MALDI-TOF mass spectrometry and identification of new bacteria species in air samples from Makkah, Saudi Arabia.

Project description:BACKGROUND: During the Hajj season, respiratory symptoms are very common among pilgrims. Here, we investigated the viable bacterial population in air samples collected around the slaughterhouses used during the Hajj. METHODS AND RESULTS: We collected air samples on three days from four different sites: slaughterhouses at Al-Kakia, Al-Meaisim and Al-Sharaia, and from a waste disposal area designated for the remnants of slaughter. Samples were cultured on blood agar plates for 48 h, and bacterial isolates were identified using MALDI-TOF MS. A dendrogram using the spectra of the unidentified bacterial species was constructed, and PCR amplification and sequencing of the 16S rRNA gene was performed for one isolate per cluster. In total, 2500 colonies appeared on the nutrient agar plates, and 244 were purified for further analysis. Good identification was obtained for 202 (83%) isolates by MALDI-TOF MS. The most common genera were Bacillus (n=94, 45%) and Staphyloccocus (n=55, 26%). Poor identification was obtained for 42 (17%) isolates, and their spectra clustering revealed that these isolates belonged to 10 species. Four of these were considered to be new species. CONCLUSIONS: During the Hajj, the air was contaminated by many environmental bacterial agents, and MALDI-TOF MS was successfully adapted for their rapid identification.

Ecological studies of Commiphora genus (myrrha) in Makkah region, Saudi Arabia.

Project description:Commiphora, myrrha, is a pantropical genus and perform well in arid and semi-arid environments. This genus has economic importance. Distribution of Commiphora species and their associated species in Saudi Arabia has not been studied to date. The current study report on (a) characterization and distribution of plant communities including Commiphora species and (b) assessment of factors influencing ecological preferences of these species. Five species of Commiphora are recorded inhabiting mountain slopes, steep escarpments or hills consisting of igneous rocks, either granites or basalt with drought prone shallow soil. One hundred and twenty-six plant species belonging to 95 genera and 35 families were found associated with different Commiphora species. Therophytes showed the most frequent life form class and Sudanian region elements recorded the highest phytogeographical units (28%) followed by Tropical elements. Field study showed that Commiphora gileadensis and C. quadricincta preferred granite and basalt rocks exposed to erosion, while C. myrrah, C. kataf and C. habessinica grow on resistant coarse pink granite. The analysis of 240 sampling stands with TWINSPAN revealed the vegetation of Commiphora habitats into eight vegetation groups; each group represented a distinct microhabitat. Dendrogram obtained from a hierarchical classification showed that habitats of C. gileadinsis and C. quadricincta are more similar than those of other species. This similarity was confirmed by Jaccard and Sorenson similarity indices and by Pearson correlation coefficient. This investigation compiled the information/data to facilitate future range management of Commiphora species.

Project description:We report here the main characteristics of 'Arabia massiliensis' strain Marseille-P3078T gen. nov., sp. nov., 'Gordonibacter massiliensis' Marseille-P2775T sp. nov. and 'Bacilliculturomica massiliensis' strain Marseille-P3303 gen. nov., sp. nov. The culturomics approach combined with taxonogenomics was used to characterize these strains, which were all isolated from a faecal specimen of a 50-year-old Saudi Bedouin woman.

Pseudomonas atagosis sp. nov., and Pseudomonas akappagea sp. nov., New Soil Bacteria Isolated from Samples on the Volcanic Island Izu Oshima, Tokyo.

Project description:During the exploration of microbial natural resources, two strains of Pseudomonas, PS14T and PS24T, were isolated from samples taken from Izu Oshima, a volcanic island located 120 km southwest of central Tokyo. Phylogenetic analysis based on 16S rRNA gene sequences showed that PS14T was most similar to Pseudomonas baetica a390T (99.6%) and Pseudomonas helmanticensis OHA11T (99.5%), and that PS24T was most similar to Pseudomonas qingdaonensis JJ3T (98.8%) and Pseudomonas lutea OK2T (98.7%). The major fatty acids of these two strains were C16:0 and C17:0 cyclo, summed feature 3 (C16:1 ω6c and/or C16:1 ω7c), and summed feature 8 (C18:1 ω7c and/or 18:1 ω6c). The phylogenetic analyses, DNA-DNA hybridization results and phenotypic traits indicated that PS14T and PS24T constitute two novel species, Pseudomonas atagosis sp. nov. (type strain PS14T = CECT 9940T, = LMG 31496T) and Pseudomonas akappagea sp. nov. (type strain PS24T = CECT 9941T, = LMG 31497T), respectively. The sequence data of the draft genomes of PS14T and PS24T were deposited in the GenBank database under accession numbers VXCA00000000 and VXCP00000000, respectively, and the sequence data of their 16S rRNA genes were deposited in the GenBank database under accession numbers MN396717 and MN382268, respectively.

Assessment of knowledge about childhood autism spectrum disorder among healthcare workers in Makkah- Saudi Arabia.

Project description:Objective:To measure the knowledge of healthcare professionals about increasingly prevalent Autism Spectrum Disorder (ASD) along with perceptions around its management and prognosis and comparison across various specialties. Methods:This Cross sectional survey based comparative analysis took place at Maternity and Children Hospital and King Faisal Hospital Makkah from December 2017 to May 2018. The validated self-administered "Knowledge about childhood autism among health workers" questionnaire was used along with additional questions regarding perceptions about ASD. The mean and mean percent scores were calculated. Chi squared test and ANOVA were applied to find the association between quantitative and qualitative variables respectively. Results:Out of 162 participants, 153 returned the questionnaire and 147 were included in final analysis. Physicians constituted 81.6% (120) of participants. The mean score for participants was 9.80(S.E.M ±0.32) where non-physicians yielded higher mean score (11.2±4.41) as compared to physicians (9.6±3.28) (p=0.113). Psychiatrists had highest score of 16/19 while general physicians had lowest (6/19). Participants with more years of experience had higher mean scores (p-value = 0.01). About 72.10% (106) of participants opted for medication as a treatment option. Nearly 38.1% (56) of participants were skeptical about improvement of ASD with early interventions. Conclusion:There is a lack of knowledge about ASD amongst healthcare professionals in Saudi Arabia. Experienced professionals working with ASD children can be utilized to deliver targeted trainings nationwide.
