Characterization of Two Species of Acremonium (Unrecorded in Korea) from Soil Samples: A. variecolor and A. persicinum.

ABSTRACT: During a survey of fungal diversity of the order Hypocreales in Korea, two Acremonium isolates, CNUFC-1YSRS2-4 and CNUFC-GSNPF3-1, were isolated from soils collected on a bank of the Yeongsan River, Naju, and in a forest on the Mt. Daegak located on Sinsi Island, Gunsan, South Korea, respectively. Based on the morphological characteristics and sequence analysis of the internal transcribed spacer and D1/D2 domains of 28S ribosomal DNA, the isolates CNUFC-1YSRS2-4 and CNUFC-GSNPF3-1 were identified as A. variecolor and A. persicinum, respectively. These 2 species represent novel Hypocreales isolates in Korea.

Isolation and Characterization of Four Unrecorded Mucor Species in Korea.

Project description:During an investigation of fungi of the order Mucorales from freshwater and sediment samples in Korea, we isolated six strains, NNIBRFG6649, NNIBRFG6255, NNIBRFG1498, CNUFC-YJ13, CNUFC-YR7, and NNIBRFG2739. The morphology and phylogeny of these strains were analyzed. Based on the morphological characteristics and molecular data from internal transcribed spacer (ITS) region, the isolates NNIBRFG6649 and NNIBRFG6255 were identified as Mucor abundans, and M. aligarensis, respectively. The isolates NNIBRFG1498 and CNUFC-YJ13 were identified as M. moelleri, whereas the isolates CNUFC-YR7 and NNIBRFG2739 were identified as M. heterogamus. To the best of our knowledge, M. abundans, M. aligarensis, M. moelleri, and M. heterogamus have not yet been reported in Korea.

Two Previously Unrecorded Fungal Species Isolated from Muui Island in Korea.

Project description:Fungi are cosmopolitan and they occupy diverse niches as consumers, producers, and decomposers. They play critical roles in the environment by enabling nutrient cycling and generating a plethora of secondary metabolites. This study aimed to identify and characterize fungal strains isolated from diverse sources on Muui Island, Republic of Korea. In 2023, a total of 86 fungal strains were collected and examined. Investigation of the morphological features and phylogenetic analyses of multiple barcode loci identified one putative novel species and two species previously unrecorded in the Republic of Korea: Colletotrichum sp., Colletotrichum guizhouense, and Fusarium brachygibbosum. This study provides a comprehensive description of their molecular phylogenies and morphological characteristics. These findings will contribute to the existing knowledge about fungal species in the Republic of Korea and future research on the fungal diversity on Muui Island.

Four Unrecorded <i>Aspergillus</i> Species from the Rhizosphere Soil in South Korea.

Project description:The genus Aspergillus is commonly isolated from various marine and terrestrial environments; however, only a few species have been studied in rhizosphere soil. As part of the Korean indigenous fungal excavation project, we investigated fungal diversity from rhizosphere soil, focusing on Aspergillus species. A total of 13 strains were isolated from the rhizosphere soil of three different plants. Based on phylogenetic analysis of β-tubulin and calmodulin and morphological characteristics, we identified five Aspergillus species. A. calidoustus and A. pseudodeflectus were commonly isolated from the rhizosphere soil. Four species were confirmed as unrecorded species in Korea: A. calidoustus, A. dimorphicus, A. germanicus, and A. pseudodeflecuts. The detailed morphological descriptions of these unrecorded species are provided.

Characterization of Three Species of Sordariomycetes Isolated from Freshwater and Soil Samples in Korea.

Project description:During a survey of fungal diversity in the class Sordariomycetes, 3 fungal strains, CNUFC-KMHY6-1, CNUFC-MSW24-2-11, and CNUFC-GW2S-4 were isolated from soil and freshwater samples, respectively in Korea. The strains were analyzed both morphologically and phylogenetically on the basis of internal transcribed spacer and RNA polymerase II second largest subunit gene sequences. On the basis of their morphology and phylogeny, CNUFC-KMHY6-1, CNUFC-MSW24-2-11, and CNUFC-GW2S-4 isolates were identified as Arcopilus aureus, Memnoniella echinata, and Stachybotrys sansevieriae, respectively. To the best of our knowledge, Ar. aureus and M. echinata have not been previously recorded in Korea, and this is the first report of S. sansevieriae from freshwater niche.

Seventeen Unrecorded Species from Gayasan National Park in Korea.

