Nanostar Clustering Improves the Sensitivity of Plasmonic Assays.

ABSTRACT: Star-shaped Au nanoparticles (Au nanostars, AuNS) have been developed to improve the plasmonic sensitivity, but their application has largely been limited to single-particle probes. We herein describe a AuNS clustering assay based on nanoscale self-assembly of multiple AuNS and which further increases detection sensitivity. We show that each cluster contains multiple nanogaps to concentrate electric fields, thereby amplifying the signal via plasmon coupling. Numerical simulation indicated that AuNS clusters assume up to 460-fold higher field density than Au nanosphere clusters of similar mass. The results were validated in model assays of protein biomarker detection. The AuNS clustering assay showed higher sensitivity than Au nanosphere. Minimizing the size of affinity ligand was found important to tightly confine electric fields and improve the sensitivity. The resulting assay is simple and fast and can be readily applied to point-of-care molecular detection schemes.

Plasmonic nanostar photocathodes for optically-controlled directional currents.

Project description:Plasmonic nanocathodes offer unique opportunities for optically driving, switching, and steering femtosecond photocurrents in nanoelectronic devices and pulsed electron sources. However, angular photocurrent distributions in nanoplasmonic systems remain poorly understood and are therefore difficult to anticipate and control. Here, we provide a direct momentum-space characterization of multiphoton photoemission from plasmonic gold nanostars and demonstrate all-optical control over these currents. Versatile angular control is achieved by selectively exciting different tips on single nanostars via laser frequency or linear polarization, thereby rotating the tip-aligned directional photoemission as observed with angle-resolved 2D velocity mapping and 3D reconstruction. Classical plasmonic field simulations combined with quantum photoemission theory elucidate the role of surface-mediated nonlinear excitation for plasmonic field enhancements highly concentrated at the sharp tips (Rtip = 3.4 nm). We thus establish a simple mechanism for femtosecond spatiotemporal current control in designer nanosystems.

Feasibility of NAD(P)/NAD(P)H as redox agents in enzymatic plasmonic gold nanostar assays for galactose quantification.

Project description:Plasmonic colorimetric sensors have emerged as powerful analytical tools in biochemistry due to their localized surface plasmon resonance extinction in the visible range. Here, we describe the feasibility of NAD(P)/NAD(P)H as redox agents in enzymatic plasmonic gold nanostar (AuNS) assays for galactose quantification using three model enzymes, GalDH, AR and GalOx, immobilized separately on polyvinylpyrrolidone-capped AuNS scaffolds. These highly specific, sensitive and selective bioassays induce the transformation of AuNS into quasi-spherical nanoparticles during the biorecognition of galactose in water and synthetic blood matrices. As a result, using our inexpensive and simple AuNS plasmon bioassays, the presence of galactose may be detected spectrophotometrically and by the naked eye.

A Plasmonic Gold Nanostar Theranostic Probe for In Vivo Tumor Imaging and Photothermal Therapy.

Project description:Nanomedicine has attracted increasing attention in recent years, because it offers great promise to provide personalized diagnostics and therapy with improved treatment efficacy and specificity. In this study, we developed a gold nanostar (GNS) probe for multi-modality theranostics including surface-enhanced Raman scattering (SERS) detection, x-ray computed tomography (CT), two-photon luminescence (TPL) imaging, and photothermal therapy (PTT). We performed radiolabeling, as well as CT and optical imaging, to investigate the GNS probe's biodistribution and intratumoral uptake at both macroscopic and microscopic scales. We also characterized the performance of the GNS nanoprobe for in vitro photothermal heating and in vivo photothermal ablation of primary sarcomas in mice. The results showed that 30-nm GNS have higher tumor uptake, as well as deeper penetration into tumor interstitial space compared to 60-nm GNS. In addition, we found that a higher injection dose of GNS can increase the percentage of tumor uptake. We also demonstrated the GNS probe's superior photothermal conversion efficiency with a highly concentrated heating effect due to a tip-enhanced plasmonic effect. In vivo photothermal therapy with a near-infrared (NIR) laser under the maximum permissible exposure (MPE) led to ablation of aggressive tumors containing GNS, but had no effect in the absence of GNS. This multifunctional GNS probe has the potential to be used for in vivo biosensing, preoperative CT imaging, intraoperative detection with optical methods (SERS and TPL), as well as image-guided photothermal therapy.

