Supplements are increasingly important to the scientific record, particularly in genomics. However, they are often underutilized. Optimally, supplements should make results findable, accessible, interoperable, and reusable (i.e., "FAIR"). Moreover, properly off-loading to them the data and detail in a paper could make the main text more readable. Project description:Reticular materials are of high interest for diverse applications, ranging from catalysis and separation to gas storage and drug delivery. These open, extended frameworks can be tailored to the intended application through crystal-structure design. Implementing these materials in application settings, however, requires structuring beyond their lattices, to interface the functionality at the molecular level effectively with the macroscopic world. To overcome this barrier, efforts in expressing structural control across molecular, nano-, meso-, and bulk regimes is the essential next step. In this Review, we give an overview of recent advances in using self-assembly as well as externally controlled tools to manufacture reticular materials over all the length scales. We predict that major research advances in deploying these two approaches will facilitate the use of reticular materials in addressing major needs of society.

Project description:Metal additive manufacturing (AM) enables unparalleled design freedom for the development of optimized devices in a plethora of applications. The requirement for the use of nonconventional aluminum alloys such as AlSi10Mg has made the rational micro/nanostructuring of metal AM challenging. Here, the techniques are developed and the fundamental mechanisms governing the micro/nanostructuring of AlSi10Mg, the most common metal AM material, are investigated. A surface structuring technique is rationally devised to form previously unexplored two-tier nanoscale architectures that enable remarkably low adhesion, excellent resilience to condensation flooding, and enhanced liquid-vapor phase transition. Using condensation as a demonstration framework, it is shown that the two-tier nanostructures achieve 6× higher heat transfer coefficient when compared to the best filmwise condensation. The study demonstrates that AM-enabled nanostructuring is optimal for confining droplets while reducing adhesion to facilitate droplet detachment. Extensive benchmarking with past reported data shows that the demonstrated heat transfer enhancement has not been achieved previously under high supersaturation conditions using conventional aluminum, further motivating the need for AM nanostructures. Finally, it has been demonstrated that the synergistic combination of wide AM design freedom and optimal AM nanostructuring method can provide an ultracompact condenser having excellent thermal performance and power density.

Project description:Colloidal dispersions of cellulose nanofibrils (CNFs) are viable alternatives to cellulose II dissolutions used for filament spinning. The porosity and water vapor affinity of CNF filaments make them suitable for controlled breathability. However, many textile applications also require water repellence. Here, we investigated the effects of postmodification of wet-spun CNF filaments via chemical vapor deposition (CVD). Two organosilanes with different numbers of methyl substituents were considered. Various surface structures were achieved, either as continuous, homogeneous coating layers or as three-dimensional, hairy-like assemblies. Such surface features reduced the surface energy, which significantly affected the interactions with water. Filaments with water contact angles of up to 116° were obtained, and surface energy measurements indicated the possibility of developing amphiphobicity. Dynamic vapor sorption and full immersion experiments were carried out to inquire about the interactions with water, whether in the liquid or gas forms. Mechanical tests revealed that the wet strength of the modified filaments were almost 3 times higher than that of the unmodified precursors. The hydrolytic and mechanical stabilities of the adsorbed layers were also revealed. Overall, our results shed light on the transformation of aqueous dispersions of CNFs into filaments that are suited for controlled interactions with water via concurrent hydrolysis and condensation reactions in CVD, while maintaining the moisture buffering capacity and breathability of related structures.

