Origin of the blueshift of water molecules at interfaces of hydrophilic cyclic compounds.

ABSTRACT: Water molecules at interfaces of materials exhibit enigmatic properties. A variety of spectroscopic studies have observed a high-frequency motion in these water molecules, represented by a blueshift, at both hydrophobic and hydrophilic interfaces. However, the molecular mechanism behind this blueshift has remained unclear. Using Raman spectroscopy and ab initio molecular dynamics simulations, we reveal the molecular mechanism of the blueshift of water molecules around six monosaccharide isomers. In the first hydration shell, we found weak hydrogen-bonded water molecules that cannot have a stable tetrahedral water network. In the water molecules, the vibrational state of the OH bond oriented toward the bulk solvent strongly contributes to the observed blueshift. Our work suggests that the blueshift in various solutions originates from the vibrational motions of these observed water molecules.

Recognition of Hydrophilic Cyclic Compounds by a Water-Soluble Cavitand.

Project description:A water-soluble deep cavitand bearing amides on the upper rim and trimethyl ammonium groups on the feet was synthesized. The open-ended cavity is stabilized by the intramolecular hydrogen bonds formed between the adjacent amides, and the introduction of trimethylammonium imparts to the cavitand good solubility in water. The cavitand exhibits high binding affinity and selectivity to hydrophilic molecules in water. With certain guests, such as cyclohexyl alcohols, amines and acids, the recognition involves the synergistic action of hydrogen bonding with hydrophobic effects. The binding phenomena are interpreted in terms of a fixed solvent cage presented by the host to the guest.

Origin of proton affinity to membrane/water interfaces.

Project description:Proton diffusion along biological membranes is vitally important for cellular energetics. Here we extended previous time-resolved fluorescence measurements to study the time and temperature dependence of surface proton transport. We determined the Gibbs activation energy barrier ?G ‡r that opposes proton surface-to-bulk release from Arrhenius plots of (i) protons' surface diffusion constant and (ii) the rate coefficient for proton surface-to-bulk release. The large size of ?G ‡r disproves that quasi-equilibrium exists in our experiments between protons in the near-membrane layers and in the aqueous bulk. Instead, non-equilibrium kinetics describes the proton travel between the site of its photo-release and its arrival at a distant membrane patch at different temperatures. ?G ‡r contains only a minor enthalpic contribution that roughly corresponds to the breakage of a single hydrogen bond. Thus, our experiments reveal an entropic trap that ensures channeling of highly mobile protons along the membrane interface in the absence of potent acceptors.

Lateral diffusion of single polymer molecules at interfaces between water and oil.

Project description:Lateral diffusion of polymer molecules at the interfaces between immiscible oil and water is investigated at the single molecular level. The interfaces between water and alkanes are chosen as the model systems and polyethylene oxide (PEO) is the model polymer. Fluorescence correlation spectroscopy is used to measure the interfacial diffusion of fluorescence-labeled PEO with its molecular weight ranging over more than an order of magnitude. It is discovered that the interfacial diffusion coefficient scales with the molecular weight by the exponent of -0.5. Detailed analysis shows that the PEO chain takes an ideal two-dimensional random coil conformation at these fluidic interfaces and the bigger contribution from water's hydrodynamic friction is discovered.

Origin of subdiffusion of water molecules on cell membrane surfaces.

Project description:Water molecules play an important role in providing unique environments for biological reactions on cell membranes. It is widely believed that water molecules form bridges that connect lipid molecules and stabilize cell membranes. Using all-atom molecular dynamics simulations, we show that translational and rotational diffusion of water molecules on lipid membrane surfaces exhibit subdiffusion and aging. Moreover, we provide evidence that both divergent mean trapping time (continuous-time random walk) and long-correlated noise (fractional Brownian motion) contribute to this subdiffusion. These results suggest that subdiffusion on cell membranes causes the water retardation, an enhancement of cell membrane stability, and a higher reaction efficiency.

Hydrophilic fluorinated molecules for spectral 19F MRI.

Project description:Fluorine-19 (19F) Magnetic Resonance Imaging (MRI) is an emerging modality for molecular imaging and cell tracking. The hydrophobicity of current exogenous probes, perfluorocarbons (PFCs) and perfluoropolyethers (PFPEs), limits the formulation options available for in vivo applications. Hydrophilic probes permit more formulation flexibility. Further, the broad Nuclear Magnetic Resonance (NMR) chemical shift range of organofluorine species enables multiple probes with unique 19F MR signatures for simultaneous interrogation of distinct molecular targets in vivo. We report herein a flexible approach to stable liposomal formulations of hydrophilic fluorinated molecules (each bearing numerous magnetically equivalent 19F atoms), with 19F encapsulation of up to 22.7?mg/mL and a per particle load of 3.6?×?106 19F atoms. Using a combination of such probes, we demonstrate, with no chemical shift artifacts, the simultaneous imaging of multiple targets within a given target volume by spectral 19F MRI.

