Highly heterogeneous soil bacterial communities around Terra Nova Bay of Northern Victoria Land, Antarctica.

Project description:Given the diminished role of biotic interactions in soils of continental Antarctica, abiotic factors are believed to play a dominant role in structuring of microbial communities. However, many ice-free regions remain unexplored, and it is unclear which environmental gradients are primarily responsible for the variations among bacterial communities. In this study, we investigated the soil bacterial community around Terra Nova Bay of Victoria Land by pyrosequencing and determined which environmental variables govern the bacterial community structure at the local scale. Six bacterial phyla, Actinobacteria, Proteobacteria, Acidobacteria, Chloroflexi, Cyanobacteria, and Bacteroidetes, were dominant, but their relative abundance varied greatly across locations. Bacterial community structures were affected little by spatial distance, but structured more strongly by site, which was in accordance with the soil physicochemical compositions. At both the phylum and species levels, bacterial community structure was explained primarily by pH and water content, while certain earth elements and trace metals also played important roles in shaping community variation. The higher heterogeneity of the bacterial community structure found at this site indicates how soil bacterial communities have adapted to different compositions of edaphic variables under extreme environmental conditions. Taken together, these findings greatly advance our understanding of the adaption of soil bacterial populations to this harsh environment.

Growth Forms and Functional Guilds Distribution of Soil Fungi in Coastal Versus Inland Sites of Victoria Land, Antarctica.

Project description:In Victoria Land, Antarctica, ice-free areas are restricted to coastal regions and dominate the landscape of the McMurdo Dry Valleys. These two environments are subjected to different pressures that determine the establishment of highly adapted fungal communities. Within the kingdom of fungi, filamentous, yeasts and meristematic/microcolonial growth forms on one side and different lifestyles on the other side may be considered adaptive strategies of particular interest in the frame of Antarctic constraints. In this optic, soil fungal communities from both coastal and Dry Valleys sites, already characterized thorough ITS1 metabarcoding sequencing, have been compared to determine the different distribution of phyla, growth forms, and lifestyles. Though we did not find significant differences in the richness between the two environments, the communities were highly differentiated and Dry Valleys sites had a higher evenness compared to coastal ones. Additionally, the distribution of different growth forms and lifestyles were well differentiated, and their diversity and composition were likely influenced by soil abiotic parameters, among which soil granulometry, pH, P, and C contents were the potential main determinants.

Antarctic Salt-Cones: An Oasis of Microbial Life? The Example of Boulder Clay Glacier (Northern Victoria Land).

Project description:The evaporation of a localized, highly saline water body of the Boulder Clay debris-covered glacier, in the Northern Victoria Land, probably generated the accumulation of mirabilite (Na2SO4 × 10H2O) and thenardite (Na2SO4) in a glacier salt-cone. Such an extremely cold and salty environment resembles the conditions on Mars, so it can be considered a terrestrial analog. The study was aimed at gaining a first glimpse at the prokaryotic community associated with Antarctic mirabilite and thenardite minerals and also to find clues about the origin of the salts. For this purpose, samples were analyzed by a next generation approach to investigate the prokaryotic (Bacteria and Archaea) diversity. Phylogenetic analysis allowed the identification of Bacteroidota, Actinobacteriota, Firmicutes, and Gammaproteobacteria as the main bacterial lineages, in addition to Archaea in the phylum Halobacterota. The genera Arthrobacter, Rhodoglobus, Gillisia, Marinobacter and Psychrobacter were particularly abundant. Interestingly, several bacterial and archaeal sequences were related to halotolerant and halophilic genera, previously reported in a variety of marine environments and saline habitats, also in Antarctica. The analyzed salt community also included members that are believed to play a major role in the sulfur cycle.

A thin ice layer segregates two distinct fungal communities in Antarctic brines from Tarn Flat (Northern Victoria Land).

Project description:Brines are hypersaline solutions which have been found within the Antarctic permafrost from the Tarn Flat area (Northern Victoria Land). Here, an investigation on the possible presence and diversity of fungal life within those peculiar ecosystems has been carried out for the first time. Brines samples were collected at 4- and 5-meter depths (TF1 and TF2, respectively), from two brines separated by a thin ice layer. The samples were analyzed via Illumina MiSeq targeting the ITS region specific for both yeasts and filamentous fungi. An unexpected high alpha diversity was found. Beta diversity analysis revealed that the two brines were inhabited by two phylogenetically diverse fungal communities (Unifrac value: 0.56, p value < 0.01; Martin's P-test p-value < 0.001) characterized by several specialist taxa. The most abundant fungal genera were Candida sp., Leucosporidium sp., Naganishia sp. and Sporobolomyces sp. in TF1, and Leucosporidium sp., Malassezia sp., Naganishia sp. and Sporobolomyces sp. in TF2. A few hypotheses on such differentiation have been done: i) the different chemical and physical composition of the brines; ii) the presence in situ of a thin layer of ice, acting as a physical barrier; and iii) the diverse geological origin of the brines.

