Contracted time and expanded space: The impact of circumnavigation on judgements of space and time.

ABSTRACT: The ability to estimate distance and time to spatial goals is fundamental for survival. In cases where a region of space must be navigated around to reach a location (circumnavigation), the distance along the path is greater than the straight-line Euclidean distance. To explore how such circumnavigation impacts on estimates of distance and time, we tested participants on their ability to estimate travel time and Euclidean distance to learned destinations in a virtual town. Estimates for approximately linear routes were compared with estimates for routes requiring circumnavigation. For all routes, travel times were significantly underestimated, and Euclidean distances overestimated. For routes requiring circumnavigation, travel time was further underestimated and the Euclidean distance further overestimated. Thus, circumnavigation appears to enhance existing biases in representations of travel time and distance.

Doubly N-confused and ring-contracted [24]hexaphyrin Pd-complexes as stable antiaromatic N-confused expanded porphyrins.

Project description:As isomers of the regular porphyrins, N-confused porphyrins have attracted extensive attention of chemists because of their unique chemical structures, chemical reactivities, and physical properties, which result in their promising applications in the fields of catalytic chemistry, biochemistry and material science. Typically, N-confused porphyrins are synthesized via acid catalyzed condensation and following oxidation during which lactams are often formed as the byproducts. Here we report doubly N-confused and ring-contracted [24]hexaphyrin(1.1.0.1.1.0) mono- and bis-Pd-complexes as stable antiaromatic N-confused expanded porphyrins, which are synthesized through Pd-catalyzed Suzuki-Miyaura coupling of 1,14-dibromotripyrrin. These macrocycles show a paratropic ring currents, an ill-defined Soret band, a red-shifted weak absorption tail, and a small HOMO-LUMO gap. NBS bromination of the bis Pd-complex give its mono- and dibromides regioselectively, which are effectively used to synthesize a [24]hexaphyrin dimer and a NiII porphyrin-[24]hexaphyrin-NiII porphyrin triad, respectively.

Engineered CRISPR-Cas9 nuclease with expanded targeting space.

Project description:The RNA-guided endonuclease Cas9 cleaves its target DNA and is a powerful genome-editing tool. However, the widely used Streptococcus pyogenes Cas9 enzyme (SpCas9) requires an NGG protospacer adjacent motif (PAM) for target recognition, thereby restricting the targetable genomic loci. Here, we report a rationally engineered SpCas9 variant (SpCas9-NG) that can recognize relaxed NG PAMs. The crystal structure revealed that the loss of the base-specific interaction with the third nucleobase is compensated by newly introduced non-base-specific interactions, thereby enabling the NG PAM recognition. We showed that SpCas9-NG induces indels at endogenous target sites bearing NG PAMs in human cells. Furthermore, we found that the fusion of SpCas9-NG and the activation-induced cytidine deaminase (AID) mediates the C-to-T conversion at target sites with NG PAMs in human cells.

Chemical Multiverse: An Expanded View of Chemical Space.

Project description:Technological advances and practical applications of the chemical space concept in drug discovery, natural product research, and other research areas have attracted the scientific community's attention. The large- and ultra-large chemical spaces are associated with the significant increase in the number of compounds that can potentially be made and exist and the increasing number of experimental and calculated descriptors, that are emerging that encode the molecular structure and/or property aspects of the molecules. Due to the importance and continued evolution of compound libraries, herein, we discuss definitions proposed in the literature for chemical space and emphasize the convenience, discussed in the literature to use complementary descriptors to obtain a comprehensive view of the chemical space of compound data sets. In this regard, we introduce the term chemical multiverse to refer to the comprehensive analysis of compound data sets through several chemical spaces, each defined by a different set of chemical representations. The chemical multiverse is contrasted with a related idea: consensus chemical space.

Time-expanded phase-sensitive optical time-domain reflectometry.

Project description:Phase-sensitive optical time-domain reflectometry (ΦOTDR) is a well-established technique that provides spatio-temporal measurements of an environmental variable in real time. This unique capability is being leveraged in an ever-increasing number of applications, from energy transportation or civil security to seismology. To date, a wide number of different approaches have been implemented, providing a plethora of options in terms of performance (resolution, acquisition bandwidth, sensitivity or range). However, to achieve high spatial resolutions, detection bandwidths in the GHz range are typically required, substantially increasing the system cost and complexity. Here, we present a novel ΦOTDR approach that allows a customized time expansion of the received optical traces. Hence, the presented technique reaches cm-scale spatial resolutions over 1 km while requiring a remarkably low detection bandwidth in the MHz regime. This approach relies on the use of dual-comb spectrometry to interrogate the fibre and sample the backscattered light. Random phase-spectral coding is applied to the employed combs to maximize the signal-to-noise ratio of the sensing scheme. A comparison of the proposed method with alternative approaches aimed at similar operation features is provided, along with a thorough analysis of the new trade-offs. Our results demonstrate a radically novel high-resolution ΦOTDR scheme, which could promote new applications in metrology, borehole monitoring or aerospace.

The Impact of Space and Time on the Functional Output of the Genome.

