Wavelength-dependent photocycle activity of xanthorhodopsin in the visible region.

ABSTRACT: Xanthorhodopsin (xR) is a dual-chromophore proton-pump photosynthetic protein comprising one retinal Schiff base and one light-harvesting antenna salinixanthin (SX). The excitation wavelength-dependent transient population of the intermediate M demonstrates that the excitation of the retinal at 570 nm leads to the highest photocycle activity and the excitations of SX at 460 and 430 nm reduce the activity to ca. 37% relatively, suggesting an energy transfer pathway from the S₂ state of the SX to the S₁ state of the retinal and a quick internal vibrational relaxation in the S₂ state of SX prior to the energy transfer from SX to retinal.

Infrared and visible absolute and difference spectra of bacteriorhodopsin photocycle intermediates.

Project description:We have used new kinetic fitting procedures to obtain infrared (IR) absolute spectra for intermediates of the main bacteriorhodopsin (bR) photocycle(s). The linear-algebra-based procedures of Hendler et al. (J. Phys. Chem. B, 105, 3319-3228 (2001)) for obtaining clean absolute visible spectra of bR photocycle intermediates were adapted for use with IR data. This led to isolation, for the first time, of corresponding clean absolute IR spectra, including the separation of the M intermediate into its M(F) and M(S) components from parallel photocycles. This in turn permitted the computation of clean IR difference spectra between pairs of successive intermediates, allowing for the most rigorous analysis to date of changes occurring at each step of the photocycle. The statistical accuracy of the spectral calculation methods allows us to identify, with great confidence, new spectral features. One of these is a very strong differential IR band at 1650 cm(-1) for the L intermediate at room temperature that is not present in analogous L spectra measured at cryogenic temperatures. This band, in one of the noisiest spectral regions, has not been identified in any previous time-resolved IR papers, although retrospectively it is apparent as one of the strongest L absorbance changes in their raw data, considered collectively. Additionally, our results are most consistent with Arg82 as the primary proton-release group (PRG), rather than a protonated water cluster or H-bonded grouping of carboxylic residues. Notably, the Arg82 deprotonation occurs exclusively in the M(F) pathway of the parallel cycles model of the photocycle.

SARS-CoV-2 inactivation by ultraviolet radiation and visible light is dependent on wavelength and sample matrix.

Project description:Numerous studies have demonstrated that SARS-CoV-2 can be inactivated by ultraviolet (UV) radiation. However, there are few data available on the relative efficacy of different wavelengths of UV radiation and visible light, which complicates assessments of UV decontamination interventions. The present study evaluated the effects of monochromatic radiation at 16 wavelengths from 222 nm through 488 nm on SARS-CoV-2 in liquid aliquots and dried droplets of water and simulated saliva. The data were used to generate a set of action spectra which quantify the susceptibility of SARS-CoV-2 to genome damage and inactivation across the tested wavelengths. UVC wavelengths (≤280 nm) were most effective for inactivating SARS-CoV-2, although inactivation rates were dependent on sample type. Results from this study suggest that UV radiation can effectively inactivate SARS-CoV-2 in liquids and dried droplets, and provide a foundation for understanding the factors which affect the efficacy of different wavelengths in real-world settings.

Lethal effects of short-wavelength visible light on insects.

Project description:We investigated the lethal effects of visible light on insects by using light-emitting diodes (LEDs). The toxic effects of ultraviolet (UV) light, particularly shortwave (i.e., UVB and UVC) light, on organisms are well known. However, the effects of irradiation with visible light remain unclear, although shorter wavelengths are known to be more lethal. Irradiation with visible light is not thought to cause mortality in complex animals including insects. Here, however, we found that irradiation with short-wavelength visible (blue) light killed eggs, larvae, pupae, and adults of Drosophila melanogaster. Blue light was also lethal to mosquitoes and flour beetles, but the effective wavelength at which mortality occurred differed among the insect species. Our findings suggest that highly toxic wavelengths of visible light are species-specific in insects, and that shorter wavelengths are not always more toxic. For some animals, such as insects, blue light is more harmful than UV light.

