ABSTRACT: This paper reports an isolation and identification of novel poly-?-hydroxybutyrate (PHB) degrading bacterium Stenotrophomonas sp. RZS 7 and studies on its extracellular PHB degrading depolymerase enzyme. The bacterium isolated from soil samples of plastic contaminated sites of municipal area in Shahada, Maharashtra, Western India. It was identified as Stenotrophomonas sp. RZS 7 based on polyphasic approach. The bacterium grew well in minimal salt medium (MSM) and produced a zone (4.2 mm) of PHB hydrolysis on MSM containing PHB as the only source of nutrient. An optimum yield of enzyme was obtained on the fifth day of incubation at 37 °C and at pH 6.0. Further increase in enzyme production was recorded with Ca2+ ions, while other metal ions like Fe2+ (1 mM) and chemical viz. mercaptoethanol severally affected the production of enzyme.

Project description:There are numerous reports on poly-β-hydroxybutyrate (PHB) depolymerases produced by various microorganisms isolated from various habitats, however, reports on PHB depolymerase production by an isolate from plastic rich sites scares. Although PHB has attracted commercial significance, the inefficient production and recovery methods, inefficient purification of PHB depolymerase and lack of ample knowledge on PHB degradation by PHB depolymerase have hampered its large scale commercialization. Therefore, to ensure the biodegradability of biopolymers, it becomes imperative to study the purification of the biodegrading enzyme system. We report the production, purification, and characterization of extracellular PHB depolymerase from Stenotrophomonas sp. RZS7 isolated from a dumping yard rich in plastic waste. The isolate produced extracellular PHB depolymerase in the mineral salt medium (MSM) at 30°C during 4 days of incubation under shaking. The enzyme was purified by three methods namely ammonium salt precipitation, column chromatography, and solvent purification. Among these purification methods, the enzyme was best purified by column chromatography on the Octyl-Sepharose CL-4B column giving optimum yield (0.7993 Umg-1mL-1). The molecular weight of purified PHB depolymerase was 40 kDa. Studies on the assessment of biodegradation of PHB in liquid culture medium and under natural soil conditions confirmed PHB biodegradation potential of Stenotrophomonas sp. RZS7. The results obtained in Fourier-Transform Infrared (FTIR) analysis, High-Performance Liquid Chromatography (HPLC) study and Gas Chromatography Mass-Spectrometry (GC-MS) analysis confirmed the biodegradation of PHB in liquid medium by Stenotrophomonas sp. RZS7. Changes in surface morphology of PHB film in soil burial as observed in Field Emission Scanning Electron Microscopy (FESEM) analysis confirmed the biodegradation of PHB under natural soil environment. The isolate was capable of degrading PHB and it resulted in 87.74% biodegradation. A higher rate of degradation under the natural soil condition is the result of the activity of soil microbes that complemented the biodegradation of PHB by Stenotrophomonas sp. RZS7.

Project description:BackgroundAgricultural plastic mulches offer great benefits such as higher yields and lower pesticide use. Yet, plastic mulches may disintegrate over time and fragment into smaller debris. Such plastic debris is expected to remain in the field after removal of the plastic mulch and thus contributes to soil contamination with plastics.MethodTo investigate this, we collected soil samples at 0-10 cm and 10-40 cm depth from three fields covered with black mulch film for three consecutive years. Three fields without any reported plastic use served as control. Visual plastic debris > 1 cm (macroplastics) was collected from the soil surface. Mesoplastics between 2 mm and 1 cm were density separated from the sampled soil using saturated NaCl solution and analyzed by Fourier-transform infrared spectroscopy. Debris ≤ 2 mm (microplastics) was dispersed from 50 g soil using sodium hexametaphosphate solution followed by the aforementioned density separation. The separated polyethylene (PE), polypropylene (PP), and polystyrene (PS) were quantified via solvent-based pyrolysis-gas chromatography/mass spectrometry (Py-GC/MS).ResultsWith 89-206 fragments ha-1, the majority of macroplastics were located in fields previously covered with mulch films. 80% of the collected specimen were identified as black PE film. The number of mesoplastics in plastic-mulched soil was 2.3 particles kg-1, while only 1.0 particles kg-1 were found in the reference fields. Py-GC/MS revealed microplastic levels of up to 13 mg kg-1. The PE content was significantly higher in plastic-mulched fields than in reference fields.DiscussionAlthough the identified plastic levels are lower than those reported in comparable studies, our results still suggest that plastic mulching functions as a source of plastic debris in agricultural systems. Due to its severely restricted degradability, these plastics are likely to accumulate in soil in the long term and further fragment into smaller and smaller debris.

Project description:Large amount of phthalate esters (PAEs) used as plasticizers in polyvinyl chloride (PVC) products has caused ubiquitous contamination to the environment and potential ecology security risk all around the world, especially in places plastic films were indispensably utilized due to the widely proposing of facility agriculture in China. A case of PAEs contamination in four suburb areas of Nanjing was analyzed and discussed in this study. A new frame work has been put forward based on multi-criteria evaluation model and mathematical method of catastrophe theory, using farming work, laboratory determination and relevant environmental standards to measure the ecology security risk of PAEs in study areas. The factors were selected based on the availability of the data and the local conditions. The assessment model involves the contamination status of PAEs in soil and vegetables, the contamination effects of PAEs to human and soil organisms and the contamination source of PAEs from plastic films and other products in the four study facility agriculture areas. An evaluation system of the model was composed of thirteen mesosphere indicators and twenty-five underlying indicators including total PAEs concentration in soils, single PAE concentration in soils, total PAEs concentrations in roots, leafy, solanaceous and stem vegetables, PAE human risks, soil microbial counts, microorganism diversity indices, atmospheric deposition of PAEs, whether sewage wastewater irrigation, planting mode of the facility agriculture areas and climate condition of study areas. The modified evaluation system was used in the assessment of ecology security of the same place based on the data of 2012, and the results suggested that the ecology security indicators were reliable and were agree well with the practical situation of the study areas. The results could provide guidance for the application of health risk assessment of soil environment for the strong objectivity of catastrophe theory compared with other evaluation methods.
