Prefrontal cortex modulates sensory signals in extrastriate visual cortex, in part via its direct projections from the frontal eye field (FEF), an area involved in selective attention. Project description:Visually guided behavior relies on the integration of sensory input and information held in working memory (WM). Yet it remains unclear how this is accomplished at the level of neural circuits. We studied the direct visual cortical inputs to neurons within a visuomotor area of prefrontal cortex in behaving monkeys. We show that the efficacy of visual input to prefrontal cortex is gated by information held in WM. Surprisingly, visual input to prefrontal neurons was found to target those with both visual and motor properties, rather than preferentially targeting other visual neurons. Furthermore, activity evoked from visual cortex was larger in magnitude, more synchronous, and more rapid, when monkeys remembered locations that matched the location of visual input. These results indicate that WM directly influences the circuitry that transforms visual input into visually guided behavior.

Project description:Spatial working memory, the caching of behaviourally relevant spatial cues on a timescale of seconds, is a fundamental constituent of cognition. Although the prefrontal cortex and hippocampus are known to contribute jointly to successful spatial working memory, the anatomical pathway and temporal window for the interaction of these structures critical to spatial working memory has not yet been established. Here we find that direct hippocampal-prefrontal afferents are critical for encoding, but not for maintenance or retrieval, of spatial cues in mice. These cues are represented by the activity of individual prefrontal units in a manner that is dependent on hippocampal input only during the cue-encoding phase of a spatial working memory task. Successful encoding of these cues appears to be mediated by gamma-frequency synchrony between the two structures. These findings indicate a critical role for the direct hippocampal-prefrontal afferent pathway in the continuous updating of task-related spatial information during spatial working memory.

Project description:Ample research in visual working memory (VWM) has demonstrated that the memorized items are maintained in integrated spatial configurations, even when the spatial context is task irrelevant. These insights were obtained in studies in which participants were provided with the information they memorized. However, the encoding of provided information is only one aspect of memory. In everyday life, individuals often construct their own memory representations, an aspect of memory we have previously termed self-initiated (SI) working memory. In this study, we employed a SI VWM task in which participants selected the visual targets they memorized. The spatial locations of the targets were task irrelevant. Nevertheless, we were interested to see whether participants would construct spatially structured memory representations, which would suggest that they intended to maintain the visual targets as integrated spatial configurations. The results of two experiments demonstrated that participants constructed spatially structured configurations relative to random displays. Specifically, participants selected visual targets in close spatial proximity and constructed spatial sequences with short distances and fewer path crossings. When asked to construct configurations for a hypothetical competitor in a memory contest, participants disrupted the spatial structure by selecting visual targets that were further apart and by increasing the distances between them, which suggests that these characteristics were under their control. At the end of each experiment, participants provided verbal descriptions of the strategies they used to construct the memory displays. While the spatial structure of the SI memory representations was robust, it was absent from the participants' explicit descriptions, which focused on non-spatial strategies. Participants reported selecting items based, most frequently, on semantic categories and visual features. Taken together, these results demonstrated that participants had access to the metacognitive knowledge on the spatial structure of VWM representations, knowledge they manipulated to construct memory representations that enhanced or disrupted memory performance. While having a profound impact on behavior, this metacognitive knowledge on spatial structure remained implicit, as it was absent from the participants' verbal reports. Viewed from a larger perspective, this study explores how individuals interact with the world by actively structuring their surroundings to maximize cognitive performance.

Project description:Although the contents of working memory can be decoded from visual cortex activity, these representations may play a limited role if they are not robust to distraction. We used model-based fMRI to estimate the impact of distracting visual tasks on working memory representations in several visual field maps in visual and frontoparietal association cortex. Here, we show distraction causes the fidelity of working memory representations to briefly dip when both the memorandum and distractor are jointly encoded by the population activities. Distraction induces small biases in memory errors which can be predicted by biases in neural decoding in early visual cortex, but not other regions. Although distraction briefly disrupts working memory representations, the widespread redundancy with which working memory information is encoded may protect against catastrophic loss. In early visual cortex, the neural representation of information in working memory and behavioral performance are intertwined, solidifying its importance in visual memory.

Project description:Working memory (WM) enables the storage and manipulation of limited amounts of information over short periods. Prominent models posit that increasing the number of remembered items decreases the spiking activity dedicated to each item via mutual inhibition, which irreparably degrades the fidelity of each item's representation. We tested these models by determining if degraded memory representations could be recovered following a post-cue indicating which of several items in spatial WM would be recalled. Using an fMRI-based image reconstruction technique, we identified impaired behavioral performance and degraded mnemonic representations with elevated memory load. However, in several cortical regions, degraded mnemonic representations recovered substantially following a post-cue, and this recovery tracked behavioral performance. These results challenge pure spike-based models of WM and suggest that remembered items are additionally encoded within latent or hidden neural codes that can help reinvigorate active WM representations.

