Mangrove endophyte promotes reforestation tree (Acacia polyphylla) growth.

ABSTRACT: Mangroves are ecosystems located in the transition zone between land and sea that serve as a potential source of biotechnological resources. Brazil's extensive coast contains one of the largest mangrove forests in the world (encompassing an area of 25,000km2 along all the coast). Endophytic bacteria were isolated from the following three plant species: Rhizophora mangle, Laguncularia racemosa and Avicennia nitida. A large number of these isolates, 115 in total, were evaluated for their ability to fix nitrogen and solubilize phosphorous. Bacteria that tested positive for both of these tests were examined further to determine their level of indole acetic acid production. Two strains with high indole acetic acid production were selected for use as inoculants for reforestation trees, and then the growth of the plants was evaluated under field conditions. The bacterium Pseudomonas fluorescens (strain MCR1.10) had a low phosphorus solubilization index, while this index was higher in the other strain used, Enterobacter sp. (strain MCR1.48). We used the reforestation tree Acacia polyphylla. The results indicate that inoculation with the MCR1.48 endophyte increases Acacia polyphylla shoot dry mass, demonstrating that this strain effectively promotes the plant's growth and fitness, which can be used in the seedling production of this tree. Therefore, we successfully screened the biotechnological potential of endophyte isolates from mangrove, with a focus on plant growth promotion, and selected a strain able to provide limited nutrients and hormones for in plant growth.

Project description:The abandoned pond-to-mangrove restoration project provides greater advantages than tidal flats afforestation in restoring mangrove ecosystem services and will be the primary method for mangrove restoration in the future. The existing methods for abandoned pond-to-mangrove restoration include artificial restoration through 'dike-breaking, filling with imported soil and tree planting' and natural restoration through 'dike-breaking and natural succession'. However, little is known about which restoration strategy (natural or artificial restoration) provides more benefits to the biodiversity of mangrove macrobethos. Given a prevailing view suggested that artificial restoration should be the preferred approach for accelerating recovery of biodiversity and vegetation structure in tropical regions, we hypothesised higher macrobenthic biodiversity and more complex community structure in artificial restoration than in natural restoration. To test this hypothesis, macrobenthic biodiversity and ecological processes were monitored in a typical abandoned pond-to-mangrove area of Dongzhaigang Bay, China, where artificial and natural restoration methods were used concurrently. Differences in macrobenthic biodiversity, community structure and ecological processes were compared using diversity indices, complex network analysis and null models. Similar species composition and ecological niche overlap and width among macrobenthos were observed at artificial and natural restoration sites. The biotic heterogeneity and interaction among macrobenthos were higher at the natural restoration sites than at the artificial restoration sites. Macrobenthos community assembly at natural and artificial restoration sites was both determined by deterministic processes, with environmental filtering dominating, which explained 52% and 54% of the variations in macrobenthic community structures respectively. Although our findings did not validate the research hypothesis, higher biotic heterogeneity and species interaction among macrobenthos could support natural restoration as the primary method for abandoned pond-to-mangrove projects, because it is a nature-based solution for mangrove restoration.

Project description:IntroductionPolyphyllin from Paris polyphylla var. yunnanensis exhibits anti-inflammatory, analgesic, antibacterial, and antiviral properties. However, the current production of polyphyllin can barely meet market demand. To improve the content of polyphyllin produced by P. polyphylla, two endophyte strains, Bacillus cereus LgD2 and Fusarium oxysporum TPB, were isolated from Paris fargesii Franch. and inoculated in the roots of P. polyphylla. Both symbiotic strains significantly promoted the accumulation of saponins in P. polyphylla.MethodsThe content of polyphyllin in rhizomes of P. polyphylla treated with TPB with LgD2 strain was determined using High Performance Liquid Chromatography and the expressed genes were analyzed by RNA-seq. Gene Ontology and Kyoto Encyclopedia of Genes annotations were performed on the differentially expressed genes, a clustering tree of UDP-glycosyltransferase (UGT) and cytochrome P450 (CYP450) gene families was constructed, and UGT and CYP450 involved in the biosynthesis of polyphyllin were predicted using weighted correlation network analysis (WGCNA).ResultsRNA-seq and qRT-PCR analyses showed that endophytic inoculation did not promote polyphyllin accumulation by enhancing the upstream terpene biosynthesis pathway, but probably by up-regulating the downstream CYP450 and UGT genes associated with polyphyllin biosynthesis. Genomes enrichment analyses of differentially expressed genes indicated that inoculation with LgD2 and TPB played a positive role in promoting the defense against pathogenic bacteria, enhancing the biosynthesis of carbohydrates, attenuating the process of nitrogen metabolism, and maintaining the equilibrium of the redox reaction homeostasis, potentially indirectly enhancing the polyphyllin yield of P. polyphylla. By combining differentially expressed genes screening, WGCNA, and phylogenetic tree analyses, 17 CYP450 and 2 UGT candidate genes involved in the biosynthesis of polyphyllin I, polyphyllin II, polyphyllin VII, polyphyllin D, and polyphyllin H were identified. These results suggest that endophytes probably effectively promote the accumulation of polyphyllin by regulating key downstream genes in biosynthetic pathways.DiscussionThis study provides a new approach for investigating the regulatory mechanisms of endophytes that promote the production and accumulation of polyphyllin in P. polyphylla, providing a basis for further elucidating the mechanisms of plant-endophyte interactions.
