The prior genus-level classification of Cardiophorinae had never been assessed phylogenetically, and not revised since 1906. A phylogeny for Cardiophorinae and Negastriinae is inferred by Bayesian analyses of 163 adult morphological characters to revise the generic classification. Parsimony analysis is also performed to assess the sensitivity of the Bayesian results to the choice of optimality criterion. Fossils improve phylogenetic analyses of morphological characters.

Project description:Fossils provide our only direct window into evolutionary events in the distant past. Incorporating them into phylogenetic hypotheses of living clades can help time-calibrate divergences, as well as elucidate macroevolutionary dynamics. However, the effect fossils have on phylogenetic reconstruction from morphology remains controversial. The consequences of explicitly incorporating the stratigraphic ages of fossils using tip-dated inference are also unclear. Here, we use simulations to evaluate the performance of inference methods across different levels of fossil sampling and missing data. Our results show that fossil taxa improve phylogenetic analysis of morphological datasets, even when highly fragmentary. Irrespective of inference method, fossils improve the accuracy of phylogenies and increase the number of resolved nodes. They also induce the collapse of ancient and highly uncertain relationships that tend to be incorrectly resolved when sampling only extant taxa. Furthermore, tip-dated analyses under the fossilized birth-death process outperform undated methods of inference, demonstrating that the stratigraphic ages of fossils contain vital phylogenetic information. Fossils help to extract true phylogenetic signals from morphology, an effect that is mediated by both their distinctive morphology and their temporal information, and their incorporation in total-evidence phylogenetics is necessary to faithfully reconstruct evolutionary history.

Congruence Between Molecular Data and Morphology: Phylogenetic Position of Senodoniini (Coleoptera: Elateridae).

Project description:Senodoniini is a small lineage of click beetles currently comprising 21 species in two genera, distributed in the Himalayas and East and Southeast Asia. The definition and limits of this group have changed considerably during its history. Recent authors treat Senodoniini as a tribe within Dendrometrinae, usually close to Dimini, but this placement has never been rigorously tested. Here, we shed new light on the systematic position and limits of Senodoniini by performing a combined phylogenetic analysis of two nuclear and two mitochondrial molecular markers. Our results recovered Senodoniini not monophyletic, and placed them into the Lissominae complex, where they formed a clade with Austrelater Calder & Lawrence (Protelaterini). Molecular phylogeny is in agreement with the adult morphology. Additionally, we examined the morphology of a monotypic genus Rostricephalus Fleutiaux from Southeast Asia, which was previously classified in various Elateridae groups including Senodoniini, and its position was always uncertain. This genus shares morphological characters with Protelaterini. We provide morphological redescriptions as well as the figures of main diagnostic characters for Senodonia Laporte, Sossor Candèze, and Rostricephalus. Based on our results, we place these genera to Lissominae: Protelaterini, and hence synonymize Senodoniini Schenkling with Protelaterini Schwarz.

Two new Clitocella species from North China revealed by phylogenetic analyses and morphological characters.

Project description:Two new species of Clitocella are proposed based on morphological and phylogenetic investigations. Clitocellaborealichinensis sp. nov. is closely related to C.orientalis but distinguished from the latter by its slightly smaller basidiospores and hyphae of pileipellis with pale brown to brown intracellular or parietal pigment. Clitocellacolorata sp. nov. is closely related to C.popinalis and C.mundula in macromorphology but is differentiated from C.popinalis by its slightly smaller basidiospores and the difference in genetic profile, and from C.mundula by its relatively colorful pileus (white to yellowish white, grayish white to grayish brown, pink white). Phylogenetic analyses based on sequence data from five different loci (ITS, nrLSU, tef1, rpb2 and atp6) support the taxonomic position of the two new species in the genus Clitocella. The illustrations and descriptions for the new taxa are provided.

Phylogenetic analyses and morphological characters reveal two new species of Ganoderma from Yunnan province, China.

Project description:Ganodermadianzhongense sp. nov. and G.esculentum sp. nov. are proposed as two new species based on both phenotypic and genotypic evidences. Ganodermadianzhongense is characterized by the stipitate basidiomata, laccate and oxblood red pileus, gray white pore surface, duplex context and broadly ellipsoid basidiospores (9.0-12.5 × 6.5-9.0 μm) with coarse interwall pillars. Ganodermaesculentum is characterized by its basidiomata with slender stipe, white pore surface, homogeneous pileus context, and slightly truncate, narrow basidiospores (8.0-12.5 × 5.0-8.0 µm). Phylogenetic analyses were carried out based on the internal transcribed spacer (ITS), translation elongation factor 1-α (TEF1-α) and the second subunit of RNA polymerase II (RPB2) sequence data. The illustrations and descriptions for the new taxa are provided.

Project description:We determined the complete mitogenome sequence of Melanotus cribricollis (Faldermann) (GenBank accession no. MK792748). The mitogenome is 15,908 bp in length, consisting of 13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes, and one non-coding control region. The overall nucleotide composition was 40.8% A, 31.5% T, 17.4% C, and 10.3% G, with 72.3% of AT, respectively. Phylogenetic analysis based on the 13 coding protein genes nucleotide sequences revealed that M. cribricollis clustered with the same genus species M. villosus, and the three genus Melanotus Eschscholtz, Agriotes Eschscholtz and Adrastus Eschscholtz had a close relationship.

Phylogenetic Status and Morphological Characters of Rhabditolaimus anoplophorae (Rhabditida: Diplogastridae).

Project description:Rhabditolaimus anoplophorae Kanzaki and Futai was re-isolated from its type host (carrier), the cerambycid beetle Anoplophora malasiaca, collected in an experimental field of the Forestry and Forest Products Research Institute, Tsukuba, Ibaraki, Japan. The nematode was cultured on nematode growth medium plates seeded with Escherichia coli OP50, and its morphological characters and molecular profile were examined to modernize the description. Scanning electron microscopic and light microscopy revealed the presence of four stomatal flaps, a very long gymnostom, a single ventral papilla in males, and a horizontal slit-like vulval opening in females. The positions of the deirids, hemizonids, phasmids, and rectal glands are additionally described, and the absence of a male bursa was confirmed. Phylogenetically, the genus forms a well-supported clade in the family Diplogastridae. Rhabditolaimus anoplophorae is a member of the monophyletic Rhabditolaimus clade and is closely related to R. leuckarti and several undescribed species.

MN306531) using next-generation sequencing (NGS) method. The mitogenome is 15,842 bp in length, consisting of 13 protein-coding genes (PCGs), two ribosomal RNA genes (rRNAs), 22 transfer RNA genes (tRNAs), and one non-coding control region. The overall nucleotide composition was 41.6% A, 31.1% T, 16.9% C, and 10.4% G, with 72.7% AT, respectively. The gene arrangement is consistent with the typical insect mitochondrial genome. Phylogenetic analysis revealed that C. vittiger clustered into a clade with Chiagosnius sulcicollis with high bootstrap support.