Visible and infrared three-wavelength modulated multi-directional actuators.

Project description:In recent years, light-guided robotic soft actuators have attracted intense scientific attention and rapidly developed, although it still remains challenging to precisely and reversibly modulate the moving directions and shape morphing modes of soft actuators with ease of stimulating operation. Here we report a strategy of building a multi-stimuli-responsive liquid crystal elastomer soft actuator system capable of performing not only multi-directional movement, but also different shape morphing modes. This strategy is based on the selective stimulation of specific domains of the hierarchical structured actuator through the modulation of three wavelength bands (520, 808, 980 nm) of light stimulus, which release the actuation system from light scanning position/direction restriction. Three near-infrared dual-wavelength modulated actuators and one visible/infrared tri-wavelength modulated multi-directional walker robot are demonstrated in this work. These devices have broad application prospects in robotic and biomimetic technology.

Wavelength-Orthogonal Stiffening of Hydrogel Networks with Visible Light.

Project description:Herein, we introduce the wavelength-orthogonal crosslinking of hydrogel networks using two red-shifted chromophores, i.e. acrylpyerene (AP, λactivation =410-490 nm) and styrylpyrido[2,3-b]pyrazine (SPP, λactivation =400-550 nm), able to undergo [2+2] photocycloaddition in the visible-light regime. The photoreactivity of the SPP moiety is pH-dependent, whereby an acidic environment inhibits the cycloaddition. By employing a spiropyran-based photoacid generator with suitable absorption wavelength, we are able to restrict the activation wavelength of the SPP moiety to the green light region (λactivation =520-550 nm), enabling wavelength-orthogonal activation of the AP group. Our wavelength-orthogonal photochemical system was successfully applied in the design of hydrogels whose stiffness can be tuned independently by either green or blue light.

Two bathointermediates of the bacteriorhodopsin photocycle, from time-resolved nanosecond spectra in the visible.

Project description:Time-resolved measurements were performed on wild-type bacteriorhodopsin with an optical multichannel analyzer in the spectral range 350-735 nm, from 100 ns to the photocycle completion, at four temperatures in the 5-30 degrees C range. The intent was to examine the possibility of two K-like bathochromic intermediates and to obtain their spectra and kinetics in the visible. The existence of a second K-like intermediate, termed KL, had been postulated (Shichida et al., Biochim. Biophys. Acta 1983, 723, 240-246) to reconcile inconsistencies in data in the pico- and microsecond time domains. However, introduction of KL led to a controversy, since neither its visible spectrum nor its kinetics could be confirmed. Infrared data (Dioumaev and Braiman, J. Phys. Chem. B 1997, 101, 1655-1662) revealed a state which might have been considered a homologue to KL, but it had a kinetic pattern different from that of the earlier proposed KL. Here, we characterize two distinct K-like intermediates, K(E) ("early") and K(L) ("late"), by their spectra and kinetics in the visible as revealed by global kinetic analysis. The K(E)-to-K(L) transition has a time constant of approximately 250 ns at 20 degrees C, and describes a shift from K(E) with lambda(max) at approximately 600 nm and extinction of approximately 56,000 M(-1) x cm(-1) to K(L) with lambda(max) at approximately 590 nm and extinction of approximately 50,000 M(-1) x cm(-1). The temperature dependence of this transition is characterized by an enthalpy of activation of DeltaH(++) approximately 40 kJ/mol and a positive entropy of activation of DeltaS(++)/R approximately 4. The consequences of multiple K-like states for interpreting the spectral evolution in the early stages of the photocycle are discussed.

Towards visible-wavelength passively mode-locked lasers in all-fibre format.

