) of A. theae was amplified by polymerase chain reaction and sequenced. The complete mitogenome of A. theae, encoding 37 genes, was 15,737 bp in length (Genbank no. KX365419), and consisted of 13 protein-coding genes (PCGs), 22 tRNA genes, 2 ribosomal RNA genes and an adenine (A) + thymine (T)-rich region (AT-rich region). The gene order of A. theae mitogenome was typical for Lepidoptera mitogenomes. Except for cox1, which started with CGA, all other 12 PCGs started with ATN. Eleven of the 13 PCGs ended with TAA, expect for cox1 and cox2, which ended with a single T. The maximum likelihood method and the Bayesian method were used to analyze the phylogenetic relationship among 22 representative bombycoid species with a matrix consisting of the 13 PCGs of the mitogenomes of the 22 species. The topological structures of the two phylogenetic trees we constructed were almost identical, with the results indicating that the bombycid species, including A. theae, clustered into a single clade with a bootstrap value of 58% and a posterior probability of 0.98. The phylogenetic relationship among the Bombycoidea species analyzed was Lasiocampidae + (Bombycidae + (Saturniidae + Sphingidae)) which was supported by a high bootstrap value of 100% and a posterior probability of 1.00.

A global checklist of the Bombycoidea (Insecta: Lepidoptera).

Project description:BackgroundBombycoidea is an ecologically diverse and speciose superfamily of Lepidoptera. The superfamily includes many model organisms, but the taxonomy and classification of the superfamily has remained largely in disarray. Here we present a global checklist of Bombycoidea. Following Zwick (2008) and Zwick et al. (2011), ten families are recognized: Anthelidae, Apatelodidae, Bombycidae, Brahmaeidae, Carthaeidae, Endromidae, Eupterotidae, Phiditiidae, Saturniidae and Sphingidae. The former families Lemoniidae and Mirinidae are included within Brahmaeidae and Endromidae respectively. The former bombycid subfamilies Oberthueriinae and Prismostictinae are also treated as synonyms of Endromidae, and the former bombycine subfamilies Apatelodinae and Phitditiinae are treated as families.New informationThis checklist represents the first effort to synthesize the current taxonomic treatment of the entire superfamily. It includes 12,159 names and references to their authors, and it accounts for the recent burst in species and subspecies descriptions within family Saturniidae (ca. 1,500 within the past 10 years) and to a lesser extent in Sphingidae (ca. 250 species over the same period). The changes to the higher classification of Saturniidae proposed by Nässig et al. (2015) are rejected as premature and unnecessary. The new tribes, subtribes and genera described by Cooper (2002) are here treated as junior synonyms. We also present a new higher classification of Sphingidae, based on Kawahara et al. (2009), Barber and Kawahara (2013) and a more recent phylogenomic study by Breinholt et al. (2017), as well as a reviewed genus and species level classification, as documented by Kitching (2018).

Comparative genomic analysis of intracranial germ cell tumors - the preliminary study focused on Sonic Hedgehog signaling pathway.

Project description:Aim of the study:Examination of copy number changes in a group of intracranial germ cell tumors (GCTs) with particular focus on putative aberrations of the main genes coding SHh pathway proteins. Material and methods:The study was performed on DNA isolated from fresh-frozen tumor tissue samples from eight GCTs, including six intracranial GCTs. The intracranial group consisted of three germinomas, two mature teratomas and one mixed germ cell tumor. Comparative genomic profiling analysis was carried out using microarray-CGH method (Cytosure ISCA UPD 4×180k, OGT). The results were analyzed with Feature Extraction (Agilent Technologies) and Nexus Copy Number (BioDiscovery) softwares. Results and conclusions:Chromosomal aberrations were found in two intracranial germinomas. These tumors were characterized by complex genomic profiles encompassing chromosomes 7, 8, 9, 10, 11, 12, 16, 17 and 19. Common findings were gain at 12p13.33p11.1 of 35 Mbp and gain at 17q11.1q25.3 of 55 Mbp. In one tumor, also SHh (7q36.3), SMO (7q32.1) and GLI3 (7p14.1) copy gains occurred together with 9q21.11q34.3 loss, including PTCH1, all being elements of SHh signaling pathway. Moreover, both tumors showed various copy gain of genes being ligands, regulators, receptors or target genes of SHh (MTSS1; PRKACA and FKBP8) as well as gain of genes of SHh coopting WNT pathway (WNT3, WNT5B, WNT9B in both tumors; WNT16, WNT2 in pineal lesion). Further studies on larger group are needed to characterize SHh-related gene alterations in intracranial GCTs and for searching genotype-phenotype relations.

Project description:We assembled larval transcriptome of D. arcuata using RNA-seq data from both social and solitary instars, and then conducted differential gene expression analysis between the two behavioural instars. This revealed a large number of transcripts that were differential expressed between the two behavioral states, including some transcripts coding for gene products that have been previously implicated in social behaviour in other insects.
