The formation of unusual multilayered parallel lamellae-in-lamellae in symmetric supramolecular double-comb diblock copolymers is presented. While keeping the concentration of surfactant fixed, the number of internal layers was found to increase with molecular weight M up to 34 for the largest block copolymer. Project description:Involving supramolecular chemistry in self-assembling block copolymer systems enables design of macromolecular architectures that are challenging to obtain through conventional all-covalent routes. In this work we present supramolecular double-comb triblock terpolymers in which both outer blocks are able to interact with a surfactant via hydrogen bonding and thereby form a comb-shaped architecture upon complexation. While the neat triblock terpolymer only formed a triple lamellar morphology, multiple hierarchical structures were observed in these supramolecular comb-coil-comb triblock terpolymers by simply adjusting the surfactant concentration. Structures included spheres on tetragonally packed cylinders-in-lamellae and spheres on double parallel lamellae-in-lamellae, as evidenced by electron microscopy and X-ray scattering. Incorporation of a middle coil block thus allowed an even higher macromolecular complexity than the previously reported double-comb diblock copolymers.

Project description:Supramolecular block copolymers (PS-DAT-sb-PI-Thy) were synthesized via noncovalent hydrogen bonding between well-defined thymine end-functionalized polyisoprene (PI-Thy) and diaminotriazine (DAT) end-functionalized polystyrene (PS-DAT). Three covalently linked block copolymers were also synthesized for comparison with the noncovalent supramolecular block copolymers. The complementary DAT/Thy interaction resulted in the microphase separation of the supramolecular block copolymer system. Detailed characterization of all functionalized homopolymers and block copolymers was carried out via proton nuclear magnetic resonance (1H NMR) spectroscopy, gel permeation chromatography, matrix-assisted laser desorption/ionization-time of flight mass spectrometry, and differential scanning calorimetry. The self-assembly process of supramolecular block copolymers was evidenced by transmission electron microscopy. Small-angle X-ray scattering was also performed to study the microphase separation of supramolecular and covalently linked block copolymers. Comparison of microphase separation images of supramolecular block copolymers and the corresponding covalently linked analogues reveals differences in d-spacing and microdomain shape.

Project description:We report the formation of a double-gyroid network morphology in normal-tapered poly(isoprene-b-isoprene/styrene-b-styrene) [P(I-IS-S)] and inverse-tapered poly(isoprene-b- styrene/isoprene-b-styrene) [P(I-SI-S)] diblock copolymers. Our tapered diblock copolymers with overall poly(styrene) volume fractions of 0.65 (normal-tapered) and 0.67 (inverse-tapered), and tapered regions comprising 30 volume percent of the total polymer, were shown to self-assemble into the double-gyroid network morphology through a combination of small angle X-ray scattering (SAXS) and transmission electron microscopy (TEM). The block copolymers were synthesized by anionic polymerization, where the tapered region between the pure poly(isoprene) and poly(styrene) blocks was generated using a semi-batch feed with programmed syringe pumps. The overall composition of these tapered copolymers lies within the expected network-forming region for conventional poly(isoprene-b-styrene) [P(I-S)] diblock copolymers. Dynamic mechanical analysis (DMA) clearly demonstrated that the order-disorder transition temperatures (T(ODT)'s) of the network-forming tapered block copolymers were depressed when compared to the T(ODT) of their non-tapered counterpart, with the P(I-SI-S) showing the greater drop in T(ODT). These results indicate that it is possible to manipulate the copolymer composition profile between blocks in a diblock copolymer, allowing significant control over the T(ODT), while maintaining the ability to form complex network structures.

