Resurrection of Bronchocela burmana Blanford, 1878 for the Green Crested Lizard (Squamata, Agamidae) of southern Myanmar.

ABSTRACT: Recent fieldwork in southern Tanintharyi revealed the presence of a small Green Crested Lizard in the wet evergreen forest. We generated mtDNA sequence data (ND2) that demonstrates that this population's nearest relative is Bronchocela rayaensis Grismer et al., 2015 of Pulau Langkawi, northwestern Peninsular Malaysia and Phuket Island. Morphologically the Burmese Bronchocela shares many features with Bronchocela rayaensis, which potentially would make this recently described Thai-Malay species a synonym of Bronchocela burmana Blanford, 1878; however, we interpret the genetic and morphological differences to reflect evolutionary divergence and recommend the recognition of both species.

Project description:The whole mitogenome can prove useful tools for phylogenetic reconstruction and efficiently recover with reasonable taxon sampling. Calotes emma is widely distributed and arboreal in habits. However, studies of C. emma are still very limited, including population genetics and evolutionary biology. In this study, we reported the complete mitochondrial genome of the C. emma by next-generation sequencing for future more researches on systematics and evolution of C. emma from the perspective of mitochondrial DNA. The length of mitogenome was 17,688 bp, including 13 protein-coding genes (PCGs), 2 ribosomal RNA (rRNA) genes, 22 tRNA genes and a control region. The phylogenetic tree recovered the monophyly of the Calotes and revealed that newly sequenced C. emma well supported as the sister taxon to C. mystaceus by very high posterior probabilities (1.0). The complete mitochondrial genome of C.emma in this study will be helpful for understanding the phylogenetic systematics and relationships, and molecular evolution of Calotes in Agamidae.

Acanthosaura aurantiacrista (Squamata: Agamidae), a new long horn lizard from northern Thailand.

Project description:BackgroundIn Thailand, five species of Acanthosaura have been recorded so far, including Acanthosaura armata from the southern region, A. cardamomensis from the eastern region, A. crucigera from the western region, A. lepidogaster from the northern region and A. phuketensis from the Phuket Island and south-western region. However, comprehensive studies of diversity patterns and distribution of Acanthosaura are still lacking in some areas and need further information for designating areas of special conservation importance and nature protection planning in Thailand.New informationAcanthosaura aurantiacrista is a new species of long-horned lizard of the genus Acanthosaura from northern Thailand. It is distinguished from all other species of Acanthosaura by a dagger-like nuchal spine with yellowish-orange colouration in females, bright yellow colouration in males and a combination of other morphological characters: a greater tail length to snout-vent length ratio; a larger postorbital spine, nuchal spine, dorsal spine and occipital spine compared to its head length; a smaller diastema to snout-vent length ratio; a greater number of subdigital lamellae on the fourth finger and fourth toe; and a larger gular pouch than other Acanthosaura species. Analysis of mitochondrial ND2 gene sequences revealed a sister clade between the A. aurantiacrista lineage and the A. crucigera lineage with a 100% probability of divergence, according to Bayesian analysis and strong support value for Maximum Likelihood analysis. The pairwise distance ranged from 13.8-15.0% between A. aurantiacrista and A. cardamomensis, 10.9-14.5% between A. aurantiacrista and A. crucigera and 0-1.2% amongst A. aurantiacrista populations. The discovery of this lizard increases the known endemic herpetological diversity and underscores the importance of conservation in the mountain rainforest region of northern Thailand.

Male reproductive cycle in a population of the endemic butterfly lizard, Leiolepis ocellata Peters, 1971 (Squamata: Agamidae) from northern Thailand

