Project description:Speech processing is built upon encoding by the auditory nerve and brainstem, yet we know very little about how these processes unfold in specific subcortical structures. These structures are deep and respond quickly, making them difficult to study during ongoing speech. Recent techniques have begun to address this problem, but yield temporally broad responses with consequently ambiguous neural origins. Here, we describe a method that pairs re-synthesized 'peaky' speech with deconvolution analysis of electroencephalography recordings. We show that in adults with normal hearing the method quickly yields robust responses whose component waves reflect activity from distinct subcortical structures spanning auditory nerve to rostral brainstem. We further demonstrate the versatility of peaky speech by simultaneously measuring bilateral and ear-specific responses across different frequency bands and discuss the important practical considerations such as talker choice. The peaky speech method holds promise as a tool for investigating speech encoding and processing, and for clinical applications.

Project description:HypothesisThe hypothesis tested was that forward masking of the speech-evoked auditory brainstem response (sABR) increases peak latency as an inverse function of masker-signal interval (Δt), and that the overall persistence of forward masking is age dependent.BackgroundOlder listeners exhibit deficits in forward masking. If forward-masked sABRs provide an objective measure of the susceptibility of speech sounds to prior stimulation, then this provides a novel approach to examining the age dependence of temporal processing.MethodsA /da/ stimulus forward masked by speech-shaped noise (Δt = 4-64 ms) was used to measure sABRs in 10 younger and nine older participants. Forward masking of subsegments of the /da/ stimulus (Δt = 16 ms) and click trains (Δt = 0-64 ms) was also measured.ResultsForward-masked sABRs from young participants showed an increase in latency with decreasing Δt for the initial peak. Latency shifts for later peaks were smaller and more uniform. None of the peak latencies returned to baseline by Δt = 64 ms. Forward-masked /da/ subsegments showed peak latency shifts that did not depend simply on peak position, while forward-masked click trains showed latency shifts that were dependent on click position. The sABRs from older adults were less robust but confirmed the viability of the approach.ConclusionForward masking of the sABR provides an objective measure of the susceptibility of the auditory system to prior stimulation. Failure of recovery functions to return to baseline suggests an interaction between forward masking by the prior masker and temporal effects within the stimulus itself.

Project description:Rapid recognition and categorization of sounds are essential for humans and animals alike, both for understanding and reacting to our surroundings and for daily communication and social interaction. For humans, perception of speech sounds is of crucial importance. In real life, this task is complicated by the presence of a multitude of meaningful non-speech sounds. The present behavioural, magnetoencephalography (MEG) and functional magnetic resonance imaging (fMRI) study was set out to address how attention to speech versus attention to natural non-speech sounds within complex auditory scenes influences cortical processing. The stimuli were superimpositions of spoken words and environmental sounds, with parametric variation of the speech-to-environmental sound intensity ratio. The participants' task was to detect a repetition in either the speech or the environmental sound. We found that specifically when participants attended to speech within the superimposed stimuli, higher speech-to-environmental sound ratios resulted in shorter sustained MEG responses and stronger BOLD fMRI signals especially in the left supratemporal auditory cortex and in improved behavioural performance. No such effects of speech-to-environmental sound ratio were observed when participants attended to the environmental sound part within the exact same stimuli. These findings suggest stronger saliency of speech compared with other meaningful sounds during processing of natural auditory scenes, likely linked to speech-specific top-down and bottom-up mechanisms activated during speech perception that are needed for tracking speech in real-life-like auditory environments.

Project description:Although we have convincing evidence that attention to auditory stimuli modulates neuronal responses at or before the level of primary auditory cortex (A1), the underlying physiological mechanisms are unknown. We found that attending to rhythmic auditory streams resulted in the entrainment of ongoing oscillatory activity reflecting rhythmic excitability fluctuations in A1. Strikingly, although the rhythm of the entrained oscillations in A1 neuronal ensembles reflected the temporal structure of the attended stream, the phase depended on the attended frequency content. Counter-phase entrainment across differently tuned A1 regions resulted in both the amplification and sharpening of responses at attended time points, in essence acting as a spectrotemporal filter mechanism. Our data suggest that selective attention generates a dynamically evolving model of attended auditory stimulus streams in the form of modulatory subthreshold oscillations across tonotopically organized neuronal ensembles in A1 that enhances the representation of attended stimuli.