Electrification at water-hydrophobe interfaces.

Project description:The mechanisms leading to the electrification of water when it comes in contact with hydrophobic surfaces remains a research frontier in chemical science. A clear understanding of these mechanisms could, for instance, aid the rational design of triboelectric generators and micro- and nano-fluidic devices. Here, we investigate the origins of the excess positive charges incurred on water droplets that are dispensed from capillaries made of polypropylene, perfluorodecyltrichlorosilane-coated glass, and polytetrafluoroethylene. Results demonstrate that the magnitude and sign of electrical charges vary depending on: the hydrophobicity/hydrophilicity of the capillary; the presence/absence of a water reservoir inside the capillary; the chemical and physical properties of aqueous solutions such as pH, ionic strength, dielectric constant and dissolved CO2 content; and environmental conditions such as relative humidity. Based on these results, we deduce that common hydrophobic materials possess surface-bound negative charge. Thus, when these surfaces are submerged in water, hydrated cations form an electrical double layer. Furthermore, we demonstrate that the primary role of hydrophobicity is to facilitate water-substrate separation without leaving a significant amount of liquid behind. These results advance the fundamental understanding of water-hydrophobe interfaces and should translate into superior materials and technologies for energy transduction, electrowetting, and separation processes, among others.

Microarray analysis of Caenorhabditis elegans exposed to three hydrophilic genotoxicants in water

Project description:In this study, we exposed Caenorhabditis elegans wild types N2 to two concentrations of the model genotoxicants formaldehyde (HCHO), N-ethyl-N-nitrosourea (ENU), and methyl methanesulfonate (MMS).We performed a concentration-response test to determine the non-toxic concentration range for studying the transcriptional effects of compounds. In microarray experiments, we studied two concentrations (1 mM and 5 mM) for each compound and a control of M9 medium. Age synchronized worms at developmental L4 larval stage were exposed to treatment for four hours. After flash freezing the samples, RNA was isolated, labeled and hybridized on oligo microarray (Agilent) slides.

Lipogels for Encapsulation of Hydrophilic Proteins and Hydrophobic Small Molecules.

Project description:Lipid-polymer hybrid materials have the potential to exhibit enhanced stability and loading capabilities in comparison to parent liposome or polymer materials. However, complexities lie in formulating and characterizing such complex nanomaterials. Here we describe a lipid-coated polymer gel (lipogel) formulated using a single-pot methodology, where self-assembling liposomes template a UV-curable polymer gel core. Using fluorescently labeled lipids, protein, and hydrophobic molecules, we characterized their formation, purification, stability, and encapsulation efficiency via common instrumentation methods such as dynamic light scattering (DLS), matrix-assisted laser desorption ionization-mass spectrometry (MALDI-MS), UV-vis spectroscopy, fluorescence spectroscopy, and single-particle total internal reflection fluorescence (TIRF) microscopy. In addition, we confirmed that these dual-guest-loaded lipogels are stable in solution for several months. The simplicity of this complete aqueous formation and noncovalent dual-guest encapsulation holds potential as a tunable nanomaterial scaffold.

Trapping and assembly of living colloids at water-water interfaces.

Project description:We study the assembly of inert and living colloids in a two-phase water-water system that provides an environment that can sustain bacteria, providing a new structure with rich potential to confine and structure microbial communities. The water-water system, formed via phase separation of a casein and xanthan mixture, forms a 3-D structure of coexisting casein-rich and xanthan-rich phases. Fluorescent labelling and confocal microscopy reveal the attachment of these living colloids, including Escherichia coli and Pseudomonas aeruginosa, at the interface between the two phases. Inert colloids also become trapped at the interfaces, suggesting that the observed attachment can be attributed to capillarity. Over time, these structures coarsen and eventually degrade, illustrating the dynamic nature of these systems. This system lays the foundation for future studies of the interplay of physicochemical properties of the fluid interfaces and bulk phases and microbial responses they provoke to induce complex spatial organization, to study species which occupy distinct niches, and to optimize efficient microbial cross-feeding or protection from competitors.

Kinetic photovoltage along semiconductor-water interfaces.

Project description:External photo-stimuli on heterojunctions commonly induce an electric potential gradient across the interface therein, such as photovoltaic effect, giving rise to various present-day technical devices. In contrast, in-plane potential gradient along the interface has been rarely observed. Here we show that scanning a light beam can induce a persistent in-plane photoelectric voltage along, instead of across, silicon-water interfaces. It is attributed to the following movement of a charge packet in the vicinity of the silicon surface, whose formation is driven by the light-induced potential change across the capacitive interface and a high permittivity of water with large polarity. Other polar liquids and hydrogel on silicon also allow the generation of the in-plane photovoltage, which is, however, negligible for nonpolar liquids. Based on the finding, a portable silicon-hydrogel array has been constructed for detecting the shadow path of a moving Cubaris. Our study opens a window for silicon-based photoelectronics through introducing semiconductor-water interfaces.
