Project description:Implementation of the European Union Renewable Energy Directive has triggered exponential growth in trading of pelletized wood fibers. Over 18 million tons of wood pellets were traded by EU member countries in 2018 of which a third were imported from the US. Concerns exist about negative impacts on US forests but systematic assessments are currently lacking. We assessed variability in fundamental attributes for timberland structure and carbon stocks within 123 procurement landscapes of wood pellet mills derived from over 38 thousand forest inventory plots in the eastern US from 2005 to 2017. We found more carbon stocks in live trees, but a fewer number of standing-dead trees, associated with the annual operation of large-scale wood pellet mills. In the US coastal southeast-where US pellet exports to the EU originate-there were fewer live and growing-stock trees and less carbon in soils with every year of milling operation than in the rest of the eastern US-which supplies the domestic market. Greater overlap of mills' procurement areas exhibited discernible increments across selected carbon stocks. These trends likely reflect more intensive land management practices. Localized forest impacts associated with the wood pellet industry should continue to be monitored.

Project description:The European market for wood pellets used in small-scale heating systems has been expanding significantly over the past decade. For an analysis of market efficiency in the Journal Energy with the title "The European wood pellets for heating market - price developments, trade and market efficiency" wood pellet prices have been collected as well as trade flows downloaded for the trade relations between Austria, Germany, Italy and France. Only since January 2012 monthly wood pellet trade data is published by Eurostat. This, now monthly expanding data-set provides new opportunities for analysing the development of this important renewable energy commodity. Furthermore, national wood pellet prices published by national authorities and interest groups are improving in quality in the recent years. The collection and combination of these data-sets are a chance for novel econometric analysis. This paper presents valuable tools and processes to acquire and prepare this data and connects to a data and code repository for downloading the resources described in this and the related Journal Energy publication.

Project description:The effects of bedding material on air quality are important amongst horses worldwide. Respiratory diseases, especially equine asthma, are highly prevalent with air hygiene playing a major role on the pathophysiology of these diseases. The objective of our study was to investigate the effects of four bedding materials on the respiratory signs, tracheal mucus score, and tracheal wash (TW) and bronchoalveolar lavage fluid (BALF) cytology in healthy adult horses. The study design was a prospective controlled cross-over study, and the subjects were healthy adult riding school horses (n = 32) from a single stable. Wood pellet, straw pellet, and loosely stored peat (Peat 3) were compared to peat packed in plastic-covered bales (Peat 2). Lower airway endoscopy and sampling (TW and BALF) for cytological examination were performed after each 35-day bedding period. The tracheal mucus scores (P = 0.014) and respiratory rate (P = 0.026) were higher during the straw pellet period compared to the Peat 2 period. The respiratory rate was lower during the wood pellet period compared to the Peat 2 period (P = 0.004). The TW neutrophil percentage during the straw pellet period was higher compared to the Peat 2 period (P = 0.0003). The BALF neutrophil percentage was higher during the straw pellet period (P = 0.005) and during the Peat 3 period compared to the Peat 2 period (P = 0.04). We conclude that baled peat (Peat 2) caused lower neutrophil percentages in the airway samples compared to straw pellet and loosely stored peat (Peat 3). No difference was observed between Peat 2 and wood pellet. The information gained from this study may assist veterinarians and horse owners in selecting appropriate bedding materials, especially for horses with equine asthma.

Project description:Poultry immunity, health, and production are several factors that challenge the future growth of the poultry industry. Consumer confidence, product quality and safety, types of products, and the emergence and re-emergence of diseases will continue to be major challenges to the current situation and the strategic future of the industry. Foodborne and zoonotic diseases are strictly linked with poultry. Eradication, elimination, and/or control of foodborne and zoonotic pathogens present a major challenge to the poultry industry. In addition, the public health hazards from consuming foods with high antibiotic residues will remain a critical issue. The theory of poultry production described in this review will not be limited to considering disease control. Rather, it will also incorporate the interconnection of the animals' health, welfare, and immunity. It is essential to know that chickens are not susceptible to intranasal infection by the SARS-CoV-2 (COVID-19) virus. Nevertheless, the COVID-19 pandemic will affect poultry consumption, transport, and the economics of poultry farming. It will also take into consideration economic, ethical, social dimensions, and the sustenance of the accomplishment of high environmental security. Stockholders, veterinarians, farmers, and all the partners of the chain of poultry production need to be more involved in the current situation and the strategic future of the industry to fulfill human demands and ensure sustainable agriculture. Thus, the present review explores these important tasks.

Project description:Microglia are the tissue macrophages of the central nervous system (CNS) and the first to respond to CNS dysfunction and disease. Gene expression profiling of microglia during development, under homeostatic conditions and in the diseased CNS provided insight in microglia functions and changes thereof. Single cell sequencing studies further contributed to our understanding of microglia heterogeneity in relation to age, sex and CNS disease. Recently, single nucleus gene expression profiling was performed on (frozen) CNS tissue. Transcriptomic profiling of CNS tissues by (single) nucleus RNA-sequencing has the advantage that it can be applied to archived and well stratified frozen specimens. Here, we give an overview of the significant advances recently made in microglia transcriptional profiling. In addition, we present matched cellular and nuclear microglia RNA-seq datasets we generated from mouse and human CNS tissue to compare cellular versus nuclear transcriptomes from fresh and frozen samples. We demonstrate that microglia can be similarly profiled with cell and nucleus profiling, and importantly also with nuclei isolated from frozen tissue. Nuclear microglia transcriptomes are a reliable proxy for cellular transcriptomes. Interestingly, lipopolysaccharide- (LPS)-induced changes in gene expression were even more pronounced in the nuclear transcriptome. In addition, heterogeneity in microglia observed in fresh samples is similarly detected in frozen nuclei of the same donor. Together, these results show that microglia nuclear RNAs obtained from frozen CNS tissue are a reliable proxy for microglia gene expression and cellular heterogeneity and may prove an effective strategy to study of the role of microglia in neuropathology.

