Stability and Plasticity of Contextual Modulation in the Mouse Visual Cortex.

ABSTRACT: Activity of neurons in primary visual cortex is shaped by sensory and behavioral context. However, the long-term stability of the influence of contextual factors in the mature cortex remains poorly understood. To investigate this, we used two-photon calcium imaging to track the influence of surround suppression and locomotion on individual neurons over 14 days. We found that highly active excitatory neurons and parvalbumin-positive (PV+) interneurons exhibited relatively stable modulation by visual context. Similarly, most neurons exhibited a stable yet distinct degree of modulation by locomotion. In contrast, less active excitatory neurons exhibited plasticity in visual context influence, resulting in increased suppression. These findings suggest that the mature visual cortex possesses stable subnetworks of neurons, differentiated by cell type and activity level, which have distinctive and stable interactions with sensory and behavioral contexts, as well as other less active and more labile neurons, which are sensitive to visual experience.

A Disinhibitory Circuit for Contextual Modulation in Primary Visual Cortex.

Project description:Context guides perception by influencing stimulus saliency. Accordingly, in visual cortex, responses to a stimulus are modulated by context, the visual scene surrounding the stimulus. Responses are suppressed when stimulus and surround are similar but not when they differ. The underlying mechanisms remain unclear. Here, we use optical recordings, manipulations, and computational modeling to show that disinhibitory circuits consisting of vasoactive intestinal peptide (VIP)-expressing and somatostatin (SOM)-expressing inhibitory neurons modulate responses in mouse visual cortex depending on similarity between stimulus and surround, primarily by modulating recurrent excitation. When stimulus and surround are similar, VIP neurons are inactive, and activity of SOM neurons leads to suppression of excitatory neurons. However, when stimulus and surround differ, VIP neurons are active, inhibiting SOM neurons, which leads to relief of excitatory neurons from suppression. We have identified a canonical cortical disinhibitory circuit that contributes to contextual modulation and may regulate perceptual saliency.

Plasticity and stability of visual field maps in adult primary visual cortex.

Project description:It is important to understand the balance between cortical plasticity and stability in various systems and across spatial scales in the adult brain. Here we review studies of adult plasticity in primary visual cortex (V1), which has a key role in distributing visual information. There are claims of plasticity at multiple spatial scales in adult V1, but a number of inconsistencies in the supporting data raise questions about the extent and nature of such plasticity. Our understanding of the extent of plasticity in V1 is further limited by a lack of quantitative models to guide the interpretation of the data. These problems limit efforts to translate research findings about adult cortical plasticity into significant clinical, educational and policy applications.

Modulation of visual responses by behavioral state in mouse visual cortex.

Project description:Studies of visual processing in rodents have conventionally been performed on anesthetized animals, precluding examination of the effects of behavior on visually evoked responses. We have now studied the response properties of neurons in primary visual cortex of awake mice that were allowed to run on a freely rotating spherical treadmill with their heads fixed. Most neurons showed more than a doubling of visually evoked firing rate as the animal transitioned from standing still to running, without changes in spontaneous firing or stimulus selectivity. Tuning properties in the awake animal were similar to those measured previously in anesthetized animals. Response magnitude in the lateral geniculate nucleus did not increase with locomotion, demonstrating that the striking change in responsiveness did not result from peripheral effects at the eye. Interestingly, some narrow-spiking cells were spontaneously active during running but suppressed by visual stimuli. These results demonstrate powerful cell-type-specific modulation of visual processing by behavioral state in awake mice.

Plasticity in Adult Mouse Visual Cortex Following Optic Nerve Injury.

Project description:Optic nerve (ON) injury is an established model of axonal injury which results in retrograde degeneration and death of retinal ganglion cells as well anterograde loss of transmission and Wallerian degeneration of the injured axons. While the local impact of ON crush has been extensively documented we know comparatively little about the functional changes that occur in higher visual structures such as primary visual cortex (V1). We explored the extent of adult cortical plasticity using ON crush in aged mice. V1 function of the contralateral hemisphere was assessed longitudinally by intrinsic signal imaging and 2-photon calcium imaging before and after ON crush. Functional imaging demonstrated an immediate shift in V1 ocular dominance towards the ipsilateral, intact eye, due to the expected almost complete loss of responses to contralateral eye stimulation. Surprisingly, within 2 weeks we observed a delayed increase in ipsilateral eye responses. Additionally, spontaneous activity in V1 was reduced, similar to the lesion projection zone after retinal lesions. The observed changes in V1 activity indicate that severe ON injury in adulthood evokes cortical plasticity not only cross-modally but also within the visual cortex; this plasticity may be best compared with that seen after retinal lesions.

Rapid and cumulative adult plasticity in the mouse visual cortex.

Project description:Experience-dependent neural plasticity enables the brain to adapt to diverse and dynamic environments by reshaping circuits. In the adult visual system, this plasticity can be elicited by repeated sensory stimuli; however, its temporal dynamics and underlying mechanisms remain unclear. Here, we investigated the regulation of visual response potentiation induced by repeated light flashes in the primary visual cortex of awake adult mice. Our findings revealed two distinct temporal phases of potentiation: a rapid phase occurring within seconds and a cumulative phase developing over hours to days. Notably, the identification of this rapid phase phenomenon adds to and refines the prevailing view that visual plasticity in the adult cortex is predominantly slow. Additionally, exposure to visual stimuli enhanced spontaneous slow-wave activity in the visual cortex during non-REM sleep. This plasticity was significantly impaired in Grin2a (NR2A) knockout mice, a model of schizophrenia, which mirrors visual plasticity deficits observed in human patients. The dual temporal characteristics of flash-evoked visual plasticity likely reflect multifaceted aspects of adult brain functionality, encompassing processes related to memory, learning, and neurological disorders. This model of visual plasticity in defined neural circuits provides a simplified yet robust and extensible framework for exploring the neural mechanisms underlying adaptive and maladaptive behavioral changes.

