Curvularia microspora sp. nov. associated with leaf diseases of Hippeastrum striatum in China.

ABSTRACT: An undescribed Curvularia sp. was isolated from the leaf spot disease of Barbados Lily (Hippeastrum striatum (Lam.) Moore). Phylogenetic analyses of combined ITS, 28S, GPD1 and TEF1 sequence data place nine strains of this species in the trifolii-clade, but they clustered together as an independent lineage with strong support. This species was morphologically compared with related species in the trifolii-clade. Based on differences in morphology and phylogeny, it is concluded that this species is a new taxon, introduced as Curvularia microsporasp. nov. Pathogenicity testing determined the new species to be pathogenic on H. striatum.

Diseases of Cymbopogon citratus (Poaceae) in China: Curvularia nanningensis sp. nov.

Project description:Five Curvularia strains isolated from diseased leaves of lemongrass (Cymbopogon citratus) in Guangxi Province, China, were examined. NCBI-Blast searches of ITS sequences suggested a high degree of similarity (99-100%) to Curvularia akaii, C. akaiiensis, C. bothriochloae, C. heteropogonis and C. sichuanensis. To accurately identify these strains, we further analysed their morphology and phylogenetic relationships based on combinations of ITS, GAPDH, and tef1 gene sequences. Morphological observations indicated that the key character differing from similar species was conidial size, whereas phylogenetic analyses indicated that the five strains represent one species that is also distinct from C. akaii, C. akaiiensis and C. bothriochloae by conidial size and conidiophore length. Thus, the strains examined are found to represent a new species described herein as Curvularia nanningensis. The pathogenicity test on the host and detached leaves confirmed the new species to be pathogenic on Cymbopogon citratus leaves. Standardised requirements for reliable identification of Curvularia pathogens are also proposed.

Curvularia lunata and Curvularia Leaf Spot of Maize in China.

Project description:Curvularia leaf spot (CLS), primarily caused by Curvularia lunata (Wakker) Boedijn (C. lunata), is widely distributed in maize production regions in China. It occurs in all the developmental stages of maize and causes economic losses. The epidemic and yield loss estimation models have been constructed for the disease. C. lunata has obvious virulence differentiation and produces multiple virulence factors. CLS is managed by application of chemical and biological agents and by quantitative resistance conferred by 5 to 6 quantitative trait loci (QTL). This review summarizes research on the understanding of CLS biological characteristics, virulence factors of C. lunata, host resistance genetics, and disease management strategies in China.

Phaeotubakialithocarpicola gen. et sp. nov. (Tubakiaceae, Diaporthales) from leaf spots in China.

Project description:Tubakiaceae represents a distinct lineage of Diaporthales, including its type genus Tubakia and nine additional known genera. Tubakiaceous species are commonly known as endophytes in leaves and twigs of many tree species, but can also be plant pathogens causing conspicuous leaf symptoms. In the present study, isolates were obtained from diseased leaves of Lithocarpusglaber collected in Guangdong Province, China. The identification was conducted based on morphology and phylogeny of combined loci of 28S nrRNA gene (LSU), internal transcribed spacer regions and intervening 5.8S nrRNA gene (ITS) of the nrDNA operon, translation elongation factor 1-alpha (tef1) and beta tubulin (tub2). As a result, a distinct clade in Tubakiaceae was revealed named Phaeotubakialithocarpicola gen. et sp. nov., which was distinguished from the other tubakiaceous taxa by its dark brown conidiogenous cells and conidia.

Passalora stromatica sp. nov. associated with leaf spots of Tithonia diversifolia in Brazil.

Project description:Tithonia diversifolia, a member of Compositae native to Central America that produces showy sunflower-like flowers, became an invasive weed in other continents after it was introduced as an ornamental. Little is known about fungal pathogens infecting this plant. Knowledge of its mycobiota is of interest for future biocontrol programmes for T. diversifolia. In Brazil, a cercosporoid hyphomycete was found associated with intense leaf-spotting of this plant. Based on morphological and molecular data it was recognized as representing a new species of Passalora, and the name Passalora stromatica sp.nov. is introduced here for this taxon. This fungus is described and illustrated herein. It is possible that this fungus is playing a role in Brazil in reducing the invasiveness of T. diversifolia as, contrarily to what has been reported for countries in Africa and Asia, it remains mostly as a garden escape or rural plant in Brazil.

Lasiodiplodia fici sp. nov., Causing Leaf Spot on Ficus altissima in China.