Project description:Complementary complexation between 2,2':6',2″-terpyridine (tpy) and 6,6″-dianthracenyl-substituted tpy in the presence of Zn(II) ions provided an efficient strategy for construction of metallo-supramolecular diblock copolymers. To synthesize well-defined tpy-modified polystyrenes (PSs), an Fe(II) bis(tpy) complex bearing α-bromoester as a metallo-initiator was applied to atom transfer radical polymerization (ATRP) to avoid poisoning the Cu(I) catalyst. Subsequently, a series of tpy-functionalized PSs was obtained after the decomplexation of <tpy-Fe(II)-tpy> junction by tetrakis(triethylammonium) ethylenediaminetetraacetate (TEA-EDTA) under mild conditions. The metallo-supramolecular poly(3-hexylthiophene) (P3HT)-block-PS diblock copolymers were prepared by simply mixing the corresponding terminally tpy-modified homopolymers with Zn(II) ions, and further characterized by 1H NMR and diffusion ordered spectroscopy (DOSY) experiments. The approach using metallo-initiators for ATRP offers an opportunity to construct tpy-functionalized polymers with controllable molecular weights and low polydispersities. Through the spontaneous heteroleptic complexation, a variety of metallo-supramolecular diblock copolymers with tunable block ratios can be easily constructed.

Project description:We have developed a hierarchical process that combines linear triblock copolymers into concentric globular subunits through strong chemical bonds and is followed by their supramolecular assembly via weak noncovalent interactions to afford one-dimensionally assembled, dynamic cylindrical nanostructures. The molecular brush architecture forces triblock copolymers to adopt intramolecular interactions within confined frameworks and then drives their intermolecular interactions in the mixtures of organic solvent and water. In contrast, the triblock copolymers, when not preconnected into the molecular brush architectures, organize only into globular assemblies.

Project description:In this study, a graft copolymer, poly(N-(2-hydroxypropyl) methacrylamide dilactate)-co-(N-(2-hydroxypropyl) methacrylamide-co-histidine)-graft-poly(d,l-lactide), and a diblock copolymer, methoxy poly(ethylene glycol)-b-poly(d,l-lactide), were assembled into a mixed micellar system to encapsulate the anticancer drug doxorubicin (Dox). This mixed micellar system possesses the hydrophobic lactide segment of both copolymers, which reinforces its stability in physiological milieus; the histidine molecules appended on the graft copolymer provide the desired pH-responsive behavior to release Dox during internalization in cancer cells. The results demonstrate that the two copolymers were successfully prepared, and their ratios in the mixed micelles were optimized on the basis of the results of the stability tests. Under acidic conditions, the mixed micelles swell and are able to release their payloads. Therefore, the in vitro results indicate that the Dox in the mixed micelles is released effectively in response to the environmental pH of the mimetic internalization process, increasing cancer cells' sensitivity toward Dox. The mixed micelles display low cytotoxicity due to the degradability of the polymers. The in vivo images show that the high stability of the mixed micelles ensures a high tumor accumulation. This selective tumor accumulation results in an excellent inhibition of in vivo tumor growth and a high rate of apoptosis in cancerous tissues, with low toxicity. This highly stable, mixed micellar system with a pH-dependent drug release, which enables the precise delivery of drugs to the tumor lesions, is feasible to employ clinically in cancer therapy.

Project description:A series of diblock copolymers containing an endosomal-releasing segment composed of diethylaminoethyl methacrylate (DEAEMA) and butyl methacrylate (BMA) were synthesized via reversible addition-fragmentation chain transfer (RAFT) polymerization. The materials were designed to condense plasmid DNA (pDNA) through electrostatic interactions with a cationic poly(N,N-dimethylaminoethyl methacrylate) (DMAEMA) first block. The pDMAEMA was employed as a macro chain transfer agent (macroCTA) for the synthesis of a series in which the relative feed ratios of DEAEMA and BMA were systematically varied from 20% to 70% BMA. The resultant diblock copolymers exhibited low polydispersity (PDI ≤ 1.06) with similar molecular weights (M(n) = 19.3-23.1 kDa). Dynamic light scattering (DLS) measurements in combination with (1)H NMR D(2)O studies demonstrated that the free copolymers assemble into core-shell micelles at physiological pH. Reduction of the solution pH to values representative of endosomal/lysosomal compartments induced an increase in the net cationic charge of the core through protonation of the DEAEMA residues. This protonation promotes micelle destabilization and exposure of the hydrophobic BMA residues that destabilize biological membranes. The pH value at which this micelle-to-unimer transition occurred was dependent on the hydrophobic content of the copolymer, with higher BMA-containing copolymer compositions exhibiting pH-induced transitions to the membrane-destabilizing state at successively lower pH values. The ability of the diblock copolymers to deliver pDNA was subsequently investigated using a GFP expression vector in two monocyte cell lines. High levels of DNA transfection were observed for the copolymer compositions exhibiting the sharpest pH transitions and membrane destabilizing activities, demonstrating the importance of tuning the endosomal-releasing segment composition.

