Qingryllus jiguanshanensis sp. n. from Sichuan, China, the second species of Qingryllus (Orthoptera, Gryllidae).

ABSTRACT: A second species of Qingryllus Chen & Zheng is described and illustrated, Q. jiguanshanensissp. n., from Sichuan, China. This new species is similar to Q. striofemorus Chen & Zheng, 1995, but differs from the latter by the posterior margin of pronotum being distinctly widened, the veins of tegmina yellowish-white only on lateral side of dorsal area, and the epiphallus distinctly widened posteriorly. A distribution map of the species of this genus and habitus photographs of the new species are presented.

Pseudolebinthus lunipterus sp. nov.: a striking deaf and mute new cricket from Malawi (Orthoptera, Gryllidae, Eneopterinae).

Project description:This article presents an intriguing new cricket species of the tribe Xenogryllini discovered in Northern Malawi. This is the first case of mute and deaf species in the subfamily Eneopterinae; it shows no stridulatory apparatus on short male forewings and no tympana on either side of fore tibiae in both sexes. We introduce the new species and its complete mitogenome and assess phylogenetic relationships based on molecular data obtained from next-generation sequencing genome skimming method. Phylogenetic analyses place the new species within the genus Pseudolebinthus in Xenogryllini, as the sister species of Pseudolebinthus gorochovi Robillard. We describe Pseudolebinthus lunipterus sp. nov., provide illustrations of main morphology, male and female genitalia, photographs of living specimens and information about habitat and update the identification key for species of genus Pseudolebinthus. We discuss the differences between the new species and related taxa and the striking loss of acoustic communication in this cricket.

Study of four Neotropical species of tree crickets Oecanthus Serville, 1831 (Orthoptera, Gryllidae) using cytogenetic and molecular markers.

Project description:Karyotypes in the worldwide subfamily Oecanthinae show variations in diploid number, chromosome morphology, and sex-chromosome system. This study described the chromosome set and phylogenetic relationships of four Neotropical species, Oecanthus lineolatus, O. valensis, O. pallidus, and O. pictus. We used classical cytogenetics and Bayesian Inference for phylogenetic reconstruction, using the mitochondrial genes COI, 12S rRNA, and 16S rRNA; and analyzed the phylogenetic patterns of changes in chromosome numbers, using ChromEvol. We observed differences in chromosome number among species and two different sex-chromosome systems. Oecanthus pictus showed 2n = 21, X0♂/22, XX♀; O. lineolatus, 2n = 20, XY♂/XX♀; and O. valensis and O. pallidus, 2n = 18, XY♂/XX♀. The karyotype of Oecanthus was asymmetric, one group with large chromosomes and variation in heterochromatin distribution, and another with small acrocentric chromosomes. The phylogenetic tree recovered two main groups: one with the Palearctic species and another with species from different bioregions, but with low posterior probability. The Neotropical species grouped separately, O. valensis and O. pictus with Nearctic and Ethiopian species, and O. pallidus and O. lineolatus in another, well-supported clade. Together, the phylogenic and chromosome data suggest descending dysploidy events during the evolution of the group.

Molecular cloning, characterization, and expression analysis of an ecdysone receptor homolog in Teleogryllus emma (Orthoptera: Gryllidae).

Project description:Ecdysteroids are steroid hormones that play important roles in the regulation of Arthropoda animal growth development, larvae ecdysis, and reproduction. The effect of ecdysteroids is mediated by ecdysteroid receptor (EcR). The ecdysone receptor (EcR) belongs to the superfamily of nuclear receptors (NRs) that are ligand-dependent transcription factors. Ecdysone receptor is present only in invertebrates and plays a critical role in regulating the expression of a series of genes during development and reproduction. Here, we isolated and characterized cDNA of the cricket Teleopgryllus emma (Ohmachi & Matsuura) (Orthoptera: Gryllidae) and studied mRNA expression pattern using real time-polymerase chain reaction. The full-length cDNA of T. emma EcR, termed TeEcR, is 2,558 bp and contains a 5'-untranslated region of 555 bp and a 3'-untranslated region of 407 bp. The open reading frame of TeEcR encodes deduced 531-amino acid peptides with a predicted molecular mass of 60.7 kDa. The amino acid sequence of T. emma EcR was similar to that of known EcR especially in the ligand-binding domain of insect EcR. Real-time quantitative reverse transcription-polymerase chain reaction was performed to compare TeEcR mRNA expression level at the whole body and gonad during T. emma development. The data revealed that TeEcR mRNA is differentially expressed during T. emma development, with the highest expression level in late-instar larvae of the body and lowest in third instar. The levels of TeEcR transcripts also vary among gonads development, and levels in ovaries were higher than in testes at every developmental stage. These results suggest that TeEcR may have potential significance to regulate the morphological structure and gonad development of T. emma, due to its expression in different developmental periods.

The complete mitochondrial genome of giant cricket, Tarbinskiellus portentosus (Orthoptera: Gryllidae) and its curation.

Project description:Tarbinskiellus portentosus, commonly known as giant cricket one of the important edible cricket species. However, the genetic information of these species is still limited. Therefore, we have assembled and annotated the first mitochondrial genome of T. portentosus. The mitogenome is 15710 bp long and has GC content of 27.19%. The nucleotide composition is similar with other insect mitogenomes (A 40.6%; T 32.2%; C 17.3%; G 9.9%). The gene organization in the mitogenome of T. portentosus is identical to the mitogenome of other cricket species. The complete mitogenome of T. portentosus consisted 37 genes including 13 protein coding genes, 22 tRNA genes, and two rRNA genes. The newly assembled mitogenome will help molecular biology research on edible crickets. Since mitogenome genes are traditionally used for DNA barcoding and phylogenetic analysis, comparative analysis of T. portentosus mitogenome with other related cricket species will also aid researchers in developing universal primers for species identification toward food security. Apart from the main goal of providing full mitogenome of T. portentosus, paper also provides conceptual workflow based on de novo assembly and its correction for final mitogenome construction.

Rubusdianchuanensis sp. nov. (Rosaceae) from Sichuan and Yunnan, southwest China.

Project description:Rubusdianchuanensis, a new name for the species previously named as R.vicarius by W. O. Focke in 1911, is proposed. A detailed description, illustrations, and remarks on its phenology, ecology, and geographic distribution are provided. This raspberry (subg. Idaeobatus) is endemic to China and was only found in Sichuan and Yunnan, southwest China. Morphologically, it is most similar to R.ovatisepalus but clearly differs from the latter by having leaf densely white or grey tomentose abaxially, usually leaf-like bracts at the base of inflorescence, 1-6 cm long pedicels, and triangular-ovate sepals with acute to caudate apex.

Primulaxilingensis (Primulaceae), a new species from Sichuan, China.

Project description:A new species, Primulaxilingensis K.Huang & Z.X.Fu, sp. nov. (Primulaceae), is described and illustrated. In gross morphology, it is clearly allied to section Minutissimae on account of having stolons, being glabrous, leaf rosette less than or equal to corolla, flower solitary and bract not swollen at base. The new species is easily distinguished by the combination of scape densely yellow farinose, leaf apex acute, rarely broadly obtuse, corolla pale purplish blue and style 3.0-6.0 mm above base of corolla tube, stamens reaching the corolla tube mouth in thrum flower. In addition, the distribution map, morphological comparison of related species and conservation status of the new species are also provided.
