Tree co-occurrence and transcriptomic response to drought.

ABSTRACT: The distribution and co-occurrence of species are partly the outcome of their interactions with environmental drivers. Drought is a key driver related to the distribution of plant species. Drought events continue to increase in frequency and severity and identifying those aspects of plant function that are related to drought is critical. Here, we perform a community-level analysis of gene expression in relation to experimental drought and relate the similarity in gene set enrichment across species to their natural co-occurrence. Species with similar gene set enrichment in response to experimental drought tend to non-randomly co-occur in a natural stand. We demonstrate that similarity in the transcriptomic response of species to drought is a significantly better indicator of natural co-occurrence than measures of functional trait similarity and phylogenetic relatedness and that transcriptomics has the capacity to greatly enhance ecological investigations of species distributions and community structure.

Project description:BackgroundSesame is an important oil crop due to its high oil, antioxidant, and protein content. Drought stress is a major abiotic stress that affects sesame production as well as the quality of sesame seed. To reveal the adaptive mechanism of sesame in response to water deficient conditions, transcriptomic and metabolomics were applied in drought-tolerant (DT) and drought-susceptible (DS) sesame genotypes.ResultsTranscriptomic analysis reveals a set of core drought-responsive genes (684 up-regulated and 1346 down-regulated) in sesame that was robustly differently expressed in both genotypes. Most enriched drought-responsive genes are mainly involved in protein processing in endoplasmic reticulum, plant hormone signal transduction photosynthesis, lipid metabolism, and amino acid metabolism. Drought-susceptible genotype was more disturbed by drought stress at both transcriptional and metabolic levels, since more drought-responsive genes/metabolites were identified in DS. Drought-responsive genes associated with stress response, amino acid metabolism, and reactive oxygen species scavenging were more enriched or activated in DT. According to the partial least-squares discriminate analysis, the most important metabolites which were accumulated under drought stress in both genotypes includes ABA, amino acids, and organic acids. Especially, higher levels of ABA, proline, arginine, lysine, aromatic and branched chain amino acids, GABA, saccharopine, 2-aminoadipate, and allantoin were found in DT under stress condition. Combination of transcriptomic and metabolomic analysis highlights the important role of amino acid metabolism (especially saccharopine pathway) and ABA metabolism and signaling pathway for drought tolerance in sesame.ConclusionThe results of the present study provide valuable information for better understanding the molecular mechanism underlying drought tolerance of sesame, and also provide useful clues for the genetic improvement of drought tolerance in sesame.

Project description:In the present study we analyzed the responses of wheat to mild salinity and drought with special emphasis on the so far unclarified interaction of these important stress factors by using high-throughput phenotyping approaches. Measurements were performed on 14 genotypes of different geographic origin (Austria, Azerbaijan, and Serbia). The data obtained by non-invasive digital RGB imaging of leaf/shoot area reflect well the differences in total biomass measured at the end of the cultivation period demonstrating that leaf/shoot imaging can be reliably used to predict biomass differences among different cultivars and stress conditions. On the other hand, the leaf/shoot area has only a limited potential to predict grain yield. Comparison of gas exchange parameters with biomass accumulation showed that suppression of CO2 fixation due to stomatal closure is the principal cause behind decreased biomass accumulation under drought, salt and drought plus salt stresses. Correlation between grain yield and dry biomass is tighter when salt- and drought stress occur simultaneously than in the well-watered control, or in the presence of only salinity or drought, showing that natural variation of biomass partitioning to grains is suppressed by severe stress conditions. Comparison of yield data show that higher biomass and grain yield can be expected under salt (and salt plus drought) stress from those cultivars which have high yield parameters when exposed to drought stress alone. However, relative yield tolerance under drought stress is not a good indicator of yield tolerance under salt (and salt plus drought) drought stress. Harvest index of the studied cultivars ranged between 0.38 and 0.57 under well watered conditions and decreased only to a small extent (0.37-0.55) even when total biomass was decreased by 90% under the combined salt plus drought stress. It is concluded that the co-occurrence of mild salinity and drought can induce large biomass and grain yield losses in wheat due to synergistic interaction of these important stress factors. We could also identify wheat cultivars, which show high yield parameters under the combined effects of salinity and drought demonstrating the potential of complex plant phenotyping in breeding for drought and salinity stress tolerance in crop plants.

Project description:The present study aimed at identifying and mapping key genes expressed in root tissues involved in drought and salinity tolerance/resistance conserved among different fruit tree species. Twenty-six RNA-Seq samples were analyzed from six published studies in five plant species (Olea europaea, Vitis riparia Michx, Prunus mahaleb, Prunus persica, Phoenix dactylifera). This meta-analysis used a bioinformatic pipeline identifying 750 genes that were commonly modulated in three salinity studies and 683 genes that were commonly regulated among three drought studies, implying their conserved role in resistance/tolerance/response to these environmental stresses. A comparison was done on the genes that were in common among both salinity and drought resulted in 82 genes, of which 39 were commonly regulated with the same trend of expression (23 were upregulated and 16 were downregulated). Gene set enrichment and pathway analysis pointed out that pathways encoding regulation of defense response, drug transmembrane transport, and metal ion binding are general key molecular responses to these two abiotic stress responses. Furthermore, hormonal molecular crosstalk plays an essential role in the fine-tuning of plant responses to drought and salinity. Drought and salinity induced a different molecular "hormonal fingerprint". Dehydration stress specifically enhanced multiple genes responsive to abscisic acid, gibberellin, brassinosteroids, and the ethylene-activated signaling pathway. Salt stress mostly repressed genes encoding for key enzymes in signaling proteins in auxin-, gibberellin-(gibberellin 2 oxidase 8), and abscisic acid-related pathways (aldehyde oxidase 4, abscisic acid-responsive element-binding protein 3). Abiotic stress-related genes were mapped into the chromosome to identify molecular markers usable for the improvement of these complex quantitative traits. This meta-analysis identified genes that serve as potential targets to develop cultivars with enhanced drought and salinity resistance and/or tolerance across different fruit tree crops in a biotechnological sustainable way.
