ABSTRACT: Here, we report the full-genome sequence of Milolii virus, a novel single-stranded (positive-sense) RNA virus discovered from Tapinoma melanocephalum ants in Hawaii. The genome is 10,475 nucleotides long, encoding a polyprotein of 3,304 amino acids.

Complete mitochondrial genome sequence of Tapinoma melanocephalum (Hymenoptera: Formicidae).

Project description:Tapinoma melanocephalum is a ubiquitous invasive species and widely distributed in subtropical and tropical regions around the world. Here, we sequenced and annotated the complete mitochondrial genome (mitogenome) of T. melanocephalum. This mitogenome was 15,499 bp long and encoded 13 protein-coding genes (PCGs), 22 transfer RNA genes (tRNAs), and 2 ribosomal RNA unit genes (rRNAs). Compared to other Formicidae species, gene order of T. melanocephalum was not conserved and one tRNA cluster trnW-trnC-trnY converted to trnW-trnY-trnC. The whole mitogenome exhibited heavy AT nucleotide bias (79.5%). All PCGs started with the standard ATN codons. Except for cox1 and nad5 end with the incomplete codon T-, all PCGs terminated with the stop codon TAA. Phylogenetic analysis showed that T. melanocephalum got together with three same subfamily Dolichoderinae species and one Dorylinae species, indicating the close relationship of Dolichoderinae and Dorylinae.

Paving Behavior in Ants and Its Potential Application in Monitoring Two Urban Pest Ants, Solenopsis invicta and Tapinoma melanocephalum.

Project description:Our previous study discovered that two urban pest ants, red imported fire ants, Solenopsis invicta Buren (Formicidae: Myrmicinae), and ghost ants, Tapinoma melanocephalum (Fabricius) (Formicidae: Dolichoderinae), can pave viscose surfaces with particles to facilitate food search and transport. We hypothesize that this paving behavior can be applied to monitor S. invicta and T. melanocephalum. In the present study, 3998 adhesive tapes, each with a food source (sausage), were placed in 20 locations around Guangzhou, China (181-224 tapes per location), and their efficiency to detect S. invicta and T. melanocephalum was compared with two traditional ant-monitoring methods, baiting and pitfall trapping. Overall, S. invicta was detected by 45.6% and 46.4% of baits and adhesive tapes, respectively. In each location, the percentage of adhesive tapes detecting S. invicta and T. melanocephalum was similar when compared to baits and pitfall traps. However, significantly more non-target ant species showed up on bait and pitfall traps. Seven non-target ant species-Pheidole parva Mayr (Formicidae: Myrmicinae), Pheidole nodus Smith (Formicidae: Myrmicinae), Pheidole sinica Wu & Wang (Formicidae: Myrmicinae), Pheidole yeensis Forel (Formicidae: Myrmicinae), Carebara affinis (Jerdon) (Formicidae: Myrmicinae), Camponotus nicobarensis Mayr (Formicidae: Formicinae), and Odontoponera transversa (Smith) (Formicidae: Ponerinae)-also showed tape paving behavior, but they can be easily distinguished morphologically from S. invicta and T. melanocephalum. Our study showed that the paving behavior occurs in different subfamilies of ants (i.e., myrmicinae, dolichoderinae, formicinae, and ponerinae). In addition, paving behavior can potentially be used to develop more specific monitoring methods for S. invicta and T. melanocephalum in urban areas in southern China.

Density-dependent benefits in ant-hemipteran mutualism? The case of the ghost ant Tapinoma melanocephalum (Hymenoptera: Formicidae) and the invasive mealybug Phenacoccus solenopsis (Hemiptera: Pseudococcidae).

Project description:Although density-dependent benefits to hemipterans from ant tending have been measured many times, few studies have focused on integrated effects such as interactions between ant tending, natural enemy density, and hemipteran density. In this study, we tested whether the invasive mealybug Phenacoccus solenopsis is affected by tending by ghost ants (Tapinoma melanocephalum), the presence of parasitoids, mealybug density, parasitoid density and interactions among these factors. Our results showed that mealybug colony growth rate and percentage parasitism were significantly affected by ant tending, parasitoid presence, and initial mealybug density separately. However, there were no interactions among the independent factors. There were also no significant interactions between ant tending and parasitoid density on either mealybug colony growth rate or percentage parasitism. Mealybug colony growth rate showed a negative linear relationship with initial mealybug density but a positive linear relationship with the level of ant tending. These results suggest that benefits to mealybugs are density-independent and are affected by ant tending level.

Roadside Survey of Ants on Oahu, Hawaii.

Project description:Hawaii is home to over 60 ant species, including five of the six most damaging invasive ants. Although there have been many surveys of ants in Hawaii, the last island-wide hand-collection survey of ants on Oahu was conducted in 1988-1994. In 2012, a timed hand-collection of ants was made at 44 sites in a systematic, roadside survey throughout Oahu. Ants were identified and species distribution in relation to elevation, precipitation and soil type was analyzed. To assess possible convenience sampling bias, 15 additional sites were sampled further from roads to compare with the samples near roads. Twenty-four species of ants were found and mapped; Pheidole megacephala (F.), Ochetellus glaber (Mayr), and Technomyrmex difficilis Forel were the most frequently encountered ants. For six ant species, a logistic regression was performed with elevation, average annual precipitation, and soil order as explanatory variables. O. glaber was found in areas with lower precipitation around Oahu. Paratrechina longicornis (Latrielle) and Tetramorium simillimum (Smith, F.) were found more often in lower elevations and in areas with the Mollisol soil order. Elevation, precipitation, and soil type were not significant sources of variation for P. megacephala, Plagiolepis alluaudi Emery, and T. difficilis. P. megacephala was associated with fewer mean numbers of ants where it occurred. Ant assemblages near and far from roads did not significantly differ. Many species of ants remain established on Oahu, and recent invaders are spreading throughout the island. Mapping ant distributions contributes to continued documentation and understanding of these pests.

The HVAC-RV1 genome is most similar to those of picorna-like viruses identified in arthropods but encodes a small domain observed only in negative-sense single-stranded RNA viruses.
