Project description:Functional connectivity (FC) has become a leading method for resting-state functional magnetic resonance imaging (rs-fMRI) analysis. However, the majority of the previous studies utilized pairwise, temporal synchronization-based FC. Recently, high-order FC (HOFC) methods were proposed with the idea of computing "correlation of correlations" to capture high-level, more complex associations among the brain regions. There are two types of HOFC. The first type is topographical profile similarity-based HOFC (tHOFC) and its variant, associated HOFC (aHOFC), for capturing different levels of HOFC. Instead of measuring the similarity of the original rs-fMRI signals with the traditional FC (low-order FC, or LOFC), tHOFC measures the similarity of LOFC profiles (i.e., a set of LOFC values between a region and all other regions) between each pair of brain regions. The second type is dynamics-based HOFC (dHOFC) which defines the quadruple relationship among every four brain regions by first calculating two pairwise dynamic LOFC "time series" and then measuring their temporal synchronization (i.e., temporal correlation of the LOFC fluctuations, not the BOLD fluctuations). Applications have shown the superiority of HOFC in both disease biomarker detection and individualized diagnosis than LOFC. However, no study has been carried out for the assessment of test-retest reliability of different HOFC metrics. In this paper, we systematically evaluate the reliability of the two types of HOFC methods using test-retest rs-fMRI data from 25 (12 females, age 24.48 ± 2.55 years) young healthy adults with seven repeated scans (with interval = 3-8 days). We found that all HOFC metrics have satisfactory reliability, specifically (1) fair-to-good for tHOFC and aHOFC, and (2) fair-to-moderate for dHOFC with relatively strong connectivity strength. We further give an in-depth analysis of the biological meanings of each HOFC metric and highlight their differences compared to the LOFC from the aspects of cross-level information exchanges, within-/between-network connectivity, and modulatory connectivity. In addition, how the dynamic analysis parameter (i.e., sliding window length) affects dHOFC reliability is also investigated. Our study reveals unique functional associations characterized by the HOFC metrics. Guidance and recommendations for future applications and clinical research using HOFC are provided. This study has made a further step toward unveiling more complex human brain connectome.

Project description:Recently, magnetic resonance imaging (MRI) has been widely used to investigate the structures and functions of the human brain in health and disease in vivo. However, there are growing concerns about the test-retest reliability of structural and functional measurements derived from MRI data. Here, we present a test-retest dataset of multi-modal MRI including structural MRI (S-MRI), diffusion MRI (D-MRI) and resting-state functional MRI (R-fMRI). Fifty-seven healthy young adults (age range: 19-30 years) were recruited and completed two multi-modal MRI scan sessions at an interval of approximately 6 weeks. Each scan session included R-fMRI, S-MRI and D-MRI data. Additionally, there were two separated R-fMRI scans at the beginning and at the end of the first session (approximately 20 min apart). This multi-modal MRI dataset not only provides excellent opportunities to investigate the short- and long-term test-retest reliability of the brain's structural and functional measurements at the regional, connectional and network levels, but also allows probing the test-retest reliability of structural-functional couplings in the human brain.

Project description:SignificanceWe recently developed a novel Bayesian adaptive method, qReading, to measure reading function. The qReading method has both the efficiency and excellent test-retest reliability in normally sighted young adults to make it an excellent candidate for future studies of its value in diagnosis and longitudinal evaluation of treatment and/or rehabilitation outcomes.PurposeA novel Bayesian adaptive method, qReading, was recently developed to measure reading function. Here we performed a systematic assessment of the test-retest reliability of the qReading method.MethodsThe variability of five repeated measurements of the reading curve was examined in two settings: within session and between sessions. For the within-session design, we considered two subpopulations: naive observers and experienced observers. All observers were normally sighted young adults. For each set of data, in addition to examining the intrinsic precision of the qReading method (the half width of the credible interval of the posterior distribution of the estimated performance), we computed four metrics to assess repeatability: standard deviation, Bland-Altman coefficient of repeatability, correlation coefficient, and Fractional Rank Precision.ResultsExtrinsic factors such as observer, time interval between repeated measures, and observer experience all contribute to the variation across measurements. Nevertheless, the four metrics consistently show that the variability across five repeated measurements is small for each set of data. This is true even without taking learning effects into account (standard deviations, ≤0.092 log10 units; Bland-Altman coefficient of repeatability, ≤0.15 (log10)2 units; correlation coefficient, ≥0.91; and Fractional Rank Precision, ≥0.81).ConclusionsThe qReading method has excellent test-retest reliability in normally sighted young adults.

