Microsatellites for Phytolacca acinosa (Phytolaccaceae), a traditional medicinal herb.

ABSTRACT: Premise of the study:Phytolacca acinosa (Phytolaccaceae) is a traditional Chinese herb with multiple medicinal uses and is an important pigment source. Due to excessive human harvesting, the population numbers and sizes have decreased dramatically. Methods and Results:Using an enriched genomic library, we developed and characterized 15 microsatellite primers for P. acinosa, 13 of which were polymorphic. The number of alleles varied from two to seven. The observed heterozygosity and expected heterozygosity per locus ranged from 0.267 to 1.000 and 0.331 to 0.743, respectively. All of the primers that were developed were also successfully applied in P. americana. Conclusions:These markers should be useful in probing the genetic diversity, genetic structure, and mating systems of P. acinosa, which could provide information about protecting and sustainably harvesting this species.

Screening of molluscicidal strain against Oncomelania hupensis from the rhizosphere of medicinal plant Phytolacca acinosa Roxb.

Project description:The research aimed to screen and exploit molluscicidal microorganisms against Oncomelania hupensis, from the rhizosphere of medicinal plant, Phytolacca acinosa Roxb., and one strain named as SL-30 was obtained with excellent molluscicidal activity. The freeze-dried powder of exocellular broth (EXB) of SL-30 could kill 100% of snails at a concentration of 48 mg/l for a submerged period of 24 h, with stabile molluscicidal activity at a temperature lower than 60°C; furthermore, it could be gradually degraded after exposure to illumination for 15 days. The freeze-dried powder of SL-30's EXB was safe to fresh fish and shrimp, even at a concentration beyond LC(90) of 24-h exposure period. The glycogen and total protein content of soft tissues of snails decreased after treating with SL-30's EXB, and glycogen content's decreasing rate had a linear relationship with molluscicidal activity. Finally, phylogenetic analysis based on ITS sequence showed that strain SL-30 had a higher similarity to Aspergillus fumigatus with bootstrap value 98%; accordingly, it was identified as a species of Aspergillus.

Transcriptome Analysis Reveals Novel Insights into the Hyperaccumulator <i>Phytolacca acinosa</i> Roxb. Responses to Cadmium Stress.

Project description:Cadmium (Cd) is a highly toxic heavy metal that causes serious damage to plant and human health. Phytolacca acinosa Roxb. has a large amount of aboveground biomass and a rapid growth rate, and it has been identified as a novel type of Cd hyperaccumulator that can be harnessed for phytoremediation. However, the molecular mechanisms underlying the response of P. acinosa to Cd2+ stress remain largely unclear. In this study, the phenotype, biochemical, and physiological traits of P. acinosa seeds and seedlings were analyzed under different concentrations of Cd2+ treatments. The results showed higher Cd2+ tolerance of P. acinosa compared to common plants. Meanwhile, the Cd2+ content in shoots reached 449 mg/kg under 10 mg/L Cd2+ treatment, which was obviously higher than the threshold for Cd hyperaccumulators. To investigate the molecular mechanism underlying the adaptability of P. acinosa to Cd stress, RNA-Seq was used to examine transcriptional responses of P. acinosa to Cd stress. Transcriptome analysis found that 61 genes encoding TFs, 48 cell wall-related genes, 35 secondary metabolism-related genes, 133 membrane proteins and ion transporters, and 96 defense system-related genes were differentially expressed under Cd2+ stress, indicating that a series of genes were involved in Cd2+ stress, forming a complex signaling regulatory mechanism. These results provide new scientific evidence for elucidating the regulatory mechanisms of P. acinosa response to Cd2+ stress and new clues for the molecular breeding of heavy metal phytoremediation.

The complete chloroplast genome of <i>Epimedium brevicornu</i> (Berberidaceae), a traditional Chinese medicinal herb.

Project description:Epimedii Folium has been used as a common traditional Chinese medicine for more than 2000 years in China. In this study, we assembled the complete chloroplast (cp) genome of Epimedium brevicornu. The whole cp genome of E. brevicornu is 158,658 bp in length, comprising a pair of inverted repeat (IR) regions (27,699 bp) separated by a large single copy (LSC) region (86,558 bp) and a small single copy (SSC) region (16,702bp). The E. brevicornu cp genome contains 129 genes, of which 84 protein-coding genes, 37 tRNA genes, and eight rRNA genes. Phylogenetic analysis shows that E. brevicornu is closely clustered with E. wushanense, E. lishihchenii, and E. sagittatum. The published E. brevicornu chloroplast genome will provide useful information for the phylogenetic and evolutionary study on Epimedium family of Berberidaceae.