Project description:Over the past two decades, it has become clear that the multiscale spatial and temporal organization of the genome has important implications for nuclear function. This review centers on insights gained from recent advances in light microscopy on our understanding of transcription. We discuss spatial and temporal aspects that shape nuclear order and their consequences on regulatory components, focusing on genomic scales most relevant to function. The emerging picture is that spatiotemporal constraints increase the complexity in transcriptional regulation, highlighting new challenges, such as uncertainty about how information travels from molecular factors through the genome and space to generate a functional output.

UO<sub>2</sub> <sup>2+</sup>-mediated ring contraction of pyrihexaphyrin: synthesis of a contracted expanded porphyrin-uranyl complex.

Project description:A new mixed hexaphyrin, pyrihexaphyrin (0.1.0.0.1.0) (1), was prepared via an acid catalyzed cyclization between 5,5'-(pyridine-2,6-diyl)bis(pyrrole-2-carbaldehyde) (2) and terpyrrole (3). This expanded porphyrin undergoes a ring contraction upon metallation with uranyl silylamide [UO2[N(SiMe3)2]2] under anaerobic conditions followed by purification over basic aluminum oxide exposed to air. The uranyl-contracted pyrihexaphyrin (0.0.0.0.1.0) complex (4) produced as a result contains a unique structural architecture and possesses a formally 22 π-electron globally aromatic periphery, as inferred from NMR spectroscopy, single crystal X-ray diffraction, and computational analyses. Support for the proposed contraction mechanism came from experimental data and DFT calculations. Proton NMR and mass spectroscopic analysis provided the first insight into expanded porphyrin-mediated activation of the uranyl dication (UO2 2+).

Diel niche variation in mammals associated with expanded trait space.

Project description:Mammalian life shows huge diversity, but most groups remain nocturnal in their activity pattern. A key unresolved question is whether mammal species that have diversified into different diel niches occupy unique regions of functional trait space. For 5,104 extant mammals we show here that daytime-active species (cathemeral or diurnal) evolved trait combinations along different gradients from those of nocturnal and crepuscular species. Hypervolumes of five major functional traits (body mass, litter size, diet, foraging strata, habitat breadth) reveal that 30% of diurnal trait space is unique, compared to 55% of nocturnal trait space. Almost half of trait space (44%) of species with apparently obligate diel niches is shared with those that can switch, suggesting that more species than currently realised may be somewhat flexible in their activity patterns. Increasingly, conservation measures have focused on protecting functionally unique species; for mammals, protecting functional distinctiveness requires a focus across diel niches.

The impact of psychological stress on men's judgements of female body size.

Project description:BACKGROUND: Previous work has suggested that the experience of psychological stress may influence physical attractiveness ideals, but most evidence in favour of this hypothesis remains archival. The objective of this study was to experimentally investigate the impact of stress on men's judgements of female body size. METHODS: Men were randomly assigned to either an experimental group, in which they took part in a task that heightened stress (experimental group, n = 41) or in which they did not take part in such a task (control group, n = 40). Both groups rated the attractiveness of female bodies varying in size from emaciated to obese, completed a measure of appetite sensation, and had their body mass indices (BMIs) measured. RESULTS: Between-groups analyses showed that the experimental group was matched with the control group in terms of mean age, BMI, and appetite sensation. Further analyses showed that men in the experimental group rated a significantly heavier female body size as maximally attractive than the control group. Men in the experimental group also rated heavier female bodies as more attractive and idealised a wider range of female figures than did the control group. CONCLUSION: This study found that the experience of stress was associated with a preference among men for heavier female body sizes. These results indicate that human attractiveness judgements are sensitive to variations in local ecologies and reflect adaptive strategies for dealing with changing environmental conditions.

Space-time structure and wavevector anisotropy in space plasma turbulence.

Project description:Space and astrophysical plasmas often develop into a turbulent state and exhibit nearly random and stochastic motions. While earlier studies emphasize more on understanding the energy spectrum of turbulence in the one-dimensional context (either in the frequency or the wavenumber domain), recent achievements in plasma turbulence studies provide an increasing amount of evidence that plasma turbulence is essentially a spatially and temporally evolving phenomenon. This review presents various models for the space-time structure and anisotropy of the turbulent fields in space plasmas, or equivalently the energy spectra in the wavenumber-frequency domain for the space-time structures and that in the wavevector domain for the anisotropies. The turbulence energy spectra are evaluated in different one-dimensional spectral domains; one speaks of the frequency spectra in the spacecraft observations and the wavenumber spectra in the numerical simulation studies. The notion of the wavenumber-frequency spectrum offers a more comprehensive picture of the turbulent fields, and good models can explain the one-dimensional spectra in the both domains at the same time. To achieve this goal, the Doppler shift, the Doppler broadening, linear-mode dispersion relations, and sideband waves are reviewed. The energy spectra are then extended to the wavevector domain spanning the directions parallel and perpendicular to the large-scale magnetic field. By doing so, the change in the spectral index at different projections onto the one-dimensional spectral domain can be explained in a simpler way.