Project description:BACKGROUND:No previous studies have investigated the test-retest reliability of tip, key, and palmar pinch force sense in healthy adults. The present study explores the test-retest reliability of tip, key, and palmar pinch force sense for different force levels in healthy adults during an ipsilateral force reproduction task. METHODS:Fifty-six healthy subjects were instructed to produce varying levels of reference forces (10, 30, and 50% maximal voluntary isometric contraction (MVIC)) using three types of pinches (tip pinch, palmar pinch, and key pinch) and to reproduce these forces using the same hand. The subjects were tested twice by the same experienced testers, 1?week apart. RESULTS:Based on the high values of the intraclass correlation coefficient (ICC), the tip pinch (0.783-0.895) and palmar pinch (0.752-0.903) force sense tests demonstrated good reliability for all the variables. The ICCs for the key pinch (0.712-0.881) indicated fair to good relative test-retest reliability. CONCLUSION:1) This study demonstrates that high test-retest reliability of tip, key, and palmar pinch force sense in healthy adults can be achieved using standardized positioning and the proposed approach. 2) According to the reliability measurements, 30 and 50% maximal voluntary isometric contraction (MVIC) are the most reliable pinch force sense levels.

Project description:Delay discounting (DD) indexes an individual's preference for smaller immediate rewards over larger delayed rewards, and is considered a form of cognitive impulsivity. Cross-sectional studies have demonstrated that DD peaks in adolescence; longitudinal studies are needed to validate this putative developmental trend, and to determine whether DD assesses a temporary state, or reflects a more stable behavioral trait. In this study, 140 individuals aged 9-23 completed a delay discounting (DD) task and cognitive battery at baseline and every-two years thereafter, yielding five assessments over approximately 10 years. Models fit with the inverse effect of age best approximated the longitudinal trajectory of two DD measures, hyperbolic discounting (log[k]) and area under the indifference-point curve (AUC). Discounting of future rewards increased rapidly from childhood to adolescence and appeared to plateau in late adolescence for both models of DD. Participants with greater verbal intelligence and working memory displayed reduced DD across the duration of the study, suggesting a functional interrelationship between these domains and DD from early adolescence to adulthood. Furthermore, AUC demonstrated good to excellent reliability across assessment points that was superior to log(k), with both measures demonstrating acceptable stability once participants reached late adolescence. The developmental trajectories of DD we observed from childhood through young adulthood suggest that DD may index cognitive control more than reward sensitivity, and that despite modest developmental changes with maturation, AUC may be conceptualized as a trait variable related to cognitive control vs impulsivity.

Project description:Neural and behavioral mechanisms during approach-avoidance conflict decision-making are relevant across various psychiatric disorders, particularly anxiety disorders. Studies using approach-avoidance conflict paradigms in healthy adults have identified preliminary neural mechanisms, but findings must be replicated and demonstrated as reliable before further application. This study sought to replicate previous findings and examine test-retest reliability of behavioral (approach behavior, reaction time) and neural (regions of interest [ROIs]) responses during an approach-avoidance conflict task conducted during functional magnetic resonance imaging (fMRI). Thirty healthy adults completed an approach-avoidance conflict task during fMRI on two occasions (mean interval: 17 days; range: 11-32). Effects of task condition during three task phases (decision-making, affective outcome and monetary reward) and intraclass correlation coefficients (ICCs) were calculated across time points. Results replicated that approach behavior was modulated by conflict during decision-making. ROI activations were replicated such that dorsal anterior cingulate cortex (dACC) was modulated by conflict during decision-making, and dACC, striatum, and anterior insula were modulated by valence during affective outcomes (p's <.0083). Approach behavior during conflict demonstrated excellent reliability (ICCs ≥.77). Activation of dACC during conflict decision-making and anterior insula during negative outcomes demonstrated fair reliability (ICCs = .51 and .54), and dACC and striatum activation demonstrated good reliability during negative outcomes (ICCs = .63 and .69). Two additional ROIs (amygdala, left dorsolateral prefrontal cortex) showed good reliability during negative outcomes (ICCs ≥.60). These results characterize several specific behavioral and neuroimaging responses that are replicable and sufficiently reliable during approach-avoidance conflict decision-making to support future utility.

Project description:Prior teleneuropsychological research has assessed the reliability between in-person and remote administration of cognitive assessments. Few, if any, studies have examined the test-retest reliability of cognitive assessments conducted in sequential clinic-to-home or home-to-home teleneuropsychological evaluations - a critical issue given the state of clinical practice during the COVID-19 pandemic. This study examined this key psychometric question for several cognitive tests administered over repeated videoconferencing visits 4-6 months apart in a sample of healthy English-speaking adults.A total of 44 participants (ages 18-75) completed baseline and follow-up cognitive testing 4-6 months apart. Testing was conducted in a home-to-home setting over HIPAA-compliant videoconferencing meetings on participants' audio-visual enabled laptop or desktop computers. The following measures were repeated at both virtual visits: the Controlled Oral Word Association Test (FAS), Category Fluency (Animals), and Digit Span Forward and Backward from the Wechsler Adult Intelligence Scale, Fourth Edition. Intraclass correlation coefficients (ICC), Pearson correlations, root mean square difference (RMSD), and concordance correlation coefficients (CCC) were calculated as test-retest reliability metrics, and practice effects were assessed using paired-samples t-tests.Some tests exhibited small practice effects, and test-retest reliability was marginal or worse for all measures except FAS, which had adequate reliability (based on ICC and r). Reliability estimates with RMSD suggested that change within +/- 1 SD on these measures may reflect typical test-retest variability.The included cognitive measures exhibited questionable reliability over repeated home-to-home videoconferencing evaluations. Future teleneuropsychology test-retest reliability research is needed with larger, more diverse samples and in clinical populations.