Project description:High temperatures and the seasonality in tropical ecosystems favours plant pathogens, which result in many fungal diseases. Among these, diseases caused by Botryosphaeriaceae species are prominent as dieback, canker and leaf spots. In this research, we isolated one leaf-spot-causing Botryosphaeriaceae species from Ficus altissima leaves, which were collected in Guangzhou, Guangdong Province, China. Isolation and identification of the pathogen were based on morphological and molecular aspects. Based on multigene phylogenetic analysis of combined internal transcribed spacer (ITS), translation elongation factor 1-α gene (tef1) and beta-tubulin gene (tub2), the fungus associated with leaf spots on F. altissima is described as Lasiodiplodia fici, a novel species. Pathogenicity assays were conducted by inoculating the fungus onto detached shoots and plants under controlled environmental conditions. The results revealed that the L. fici isolates can infect the plant tissues under stress conditions by developing disease symptoms on detached shoots within three days. However, when it was inoculated onto the leaves of the host and grown in natural conditions, the progression of the disease was slow. The putative pathogen was re-isolated, and Koch's assumptions were satisfied. This is the first report of Lasiodiplodia species causing disease on Ficus altissima. Results from the present study will provide additional knowledge on fungal pathogens associated with forest and ornamental plant species.

Phyllosticta rizhaoensis sp. nov. causing leaf blight of Ophiopogon japonicus in China.

Project description:Ophiopogon japonicus (Asparagaceae) is a perennial grass species which can be cultivated as an ornamental and medicinal plant. From April 2021 to September 2022, a serious leaf blight disease of O. japonicus was discovered in Rizhao City, Shandong Province, China. The initial disease symptoms were small yellow spots, finally developing as tip blight, often associated with many small, black, semi-immersed pycnidial conidiomata formed in lesions. To obtain isolates of the causal agent for this disease, samples were randomly collected from O. japonicus diseased leaves in Rizhao City. In total 97 Phyllosticta isolates were obtained from samples, and studied using morphological features and multi-locus phylogenetic analyses of a combined dataset using the internal transcribed spacers (ITS), the 28S large subunit of ribosomal RNA (LSU), and partial translation elongation factor 1-alpha (tef), actin (act) and glyceraldehyde-3-phosphate dehydrogenase (gapdh) loci. Phylogenetically, these Phyllosticta isolates formed a clade in the P. concentrica species complex, and clustered with P. pilospora and P. spinarum. Morphologically, isolates in this clade differed from P. pilospora and P. spinarum by the size of conidiogenous cells and conidia, and the absence of an apical conidial appendage. As a result, these isolates were described as a novel species Phyllosticta rizhaoensis. Pathogenicity was confirmed using Koch's postulates, which showed that P. rizhaoensis could induce leaf blight symptoms on O. japonicus in China. Molecular phylogeny and morphology reveal two new graminicolous species, Curvularia aurantiasp. nov. and C. vidyodayana sp. nov. with new records of Curvularia spp. from Sri Lanka.

Project description:Despite being a small island, Sri Lanka is rich in fungal diversity. Most of the fungi from Sri Lanka have been identified as pathogens of vegetables, fruits, and plantation crops to date. The pleosporalean genus Curvularia (Dothideomycetes) includes phytopathogenic, saprobic, endophytic, and human/animal opportunistic pathogenic fungal species. The majority of the plant-associated Curvularia species are known from poaceous hosts. During the current study, 22 geographical locations of the country were explored and collections were made from 10 different poaceous hosts. Morphology and molecular phylogeny based on three loci, including nuclear internal transcribed spacers 1 and 2 with 5.8S nrDNA (ITS), glyceraldehyde-3-phosphate dehydrogenase (gapdh), and translation elongation factor 1-α (tef1) supported the description of two new species of fungi described herein as C. aurantia sp. nov. and C. vidyodayana sp. nov. Moreover, novel host-fungal association records for C. chiangmaiensis, C. falsilunata, C. lonarensis, C. plantarum, and C. pseudobrachyspora are updated herein. In addition, five species within the genus Curvularia, viz., C. asiatica, C. geniculata, C. lunata, C. muehlenbeckiae, and C. verruculosa represent new records of fungi from Sri Lanka. Dwiroopa punicae sp. nov. (Dwiroopaceae fam. nov., Diaporthales), associated with leaf spot and fruit rot of pomegranate (Punica granatum).

Project description:During a survey of diseases affecting pomegranate in the southeastern USA we identified a unique species of Diaporthales causing leaf spotting and fruit rot. Objectives of this study were to provide a morphological description of the putative new species, use DNA sequence data of three gene loci (LSU, ITS and rpb2) to accurately place the fungus within the Diaporthales, and to prove Koch's postulates. Morphological and phylogenetic comparisons confirmed the fungus to represent a new species and family, for which the names Dwiroopa punicae sp. nov. and Dwiroopaceae fam. nov. are proposed. This is the first report of a species of Dwiroopa being pathogenic to Punica granatum.