Project description:Selective attention to speech versus nonspeech signals in complex auditory input could produce top-down modulation of cortical regions previously linked to perception of spoken, and even visual, words. To isolate such top-down attentional effects, we contrasted 2 equally challenging active listening tasks, performed on the same complex auditory stimuli (words overlaid with a series of 3 tones). Instructions required selectively attending to either the speech signals (in service of rhyme judgment) or the melodic signals (tone-triplet matching). Selective attention to speech, relative to attention to melody, was associated with blood oxygenation level-dependent (BOLD) increases during functional magnetic resonance imaging (fMRI) in left inferior frontal gyrus, temporal regions, and the visual word form area (VWFA). Further investigation of the activity in visual regions revealed overall deactivation relative to baseline rest for both attention conditions. Topographic analysis demonstrated that while attending to melody drove deactivation equivalently across all fusiform regions of interest examined, attending to speech produced a regionally specific modulation: deactivation of all fusiform regions, except the VWFA. Results indicate that selective attention to speech can topographically tune extrastriate cortex, leading to increased activity in VWFA relative to surrounding regions, in line with the well-established connectivity between areas related to spoken and visual word perception in skilled readers.

Project description:BackgroundSome evidence suggests that young adults exhibit a selective laterality of auditory brainstem response (ABR) elicited with speech stimuli. Little is known about such an auditory laterality in older adults.ObjectiveThe aim of this study was to investigate possible asymmetric auditory brainstem processing between right and left ear presentation in older adults.MethodsSixty-two older adults presenting with normal hearing thresholds according to their age and who were native speakers of Quebec French participated in this study. ABR was recorded using click and a 40-ms /da/ syllable. ABR was elicited through monaural right and monaural left stimulation. Latency and amplitude for click-and speech-ABR components were compared between right and left ear presentations. In addition, for the /da/ syllable, a fast Fourier transform analysis of the sustained frequency-following response (FFR) of the vowel was performed along with stimulus-to-response and right-left ear correlation analyses.ResultsNo significant differences between right and left ear presentation were found for amplitudes and latencies of the click-ABR components. Significantly shorter latencies for right ear presentation as compared to left ear presentation were observed for onset and offset transient components (V, A and O), sustained components (D and E), and voiced transition components (C) of the speech-ABR. In addition, the spectral amplitude of the fundamental frequency (F0) was significantly larger for the left ear presentation than the right ear presentation.ConclusionsResults of this study show that older adults with normal hearing exhibit symmetric encoding for click stimuli at the brainstem level between the right and left ear presentation. However, they present with brainstem asymmetries for the encoding of selective stimulus components of the speech-ABR between the right and left ear presentation. The right ear presentation of a /da/ syllable elicited reduced neural timing for both transient and sustained components compared to the left ear. Conversely, a stronger left ear F0 encoding was observed. These findings suggest that at a preattentive, sensory stage of auditory processing, older adults lateralize speech stimuli similarly to young adults.

Project description:The primary aim of this study was to investigate whether auditory brainstem response (ABR) and speech perception in noise (SPiN) were associated with occupational noise exposure in normal hearing young factory workers. Forty young adults occupationally exposed to noise and 40 non-exposed young adults (control group) from Zhejiang province in China were selected. All participants presented with normal hearing thresholds and distortion product otoacoustic emissions. Participants were evaluated with the Mandarin Bamford-Kowal-Bench (BKB) test and ABR. The latter was obtained for click stimulus at 50, 60, 70, 80, and 90 dBnHL. Peak-to-trough amplitudes and latencies for waves I and V were obtained. The ABR wave I amplitude, the wave I/V amplitude ratio, the slope of the wave I amplitude growth as a function of stimulus intensity (AMP-ISlope), and the wave V latency shift with ipsilateral noise (LAT-VSlope) were used as ABR outcomes. Finally, equivalent continuous average sound pressure level normalized to 8 h (LAeq.8h) and cumulative noise exposure (CNE) were obtained for noise-exposed participants. No significant differences between groups were found for any ABR outcomes. Noise-exposed participants exhibited worse BKB scores than control group participants. A multivariate regression model showed that 23.3% of the variance in BKB scores was explained by group category (exposed vs. non-exposed) and hearing thresholds. However, since none of the ABR outcomes exploring cochlear synaptopathy were associated with noise exposure, we cannot conclude that cochlear synaptopathy was the contributing factor for the differences between groups for BKB scores. Factors that go beyond sensory processing may explain such results, especially given socio-economic differences between the noise-exposed and control groups. We conclude that in this sample of participants, occupational noise exposure was not associated with signs of cochlear synaptopathy as measured by ABR and BKB.

