Molecular dietary analysis of two sympatric felids in the Mountains of Southwest China biodiversity hotspot and conservation implications.

ABSTRACT: Dietary information is lacking in most of small to mid-sized carnivores due to their elusive predatory behaviour and versatile feeding habits. The leopard cat (LPC; Prionailurus bengalensis) and the Asiatic golden cat (AGC; Catopuma temminckii) are two important yet increasingly endangered carnivore species in the temperate mountain forest ecosystem in Southwest China, a global biodiversity hotspot and a significant reservoir of China's endemic species. We investigated the vertebrate prey of the two sympatric felids using faecal DNA and a next-generation sequencing (NGS)/metabarcoding approach. Forty vertebrate prey taxa were identified from 93 LPC and 10 AGC faecal samples; 37 taxa were found in the LPC diet, and 20 were detected in the AGC diet. Prey included 27 mammalian taxa, 11 birds, one lizard and one fish, with 73% (29/40) of the taxa assigned to the species level. Rodents and pikas were the most dominant LPC prey categories, whereas rodents, pheasant, fowl and ungulates were the main AGC prey. We also analysed the seasonal and altitudinal variations in the LPC diet. Our results provide the most comprehensive dietary data for these felids and valuable information for their conservation planning.

Project description:Simple Summary Camera-trapping technology has been widely applied to obtain survey data and enhance understanding of animal ecology. Ground-dwelling pheasants with limited distributions and weak dispersal capacity are prone to extinction due to disturbances and climate change in high-altitude mountain areas. The Qilian Mountains form a global biodiversity hotspot for endemic species and contain crucial areas for ecological and biodiversity conservation. The Blue Eared Pheasant (EP) and Blood Pheasant (BP) are indicator species of the environment and currently occur in the Qilian Mountain National Nature Reserve (QMNNR). Understanding their stable coexistence is key for making informed conservation and management actions. They have similar daily activity patterns but their monthly activity patterns are strikingly different. Both BP and EP prefer forest habitats but BP nests in more dense vegetation cover. Ninety-one percent of BP distribution falls within EP distribution in the QMNNR. Their areas of potential overlap are in the central and eastern parts of the QMNNR, but landscape connectivity is relatively poor. This study further improved the understanding of the basic knowledge of BP and EP coexistence. Conservation actions should give priority to those highly overlapping areas and strengthen forest landscape connectivity, as they provide irreplaceable habitats for threatened Galliformes. Abstract Studying the spatio-temporal niche partitioning among closely related sympatric species is essential for understanding their stable coexistence in animal communities. However, consideration of niche partitioning across multiple ecological dimensions is still poor for many sympatric pheasant species. Here, we studied temporal activity patterns and spatial distributions of the Blue Eared Pheasant (EP, Crossoptilon auritum) and Blood Pheasant (BP, Ithaginis cruentus) in the Qilian Mountains National Nature Reserve (QMNNR), Northwestern China, using 137 camera traps from August 2017 to August 2020. Kernel density estimation was applied to analyze diel activity patterns, and the Maxent model was applied to evaluate their suitable distributions and underlying habitat preferences. Eight Galliformes species were captured in 678 detection records with 485 records of EP and 106 records of BP over a total of 39,206 camera days. Their monthly activity frequencies demonstrate temporal partitioning but their diel activity patterns do not. Furthermore, 90.78% of BP distribution (2867.99 km2) overlaps with the distribution of EP (4355.86 km2) in the QMNNR. However, BP manifests a high dependence on forest habitats and shows larger Normalized Difference Vegetation Index (NDVI) values, while EP showed obvious avoidance of forest with NDVI greater than 0.75. Hence, differentiation in monthly activity patterns and partitioning in habitat preference might facilitate their coexistence in spatiotemporal dimensions. Conservation actions should give priority to highly overlapping areas in the center and east of the QMNNR and should strengthen forest landscape connectivity, as they provide irreplaceable habitats for these threatened and endemic Galliformes.

Project description:Previous attempts to elucidate the drivers of speciation mechanisms and spatial distribution patterns of biodiversity in mountain regions have treated different floras within a single geological region as one flora, ignoring the potential contributions of high habitat/ecosystem heterogeneity. Furthermore, current conservation strategies largely focus on forest ecosystems and/or specific flagship species, ignoring marginal ecosystems, leaving species in these ecosystems at risk. Here, we compared the spatial patterns of biodiversity and the potential drivers of these patterns in the river valley and subnival ecosystems of the Hengduan Mountains region (HDM) in southwestern China. Specifically, we compared spatial patterns of diversity, endemism, and threatened species in these ecosystems based on both traditional measurements and recent phylogenetic approaches. We then examined how those patterns were related to environmental factors and human activity in these same regions. We found that the middle-southern HDM supports the highest diversity and endemism for the river valley and subnival ecosystems; however, the distribution patterns of neo- and paleo-endemism in these two ecosystems differ. Regression models indicate that habitat diversity and paleo-climatic fluctuation are important drivers of diversity and endemism for these two ecosystems. Temperature and precipitation, however, showed different influences on the spatial patterns in different ecosystems. Categorical analysis of neo- and paleo-endemism (CANAPE) indicated that most endemism centers are not covered by current nature reserves. Moreover, the intensity of human activity is highest in the southern and southeastern HDM, which coincides with the distribution patterns of diversity, mixed-endemism and high-priority (and threatened) species. These findings suggest that different floras within a single geographic/floristic region respond differently to environmental factors and show different spatial phylogenetic patterns. We, therefore, recommend that future research into the drivers of biodiversity consider the contributions of various ecosystem types within a single geological region. This study also provides a theoretical basis for protecting habitat diversity. Our work confirms that current conservation efforts are insufficient to protect ecosystem diversity in the river valley and subnival ecosystems of the Hengduan Mountains. Therefore, we recommend the establishment of nature reserves in the regions identified in this study; furthermore, we strongly recommend improving current and establishing new management policies for biodiversity conservation in this region.