Project description:Mode-locked fibre lasers (MLFLs) are fundamental building blocks of many photonic systems used in industrial, scientific and biomedical applications. To date, 1-2 μm MLFLs have been well developed; however, passively mode-locked fibre lasers in the visible region (380-760 nm) have never been reported. Here, we address this challenge by demonstrating an all-fibre visible-wavelength passively mode-locked picosecond laser at 635 nm. The 635 nm mode-locked laser with an all-fibre figure-eight cavity uses a Pr/Yb codoped ZBLAN fibre as the visible gain medium and a nonlinear amplifying loop mirror as the mode-locking element. First, we theoretically predict and analyse the formation and evolution of 635 nm mode-locked pulses in the dissipative soliton resonance (DSR) regime by solving the Ginzburg-Landau equation. Then, we experimentally demonstrate the stable generation of 635 nm DSR mode-locked pulses with a pulse duration as short as ~96 ps, a radio-frequency signal-to-noise ratio of 67 dB and a narrow spectral bandwidth of <0.1 nm. The experimental results are in excellent agreement with our numerical simulations. In addition, we also observe 635 nm noise-like pulse operation with a wide (>1 nm) and modulated optical spectrum. This work represents an important step towards miniaturized ultrafast fibre lasers in the visible spectral region.

Patterned photostimulation via visible-wavelength photonic probes for deep brain optogenetics.

Project description:Optogenetic methods developed over the past decade enable unprecedented optical activation and silencing of specific neuronal cell types. However, light scattering in neural tissue precludes illuminating areas deep within the brain via free-space optics; this has impeded employing optogenetics universally. Here, we report an approach surmounting this significant limitation. We realize implantable, ultranarrow, silicon-based photonic probes enabling the delivery of complex illumination patterns deep within brain tissue. Our approach combines methods from integrated nanophotonics and microelectromechanical systems, to yield photonic probes that are robust, scalable, and readily producible en masse. Their minute cross sections minimize tissue displacement upon probe implantation. We functionally validate one probe design in vivo with mice expressing channelrhodopsin-2. Highly local optogenetic neural activation is demonstrated by recording the induced response-both by extracellular electrical recordings in the hippocampus and by two-photon functional imaging in the cortex of mice coexpressing GCaMP6.

Excitation wavelength independent visible color emission of carbon dots.

Project description:Carbon dots (CDs) usually emit a strong blue light and excitation wavelength dependent long wavelength lights. This significantly limits their applications because one has to use a series of different excitation light sources to get different colors and the long wavelength emissions are usually very weak. We found that one type of CDs synthesized from p-phenylenediamine could emit various long wavelength lights (green to red) independent of the excitation wavelength when dispersed in different solvents. The photoluminescence quantum yields of the same CDs were 10-35% in different solvents for different color emissions. Based on this solvent-color effect, we further mixed the same CDs with different polymers to form solid CD films for various color emissions, and these film emissions were also excitation wavelength independent. Multicolor LEDs were demonstrated with the same CDs in solution and solid film states for color displays.

Optically Graded Ultra Dark Absorber for Visible and Near-infrared Wavelength Range.

Project description:Near perfect absorbers find application in many areas including solar cells, energy harvesting and antireflection coatings for space applications. Here we report the use of optical gradation concept to fabricate a near perfect absorber on etched Si wafer. As a proof of concept, 99.4% absorption is achieved in the broad range of 300 nm to 2000 nm. Moreover, absorption capacity of optically graded surface remains higher than 99% up to beam incident angle of 50°. While carbon nanotubes (index ~1.1) are used as top layer, subsequent layers with increasing optical index across the thickness are chosen so as to satisfy zero reflection condition on multilayered assembly. Inward bending of incident beam and total internal reflection of reflected beam caused due to optical index gradient contributes to absorb the incident beam more efficiently. In addition, multiple scattering of incident beam due to the presence of multiscale feature size in graded assembly helps to absorb shorter as well as longer wavelengths of incident light. The graded assembly shows contact angle of 160° with roll-off angle equal to 5° implying that the graded absorber is not only super black but also superhydrophobic and self-cleaning in nature. The combination of properties shown by the super absorber makes it very attractive, especially for next generation solar cells to harness energy in the wavelength range of 1000 nm to 2000 nm.