Project description:The frequency-specific tone-evoked auditory brainstem response (ABR) is an indispensable tool in both the audiology clinic and research laboratory. Most frequently, the toneburst ABR is used to estimate hearing thresholds in infants, toddlers, and other patients for whom behavioral testing is not feasible. Therefore, results of the ABR exam form the basis for decisions regarding interventions and hearing habilitation with implications extending far into the child's future. Currently, responses are elicited by periodic sequences of toneburst stimuli presented serially to one ear at a time, which take a long time to measure multiple frequencies and intensities, and provide incomplete information if the infant wakes up early. Here, we describe a new method, the parallel ABR (pABR), which uses randomly timed toneburst stimuli to simultaneously acquire ABR waveforms to five frequencies in both ears. Here, we describe the pABR and quantify its effectiveness in addressing the greatest drawback of current methods: test duration. We show that in adults with normal hearing the pABR yields high-quality waveforms over a range of intensities, with similar morphology to the standard ABR in a fraction of the recording time. Furthermore, longer latencies and smaller amplitudes for low frequencies at a high intensity evoked by the pABR versus serial ABR suggest that responses may have better place specificity due to the masking provided by the other simultaneous toneburst sequences. Thus, the pABR has substantial potential for facilitating faster accumulation of more diagnostic information that is important for timely identification and treatment of hearing loss.

Project description:Selective listening to cocktail-party speech involves a network of auditory and inferior frontal cortical regions. However, cognitive and motor cortical regions are differentially activated depending on whether the task emphasizes semantic or phonological aspects of speech. Here we tested whether processing of cocktail-party speech differs when participants perform a shadowing (immediate speech repetition) task compared to an attentive listening task in the presence of irrelevant speech. Participants viewed audiovisual dialogues with concurrent distracting speech during functional imaging. Participants either attentively listened to the dialogue, overtly repeated (i.e., shadowed) attended speech, or performed visual or speech motor control tasks where they did not attend to speech and responses were not related to the speech input. Dialogues were presented with good or poor auditory and visual quality. As a novel result, we show that attentive processing of speech activated the same network of sensory and frontal regions during listening and shadowing. However, in the superior temporal gyrus (STG), peak activations during shadowing were posterior to those during listening, suggesting that an anterior-posterior distinction is present for motor vs. perceptual processing of speech already at the level of the auditory cortex. We also found that activations along the dorsal auditory processing stream were specifically associated with the shadowing task. These activations are likely to be due to complex interactions between perceptual, attention dependent speech processing and motor speech generation that matches the heard speech. Our results suggest that interactions between perceptual and motor processing of speech relies on a distributed network of temporal and motor regions rather than any specific anatomical landmark as suggested by some previous studies.

Project description:Listeners use the spatial location or change in spatial location of coherent acoustic cues to aid in auditory object formation. From stimulus-evoked onset responses in normal-hearing listeners using electroencephalography (EEG), we have previously shown measurable tuning to stimuli changing location in quiet, revealing a potential window into the cortical representations of auditory scene analysis. These earlier studies used non-fluctuating, spectrally narrow stimuli, so it was still unknown whether previous observations would translate to speech stimuli, and whether responses would be preserved for stimuli in the presence of background maskers. To examine the effects that selective auditory attention and interferers have on object formation, we measured cortical responses to speech changing location in the free field with and without background babble (+6 dB SNR) during both passive and active conditions. Active conditions required listeners to respond to the onset of the speech stream when it occurred at a new location, explicitly indicating ‘yes’ or ‘no’ to whether the stimulus occurred at a block-specific location either 30 degrees to the left or right of midline. In the aggregate, results show similar evoked responses to speech stimuli changing location in quiet compared to babble background. However, the effect of the two background environments diverges somewhat when considering the magnitude and direction of the location change and where the subject was attending. In quiet, attention to the right hemifield appeared to evoke a stronger response than attention to the left hemifield when speech shifted in the rightward direction. No such difference was found in babble conditions. Therefore, consistent with challenges associated with cocktail party listening, directed spatial attention could be compromised in the presence of stimulus noise and likely leads to poorer use of spatial cues in auditory streaming.