Project description:Cangrelor is the only currently available intravenous platelet P2Y12 receptor inhibitor. It is characterized by potent, predictable, and rapidly reversible antiplatelet effects. Cangrelor has been tested in the large CHAMPION (Cangrelor Versus Standard Therapy to Achieve Optimal Management of Platelet Inhibition) program, where it was compared with different clopidogrel regimens, and it is currently indicated for use in patients with coronary artery disease undergoing percutaneous coronary intervention. However, the uptake of cangrelor use varies across the globe and may also include patients with profiles different from those enrolled in the registration trials. These observations underscore the need to fully examine the safety and efficacy of cangrelor in postregistration studies. There are several ongoing and planned studies evaluating the use of cangrelor in real-world practice which will provide important insights to this extent. The current article provides a review on the pharmacology, clinical studies, contemporary use of cangrelor in real-world practice, a description of ongoing studies, and futuristic insights on potential strategies on how to improve outcomes of patients undergoing percutaneous coronary intervention.

Project description:Wood pellet storage safety is an important aspect for implementing woody biomass as a renewable energy source. When wood pellets are stored indoors in large quantities (tons) in poorly ventilated spaces in buildings, such as in basements, off-gassing of volatile organic compounds (VOCs) can significantly affect indoor air quality. To determine the emission rates and potential impact of VOC emissions, a series of laboratory and field measurements were conducted using softwood, hardwood, and blended wood pellets manufactured in New York. Evacuated canisters were used to collect air samples from the headspace of drums containing pellets and then in basements and pellet storage areas of homes and small businesses. Multiple peaks were identified during GC/MS and GC/FID analysis, and four primary VOCs were characterized and quantified: methanol, pentane, pentanal, and hexanal. Laboratory results show that total VOCs (TVOCs) concentrations for softwood (SW) were statistically (p < 0.02) higher than blended or hardwood (HW) (SW: 412 ± 25; blended: 203 ± 4; HW: 99 ± 8, ppb). The emission rate from HW was the fastest, followed by blended and SW, respectively. Emissions rates were found to range from 10-1 to 10-5 units, depending upon environmental factors. Field measurements resulted in airborne concentrations ranging from 67 ± 8 to 5000 ± 3000 ppb of TVOCs and 12 to 1500 ppb of aldehydes, with higher concentrations found in a basement with a large fabric bag storage unit after fresh pellet delivery and lower concentrations for aged pellets. These results suggest that large fabric bag storage units resulted in a substantial release of VOCs into the building air. Occupants of the buildings tested discussed concerns about odor and sensory irritation when new pellets were delivered. The sensory response was likely due to the aldehydes.

Project description:Good practices around model-informed drug discovery and development (MID3) aim to improve the implementation, standardization, and acceptance of these approaches within drug development and regulatory review. A survey targeted to clinical pharmacology and pharmacometric colleagues across industry, the US Food and Drug Administration (FDA), and the European Medicines Agency (EMA) was conducted to understand current and future roles of MID3. The documented standards were generally affirmed as a "good match" to current industry practice and regulatory expectations, with some identified gaps that are discussed. All have seen at least a "modest" step forward in MID3 implementation associated with greater organizational awareness and share the expectation for a future wider use and impact. The priority within organizations was identified as a limitation with respect to the future of MID3. Finally, potential solutions, including a global overarching MID3 regulatory guideline, to facilitate greater acceptance by industry and regulatory decision makers are discussed.

Project description:Wood pellets are a fuel used for heat and power generation. Wood pellets are manufactured from forest residues and byproducts of other sectors in the wood processing industry, such as sawmilling. Wood pellet production generates combustible wood dust, which presents the risk of fire and explosion. The objective of this research was to incorporate the principles of inherently safer design (ISD) for the management of combustible dust hazards associated with wood pellet production. Using bow tie analysis to explicitly consider ISD within process hazard analysis (PHA), ISD barriers were successfully identified, including the use of paved surfaces to store feedstock to minimize rocks entering the process and presenting a risk of ignition sources, the use of reduced-size silos to minimize the inventory and increase the turnover frequency, the removal of unnecessary or hazardous equipment, such as fans, following a redesign, and the relocation of hazardous equipment, such as cyclones, outside and away from personnel. A summary of example-based guidance for combustible dust hazards was collected to support additional ISD implementation within PHA as part of the process safety management (PSM). The research also highlights learnings for conducting virtual PHA workshops, as well as identifying opportunities for incorporating ISD within operating wood processing facilities through the incident investigation and risk assessment elements of PSM.