Project description:Background Fundamental knowledge on the seasonal reproductive microanatomy and endocrinology of reptiles has been collected from several studies of various species. The present study was to determine annual changes in hormonal profiles, and detailed histomorphometric and histochemical characteristics of the entire male reproductive system of the tropical agamid lizard, Leiolepis ocellata. Results Male L. ocellata individuals (n = 75) collected from the territory of two provinces (Lampang and Tak) in northern Thailand exhibited annual variation in sex hormonal, histomorphometric, and histochemical characteristics of the male reproductive system. The reproductive cycle was subdivided into eight reproductive periods (early first active, first active, resting, second recrudescent, second active, regressive, quiescent, and first recrudescent), thus displaying a bimodal pattern with two actively reproductive periods. Circulating sex hormones (testosterone, estradiol, and progesterone) peaked in the first active (February) and the second active (June–July) periods. Likewise, gonadosomatic index (GSI) and histomorphometric variables of the testes and of the genital ducts (rete testis, ductuli efferentes, ductus epididymis, and ductus deferens) revealed their highest values in the first active period. Marked increase in protein and carbohydrate production was detectable in the ductuli efferentes during the active periods. Conclusions The male reproductive cycle of L. ocellata showed a biannual pattern of the hormonal profile, and detailed histomorphometric and histochemical characteristics of the entire reproductive system. Hence, the present study provides improved basic knowledge on the reptilian reproductive biology with comparative viewpoints to other reptiles. The description of the first rock-dwelling species of butterfly lizard Leiolepis Cuvier, 1829 (Squamata, Agamidae) from the Khorat Plateau in northeastern Thailand.

Project description:A new species of rock-dwelling Leiolepis is described from the Khorat Plateau in northeastern Thailand. Leiolepisglaurung sp. nov. can be differentiated from all other sexual species of Leiolepis by a combination of having a black gular region with a wide medial yellow stripe, a yellow ventrum with black mottling, bright red to orange subcaudal coloration, having reduced to no expandable flanks, and having only one black transverse bar on the flanks. This is the first rocky habitat-adapted Leiolepis. Leiolepisglaurung sp. nov. demonstrates numerous ecological adaptations to survive in these rocky habitats. Leiolepis are known for their expandable flanks with bright display colors, however Leiolepisglaurung sp. nov. has reduced or no ability to expand its flanks. We hypothesize this is an adaptation to reduce their body diameter to better fit into smaller rocky burrows unlike the larger and deeper burrows constructed in looser soils by other Leiolepis species. This discovery increases the number of Leiolepis species in Thailand to six, and worldwide to 11.

Mitochondrial DNA Variability within Uromastyx ornata philbyi (Agamidae: Squamata) from Southwestern Saudi Arabia.

Project description:Approximately 2.4?kbp of mitochondrial DNA was sequenced from 9 individuals of Uromastyx ornata philbyi originating from Taif, Namas, Al-Baha, and Jazan in southwestern Saudi Arabia. The sequenced regions cover eight tRNA genes (tRNA(Gln), tRNA(Ile), tRNA(Met), tRNA(Trp), tRNA(Ala), tRNA(Asn), tRNA(Cys), and tRNA(Tyr)) and two protein-coding genes (NADH dehydrogenase subunit 2 and cytochrome b). U. ornata philbyi had an insertion of 170?bp length between tRNA(Gln) and tRNA(Ile) genes. The first 128?bp of this insertion was similar to the one identified earlier in U. ornata ornata and can be folded into a stem-and-loop structure, which was less stable in U. ornata philbyi than in U. ornata ornata, or the second tRNA(Gln) gene. The next 42?bp of the insertion was unique in U. ornata philbyi and additionally retained a stable stem-and-loop structure. Most base substitutions found in the sequenced genes were synonymous transitions rather than transversions. Tree analyses supported the sister group relationship between the two U. ornata subspecies and divided U. ornata philbyi into two groups: Taif+Namas group in the east of Sarawat and Al-Baha+Jazan group in the west of Sarawat. These molecular data are in agreement with current classification of U. ornata.

The complete mitogenome of the splendid japalure Japalura splendida (Squamata, Agamidae).

Project description:The complete mitogenome of Japalura splendida (16,673?bp in length) is determined and analyzed in this study. It contains 13 protein-coding genes, 2 rRNA genes, 22 tRNA genes, and one non-coding regions. All the genes in J. splendida are distributed on the H-strand, except for the ND6 gene and seven tRNA genes which are encoded on the L-strand. The phylogenetic tree suggests that J. splendida and Japalura flaviceps formed a sister group and reveals the order ((((Acanthosaura lepidogaster, Acanthosaura armata), ((J. splendida, J. flaviceps), Pseudocalotes microlepis))), Calotes versicolor) with substantial support for the monophyly.

A new species of the genus Acanthosaura from Yunnan, China (Squamata, Agamidae).

Project description:A new species of Acanthosaura from Yunnan, China is described based on unique morphometric and meristic external characters and a very distinctive color pattern. The fourteenth species recorded of this genus, Acanthosaura tongbiguanensis sp. nov., was previously considered A. lepidogaster although it more closely resembles A. crucigera. It can be separated from all other species of the genus by having different numbers of subdigital lamellae on the fourth finger and toe, and a different shape of the black eye patch. The new species differs genetically from investigated congeners by percentage distance of 14.46% to 23.27% (cytochrome b gene).