The first complete plastid genome sequence of P. younghushandii reported here was 151,747 bp long, with the large single copy (LSC) region of 83,181 bp, the small single copy (SSC) region of 17,372 bp and two inverted repeats (IRa and IRb) of 25,597 bp. The plastome contained 114 genes, including 80 protein-coding genes, four encoding rRNAs, and 30 encoding tRNAs. The overall GC content was 38.5%. Phylogenetic analysis of Lamiaceae based on a whole plastome matrix suggested that Phlomoides is closely related to the genus Phlomis as members of subfamily Lamioideae.

Result from phylogenetic analysis suggests that tribe Phlomideae (Phlomoides) is closely related to tribe Lamieae and tribe Leonureae in present study.

Project description:Microsatellite loci are described for Rhodiola, a medicinal herb genus widely used in traditional Chinese medicine. • A total of 17 polymorphic microsatellite primer pairs were developed using the combined biotin capture method. The number of alleles per locus ranged from one to 12 across 192 individuals from R. bupleuroides, R. crenulata, R. fastigiata, and R. sacra, and the mean observed and expected heterozygosities ranged from 0.177 to 0.412 and from 0.363 to 0.578, respectively. • The results demonstrate the potential use of this new set of microsatellite markers for genotyping individuals and estimating genetic diversity in Rhodiola.

The complete chloroplast genome is 152,468 bp in length, with large single copy (LSC 82,257 bp) and small single copy (SSC 16,599 bp) regions separated by a pair of inverted repeats (IR 26 805 bp). The genome has a total of 113 genes including 79 protein-coding genes, 30 tRNA genes, and 4 rRNA genes. Phylogenetic analysis shows that P. dubia is closely related with Sinopodophyllum hexandrum and Epimedium species. The results are of great implication for the development and utilization of P. dubia and the phylogenetic researches on Berberidaceae.

Project description:It is the first report on complete chloroplast genome of Odontosoria chinensis, a traditional Chinese medicinal herb. The genome size is 156,293?bp, containing a pair of inverted repeats (IRs) (25,198?bp) separated by a small single-copy (SSC) region (20,649?bp) and a large single-copy (LSC) region (85,248?bp), respectively. The plastome has 128 genes, including 87 protein-coding genes, 32 tRNA genes, eight rRNA genes, and one pseudogene. The overall GC content is 40.43%. Maximum likelihood tree reveals that O. chinensis is basal lineages of polypods. The study will provide powerful molecular information for further phylogenetic analysis.

Multiple High-Affinity K+ Transporters and ABC Transporters Involved in K+ Uptake/Transport in the Potassium-Hyperaccumulator Plant Phytolacca acinosa Roxb.

Project description:Potassium is an important essential element for plant growth and development. Long-term potassium deprivation can lead to a severe deficiency phenotype in plants. Interestingly, Phytolacca acinosa is a plant with an unusually high potassium content and can grow well and complete its lifecycle even in severely potassium deficient soil. In this study, we found that its stems and leaves were the main tissues for high potassium accumulation, and P. acinosa showed a strong ability of K+ absorption in roots and a large capability of potassium accumulation in shoots. Analysis of plant growth and physiological characteristics indicated that P. acinosa had an adaptability in a wide range of external potassium levels. To reveal the mechanism of K+ uptake and transport in the potassium-hyperaccumulator plant P. acinosa, K+ uptake-/transport-related genes were screened by transcriptome sequencing, and their expression profiles were compared between K+ starved plants and normal cultured plants. Eighteen members of HAK/KT/KUPs, ten members of AKTs, and one member of HKT were identified in P. acinosa. Among them, six HAKs, and two AKTs and PaHKT1 showed significantly different expression. These transporters might be coordinatively involved in K+ uptake/transport in P. acinosa and lead to high potassium accumulation in plant tissues. In addition, significantly changed expression of some ABC transporters indicated that ABC transporters might be important for K+ uptake and transport in P. acinosa under low K+ concentrations.