Project description:Visual working memory (VWM) is a remarkable skill dependent on the brain's ability to construct and hold an internal representation of the world for later comparison with an external stimulus. The prefrontal cortex (PFC) and basal ganglia (BG) interact within a cortical and subcortical network supporting VWM. We used scalp electroencephalography in groups of patients with unilateral PFC or BG lesions to provide evidence that these regions play complementary but dissociable roles in VWM. PFC patients show behavioral and electrophysiological deficits manifested by attenuation of extrastriate attention and VWM-related neural activity only for stimuli presented to the contralesional visual field. In contrast, patients with BG lesions show behavioral and electrophysiological VWM deficits independent of the hemifield of stimulus presentation but have intact extrastriate attention activity. The results support a model wherein the PFC is critical for top-down intrahemispheric modulation of attention and VWM with the BG involved in global support of VWM processes.

Project description:Human fMRI studies have documented extensively that the content of visual working memory (VWM) can be reliably decoded from fMRI voxel response patterns during the delay period in both the occipito-temporal cortex (OTC), including early visual areas (EVC), and the posterior parietal cortex (PPC).1,2,3,4 Further work has revealed that VWM signal in OTC is largely sustained by feedback from associative areas such as prefrontal cortex (PFC) and PPC.4,5,6,7,8,9 It is unclear, however, if feedback during VWM simply restores sensory representations initially formed in OTC or if it can reshape the representational content of OTC during VWM delay. Taking advantage of a recent finding showing that object representational geometry differs between OTC and PPC in perception,10 here we find that, during VWM delay, the object representational geometry in OTC becomes more aligned with that of PPC during perception than with itself during perception. This finding supports the role of feedback in shaping the content of VWM in OTC, with the VWM content of OTC more determined by information retained in PPC than by the sensory information initially encoded in OTC.

Project description:Working memory (WM) enables the maintenance and manipulation of information relevant to behavioral goals. Variability in WM ability is strongly correlated with IQ [1], and WM function is impaired in many neurological and psychiatric disorders [2, 3], suggesting that this system is a core component of higher cognition. WM storage is thought to be mediated by patterns of activity in neural populations selective for specific properties (e.g., color, orientation, location, and motion direction) of memoranda [4-13]. Accordingly, many models propose that differences in the amplitude of these population responses should be related to differences in memory performance [14, 15]. Here, we used functional magnetic resonance imaging and an image reconstruction technique based on a spatial encoding model [16] to visualize and quantify population-level memory representations supported by multivoxel patterns of activation within regions of occipital, parietal and frontal cortex while participants precisely remembered the location(s) of zero, one, or two small stimuli. We successfully reconstructed images containing representations of the remembered-but not forgotten-locations within regions of occipital, parietal, and frontal cortex using delay-period activation patterns. Critically, the amplitude of representations of remembered locations and behavioral performance both decreased with increasing memory load. These results suggest that differences in visual WM performance between memory load conditions are mediated by changes in the fidelity of large-scale population response profiles distributed across multiple areas of human cortex.

Project description:Working memory (WM) impairments are well recognized in schizophrenia patients (PSZ) and contribute to poor psycho-social outcomes in this population. Distinct neural networks underlay the ability to encode and recall visual and spatial information raising the possibility that profile of visual working memory performance may help pinpoint dysfunctional neural correlates in schizophrenia. This study assessed the resolution and associative aspects of visual working memory deficits in schizophrenia and whether these deficits arise during encoding or maintenance processes. A total of 60 participants (30 PSZ and 30 healthy controls) matched in age, gender and education assessed on a modified object in place (OiPT), a delayed non-match-to-sample (DNMST) and a delayed spatial estimation (DSET) task. Patients demonstrated lower accuracy than controls in binding visual features of the same object and recognizing novel objects as well as lower precision recalling the location of a memorized target. Moreover, response choice set size affected recognition accuracy more in PSZ than controls. However, delay duration affected spatial recall precisions, binding, and recognition accuracy equally in the two groups. Our results suggest that visual working memory (vWM) impairments in schizophrenia predominantly reflect spatial and non-spatial binding deficits, with largely preserved discrete feature information. Moreover, these impairments likely arise more during encoding than during maintenance. These binding deficits may reflect impaired effective neural functional connectivity observed in schizophrenia.

Project description:Intelligent organisms are capable of tracking objects even when they temporarily disappear from sight, a cognitive capacity commonly referred to as visual working memory (VWM). The neural basis of VWM has been the subject of significant scientific debate, with recent work focusing on the relative roles of posterior visual areas, such as the inferior temporal cortex (ITC), and the prefrontal cortex. Here we reexamined the contribution of ITC to VWM by recording from highly selective individual ITC neurons as monkeys engaged in multiple versions of an occlusion-based memory task. As expected, we found strong evidence for a role of ITC in stimulus encoding. We also found that almost half of these selective cells showed stimulus-selective delay period modulation, with a small but significant fraction exhibiting differential responses even in the presence of simultaneously visible interfering information. When we combined the informational content of multiple neurons, we found that the accuracy with which we could decode memory content increased drastically. The memory epoch analyses suggest that behaviorally relevant visual memories were reinstated in ITC. Furthermore, we observed a population-wide enhancement of neuronal response to a match stimulus compared with the same stimulus presented as a nonmatch. The single-cell enhancement preceded any match effects identified in the local field potential, leading us to speculate that enhancement is the result of neural processing local to ITC. Moreover, match enhancement was only later followed by the more commonly observed match suppression. Altogether, the data support the hypothesis that, when a stimulus is held in memory, ITC neurons are actively biased in favor of task-relevant visual representations and that this bias can immediately impact subsequent recognition events.