Project description:BackgroundMeasuring infant and young child feeding (IYCF) indicators is important in evaluating child health programs and making evidence-based decisions. With Internet and new media rapidly developing, communication apps such as WeChat (the most popular mobile social media platform in China) are widely used and can potentially be used as an alternative way to collect infant and young child feeding information. This study compares data agreement between a WeChat-based self-administered and an interviewer-administered survey on infant and young child feeding information.MethodsWe recruited 297 mothers of children aged 6-23 months in Fenxi County, Shanxi Province, China. Using the Test-Retest method, we first collected data through a self-administered survey using a WeChat-based electronic questionnaire and asked 36 questions on breastfeeding and complementary feeding knowledge, practices, and information sources. We then conducted an interviewer-administered survey using the same questionnaire and compared the data agreement between the two survey methods during the same day. Cohen's kappa score (κ) and intraclass correlation coefficients (ICC) were used for data agreement analysis for all 36 questions and six key IYCF indicators. The McNemar test was used to identify differences between the two survey methods for the six key indicators.ResultsThere was substantial or almost perfect agreement for 33 questions (κ/ICC>0.60), and slight or fair agreement for the other 3 questions (κ/ICC<0.40). Agreement of all six key IYCF indicators was substantial or almost perfect (κ = 0.78-0.94), while two indicators showed statistical differences between the two survey methods (P = 0.03 for "Minimum meal frequency" and P = 0.001 for "Minimum accepted diet"). Analysis of reasons for inconsistencies showed that 43.6% of all the inconsistencies were not caused by the self-administered survey method. The cost of the interviewer-administered survey was much higher than that of the self-administered survey: ¥45.9 (US$6.8) vs ¥19.7 (US$2.9) per questionnaire.ConclusionsThe WeChat-based self-administered method can be used for future data collection of infant and young child feeding information in China. Most of the questions and key indicators showed very good agreement without statistical differences between the two methods.

Project description:Background and objectiveAlthough the six-minute walk test (6MWT) is widely used as a measure of exercise capacity, it may not be applicable in some settings and populations. This issue has led to increased use of the two-minute walk test (2MWT) to assess exercise capacity. The main objective of this study is to establish reference equations for the two-minute walk distance (2MWD) in healthy Chinese adults aged 18-85 years.MethodsA total of 973 volunteers took part in the study. We obtained verbal consent from all participants before the test, and the study design was approved by the ethics committees of Wenzhou People's Hospital. The participants performed two 2MWTs using a standardized protocol, and the longer distance was used for further analysis. Stepwise multiple regression analysis was performed using age, height and weight as independent variables and was used to establish the reference equations for the 2MWD in the male and female groups.ResultsThe mean walking distance for all participants was 199.1±25.81 m. Age and height were identified as independent factors that influenced the 2MWD, and they explained 35% and 34% of the variance in distance for the male and female groups, respectively.ConclusionThis study resulted in determination of reference equations for predicting the 2MWD in healthy Chinese adults. These 2MWD standards will provide useful references for medical care in some settings and populations.

Project description:In vivo dopamine D2-receptor availability is frequently assessed with 11C-raclopride and positron emission tomography. Due to low signal-to-noise ratios for 11C-raclopride in areas with low D2 receptor densities, the ligand has been considered unreliable for measurements outside the dopamine-dense striatum. Intriguingly, recent studies show that extrastriatal 11C-raclopride binding potential (BPND) values are (i) reliably higher than in the cerebellum (where D2-receptor levels are negligible), (ii) correlate with behavior in the expected direction, and (iii) showed good test-retest reliability in a sample of younger adults. The present work demonstrates high seven-month test-retest reliability of striatal and extrastriatal 11C-raclopride BPND values in healthy, older adults (n = 27, age: 64-78 years). Mean 11C-raclopride BPND values were stable between test sessions in subcortical nuclei, and in frontal and temporal cortices (p > 0.05). Across all structures analyzed, intraclass correlation coefficients were high (0.85-0.96), absolute variability was low (mean: 4-8%), and coefficients of variance ranged between 9 and 25%. Furthermore, regional 11C-raclopride BPND values correlated with previously determined 18F-fallypride BPND values (ρ = 0.97 and 0.92 in correlations with and without striatal values, respectively, p < 0.01) and postmortem determined D2-receptor densities (including striatum: ρ = 0.92; p < 0.001; excluding striatum: ρ = 0.75; p = 0.067). These observations suggest that extrastriatal 11C-raclopride measurements represent a true D2 signal.
