Proximate mechanism of behavioral manipulation of an orb-weaver spider host by a parasitoid wasp.

ABSTRACT: Some ichneumonid wasps induce modifications in the web building behavior of their spider hosts to produce resistant "cocoon" webs. These structures hold and protect the wasp's cocoon during pupa development. The mechanism responsible for host manipulation probably involves the inoculation of psychotropic chemicals by the parasitoid larva during a specific developmental period. Recent studies indicate that some spiders build cocoon webs similar to those normally built immediately before ecdysis, suggesting that this substance might be a molting hormone or a precursor chemical of this hormone. Here, we report that Cyclosa spider species exhibiting modified behavior presented higher 20-OH-ecdysone levels than parasitized spiders acting normally or unparasitized individuals. We suggest that the lack of control that spiders have when constructing modified webs can be triggered by anachronic activation of ecdysis.

An RNA Virome Associated to the Golden Orb-Weaver Spider Nephila clavipes.

Project description:The golden orb-weaver spider Nephila clavipes, known for its sexual size dimorphism, is abundant and widespread in the New World. The first annotated genome of orb-weaver spiders, exploring N. clavipes, has recently been reported. The study, focused primarily on the diversity of silk specific genes, shed light into the complex evolutionary history of spiders. Furthermore, a robust transcriptome analysis provided a massive resource for N. clavipes RNA survey. Here, I present evidence of viral sequences corresponding to the first 10 extant virus species associated to N. clavipes and indeed, nephilids. The putatively new species are linked to ssRNA positive-strand viruses, such as Picornavirales, and to ssRNA negative-strand and dsRNA viruses. In addition, I detected sequence data of new strains of two recently reported arthropod viruses, which complemented and extended the corresponding sequence references. The identified viruses appear to be complete, potentially functional, and presenting the typical architecture and consistent viral domains. The intrinsic nature of the detected sequences and their absence in the recently generated genome assembly, suggest that they correspond to bona fide RNA virus sequences. The available RNA data allowed for the first time to address a tissue/organ specific analysis of virus loads/presence in spiders, suggesting a complex spatial and differential distribution of the tentative viruses, encompassing the spider brain and also silk and venom glands. Until recently, the virus landscape associated to spiders remained elusive. The discovered viruses described here provide only a fragmented glimpse of the potential magnitude of the Aranea virosphere. Future studies should focus not only on complementing and expanding these findings, but also on addressing the potential ecological role of these viruses, which might influence the biology of these outstanding arthropod species.

The complete mitochondrial genome of an orb-weaver spider Araneus angulatus (Araneae: Araneidae).

Project description:The complete mitochondrial genome of an orb-weaver spider Araneus angulatus is 14,205 bp in length and contains 13 protein-coding genes, 2 ribosomal RNAs, 22 transfer RNAs, and a control region. The A + T content of the major strand is 75.4%. Nine of the protein-coding genes are initiated by typical ATN codons, one gene (COI) starts with TTA and other three genes (ND6, COII, and COIII) begin with TTG. The truncated stop codon (T) occurs in ND3, ND4L, and ND6, whereas the rest ten genes end with the canonical stop codon (TAA and TAG). Fifteen tRNAs lack the potential to form the typically cloverleaf-shaped secondary structure. The control region is 649 bp in length and contains a long tandem repeat region. The result of phylogenetic analysis shows that the relationship of A. angulatus was close to the species in the same family Araneidae.

Age variation in the body coloration of the orb-weaver spider Alpaida tuonabo and its implications on foraging.