Label-free plasmonic nanostar probes to illuminate <i>in vitro</i> membrane receptor recognition.

Project description:Protein-ligand recognition is a key activity where chemical signals are communicated to cells to activate various biochemical pathways, which are important for understanding membrane signaling and drug interactions. Gold nanostars are highly attractive for biological applications due to their readily modified surface chemistry, facile synthesis and optical properties. The increase in electromagnetic field at their branches increases the surface enhanced Raman scattering (SERS) making them ideal candidates as label free in vitro probes that can be used to detect a variety of cellular activities. However, the use of particles in vitro is complicated by the adsorption of proteins, which forms the protein corona. In this paper we demonstrate gold nanostars as label free in vitro probes to study the interaction between αvβ3 integrin and RGD. Nanostars functionalized with cyclic-RDGFC reduced the formation of the protein corona, due to its zwitterionic nature, indicating a small peptide approach to minimizing protein absorption. Additionally, the functionalized nanostars evince a SERS response from their interaction with αvβ3 integrin representative of the amino acids present at the binding site which is also retained in a complex biological matrix. The nanostars were used in vitro to selectively detect αvβ3 integrin on the membrane of human metastatic colon cancer cells. By exploiting the intense SERS and tunable plasmon resonance properties of gold nanostars functionalized with cyclic RGDFC, we have demonstrated a label free approach to investigate the chemical interactions associated with protein-ligand binding from both purified proteins and membrane bound receptors in cells.

Enhanced Sensitivity of Delocalized Plasmonic Nanostructures.

Project description:This work reports on the observation of a delocalized surface plasmon resonance (DSPR) phenomenon in linear chains of square-shaped silver nanoparticles (NP) as a function of the chain length and the distance between the nanoparticles in the chain. Transmission spectra of the silver nanoparticle chains reveal the emergence of new, red-shifted extinction peaks that depend strongly on the spacing between the nanoparticles and the polarization of the exciting light with respect to the chain axis. As the spacing between the nanoparticles in the linear chain decreases and the number of nanoparticles in the linear chain increases, the strength of the new extinction features increase strongly. These changes can be described by a tight-binding model for the coupled chain, which indicates that the origin of the phenomenon is consistent with an increased coupling between the nanoparticles. FDTD calculations reveal that the electric field is strongly enhanced between the nanoparticles in the chain. The DSPR response is found to be much more sensitive to dielectric changes than the localized surface plasmon resonance (LSPR).

Multiplexed Biosensing of Proteins and Virions with Disposable Plasmonic Assays.

Project description:Our growing ability to tailor healthcare to the needs of individuals has the potential to transform clinical treatment. However, the measurement of multiple biomarkers to inform clinical decisions requires rapid, effective, and affordable diagnostics. Chronic diseases and rapidly evolving pathogens in a larger population have also escalated the need for improved diagnostic capabilities. Current chemical diagnostics are often performed in centralized facilities and are still dependent on multiple steps, molecular labeling, and detailed analysis, causing the result turnaround time to be over hours and days. Rapid diagnostic kits based on lateral flow devices can return results quickly but are only capable of detecting a handful of pathogens or markers. Herein, we present the use of disposable plasmonics with chiroptical nanostructures as a platform for low-cost, label-free optical biosensing with multiplexing and without the need for flow systems often required in current optical biosensors. We showcase the detection of SARS-CoV-2 in complex media as well as an assay for the Norovirus and Zika virus as an early developmental milestone toward high-throughput, single-step diagnostic kits for differential diagnosis of multiple respiratory viruses and any other emerging diagnostic needs. Diagnostics based on this platform, which we term "disposable plasmonics assays," would be suitable for low-cost screening of multiple pathogens or biomarkers in a near-point-of-care setting.

