Polarization of concave domains by traveling wave pinning.

ABSTRACT: Pattern formation is one of the most fundamental yet puzzling phenomena in physics and biology. We propose that traveling front pinning into concave portions of the boundary of 3-dimensional domains can serve as a generic gradient-maintaining mechanism. Such a mechanism of domain polarization arises even for scalar bistable reaction-diffusion equations, and, depending on geometry, a number of stationary fronts may be formed leading to complex spatial patterns. The main advantage of the pinning mechanism, with respect to the Turing bifurcation, is that it allows for maintaining gradients in the specific regions of the domain. By linking the instant domain shape with the spatial pattern, the mechanism can be responsible for cellular polarization and differentiation.

Subwavelength polarization optics via individual and coupled helical traveling-wave nanoantennas.

Project description:Light polarization control is a key factor in modern photonics. Recent advances in surface plasmon manipulation have introduced the prospect of more compact and more efficient devices for this purpose. However, the current plasmonic-based polarization optics remain much larger than the wavelength of light, which limits the design degrees of freedom. Here, we present a plasmonic traveling-wave nanoantenna using a gold-coated helical carbon nanowire end-fired with a dipolar aperture nanoantenna. Our nonresonant helical nanoantenna enables tunable polarization control by swirling surface plasmons on the subwavelength scale and taking advantage of the optical spin-orbit interaction. Four closely packed helical traveling-wave nanoantennas (HTNs) are demonstrated to locally convert an incoming light beam into four beams of tunable polarizations and intensities, with the ability to impart different polarization states to the output beams in a controllable way. Moreover, by near-field coupling four HTNs of opposite handedness, we demonstrate a subwavelength waveplate-like structure providing a degree of freedom in polarization control that is unachievable with ordinary polarization optics and current metamaterials.

Superior polarization retention through engineered domain wall pinning.

Project description:Ferroelectric materials possess a spontaneous polarization that is switchable by an electric field. Robust retention of switched polarization is critical for non-volatile nanoelectronic devices based on ferroelectrics, however, these materials often suffer from polarization relaxation, typically within days to a few weeks. Here we exploit designer-defect-engineered epitaxial BiFeO3 films to demonstrate polarization retention with virtually no degradation in switched nanoscale domains for periods longer than 1 year. This represents a more than 2000% improvement over the best values hitherto reported. Scanning probe microscopy-based dynamic switching measurements reveal a significantly increased activation field for domain wall movement. Atomic resolution scanning transmission electron microscopy indicates that nanoscale defect pockets pervade the entire film thickness. These defects act as highly efficient domain wall pinning centres, resulting in anomalous retention. Our findings demonstrate that defects can be exploited in a positive manner to solve reliability issues in ferroelectric films used in functional devices.

Cell behavior in traveling wave patterns of myxobacteria.

Project description:Cells in the early stages of starvation-induced fruiting body development migrate in a highly organized periodic pattern of equispaced accumulations that move as traveling waves. Two sets of waves are observed moving in opposite directions with the same wavelength and speed. To learn how the behavior of individual cells contributes to the wave pattern, fluorescent cells were tracked within a rippling population. These cells exhibit at least three types of organized behavior. First, most cell movement occurs along the same axis as the rippling movement. Second, there is a high degree of cell alignment parallel to the direction of rippling, as indicated by the biased movement. Third, by controlling the reversal frequency, cell movement becomes periodic in a rippling field. The periodicity of individual cells matches the period of macroscopic rippling. This last behavior is unique to a rippling population and, on the basis of Myxococcus xanthus genetic data, we conclude that this periodicity is linked to the C signal, a nondiffusible cell contact-mediated signaling molecule. When two cells moving in opposite directions meet end to end, they transmit the C signal to each other and in response reverse their gliding direction. This model of traveling waves represents a new mode of biological pattern formation that depends on cell-contact interactions rather than reaction diffusion.

A traveling-wave solution for bacterial chemotaxis with growth.

Project description:Bacterial cells navigate their environment by directing their movement along chemical gradients. This process, known as chemotaxis, can promote the rapid expansion of bacterial populations into previously unoccupied territories. However, despite numerous experimental and theoretical studies on this classical topic, chemotaxis-driven population expansion is not understood in quantitative terms. Building on recent experimental progress, we here present a detailed analytical study that provides a quantitative understanding of how chemotaxis and cell growth lead to rapid and stable expansion of bacterial populations. We provide analytical relations that accurately describe the dependence of the expansion speed and density profile of the expanding population on important molecular, cellular, and environmental parameters. In particular, expansion speeds can be boosted by orders of magnitude when the environmental availability of chemicals relative to the cellular limits of chemical sensing is high. Analytical understanding of such complex spatiotemporal dynamic processes is rare. Our analytical results and the methods employed to attain them provide a mathematical framework for investigations of the roles of taxis in diverse ecological contexts across broad parameter regimes.

Directional persistence of chemotactic bacteria in a traveling concentration wave.

Project description:Chemotactic bacteria are known to collectively migrate towards sources of attractants. In confined convectionless geometries, concentration "waves" of swimming Escherichia coli can form and propagate through a self-organized process involving hundreds of thousands of these microorganisms. These waves are observed in particular in microcapillaries or microchannels; they result from the interaction between individual chemotactic bacteria and the macroscopic chemical gradients dynamically generated by the migrating population. By studying individual trajectories within the propagating wave, we show that, not only the mean run length is longer in the direction of propagation, but also that the directional persistence is larger compared to the opposite direction. This modulation of the reorientations significantly improves the efficiency of the collective migration. Moreover, these two quantities are spatially modulated along the concentration profile. We recover quantitatively these microscopic and macroscopic observations with a dedicated kinetic model.

Dividing to unveil protein microheterogeneities: traveling wave ion mobility study.