Project description:Spiders show a repertoire of strategies to increase their foraging success. In particular, some orb-weaver spiders use attractive body colorations to lure prey. Interestingly, coloration varies with age in many species, which may result in ontogenetic variation of foraging success. By using field observations, laboratory experiments and spectrophotometric analysis, we investigated whether pale juveniles and bright adults of the orb-weaver Alpaida tuonabo use different foraging strategies due to ontogenetic variation in coloration. Field observations revealed that foraging success of juveniles and adults was influenced by web properties. However, foraging success increased with body size only in adults, supporting the idea that larger individuals produce a stronger visual signal for prey. The attractiveness of the adult coloration for prey was confirmed in the laboratory with frame-web-choice experiments, in which webs bearing a spider intercepted more bees than empty webs. Our spectrophotometric analysis suggests that the yellow coloration may produce the deceiving signal for prey. Moreover, we identified potential alternative foraging strategies: cryptic juveniles at higher heights and 'attractive' adults at lower heights. This study reveals how ontogenetic colour variation may favour the use of alternative foraging strategies in orb-weaver spiders and reduces intraspecific competition.

Parasitoid wasp usurps its host to guard its pupa against hyperparasitoids and induces rapid behavioral changes in the parasitized host.

Project description:Some parasites have an ability to fabricate the behavior of their host and impel the host to guard parasites' offspring, which is popularly called as bodyguard manipulation. Psalis pennatula larva parasitized by a braconid parasitoid wasp Microplitis pennatula exhibits some behavioral changes including the guarding of the parasitoid pupa from its natural enemies. We hypothesized that these behavioral change exhibited by the parasitized host larva are induced by the parasitoid and can be considered as an example of bodyguard manipulation. Even though hyperparasitoids are the more specialized natural enemy of parasitoids than predators, very few studies tested the success of guarding parasitoid pupa against hyperparasitoids. This study analyzed the success of guarding behavior of the parasitized host against hyperparasitoids. The onsets of parasite-induced phenotypic alterations (PIPAs) in the parasitized host were inspected to analyze whether these behavioral changes in the host larva manifests gradually or abruptly. The study concludes that parasitized host larva defends the parasitoid pupa from hyperparasitoids and the PIPAs in the parasitized host develops abruptly only after the egression of parasitoid prepupa.

Suppression of host nocifensive behavior by parasitoid wasp venom.

Project description:The parasitoid wasp Ampulex compressa envenomates the brain of its host the American cockroach (Periplaneta americana), thereby making it a behaviorally compliant food supply for its offspring. The target of venom injection is a locomotory command center in the brain called the central complex. In this study, we investigate why stung cockroaches do not respond to injuries incurred during the manipulation process by the wasp. In particular, we examine how envenomation compromises nociceptive signaling pathways in the host. Noxious stimuli applied to the cuticle of stung cockroaches fail to evoke escape responses, even though nociceptive interneurons projecting to the brain respond normally. Hence, while nociceptive signals are carried forward to the brain, they fail to trigger robust nocifensive behavior. Electrophysiological recordings from the central complex of stung animals demonstrate decreases in peak firing rate, total firing, and duration of noxious-evoked activity. The single parameter best correlated with altered noxious-evoked behavioral responses of stung cockroaches is reduced duration of the evoked response in the central complex. Our findings demonstrate how the reproductive strategy of a parasitoid wasp is served by venom-mediated elimination of aversive, nocifensive behavior in its host.

Biomaterial evolution parallels behavioral innovation in the origin of orb-like spider webs.

Project description:Correlated evolution of traits can act synergistically to facilitate organism function. But, what happens when constraints exist on the evolvability of some traits, but not others? The orb web was a key innovation in the origin of >12,000 species of spiders. Orb evolution hinged upon the origin of novel spinning behaviors and innovations in silk material properties. In particular, a new major ampullate spidroin protein (MaSp2) increased silk extensibility and toughness, playing a critical role in how orb webs stop flying insects. Here, we show convergence between pseudo-orb-weaving Fecenia and true orb spiders. As in the origin of true orbs, Fecenia dragline silk improved significantly compared to relatives. But, Fecenia silk lacks the high compliance and extensibility found in true orb spiders, likely due in part to the absence of MaSp2. Our results suggest how constraints limit convergent evolution and provide insight into the evolution of nature's toughest fibers.
