ABSTRACT: Most philosophers believe that a person can have an obligation only insofar as she is able to fulfil it, a principle generally referred to as "Ought Implies Can". Arguably, this principle reflects something basic about the ordinary concept of obligation. However, in a paper published recently in this journal, Wesley Buckwalter and John Turri presented evidence for the conclusion that ordinary people in fact reject that principle. With a series of studies, they claimed to have demonstrated that, in people's judgements, obligations persist irrespective of whether those who hold them have the ability to fulfil them. We argue in this paper that due to some problems in their design, Buckwalter & Turri's conclusions may not be warranted. We present the results of a series of studies demonstrating the problems with their design and showing that, with an improved design, people judge that obligation depends on ability after all.

Quantum correlations which imply causation.

Project description:In ordinary, non-relativistic, quantum physics, time enters only as a parameter and not as an observable: a state of a physical system is specified at a given time and then evolved according to the prescribed dynamics. While the state can, and usually does, extend across all space, it is only defined at one instant of time. Here we ask what would happen if we defined the notion of the quantum density matrix for multiple spatial and temporal measurements. We introduce the concept of a pseudo-density matrix (PDM) which treats space and time indiscriminately. This matrix in general fails to be positive for measurement events which do not occur simultaneously, motivating us to define a measure of causality that discriminates between spatial and temporal correlations. Important properties of this measure, such as monotonicity under local operations, are proved. Two qubit NMR experiments are presented that illustrate how a temporal pseudo-density matrix approaches a genuinely allowed density matrix as the amount of decoherence is increased between two consecutive measurements.

Ulcerative Colitis Seems to Imply Oral Microbiome Dysbiosis

Project description:Ulcerative colitis (UC) is a recurrent pathology of complex etiology that has been occasionally associated with oral lesions, but the overall composition of the oral microbiome in UC patients and its role in the pathogenesis of the disease are still poorly understood. In this study, the oral microbiome of UC patients and healthy individuals was compared to ascertain the possible changes in the oral microbial communities associated with UC. For this, the salivary microbiota of 10 patients diagnosed with an active phase of UC and 11 healthy controls was analyzed by 16S rRNA gene sequencing (trial ref. ISRCTN39987). Metataxonomic analysis revealed a decrease in the alpha diversity and an imbalance in the relative proportions of some key members of the oral core microbiome in UC patients. Additionally, Staphylococcus members and four differential species or phylotypes were only present in UC patients, not being detected in healthy subjects. This study provides a global snapshot of the existence of oral dysbiosis associated with UC, and the possible presence of potential oral biomarkers.

Computer Simulations Imply Forelimb-Dominated Underwater Flight in Plesiosaurs.

Project description:Plesiosaurians are an extinct group of highly derived Mesozoic marine reptiles with a global distribution that spans 135 million years from the Early Jurassic to the Late Cretaceous. During their long evolutionary history they maintained a unique body plan with two pairs of large wing-like flippers, but their locomotion has been a topic of debate for almost 200 years. Key areas of controversy have concerned the most efficient biologically possible limb stroke, e.g. whether it consisted of rowing, underwater flight, or modified underwater flight, and how the four limbs moved in relation to each other: did they move in or out of phase? Previous studies have investigated plesiosaur swimming using a variety of methods, including skeletal analysis, human swimmers, and robotics. We adopt a novel approach using a digital, three-dimensional, articulated, free-swimming plesiosaur in a simulated fluid. We generated a large number of simulations under various joint degrees of freedom to investigate how the locomotory repertoire changes under different parameters. Within the biologically possible range of limb motion, the simulated plesiosaur swims primarily with its forelimbs using an unmodified underwater flight stroke, essentially the same as turtles and penguins. In contrast, the hindlimbs provide relatively weak thrust in all simulations. We conclude that plesiosaurs were forelimb-dominated swimmers that used their hind limbs mainly for maneuverability and stability.

Metabolite concentrations, fluxes and free energies imply efficient enzyme usage.

Project description:In metabolism, available free energy is limited and must be divided across pathway steps to maintain a negative ΔG throughout. For each reaction, ΔG is log proportional both to a concentration ratio (reaction quotient to equilibrium constant) and to a flux ratio (backward to forward flux). Here we use isotope labeling to measure absolute metabolite concentrations and fluxes in Escherichia coli, yeast and a mammalian cell line. We then integrate this information to obtain a unified set of concentrations and ΔG for each organism. In glycolysis, we find that free energy is partitioned so as to mitigate unproductive backward fluxes associated with ΔG near zero. Across metabolism, we observe that absolute metabolite concentrations and ΔG are substantially conserved and that most substrate (but not inhibitor) concentrations exceed the associated enzyme binding site dissociation constant (Km or Ki). The observed conservation of metabolite concentrations is consistent with an evolutionary drive to utilize enzymes efficiently given thermodynamic and osmotic constraints.

Scalar utility theory and proportional processing: What does it actually imply?

Project description:Scalar Utility Theory (SUT) is a model used to predict animal and human choice behaviour in the context of reward amount, delay to reward, and variability in these quantities (risk preferences). This article reviews and extends SUT, deriving novel predictions. We show that, contrary to what has been implied in the literature, (1) SUT can predict both risk averse and risk prone behaviour for both reward amounts and delays to reward depending on experimental parameters, (2) SUT implies violations of several concepts of rational behaviour (e.g. it violates strong stochastic transitivity and its equivalents, and leads to probability matching) and (3) SUT can predict, but does not always predict, a linear relationship between risk sensitivity in choices and coefficient of variation in the decision-making experiment. SUT derives from Scalar Expectancy Theory which models uncertainty in behavioural timing using a normal distribution. We show that the above conclusions also hold for other distributions, such as the inverse Gaussian distribution derived from drift-diffusion models. A straightforward way to test the key assumptions of SUT is suggested and possible extensions, future prospects and mechanistic underpinnings are discussed.