Subnetwork-Specific Homeostatic Plasticity in Mouse Visual Cortex In Vivo.

Project description:Homeostatic regulation has been shown to restore cortical activity in vivo following sensory deprivation, but it is unclear whether this recovery is uniform across all cells or specific to a subset of the network. To address this issue, we used chronic calcium imaging in behaving adult mice to examine the activity of individual excitatory and inhibitory neurons in the same region of the layer 2/3 monocular visual cortex following enucleation. We found that only a fraction of excitatory neurons homeostatically recover activity after deprivation and inhibitory neurons show no recovery. Prior to deprivation, excitatory cells that did recover were more likely to have significantly correlated activity with other recovering excitatory neurons, thus forming a subnetwork of recovering neurons. These network level changes are accompanied by a reduction in synaptic inhibition onto all excitatory neurons, suggesting that both synaptic mechanisms and subnetwork activity are important for homeostatic recovery of activity after deprivation.

Achromatopsia-Visual Cortex Stability and Plasticity in the Absence of Functional Cones.

Project description:PurposeAchromatopsia is a rare inherited disorder rendering retinal cone photoreceptors nonfunctional. As a consequence, the sizable foveal representation in the visual cortex is congenitally deprived of visual input, which prompts a fundamental question: is the cortical representation of the central visual field in patients with achromatopsia remapped to take up processing of paracentral inputs? Such remapping might interfere with gene therapeutic treatments aimed at restoring cone function.MethodsWe conducted a multicenter study to explore the nature and plasticity of vision in the absence of functional cones in a cohort of 17 individuals affected by autosomal recessive achromatopsia and confirmed biallelic disease-causing CNGA3 or CNGB3 mutations. Specifically, we tested the hypothesis of foveal remapping in human achromatopsia. For this purpose, we applied two independent functional magnetic resonance imaging (fMRI)-based mapping approaches, i.e. conventional phase-encoded eccentricity and population receptive field mapping, to separate data sets.ResultsBoth fMRI approaches produced the same result in the group comparison of achromatopsia versus healthy controls: sizable remapping of the representation of the central visual field in the primary visual cortex was not apparent.ConclusionsRemapping of the cortical representation of the central visual field is not a general feature in achromatopsia. It is concluded that plasticity of the human primary visual cortex is less pronounced than previously assumed. A pretherapeutic imaging workup is proposed to optimize interventions.

Microglial P2Y12 is necessary for synaptic plasticity in mouse visual cortex.

Project description:Microglia are the resident immune cells of the brain. Increasingly, they are recognized as important mediators of normal neurophysiology, particularly during early development. Here we demonstrate that microglia are critical for ocular dominance plasticity. During the visual critical period, closure of one eye elicits changes in the structure and function of connections underlying binocular responses of neurons in the visual cortex. We find that microglia respond to monocular deprivation during the critical period, altering their morphology, motility and phagocytic behaviour as well as interactions with synapses. To explore the underlying mechanism, we focused on the P2Y12 purinergic receptor, which is selectively expressed in non-activated microglia and mediates process motility during early injury responses. We find that disrupting this receptor alters the microglial response to monocular deprivation and abrogates ocular dominance plasticity. These results suggest that microglia actively contribute to experience-dependent plasticity in the adolescent brain.

Visual Sequences Drive Experience-Dependent Plasticity in Mouse Anterior Cingulate Cortex.

Project description:Mechanisms of experience-dependent plasticity have been well characterized in mouse primary visual cortex (V1), including a form of potentiation driven by repeated presentations of a familiar visual sequence ("sequence plasticity"). The prefrontal anterior cingulate cortex (ACC) responds to visual stimuli, yet little is known about if and how visual experience modifies ACC circuits. We find that mouse ACC exhibits sequence plasticity, but in contrast to V1, the plasticity expresses as a change in response timing, rather than a change in response magnitude. Sequence plasticity is absent in ACC, but not V1, in a mouse model of a neurodevelopmental disorder associated with intellectual disability and autism-like features. Our results demonstrate that simple sensory stimuli can be used to reveal how experience functionally (or dysfunctionally) modifies higher-order prefrontal circuits and suggest a divergence in how ACC and V1 encode familiarity.

Compartmentalized dendritic plasticity in the mouse retrosplenial cortex links contextual memories formed close in time.

Project description:Events occurring close in time are often linked in memory, and recent studies suggest that such memories are encoded by overlapping neuronal ensembles. However, the role of dendritic plasticity mechanisms in linking memories is unknown. Here we show that memory linking is dependent not only on neuronal ensemble overlap in the mouse retrosplenial cortex, but also on branch-specific dendritic allocation mechanisms. The same dendritic segments are preferentially activated by two linked (but not independent) contextual memories, and spine clusters added after each of two linked (but not independent) contextual memories are allocated to the same dendritic segments. Importantly, we show that the reactivation of dendrites activated during the first context exploration is sufficient to link two contextual memories. Our results demonstrate a critical role for localized dendritic plasticity in memory integration and reveal rules governing how linked and independent memories are allocated to dendritic compartments.