Project description:Overexpression of a protein in a foreign host is often the only route toward an exhaustive characterization, especially when purification from the natural source(s) is hardly achievable. The key issue in these studies relies on quality control of the purified recombinant protein to precisely determining its identity as well as any undesirable microheterogeneities. While standard proteomics approaches preclude unbiased search for modifications, the optional technique of top-down tandem mass spectrometry (MSMS) requires the use of highly accurate and highly resolved experiments to reveal subtle sequence modifications. In the present study, the top-down MSMS approach combined with traveling wave ion mobility (TWIM) separation was evaluated for its ability to achieve high sequence coverage and to reveal subtle microheterogeneities that were hitherto only accessible with Fourier-transform ion cyclotron resonance-MS instruments. The power of this approach is herein illustrated in an in-depth analysis of both the wild type and K496C variant of the recombinant X domain (XD; aa's 459-507) of the measles virus phosphoprotein expressed in Escherichia coli . Using top-down MSMS combined with TWIM, we show that XD samples occasionally exhibit a microheterogeneity that could not be anticipated from the nucleotide sequence of the encoding constructs and that likely reflects a genetic drift, neutral or not, occurring during expression. In addition, a 1-oxyl-2,2,5,5-tetramethyl-δ3-pyrroline-3-methyl methanethiosulfonate nitroxide probe that was grafted onto the K496C XD variant was shown to undergo oxidation and/or protonation in the electrospray ionization source, leading to artifactual mass increases.

Diffusing wave paradox of phototactic particles in traveling light pulses.

Project description:Cells navigate through complex surroundings by following cues from their environment. A prominent example is Dictyostelium, which is directed by chemotaxis towards regions with higher concentrations. In the presence of traveling chemical waves, however, amoebae migrate counter to the running wave. Such behavior, referred to as diffusing wave paradox, suggests the existence of adaptation and directional memory. Here we experimentally investigate the response of phototactic self-propelled microparticles to traveling light-pulses. Despite their entirely memory-less (i.e., strictly local) response to the environment, we observe the same phenomenological behavior, i.e., particle motion counter to the pulse direction. Our findings are supported by a minimal model which considers active particle reorientations within local light gradients. The complex and robust behavior of synthetic active particles to spatially and temporally varying stimuli enables new strategies for achieving collective behavior and can be used for the design of micro-robotic systems with limited signal-processing capabilities.

Structural characterization of unsaturated phosphatidylcholines using traveling wave ion mobility spectrometry.

Project description:A number of phosphatidylcholine (PC) cations spanning a mass range of 400-1000 Da are investigated using electrospray ionization mass spectrometry coupled with traveling wave ion mobility spectrometry (TWIMS). A high correlation between mass and mobility is demonstrated with saturated phosphatidylcholine cations in N(2). A significant deviation from this mass-mobility correlation line is observed for the unsaturated PC cation. We found that the double bond in the acyl chain causes a 5% reduction in drift time. The drift time is reduced at a rate of approximately 1% for each additional double bond. Theoretical collision cross sections of PC cations exhibit good agreement with experimentally evaluated values. Collision cross sections are determined using the recently derived relationship between mobility and drift time in TWIMS stacked ring ion guide (SRIG) and compared to estimated collision cross sections using an empiric calibration method. Computational analysis was performed using the modified trajectory (TJ) method with nonspherical N(2) molecules as the drift gas. The difference between estimated collision cross sections and theoretical collision cross sections of PC cations is related to the sensitivity of the PC cation collision cross sections to the details of the ion-neutral interactions. The origin of the observed correlation and deviation between mass and mobility of PC cations is discussed in terms of the structural rigidity of these molecules using molecular dynamic simulations.

Medial entorhinal cortex activates in a traveling wave in the rat.

Project description:Traveling waves are hypothesized to support the long-range coordination of anatomically distributed circuits. Whether separate strongly interacting circuits exhibit traveling waves remains unknown. The hippocampus exhibits traveling 'theta' waves and interacts strongly with the medial entorhinal cortex (MEC). To determine whether the MEC also activates in a traveling wave, we performed extracellular recordings of local field potentials (LFP) and multi-unit activity along the MEC. These recordings revealed progressive phase shifts in activity, indicating that the MEC also activates in a traveling wave. Variation in theta waveform along the region, generated by gradients in local physiology, contributed to the observed phase shifts. Removing waveform-related phase shifts left significant residual phase shifts. The residual phase shifts covaried with theta frequency in a manner consistent with those generated by weakly coupled oscillators. These results show that the coordination of anatomically distributed circuits could be enabled by traveling waves but reveal heterogeneity in the mechanisms generating those waves.

Kerr reversal in Josephson meta-material and traveling wave parametric amplification.

Project description:Josephson meta-materials have recently emerged as very promising platform for superconducting quantum science and technologies. Their distinguishing potential resides in ability to engineer them at sub-wavelength scales, which allows complete control over wave dispersion and nonlinear interaction. In this article we report a versatile Josephson transmission line with strong third order nonlinearity which can be tuned from positive to negative values, and suppressed second order non linearity. As an initial implementation of this multipurpose meta-material, we operate it to demonstrate reversed Kerr phase-matching mechanism in traveling wave parametric amplification. Compared to previous state of the art phase matching approaches, this reversed Kerr phase matching avoids the presence of gaps in transmission, can reduce gain ripples, and allows in situ tunability of the amplification band over an unprecedented wide range. Besides such notable advancements in the amplification performance with direct applications to superconducting quantum computing and generation of broadband squeezing, the in-situ tunability with sign reversal of the nonlinearity in traveling wave structures, with no counterpart in optics to the best of our knowledge, opens exciting experimental possibilities in the general framework of microwave quantum optics, single-photon detection and quantum limited amplification.