Differential inputs to striatal cholinergic and parvalbumin interneurons imply functional distinctions.

Project description:Striatal cholinergic (ChAT) and parvalbumin (PV) interneurons exert powerful influences on striatal function in health and disease, yet little is known about the organization of their inputs. Here using rabies tracing, electrophysiology and genetic tools, we compare the whole-brain inputs to these two types of striatal interneurons and dissect their functional connectivity in mice. ChAT interneurons receive a substantial cortical input from associative regions of cortex, such as the orbitofrontal cortex. Amongst subcortical inputs, a previously unknown inhibitory thalamic reticular nucleus input to striatal PV interneurons is identified. Additionally, the external segment of the globus pallidus targets striatal ChAT interneurons, which is sufficient to inhibit tonic ChAT interneuron firing. Finally, we describe a novel excitatory pathway from the pedunculopontine nucleus that innervates ChAT interneurons. These results establish the brain-wide direct inputs of two major types of striatal interneurons and allude to distinct roles in regulating striatal activity and controlling behavior.

Humidity trends imply increased sensitivity to clouds in a warming Arctic.

Project description:Infrared radiative processes are implicated in Arctic warming and sea-ice decline. The infrared cloud radiative effect (CRE) at the surface is modulated by cloud properties; however, CRE also depends on humidity because clouds emit at wavelengths that are semi-transparent to greenhouse gases, most notably water vapour. Here we show how temperature and humidity control CRE through competing influences between the mid- and far-infrared. At constant relative humidity, CRE does not decrease with increasing temperature/absolute humidity as expected, but rather is found to be approximately constant for temperatures characteristic of the Arctic. This stability is disrupted if relative humidity varies. Our findings explain observed seasonal and regional variability in Arctic CRE of order 10?W?m(-2). With the physical properties of Arctic clouds held constant, we calculate recent increases in CRE of 1-5?W?m(-2) in autumn and winter, which are projected to reach 5-15?W?m(-2) by 2050, implying increased sensitivity of the surface to clouds.

How many cases of disease in a pedigree imply familial disease?

Project description:The ability to perform whole-exome and, increasingly, whole-genome sequencing on large numbers of individuals has led to increased efforts to identify rare genetic variants that affect the risk of both common and rare diseases. In such applications, it is important to identify families that are segregating the rare variants of interest. For rare diseases or rare familial forms of common diseases, pedigrees with multiple affected members are clearly harbouring risk variants. For more common diseases, however, it may be unclear whether a family with a few affected members is segregating a familial disease, is the result of multiple sporadic cases, or is a mixture of familial cases and phenocopies. We provide calculations for the probability that a family is harbouring familial disease, presented in general terms that admit working guidelines for selecting families for current sequencing studies. Using examples motivated by our own studies of thyroid cancer and published studies of colorectal cancer, we show that for common diseases, families with exactly two affected first-degree relatives have only a moderate probability of segregating familial disease, but this probability is higher for families with three or more affected relatives, and those families should therefore be prioritised in sequencing studies.

Brain experiments imply adaptation mechanisms which outperform common AI learning algorithms.

Project description:Attempting to imitate the brain's functionalities, researchers have bridged between neuroscience and artificial intelligence for decades; however, experimental neuroscience has not directly advanced the field of machine learning (ML). Here, using neuronal cultures, we demonstrate that increased training frequency accelerates the neuronal adaptation processes. This mechanism was implemented on artificial neural networks, where a local learning step-size increases for coherent consecutive learning steps, and tested on a simple dataset of handwritten digits, MNIST. Based on our on-line learning results with a few handwriting examples, success rates for brain-inspired algorithms substantially outperform the commonly used ML algorithms. We speculate this emerging bridge from slow brain function to ML will promote ultrafast decision making under limited examples, which is the reality in many aspects of human activity, robotic control, and network optimization.

High-Load Reovirus Infections Do Not Imply Physiological Impairment in Salmon.

Project description:The recent ubiquitous detection of PRV among salmonids has sparked international concern about the cardiorespiratory performance of infected wild and farmed salmon. Piscine orthoreovirus (PRV) has been shown to create substantial viremia in salmon by targeting erythrocytes for principle replication. In some instances, infections develop into heart and skeletal muscle inflammation (HSMI) or other pathological conditions affecting the respiratory system. Critical to assessing the seriousness of PRV infections are controlled infection studies that measure physiological impairment to critical life support systems. Respiratory performance is such a system and here multiple indices were measured to test the hypothesis that a low-virulence strain of PRV from Pacific Canada compromises the cardiorespiratory capabilities of Atlantic salmon. Contrary to this hypothesis, the oxygen affinity and carrying capacity of erythrocytes were unaffected by PRV despite the presence of severe viremia, minor heart pathology and transient cellular activation of antiviral response pathways. Similarly, PRV-infected fish had neither sustained nor appreciable differences in respiratory capabilities compared with control fish. The lack of functional harm to salmon infected with PRV in this instance highlights that, in an era of unprecedented virus discovery, detection of viral infection does not necessarily imply bodily harm and that viral load is not always a suitable predictor of disease within a host organism.
