A New Species of the Rare Genus Anguillonema Fuchs, 1938 (Nematoda: Hexatylina, Sphaerularioidea) with Its Molecular Phylogenetic Study.

ABSTRACT: Anguillonema amolensis n. sp. is described and illustrated based on its morphological, morphometric, and molecular characters. The new species is characterized by its 575 to 820 ?m long and wide body (body width at vulva = 30 to 59 ?m), irregularly ventrally curved after fixation, five to six lines in lateral fields, 6.0 to 7.5 ?m long stylet with small rounded knobs, pharynx lacking a median bulb, pharyngo-intestinal junction anterior to nerve ring and excretory pore, females with monodelphic-prodelphic reproductive system, 15 to 19 ?m long conical tail with broad rounded tip, and males absent. The new species is compared with two known species of the genus, Anguillonema poligraphi and A. crenati. Molecular phylogenetic studies of the new species using partial sequences of small subunit (SSU) rDNA revealed that it forms a clade with an unidentified nematode species and two species of the genus Howardula. In phylogenetic analyses using partial sequences of the 28S rDNA (D2-D3 segment), the new species formed a monophyletic group with species belonging to two genera Howardula and Parasitylenchus.

Project description:A new species of the genus Paurodontella, P. composticola n. sp., collected from Nazar Abad City, Alborz Province, Iran, is described and illustrated. The new species has a body length of 803-1053???m (females n?=?10) and 620 and 739???m (males n?=?2). The cuticle is weakly annulated with four lateral lines. Cephalic region is annulated and continuous with body contour. The stylet is 8.0 to 9.0???m long with asymmetrical knobs. Esophageal basal bulb is present with a small posterior extension projecting into the intestine. Excretory pore is situated at the level of esophageal basal bulb region. Post-uterine sac is 5 to 8???m long and uterus is without diverticulum. Tails of both sexes are similar, short and sub-cylindrical. Project description:BackgroundThe genus Marshallagia (Family Haemonchidae, subfamily Ostertagiinae) contains multiple species of nematodes parasitising the abomasum (or duodenum) of ruminants, in particular of Caprinae. Male specimens have been described to be polymorphic with the frequent/major morphotype initially described in the genus Marshallagia while the minor/rare morphotype was initially often placed in the genus Grossospicularia. Due to common morphological features, certain pairs of morphotypes were suggested to belong to the same species such as Marshallagia marshalli/M. occidentalis. However, molecular evidence to confirm these pairs of morphotypes belonging to the same species is missing.MethodsIn the present study, Marshallagia sp. were collected from domestic sheep in Uzbekistan. Male specimens were morphologically described with particular emphasis on the structure of the bursa copulatrix. After DNA isolation from morphologically identified specimens, PCRs targeting the ribosomal internal transcribed spacer 2 (ITS2) and mitochondrial cytochrome c oxidase subunit 1 (cox1) regions were conducted. After Sanger sequencing, maximum likelihood phylogenetic analyses and pairwise identities between sequences were calculated.ResultsThe major morphotypes of M. marshalli, M. schumakovitschi and M. uzbekistanica and the minor morphotypes M. occidentalis, M. trifida and M. sogdiana were identified and their morphology was documented in detail. ITS2 sequences showed little variation and did not allow diagnosing species. In contrast, phylogenetic analysis of cox1 sequences identified highly supported clusters and verified that M. marshalli, M. occidentalis and M. uzbekistanica are different morphotypes of the species M. marshalli while M. schumakovitschi and M. trifida represent distinct morphotypes of M. trifida. For M. sogdiana no corresponding major morphotype could be identified in the present study. Due to a large barcoding gap, comparison of cox1 sequences in terms of percent identity was sufficient to reliably assign the sequences to a particular species without phylogenetic analysis.ConclusionsThe data presented here create a framework that will allow the classification of other members of the genus in the future and underline that parallel morphological and molecular analysis of specimens is crucial to improve the taxonomy of polymorphic species.

Project description:In the summer of 2016, a field of corn (Zea mays) in Spencer County, Indiana was observed with heavily stunted plants, and from the affected roots a large number of cysts were recovered. Soil samples were submitted to one of us (JF), who extracted the nematode cysts and sent them to the USDA-ARS, Mycology and Nematology Genetic Diversity and Biology Laboratory (MNGDBL), Beltsville, MD for morphological and molecular identification. Cysts and the recovered second-stage juveniles (J2) that were examined morphologically conformed to the measurements of Vittatidera zeaphila, the goose cyst nematode originally described from Tennessee, USA in 2010. The molecular analysis of J2 showed the sample from Spencer County matched exactly with V. zeaphila according to ribosomal DNA markers ITS, 28S, and 18S, and with mitochondrial cytochrome oxidase I (COI). The nuclear marker heat shock protein 90 (Hsp90) was also analyzed for the first time from the Indiana population of V. zeaphila. Similarities to existing cyst nematode sequences are reported herein. Geographically, although the county is across the Ohio River from Kentucky, the previously reported Hickman County, Kentucky location and Indiana detection are approximately 200 miles apart. Project description:Three new species of Aporcelaimoides from natural habitats in Vietnam are studied, described and illustrated, including line drawings, LM and/or SEM pictures. Aporcelaimoidesbrevistylum sp. n. is characterized by its body 1.95-2.90 mm long, lip region offset by deep constriction and 17-18 µm broad, ventral side of mural odontostyle 11-14 µm long with aperture occupying 62-71% of its length, neck 663-767 µm long, pharyngeal expansion occupying 58-66% of total neck length, uterus a simple tube 85-182 µm long, pars refringens vaginae absent, V = 55-63, tail short and rounded (34-46 µm, c = 49-76, c' = 0.6-0.8), spicules 67-86 µm long, and one ventromedian supplement out the range of spicules. Aporcelaimoidesminor sp. n. is distinguished in having body 2.09-2.61 mm long, lip region offset by deep constriction and 19-20 µm broad, mural odontostyle 14-16 µm long at its ventral side with aperture occupying 73-84% of its length, neck 579-649 µm long, pharyngeal expansion occupying 57-66% of total neck length, uterus a simple tube 44-69 µm long, pars refringens vaginae well developed, V = 48-56, female tail very short, rounded conoid or truncate (14-26 µm, c = 90-146, c' = 0.3-0.6), and male unknown. Aporcelaimoidessilvaticum sp. n. is characterized by its body 2.09-2.60 mm long, lip region offset by depression and 17-18 µm broad, mural odontostyle 11-12 µm long at its ventral side with aperture occupying 60-66% of its length, neck 597-720 µm long, pharyngeal expansion occupying 58-64% of total neck length, uterus a simple tube 128-243 µm long, pars refringens vaginae well developed, V = 58-60, tail short and rounded (27-37 µm, c = 67-94, c' = 0.6-0.7), spicules 64-75 µm long, and two or three widely spaced ventromedian supplements bearing hiatus. The genus Aporcelaimoides is restored, its diagnosis emended, and three species of Sectonema, namely Sectonemaamazonicum, Sectonemahaguei and Sectonemamoderatum, transferred to it. An updated list of its species, a key to their identification and a tabular compendium with the most important morphometric features are also presented.
