Continuous cropping of endangered therapeutic plants via electron beam soil-treatment and neutron tomography.

ABSTRACT: Various medicinal plants are threatened with extinction owing to their over-exploitation and the prevalence of soil borne pathogens. In this study, soils infected with root-rot pathogens, which prevent continuous-cropping, were treated with an electron beam. The level of soil-borne fungus was reduced to ?0.01% by soil electron beam treatment without appreciable effects on the levels of antagonistic microorganism or on the physicochemical properties of the soil. The survival rate of 4-year-old plant was higher in electron beam-treated soil (81.0%) than in fumigated (62.5%), virgin (78%), or untreated-replanting soil (0%). Additionally, under various soils conditions, neutron tomography permitted the monitoring of plant health and the detection of root pathological changes over a period of 4-6 years by quantitatively measuring root water content in situ. These methods allow continual cropping on the same soil without pesticide treatment. This is a major step toward the environmentally friendly production of endangered therapeutic herbs.

Project description:The continuous cropping obstacles of melons have become increasingly serious in recent years. To investigate this, we explored the effects and mechanisms of Bacillus subtilis C3 in control of the continuous cropping obstacles of melon. We provide a novel interaction model of the occurrence factors of continuous cropping obstacles. The dominant pathogen isolated from melon soil was Fusarium. Their hyphae were used as food to cultivate root-knot nematodes. The main phenolic acids in melon soil promoted the growth of Fusarium and indirectly increased the number of root-knot nematodes, but they also had direct toxic effects on melon root-knot nematodes. The simultaneous inoculation of the three had the strongest inhibitory effect on melon seedlings, while the inhibitory effect of paired inoculation was weaker than that of single inoculation. Therefore, the three balance each other, forming a vicious cycle. Bacillus subtilis C3 weakened the negative effects of this cycle on melon by eliminating phenolic acids and inhibiting the growth of Fusarium and root-knot nematodes. Simultaneously, they also alleviated the continuous cropping obstacles of melon by improving the composition and structure of the rhizosphere microbial community. Our results might be useful for the effective control of the continuous cropping obstacles of melon. IMPORTANCE The soil environment, crop growth and fruit quality of melons are negatively affected by long-term continuous cropping. It is important to study the mechanism of continuous cropping obstacles and their biological control. In this study, we propose a novel interaction model of the occurrence factors of continuous cropping obstacles. The dominant phenolic acids, pathogenic fungi, and root-knot nematodes from melon soil balance each other, forming a vicious cycle. Bacillus subtilis C3 weakened the negative effects of this cycle on melon by eliminating phenolic acids and inhibiting the growth of Fusarium and root-knot nematodes. In addition, C3 also improved the composition and structure of the melon rhizosphere microbial community. These results advance the study of the occurrence mechanism of continuous cropping obstacles and demonstrate an efficient and environmentally friendly biological control scheme.

Project description:The continuous planting of soybeans leads to soil acidification, aggravation of soil-borne diseases, reduction in soil enzyme activity, and accumulation of toxins in the soil. Microorganisms in the rhizosphere play a very important role in maintaining the sustainability of the soil ecosystem and plant health. In this study, two soybean genotypes, one bred for continuous cropping and the other not, were grown in a Mollisol in northeast China under continuous cropping for 7 and 36years in comparison with soybean-maize rotation, and microbial communities in the rhizosphere composition were assessed using high-throughput sequencing technology. The results showed that short- or long-term continuous cropping had no significant effect on the rhizosphere soil bacterial alpha diversity. Short-term continuous planting increased the number of soybean cyst nematode (Heterodera glycines), while long-term continuous planting reduced these numbers. There were less soybean cyst nematodes in the rhizosphere of the tolerant genotypes than sensitive genotypes. In addition, continuous cropping significantly increased the potential beneficial bacterial populations, such as Pseudoxanthomonas, Nitrospira, and Streptomyces compared to rotation and short-term continuous cropping, suggesting that long-term continuous cropping of soybean shifts the microbial community toward a healthy crop rotation system. Soybean genotypes that are tolerant to soybean might recruit some microorganisms that enhance the resistance of soybeans to long-term continuous cropping. Moreover, the network of the two genotypes responded differently to continuous cropping. The tolerant genotype responded positively to continuous cropping, while for the sensitive genotype, topology analyses on the instability of microbial community in the rhizosphere suggested that short periods of continuous planting can have a detrimental effect on microbial community stability, although this effect could be alleviated with increasing periods of continuous planting.

Project description:Passion fruit (Passiflora edulis) is widely grown in tropical and subtropical regions, showing high economic and ornamental value. Microorganisms are indicators for the stability and health of the soil ecosystem, which can affect the yield and quality of passion fruit under continuous cropping. High-throughput sequencing and interactive analysis were used to analyse the variation of microbial communities in the noncultivated soil (NCS), cultivated soil (CS), and the rhizosphere soil of purple passion fruit (Passiflora edulis f. edulis ×Passiflora edulis f. flavicarpa, RP) and yellow passion fruit (Passiflora edulis f. flavicarpa, RY). An average of 98,001 high-quality fungal internal transcribed spacer (ITS) sequences, mainly from Ascomycota, Basidiomycota, Mortierellomycota, Mucoromycota and Glomeromycota, as well as an average of 71,299 high-quality bacterial 16S rRNA sequences, mainly from Proteobacteria, Actinobacteria, Acidobacteria, Firmicutes and Chloroflexi, were obtained per sample. It was found that the continuous cropping of passion fruit increased the richness but reduced the diversity of soil fungi, while it dramatically increased the richness and diversity of soil bacteria. In addition, during the continuous cropping, grafting different scions in the same rootstock contributed to the aggregation of differential rhizosphere microbial communities. Among fungal genera, Trichoderma showed higher abundance in RY than in RP and CS, while the opposite was observed in the pathogen Fusarium. Moreover, the co-occurrence network and potential function analyses also showed that the appearance of Trichoderma was related to Fusarium and its contribution to plant metabolism was significantly greater in RY than in RP and CS. In conclusion, the rhizosphere of yellow passion fruit may be beneficial for the enrichment of disease-resistant microbes, such as Trichoderma, which may be an important factor inducing stronger resistance to stem rot. It will help to form a potential strategy for overcoming the pathogen-mediated obstacles in passion fruit and improve its yield and quality.
