Mechanisms controlling the impact of multi-year drought on mountain hydrology.

ABSTRACT: Mountain runoff ultimately reflects the difference between precipitation (P) and evapotranspiration (ET), as modulated by biogeophysical mechanisms that intensify or alleviate drought impacts. These modulating mechanisms are seldom measured and not fully understood. The impact of the warm 2012-15 California drought on the heavily instrumented Kings River basin provides an extraordinary opportunity to enumerate four mechanisms that controlled the impact of drought on mountain hydrology. Two mechanisms intensified the impact: (i) evaporative processes have first access to local precipitation, which decreased the fractional allocation of P to runoff in 2012-15 and reduced P-ET by 30% relative to previous years, and (ii) 2012-15 was 1?°C warmer than the previous decade, which increased ET relative to previous years and reduced P-ET by 5%. The other two mechanisms alleviated the impact: (iii) spatial heterogeneity and the continuing supply of runoff from higher elevations increased 2012-15 P-ET by 10% relative to that expected for a homogenous basin, and iv) drought-associated dieback and wildfire thinned the forest and decreased ET, which increased 2016 P-ET by 15%. These mechanisms are all important and may offset each other; analyses that neglect one or more will over or underestimate the impact of drought and warming on mountain runoff.

Project description:Wheat crop is very sensitive to osmotic stress conditions. As an abiotic stress, drought may exert a considerable effect on the levels of specialized metabolites in plants. These metabolites may exert beneficial biological activities in the prevention or treatment of disorders linked to oxidative stress in plants and humans. Furthermore, osmoprotector accumulation helps wheat to increase the maintenance of osmotic balance. Therefore, identifying wheat genotypes with better drought tolerance is extremely important. In this sense, this research aimed to understand agronomic, physiological and biochemical responses of spring wheat strains and cultivars to drought stress, under field conditions, and jointly select strains via multi-trait index. We evaluated agronomic, physiological and biochemical variables in 18 genotypes under field condition. The results demonstrated that all variables were affected by the drought. Most genotypes were significantly reduced in grain yield, except VI_14774, VI_14668, VI_9007 and TBIO_ATON. The variables related to photosynthesis were also affected. An increase above 800% was observed in proline contents in genotypes under drought. Sodium and potassium also increased, mainly for VI_131313 (Na), while VI_130758 and VI_14774 presented increased K. We evaluated the antioxidant potential of the different strains and the total content of phenolic compounds. The most drought-responsive genotypes were BRS_264, VI_14050 and VI_14426. Reduced grain yield and photosynthetic variables, and increased specialized metabolism compounds are due to plant defense mechanisms against drought conditions. Furthermore, variation in genotypes can be explained by the fact that each plant presents a different defense and tolerance mechanism, which may also occur between genotypes of the same species. Four strains were selected by the multivariate index: VI_14055, VI_14001, VI_14426 and VI_1466. Such results allow us to predict which genotype(s) performed best in semi-arid environments and under climatic fluctuations.

Project description:Droughts strongly affect carbon and nitrogen cycling in grasslands, with consequences for ecosystem productivity. Therefore, we investigated how experimental grassland communities interact with groups of soil microorganisms. In particular, we explored the mechanisms of the drought-induced decoupling of plant photosynthesis and microbial carbon cycling and its recovery after rewetting. Our aim was to better understand how root exudation during drought is linked to pulses of soil microbial activity and changes in plant nitrogen uptake after rewetting. We set up a mesocosm experiment on a meadow site and used shelters to simulate drought. We performed two 13C-CO2 pulse labelings, the first at peak drought and the second in the recovery phase, and traced the flow of assimilates into the carbohydrates of plants and the water extractable organic carbon and microorganisms from the soil. Total microbial tracer uptake in the main metabolism was estimated by chloroform fumigation extraction, whereas the lipid biomarkers were used to assess differences between the microbial groups. Drought led to a reduction of aboveground versus belowground plant growth and to an increase of 13C tracer contents in the carbohydrates, particularly in the roots. Newly assimilated 13C tracer unexpectedly accumulated in the water-extractable soil organic carbon, indicating that root exudation continued during the drought. In contrast, drought strongly reduced the amount of 13C tracer assimilated into the soil microorganisms. This reduction was more severe in the growth-related lipid biomarkers than in the metabolic compounds, suggesting a slowdown of microbial processes at peak drought. Shortly after rewetting, the tracer accumulation in the belowground plant carbohydrates and in the water-extractable soil organic carbon disappeared. Interestingly, this disappearance was paralleled by a quick recovery of the carbon uptake into metabolic and growth-related compounds from the rhizospheric microorganisms, which was probably related to the higher nitrogen supply to the plant shoots. We conclude that the decoupling of plant photosynthesis and soil microbial carbon cycling during drought is due to reduced carbon uptake and metabolic turnover of rhizospheric soil microorganisms. Moreover, our study suggests that the maintenance of root exudation during drought is connected to a fast reinitiation of soil microbial activity after rewetting, supporting plant recovery through increased nitrogen availability.

Project description:Energy-starved microbes in deep marine sediments subsist at near-zero growth for thousands of years, yet the mechanisms for their subsistence are unknown because no model strains have been cultivated from most of these groups. We investigated Baltic Sea sediments with single-cell genomics, metabolomics, metatranscriptomics, and enzyme assays to identify possible subsistence mechanisms employed by uncultured Atribacteria, Aminicenantes, Actinobacteria group OPB41, Aerophobetes, Chloroflexi, Deltaproteobacteria, Desulfatiglans, Bathyarchaeota, and Euryarchaeota marine group II lineages. Some functions appeared to be shared by multiple lineages, such as trehalose production and NAD+-consuming deacetylation, both of which have been shown to increase cellular life spans in other organisms by stabilizing proteins and nucleic acids, respectively. Other possible subsistence mechanisms differed between lineages, possibly providing them different physiological niches. Enzyme assays and transcripts suggested that Atribacteria and Actinobacteria group OPB41 catabolized sugars, whereas Aminicenantes and Atribacteria catabolized peptides. Metabolite and transcript data suggested that Atribacteria utilized allantoin, possibly as an energetic substrate or chemical protectant, and also possessed energy-efficient sodium pumps. Atribacteria single-cell amplified genomes (SAGs) recruited transcripts for full pathways for the production of all 20 canonical amino acids, and the gene for amino acid exporter YddG was one of their most highly transcribed genes, suggesting that they may benefit from metabolic interdependence with other cells. Subsistence of uncultured phyla in deep subsurface sediments may occur through shared strategies of using chemical protectants for biomolecular stabilization, but also by differentiating into physiological niches and metabolic interdependencies.IMPORTANCE Much of life on Earth exists in a very slow-growing state, with microbes from deeply buried marine sediments representing an extreme example. These environments are like natural laboratories that have run multi-thousand-year experiments that are impossible to perform in a laboratory. We borrowed some techniques that are commonly used in laboratory experiments and applied them to these natural samples to make hypotheses about how these microbes subsist for so long at low activity. We found that some methods for stabilizing proteins and nucleic acids might be used by many members of the community. We also found evidence for niche differentiation strategies, and possibly cross-feeding, suggesting that even though they are barely growing, complex ecological interactions continue to occur over ultralong timescales.
