Boosting the down-shifting luminescence of rare-earth nanocrystals for biological imaging beyond 1500?nm.

ABSTRACT: In vivo fluorescence imaging in the near-infrared region between 1500-1700?nm (NIR-IIb window) affords high spatial resolution, deep-tissue penetration, and diminished auto-fluorescence due to the suppressed scattering of long-wavelength photons and large fluorophore Stokes shifts. However, very few NIR-IIb fluorescent probes exist currently. Here, we report the synthesis of a down-conversion luminescent rare-earth nanocrystal with cerium doping (Er/Ce co-doped NaYbF4 nanocrystal core with an inert NaYF4 shell). Ce doping is found to suppress the up-conversion pathway while boosting down-conversion by ~9-fold to produce bright 1550?nm luminescence under 980?nm excitation. Optimization of the inert shell coating surrounding the core and hydrophilic surface functionalization minimize the luminescence quenching effect by water. The resulting biocompatible, bright 1550?nm emitting nanoparticles enable fast in vivo imaging of blood vasculature in the mouse brain and hindlimb in the NIR-IIb window with short exposure time of 20?ms for rare-earth based probes.Fluorescence imaging in the near-infrared window between 1500-1700?nm (NIR-IIb window) offers superior spatial resolution and tissue penetration depth, but few NIR-IIb probes exist. Here, the authors synthesize rare earth down-converting nanocrystals as promising fluorescent probes for in vivo imaging in this spectral region.

Project description:The optical imaging guided tumor vessels and vascular malformation visualization by using the second near infrared emission beyond 1500 nm (NIR-II) is emerged as the next generation fluorescence imaging technique for early tumor diagnosis and identification of tumor-associated vascular features. On the other hand, developing theranostic probes for NIR-II imaging guided photothermal therapy (PTT) is of great significance, which is rarely explored. Herein, a high performance theranostic nanoplatform based on the core-shell structured NaLuF4 nanorods@polydopamine (denoted as NRs@PDA) by integrating the new advanced NIR-II imaging beyond 1500 nm with PTT function was developed for tumor-associated vascular malformation visualization and imaging-guided PTT. Methods: In this work, the hydrophilic NaLuF4 NRs@PDA therapeutic probe was synthesized by using a reverse microemulsion method. The crystal phase, morphology, emission spectra and photothermal performance of the synthesized samples were systematically characterized. The NIR-II optical imaging and photothermal properties were investigated by in vitro and in vivo experiments. Results: The NaLuF4 NRs@PDA therapeutic probe possessed efficient NIR-II emission centered at 1525 nm with high quantum yield (QY), good photo-stability and high biocompatibility. In vivo NIR-IIb imaging based on the designed probe can clearly visualize the whole-body vessel and brain vessel with high spatial resolution, especially tumor-associated vessels. In addition, in vitro and in vivo experiments also demonstrated that the designed NaLuF4 NRs@PDA probe possessed efficient photothermal conversion efficiency (40.18%) for PTT ablation of tumor. Conclusion: With the excellent NIR-II imaging ability and PTT of tumor, the designed theranostic nanoplatform successfully realize the simultaneous tumor vessel diagnosis and tumor therapy, which may provide the opportunity of designing new theranostic bioprobes with combination of the NIR-II optical imaging technique and PTT function for tumor diagnosis and therapy.

Project description:We report the synthesis and properties of the first representative of a new class of PtL2 complexes with ambidentate mixed-donor cyanoxime ligands [L = 2-cyano-2-oximino-N,N'-diethylaminoacetamide, DECO (1)]. Three differently colored polymorphs of "Pt(DECO)2" (3-5) were isolated, with the first two being crystallographically characterized. The dark-green complex [Pt(DECO)2]n (5) spontaneously forms in aqueous solution via aggregation of yellow monomeric complex 3 into the red dimer [Pt(DECO)2]2 (4), followed by further oligomerization into coordination polymer 5. A spectroscopic and light-scattering study revealed a "poker-chips"-type 1D polymeric structure of 5 in which units are held by noncovalent metallophilic interactions, forming a Pt---Pt wire. The polymer 5 shows a broad absorption at 400-900 nm and emission at unusually long wavelengths in the range of 1000-1100 nm in the solid state. The near-infrared (NIR) emission of polymer 5 is due to the formation of a small amount of nonstoichiometric mixed-valence Pt(II)/Pt(IV) species during synthesis. A featureless electron paramagnetic resonance spectrum of solid sample 5 recorded at +23 and -193 °C evidences the absence of Pt(III) states, and the compound represents a "solid solution" containing mixed-valence Pt(II)/Pt(IV) centers. Exposure of KBr pellets with 5% 5 to Br2 vapors leads to an immediate ∼30% increase in the intensity of photoluminescence at 1024 nm, which confirms the role and importance of mixed-valence species for the NIR emission. Thus, the emission is further enhanced upon additional oxidation of Pt(II) centers, which improves delocalization of electrons along the Pt---Pt vector. Other polymorph of the "Pt(DECO)2" complex--monomer--did not demonstrate luminescent properties in solutions and the solid state. An excitation scan of 5 embedded in KBr tablets revealed an emission only weakly dependent on the wavelength of excitation. The NIR emission of quasi-1D complex 5 was studied in the range of -193 to +67 °C. Data showed a blue shift of λmax and a simultaneous increase in the emission line intensity with a temperature rise, which is explained by analogy with similar behavior of known quasi-1D K2[Pt(CN)4]-based solids, quantum dots, and quantum wells with delocalized carriers. The presented finding opens a route to a new class of platinum cyanoxime based NIR emissive complexes that could be used in the design of novel NIR emitters and imaging agents.