Enhanced sensitivity in THz plasmonic sensors with silver nanowires.

Project description:We developed hybrid slot antenna structures for microbial sensing in the THz frequency range, where silver nanowires (AgNWs) were employed to increase the sensitivity. In order to fabricate the hybrid devices, we partially etched the AgNW in the slot antenna region, where we can expect the field enhancement effect at the AgNW tip. We measured the resonant-frequency shift observed upon the deposition of a polymer layer, and observed that the sensitivity increased upon the introduction of AgNWs, with an enhancement factor of more than four times (approximately six times in terms of figure-of-merit). The sensitivity increased with the AgNW density until saturation. In addition, we tested devices with PRD1 viruses, and obtained an enhancement factor of 3.4 for a slot antenna width of 3??m. Furthermore, we performed finite-difference time-domain simulations, which confirmed the experimental results. The sensitivity enhancement factor decreased with the decrease of the slot width, consistent with the experimental findings. Two-dimensional mapping of the electric field confirmed the strong field localization and enhancement at the AgNW tips.

MNase and HpaII sensitivity assays in MCF7 breast cancer cells

Project description:DNA are packaged into nucleosomes and chromatin. We performed incomplete MNase digestion of chromatin to identify nucleosome-free regions that may indicate active promoters and regulatory regions. Additionally, HpaII digestion was performed which cleaves CCGG sites when the internal C remains unmodified. The hypomethylation state and nuclease sensitivity of the chromatin are indicators of transcription regulatory regions.

Sombrero-shaped plasmonic nanoparticles with molecular-level sensitivity and multifunctionality.

Project description:We demonstrate top-down synthesis of monodisperse plasmonic nanoparticles designed to contain internal Raman hot spots. Our Raman-active nanoparticles are fabricated using nanoimprint lithography and thin-film deposition and are composed of novel internal structures with sublithographic dimensions: a disk-shaped Ag core, a Petri-dish-shaped SiO(2) base whose inner surface is coated with Ag film, and a sub-10 nm scale circular gap between the core and the base. Confocal Raman measurements and electromagnetic simulations show that Raman hot spots appear at the inside perimeter of individual nanoparticles and serve as the source of a 1000-fold improvement of minimum molecular detection level that enables detection of signals from a few molecules near hot spots. A multimodality version of these nanoparticles, which includes the functionality offered by magnetic multilayers, is also demonstrated. These results illustrate the potential of direct fabrication for creating exotic monodisperse nanoparticles, which combine engineered internal nanostructures and multilayer composite materials, for use in nanoparticle-based molecular imaging and detection.

Enhancing the Angular Sensitivity of Plasmonic Sensors Using Hyperbolic Metamaterials.

Project description:Surface plasmon resonance (SPR) sensors operate mainly on prism and grating coupling techniques, with spectral and angular scans being the two major interrogation schemes. Among them, the angular scan technique has several advantages including higher measurement precision owing to its higher signal-to-noise ratio. The currently available SPR sensor arrangements provide a maximum angular sensitivity of 500°-600° per RIU. Here, we report the study of grating coupled-hyperbolic metamaterial (GC-HMM) sensors with high angular sensitivity. The experimental studies show extraordinary angular sensitivities from visible to near infrared (NIR) wavelengths by exciting bulk plasmon polaritons associated with hyperbolic metamaterials, with a maximum of 7000° per RIU. This angular-scan plasmonic biosensor has been used for the detection of low molecular weight biomolecules such as biotin (244 Da) and high molecular weight macromolecules such as Cowpea mosaic virus (CPMV, 5.6 × 106 Da) at ultralow concentrations. The miniaturized sensing device can be integrated with microfluidic systems for the development of next-generation biosensors for lab-on-a-chip and point-of-care applications.