Project description:Polymer vesicles, or polymersomes, are promising candidates for applications in drug delivery and tissue imaging. While a vast variety of polymers have been explored for their ability to assemble into polymersomes, relatively little research on the functionalization of these polymers has been reported. We present here a novel route for the synthesis of poly(caprolactone)-b-poly(ethylene glycol) (PCL-b-PEG) diblock copolymers that allows for the insertion of functional groups at the block junctions and the assembly of functional membranes. This modular synthesis has been developed on the basis of solid-phase peptide synthesis techniques and is accomplished through the formation of two peptide bonds, one between an amine-terminated PEG and the carboxyl moiety of the functional group and the other between the functional group amine and a carboxy-terminated PCL. As a demonstration of the potential utility of the resulting vesicles, we incorporated two different amino acid functional groups at the junction. 2-Nitrophenylalanine was utilized to create UV-responsive membranes in which the vesicles were destabilized and released encapsulated contents upon irradiation. A fluorescein-conjugated lysine was also utilized to create stable fluorescent membranes in which the fluorescence was built into the polymer. This method should contribute to our ability to further develop smart, functional membranes.

Project description:The self-assembly of synthetic diblock copolymers has been extensively studied experimentally and theoretically. In contrast, self-assembly of polypeptide diblock copolymers has so far been mostly studied experimentally. We discovered that the theory developed for synthetic diblock copolymer does not fully explain the self-assembly of elastin-like polypeptide diblock copolymers, leading us to generalize the theory to make it applicable for these polypeptides. We demonstrated that elastin-like polypeptide diblocks self-assemble into weak micelles with dense cores and almost unstretched coronas, a state not previously observed for synthetic diblock copolymers. Weak micelles form if the surface tension at the core-corona interface is low compared to that expected of a micelle with a dense core. The predictions of the theory of weak micelles for the critical micelle temperature, hydrodynamic radius, and aggregation number of elastin-like polypeptide diblocks are in reasonable agreement with the experimentally measured values. The unique and unprecedented control of amphiphilicity in these recombinant peptide polymers reveals a new micellar state that has not been previously observed in synthetic diblock copolymer systems.

Project description:In this study, we synthesized amphiphilic poly(2,7⁻(9,9⁻dioctylfluorene))⁻block⁻N,N⁻(diisopropylamino)ethyl methacrylate (POF⁻b⁻PDPMAEMA) rod-coil diblock copolymers by atom transfer radical polymerization (ATRP). The structure and multifunctional sensing properties of these copolymers were also investigated. The POF rod segment length of 10 was fixed and the PDPAEMA coil segment lengths of 90 and 197 were changed, respectively. The micellar aggregates of POF10⁻b⁻PDPAEMA90 rod-coil diblock copolymer in water showed a reversible shape transition from cylinder bundles to spheres when the temperature was changed from 20 to 80 °C or the pH was changed from 11 to 2. The atomic force microscopy (AFM) and transmission electron microscopy (TEM) measurements indicated that the temperature had also an obvious influence on the micelle size. In addition, since POF10⁻b⁻PDPAEMA90 had a lower critical solution temperature, its photoluminescence (PL) intensity in water is thermoreversible. The PL spectra showed that the POF⁻b⁻PDPAEMA copolymer had a reversible on/off profile at elevated temperatures, and thus could be used as an on/off fluorescent indicator for temperature or pH. The fluorescence intensity distribution of pH switched from "off⁻on" to "on⁻off" as the temperature increased. These results showed that the POF⁻b⁻PDPAEMA copolymer has a potential application for temperature and pH sensing materials.