Project description:Structuring blends on sub-micrometer scales, especially nano-scales, has a higher potential for improving their thermomechanical properties. Here, we propose a design strategy to fabricate compatible nano-blends by manipulating the reactions between two biodegradable polymers, e.g. polybutylene succinate (PBS) and polybutylene adipate terephthalate (PBAT), with extremely low free radical contents through reactive extrusion processing. Observed by transmission electron microscopy (TEM) and atomic force microscopy (AFM), it is found that PBAT is tightly surrounded by large amounts of PBS-PBAT co-polymers and dispersed in a PBS matrix with a particle size of less than 100 nm. We show how impact strength and polymer moduli can be improved simultaneously by decreasing the small amount of dispersed phase into nano-scale (droplet or lamina structures). With 5 wt% PBAT content in the PBS-PBAT blend, the notched impact strength of PBS is increased 1200% and the Young's modulus is increased 15%. Through in situ rheological monitoring and Fourier-transform infrared spectroscopy (FTIR) studies, the reason why nano-blends can be formed in such a low amount of peroxide is illustrated. Our investigation most significantly indicates the transformation of the partially compatible PBS-PBAT micro-blend into a fully compatible PBS-PBAT through nano-structuring. This work addresses the importance of reaction rate and mechanism in favoring the formation of co-polymers rather than homo-polymer crosslinking or self-decomposition in polymer blend modification via reactive extrusion design.

Project description:Importance:Manufacturers of US Food and Drug Administration-approved prescription drugs often apply for additional indications based on randomized clinical trials. Real-world database analyses on a medication's use and outcomes in routine settings of care might help to inform decision making regarding such supplemental indications. Objective:To examine whether longitudinal data from a health care database can support the results of a randomized clinical trial that led to a supplemental indication for telmisartan. Design, Setting, and Participants:This cohort study of patients newly prescribed telmisartan or ramipril used insurance claims data from a nationwide health care database from January 1, 2003, through September 30, 2009, to compare patient outcomes. This study replicated the inclusion and exclusion criteria used in the Ongoing Telmisartan Alone and in Combination with Ramipril Global End-point Trial (ONTARGET) and used propensity score matching to balance 74 patient characteristics. Data analysis was performed from February 15, 2017, to May 24, 2017. Exposures:Telmisartan use vs ramipril use. Main Outcomes and Measures:The primary outcome was a composite of myocardial infarction, stroke, or hospitalization for congestive heart failure. Results:Of the 640 951 patients included in the study, 48 053 were newly prescribed ramipril (mean [SD] age, 68.29 [9.52] years; 31 940 male [66.5%]) and 4665 were newly prescribed telmisartan (mean [SD] age, 69.43 [9.60] years; 2413 male [51.7%]). After propensity score matching, a total of 4665 patients were newly prescribed telmisartan (mean [SD] age, 69.43 [9.60] years; 2413 [51.7%]), and 4665 patients were newly prescribed ramipril (mean [SD] age, 69.36 [9.67] years; 2343 male [50.2%]). As seen in ONTARGET, the composite risk of stroke, myocardial infarction, and hospitalization for congestive heart failure was similar for the 2 medications (hazard ratio, 1.0; 95% CI, 0.9-1.1). In addition, the study found that telmisartan was associated with a substantially decreased risk of angioedema (hazard ratio, 0.1; 95% CI, 0.03-0.56) compared with ramipril. Conclusions and Relevance:Real-world data analyses of patients receiving routine care provided findings similar to those found in the randomized clinical trial that established telmisartan's supplemental indication. In certain situations, database studies may support supplemental applications for effectiveness for already approved medications.

Project description:BackgroundHypothesis generation in molecular and cellular biology is an empirical process in which knowledge derived from prior experiments is distilled into a comprehensible model. The requirement of automated support is exemplified by the difficulty of considering all relevant facts that are contained in the millions of documents available from PubMed. Semantic Web provides tools for sharing prior knowledge, while information retrieval and information extraction techniques enable its extraction from literature. Their combination makes prior knowledge available for computational analysis and inference. While some tools provide complete solutions that limit the control over the modeling and extraction processes, we seek a methodology that supports control by the experimenter over these critical processes.ResultsWe describe progress towards automated support for the generation of biomolecular hypotheses. Semantic Web technologies are used to structure and store knowledge, while a workflow extracts knowledge from text. We designed minimal proto-ontologies in OWL for capturing different aspects of a text mining experiment: the biological hypothesis, text and documents, text mining, and workflow provenance. The models fit a methodology that allows focus on the requirements of a single experiment while supporting reuse and posterior analysis of extracted knowledge from multiple experiments. Our workflow is composed of services from the 'Adaptive Information Disclosure Application' (AIDA) toolkit as well as a few others. The output is a semantic model with putative biological relations, with each relation linked to the corresponding evidence.ConclusionWe demonstrated a 'do-it-yourself' approach for structuring and extracting knowledge in the context of experimental research on biomolecular mechanisms. The methodology can be used to bootstrap the construction of semantically rich biological models using the results of knowledge extraction processes. Models specific to particular experiments can be constructed that, in turn, link with other semantic models, creating a web of knowledge that spans experiments. Mapping mechanisms can link to other knowledge resources such as OBO ontologies or SKOS vocabularies. AIDA Web Services can be used to design personalized knowledge extraction procedures. In our example experiment, we found three proteins (NF-Kappa B, p21, and Bax) potentially playing a role in the interplay between nutrients and epigenetic gene regulation.