Microbial Assemblages in Pressurized Antarctic Brine Pockets (Tarn Flat, Northern Victoria Land): A Hotspot of Biodiversity and Activity.

Project description:Two distinct pressurized hypersaline brine pockets (named TF4 and TF5), separated by a thin ice layer, were detected below an ice-sealed Antarctic lake. Prokaryotic (bacterial and archaeal) diversity, abundances (including virus-like particles) and metabolic profiles were investigated by an integrated approach, including traditional and new-generation methods. Although similar diversity indices were computed for both Bacteria and Archaea, distinct bacterial and archaeal assemblages were observed. Bacteroidetes and Gammaproteobacteria were more abundant in the shallowest brine pocket, TF4, and Deltaproteobacteria, mainly represented by versatile sulphate-reducing bacteria, dominated in the deepest, TF5. The detection of sulphate-reducing bacteria and methanogenic Archaea likely reflects the presence of a distinct synthrophic consortium in TF5. Surprisingly, members assigned to hyperthermophilic Crenarchaeota and Euryarchaeota were common to both brines, indicating that these cold habitats host the most thermally tolerant Archaea. The patterns of microbial communities were different, coherently with the observed microbiological diversity between TF4 and TF5 brines. Both the influence exerted by upward movement of saline brines from a sub-surface anoxic system and the possible occurrence of an ancient ice remnant from the Ross Ice Shelf were the likely main factors shaping the microbial communities.

Redefining the boundary between crystalline and sedimentary rock of Eastern Dahomey Basin.

Project description:This study defines a new boundary between the crystalline and sedimentary rocks of Eastern Dahomey Basin at the southwestern part of Nigeria using a geophysical approach that combines regional aeromagnetic and ground resistivity data. Aeromagnetic data covering the entire Eastern Dahomey Basin were acquired at 500 m line spacing, 80 m mean terrain, and processed into grids of Residual Magnetic Intensity (RMI) map. Filters and corrections such as upward continuation, and reduction to equator were applied to enhance deep magnetic sources and correct for magnetic inclination and declination. Tilt Derivative Angles (TDR) was applied for edge detection. To support the aeromagnetic analysis and interpretation, 104 Vertical Electrical Sounding (VES) surveys and 8 Electrical Resistivity Tomography (ERT) data were also acquired, processed and interpreted along the basement-sedimentary rock boundary. The TDR revealed a significant trend that corresponds to the edge between the basement complex and the sediments of the Eastern Dahomey Basin. A strong match was also noticed between the VES positions and the TDR map. Areas interpreted as basement rocks from the VES stations align with positive values on the TDR maps while the sedimentary terrains have negative TDR values. Our work demonstrates that areas that were previously fixed as sedimentary terrains on geological maps belong to the crystalline basement or transition zone. A new and reliable geological boundary is hereby drawn between the basement and sedimentary rocks. Thus, providing a revised map of the Eastern Dahomey Basin.

Fingerprinting Proterozoic Bedrock in Interior Wilkes Land, East Antarctica.

Project description:Wilkes Land in East Antarctica remains one of the last geological exploration frontiers on Earth. Hidden beneath kilometres of ice, its bedrock preserves a poorly-understood tectonic history that mirrors that of southern Australia and holds critical insights into past supercontinent cycles. Here, we use new and recently published Australian and Antarctic geological and geophysical data to present a novel interpretation of the age and character of crystalline basement and sedimentary cover of interior Wilkes Land. We combine new zircon U-Pb and Hf isotopic data from remote Antarctic outcrops with aeromagnetic data observations from the conjugate Australian-Antarctic margins to identify two new Antarctic Mesoproterozoic basement provinces corresponding to the continuation of the Coompana and Madura provinces of southern Australia into Wilkes Land. Using both detrital zircon U-Pb-Hf and authigenic monazite U-Th-Pb isotopic data from glacial erratic sandstone samples, we identify the presence of Neoproterozoic sedimentary rocks covering Mesoproterozoic basement. Together, these new geological insights into the ice-covered bedrock of Wilkes Land substantially improve correlations of Antarctic and Australian geological elements and provide key constraints on the tectonic architecture of this sector of the East Antarctic Shield and its role in supercontinent reconstructions.