Project description:Macrofungi play important roles in forest ecology as wood decayers, symbionts, and pathogens of living trees. For the effective forest management, it is imperative to have a comprehensive overview of macrofungi diversity in specific areas. As a part of the National Institute of Biological Resources projects for discovering indigenous fungi in Korea, we collected macrofungi in Gayasan National Park from 2017 to 2018. These specimens were identified based on morphological characteristics and sequence analysis of internal transcribed spacer (ITS) or the nuclear large subunit rRNA (LSU) region. We discovered 17 macrofungi new to Korea: Butyrea japonica, Ceriporia nanlingensis, Coltricia weii, Coltriciella subglobosa, Crepidotus crocophyllus, Cylindrobasidium laeve, Fulvoderma scaurum, Laetiporus cremeiporus, Lentinellus castoreus, Leucogyrophana mollusca, Marasmius insolitus, Nidularia deformis, Phaeophlebiopsis peniophoroides, Phanerochaete angustocystidiata, Phlebiopsis pilatii, Postia coeruleivirens, and Tengioboletus fujianensis. We described their detailed morphological characteristics.

Molecular and Morphological Characterization of Two Novel Species Collected from Soil in Korea.

Project description:Two fungal species of ascomycetes were discovered during the screening of soil microflora from the Gangwon Province in Korea: Didymella chlamydospora sp. nov. (YW23-14) and Microdochium salmonicolor sp. nov. (NC14-294). Morphologically, YW23-14 produces smaller chlamydospores (8.0-17.0 × 7.0-15.0 µm) than D. glomerata and D. musae. The strain NC14-294 was characterized by smaller conidiogenous cells (4.9-8.8 × 2.0-3.2 µm) compared with the closest strains M. fisheri and M. phragmitis. The detailed descriptions, illustrations, and discussions regarding the morphological and phylogenetical analyses of the closely related species are provided to support the novelty of each species. Thus, YW23-14 and NC14-294 are described here as newly discovered species.

Five Previously Unrecorded Fungal Species Isolated from Marine Plastic Wastes in South Korea.

Project description:Plastic wastes have a negative impact on marine environments; however, they can be used as carbon sources and habitats by certain microbes. Microbes in the marine plastisphere can migrate worldwide through the ocean and cause serious environmental problems when they encounter suitable environments. Therefore, efforts to investigate the microbes inhabiting the marine plastisphere are increasing. In the present study, fungal strains were isolated from plastic wastes buried in Korean sea sands and mudflats and identified using molecular and morphological analyses. Five species were identified that were previously unrecorded from South Korea: Cladosporium funiculosum, Neosetophoma poaceicola, Neosetophoma rosigena, Parasarocladium gamsii, and Trichoderma fomiticola. Their molecular phylogenies and morphological characteristics are described in this study.

Discovery of Two New Talaromyces Species from Crop Field Soil in Korea.

Project description:Two new fungal species of the genus Talaromyces, Talaromyces purpurogenus and Talaromyces trachyspermus from the Trichocomaceae family, were recovered during an investigation of fungal communities in soil collected from the Gangwon-do and Jeollanam-do provinces of Korea. These two species have not been previously officially reported from Korea. In this study, detailed descriptions of internal transcribed spacer rDNA and beta-tubulin gene regions of these two fungi are presented. Morphological features of the two fungi in five agar media, potato dextrose, oatmeal, malt extract, czapek yeast extract, and yeast extract sucrose, are also reported. The species were identified on the basis of molecular and morphological analysis, and herein we present data with detailed descriptions and figures.

Molecular and Morphological Characterization of Two Novel Species from Soil and Beetles (<i>Dorcus titanus castanicolor</i>) in Korea.

Project description:Two fungal strains belonging to Ascomycota were discovered in Gyeonggi-do, Korea, during this investigation of soil microfungi and microbiota of insects. The strain KNUF-20-047 produced milky white on the back and a milky creamy center to white toward the margin on the front side of colonies. Conversely, the closest Xenoacremonium falcatus displayed a pale luteous to luteous center, white toward margins on the front side, and pale luteous or luteous pigment on the back side, whereas X. recifei produced white colonies. The conidiophores of KNUF-20-047 were slightly larger than those of X. falcatus, and the conidia were distinct from X. recifei. Strain KNUF-20-NI-005 produced light brown to subhyaline conidiophores up to 56.0 μm tall, whereas Rhinocladiella anceps displayed golden to dark brown conidiophores up to 350 μm. Strain KNUF-20-NI-005 also produced larger conidia than R. anceps but smaller than R. coryli and R. fasciculata. Moreover, the molecular phylogeny strongly supports the detailed description and illustration of each proposed species to be designated as Xenoacremonium minutisporum sp. nov. and Rhinocladiella terrigenum sp. nov. in Korea.

Four unrecorded wood decay fungi from seoul in Korea.

Project description:During the investigation of Korean indigenous fungi from Seoul, three genera-Fuscoporia, Porostereum, and Trametopsis, and four species-Fuscoporia senex, Phlebia acerina, Porostereum spadiceum, and Trametopsis cervina were found. Their morphological characteristics were examined and their identification was confirmed by molecular analysis based on internal transcribed spacer (ITS) and nuclear large subunit ribosomal DNA region sequences. These fungi are new to Korea and registered here with descriptions.