Project description:IntroductionIn African countries, respiratory infections and severe sepsis are common causes of respiratory failure and mortality in children under five years of age. Mortality and morbidity in these children could be reduced with adequate respiratory support in the emergency care setting. The purpose of this review is to describe management priorities in the emergency care of critically ill children presenting with respiratory problems. Basic and advanced respiratory support measures are described for implementation according to available resources, work load and skill-levels.MethodsWe did a focused search of respiratory support for critically ill children in resource-limited settings over the past ten years, using the search tools PubMed and Google Scholar, the latest WHO guidelines, international 'Advanced Paediatric Life Support' guidelines and paediatric critical care textbooks.ResultsThe implementation of triage and rapid recognition of respiratory distress and hypoxia with pulse oximetry is important to correctly identify critically ill children with increased risk of mortality in all health facilities in resource constrained settings. Basic, effective airway management and respiratory support are essential elements of emergency care. Correct provision of supplemental oxygen is safe and its application alone can significantly improve the outcome of critically ill children. Non-invasive ventilatory support is cost-effective and feasible, with the potential to improve emergency care packages for children with respiratory failure and other organ dysfunctions. Non-invasive ventilation is particularly important in severely under-resourced regions unable to provide intubation and invasive mechanical ventilation support. Malnutrition and HIV-infection are important co-morbid conditions, associated with increased mortality in children with respiratory dysfunction.DiscussionA multi-disciplinary approach is required to optimise emergency care for critically ill children in low-resource settings. In this context, it is important to consider aspects of training of staff, technical support and pragmatic research.

Here we present the culmination of the efforts of the modENCODE (Model Organism ENCyclopedia Of DNA Elements) and modERN (model organism Encyclopedia of Regulatory Networks) consortia to systematically assay TF binding events in vivo in two major model organisms, Drosophila melanogaster (fly) and Caenorhabditis elegans (worm). These datasets comprise 605 TFs identifying 3.6M sites in the fly and 356 TFs identifying 0.9 M sites in the worm, and represent the majority of the regulatory space in each genome. We demonstrate that TFs associate with chromatin in clusters termed “metapeaks”, that larger metapeaks have characteristics of high occupancy target (HOT) regions, and that the importance of consensus sequence motifs bound by TFs depends on metapeak size and complexity. Combining ChIP-seq data with single cell RNA-seq data in a machine learning model identifies particular TFs with a prominent role in promoting target gene expression in specific cell types, even differentiating between parent-daughter cells during embryogenesis. These data are a rich resource for the community that should fuel and guide future investigations into TF function. To facilitate data accessibility and utility, all strains expressing GFP-tagged TFs are available at the stock centers for each organism. Project description:A method for the grafting of unsymmetrical A2BC-type 5,15-bis(4-butoxyphenyl)-10-(4-carboxyphenyl)-20-(phenanthrenoimidazolyl)-porphyrin onto the surface of nanostructured aluminum oxyhydroxide modified with a single SiO2 layer (NAOM) was successfully developed. A straightforward procedure towards surface modification of NAOM allowed us to prepare a new porphyrin-containing hybrid material. The obtained 3D heterostructure was extensively characterized using XPS, TEM and diffuse reflectance spectroscopy. Structural and morphological peculiarities of the inorganic support before and after the immobilization procedure were studied and discussed in detail. The stability of the material against leaching and the porphyrin immobilization ratio ca. 14% by weight were also revealed.