Discovery of Japalura chapaensis Bourret, 1937 (Reptilia: Squamata: Agamidae) from Southeast Yunnan Province, China.

Project description:Due to a paucity of surveys in northern Indochina and lack of international collaborations among neighboring countries, recognized distributional ranges for many amphibian and reptile species end at the political borders for some countries, despite seemingly continuous suitable habitat spanning the region. Combining both morphological and genetic data, we report the first discovery of Japalura chapaensis, a rare agamid lizard believed previously to be endemic to northern Vietnam only, along the border region of southeastern Yunnan Province, China. To facilitate future research on the genus Japalura sensu lato in Indochina, we provide detailed descriptions of additional specimens of this rare species, including the first description of coloration in life and an expanded diagnosis, and discuss the species boundary of J. chapaensis with respect to its congeners.

Earliest example of a giant monitor lizard (Varanus, Varanidae, Squamata).

Project description:BACKGROUND: Varanidae is a clade of tiny (<20 mm pre-caudal length [PCL]) to giant (>600 mm PCL) lizards first appearing in the Cretaceous. True monitor lizards (Varanus) are known from diagnostic remains beginning in the early Miocene (Varanus rusingensis), although extremely fragmentary remains have been suggested as indicating earlier Varanus. The paleobiogeographic history of Varanus and timing for origin of its gigantism remain uncertain. METHODOLOGY/PRINCIPAL FINDINGS: A new Varanus from the Mytilini Formation (Turolian, Miocene) of Samos, Greece is described. The holotype consists of a partial skull roof, right side of a braincase, partial posterior mandible, fragment of clavicle, and parts of six vertebrae. A cladistic analysis including 83 taxa coded for 5733 molecular and 489 morphological characters (71 previously unincluded) demonstrates that the new fossil is a nested member of an otherwise exclusively East Asian Varanus clade. The new species is the earliest-known giant (>600 mm PCL) terrestrial lizard. Importantly, this species co-existed with a diverse continental mammalian fauna. CONCLUSIONS/SIGNIFICANCE: The new monitor is larger (longer) than 99% of known fossil and living lizards. Varanus includes, by far, the largest limbed squamates today. The only extant non-snake squamates that approach monitors in maximum size are the glass-snake Pseudopus and the worm-lizard Amphisbaena. Mosasauroids were larger, but exclusively marine, and occurred only during the Late Cretaceous. Large, extant, non-Varanus, lizards are limbless and/or largely isolated from mammalian competitors. By contrast, our new Varanus achieved gigantism in a continental environment populated by diverse eutherian mammal competitors.

Seasonal spermatogenesis in the Mexican endemic oviparous lizard, Sceloporus aeneus (Squamata: Phrynosomatidae).

Project description:Oviparous species of Sceloporus exhibit either seasonal or continuous spermatogenesis and populations from high-elevation show a seasonal pattern known as spring reproductive activity. We studied the spermatogenic cycle of a high-elevation (2700 m) population of endemic oviparous lizard, Sceloporus aeneus, that resided south of México, D.F. Histological analyses were performed on the testes and reproductive ducts from individual lizards collected monthly. This population of S. aeneus showed a seasonal pattern of spermatogenesis, with 4 successive phases common in other lizards. These include: 1) Quiescence in August, which contained solely spermatogonia and Sertoli cells; 2) Testicular recrudescence (September-January) when testes became active with mitotic spermatogonia, spermatocytes beginning meiosis, and the early stages of spermiogenesis with spermatids; 3) Maximum testicular activity occurred from March to May and is when the largest spermiation events ensued within the germinal epithelia, which were also dominated by spermatids and spermiogenic cells; 4) Testicular regression in June was marked with the number of all germs cells decreasing rapidly and spermatogonia dominated the seminiferous epithelium. February was a transitional month between recrudescence and maximum activity. The highest sperm abundance in the lumina of epididymides was during maximum testicular activity (March-May). Thus, before and after these months fewer spermatozoa were detected within the excurrent ducts as the testis transitions from recrudescence to maximum activity in February and from maximum activity to quiescence in June. Maximum spermatogenic activity corresponds with warmest temperatures at this study site. This pattern known as spring reproductive activity with a fall recrudescence was similar to other oviparous species of genus Sceloporus.
