The Cue-Approach Task as a General Mechanism for Long-Term Non-Reinforced Behavioral Change.

ABSTRACT: Recent findings show that preferences for food items can be modified without external reinforcements using the cue-approach task. In the task, the mere association of food item images with a neutral auditory cue and a speeded button press, resulted in enhanced preferences for the associated stimuli. In a series of 10 independent samples with a total of 255 participants, we show for the first time that using this non-reinforced method we can enhance preferences for faces, fractals and affective images, as well as snack foods, using auditory, visual and even aversive cues. This change was highly durable in follow-up sessions performed one to six months after training. Preferences were successfully enhanced for all conditions, except for negative valence items. These findings promote our understanding of non-reinforced change, suggest a boundary condition for the effect and lay the foundation for development of novel applications.

Spacing of cue-approach training leads to better maintenance of behavioral change.

Project description:The maintenance of behavioral change over the long term is essential to achieve public health goals such as combatting obesity and drug use. Previous work by our group has demonstrated a reliable shift in preferences for appetitive foods following a novel non-reinforced training paradigm. In the current studies, we tested whether distributing training trials over two consecutive days would affect preferences immediately after training as well as over time at a one-month follow-up. In four studies, three different designs and an additional pre-registered replication of one sample, we found that spacing of cue-approach training induced a shift in food choice preferences over one month. The spacing and massing schedule employed governed the long-term changes in choice behavior. Applying spacing strategies to training paradigms that target automatic processes could prove a useful tool for the long-term maintenance of health improvement goals with the development of real-world behavioral change paradigms that incorporate distributed practice principles.

Long-term serial position effects in cue-based inference.

Project description:An important theoretical question in decision making concerns the nature of cue-generation: What mechanism drives the generation of cues used to make inferences? Most models of decision making assume that the properties of cues, often cue validity, initiate a set of dynamic pre-decision processes. In two studies, we test how memory accessibility affects cue use by manipulating both ecological cue validity and cue accessibility in a stock-forecasting task. Cue accessibility was manipulated by the pattern of accurate cue discriminations within experiment blocks of the learning phase of the experiments. Specifically, we manipulated the serial positions in which the cues accurately discriminated while holding overall cue validity constant. At test, participants preferred cues that discriminated early in the learning phase-a kind of primacy effect. The findings suggest that cue use is influenced by memory retrieval mechanisms and that cue use is not solely determined by cue validity. The results have implications for the development of computational models of heuristic decision-making.

Vitellogenin-like A-associated shifts in social cue responsiveness regulate behavioral task specialization in an ant.

Project description:Division of labor and task specialization explain the success of human and insect societies. Social insect colonies are characterized by division of labor, with workers specializing in brood care early and foraging later in life. Theory posits that this task switching requires shifts in responsiveness to task-related cues, yet experimental evidence is weak. Here, we show that a Vitellogenin (Vg) ortholog identified in an RNAseq study on the ant T. longispinosus is involved in this process: using phylogenetic analyses of Vg and Vg-like genes, we firstly show that this candidate gene does not cluster with the intensively studied honey bee Vg but falls into a separate Vg-like A cluster. Secondly, an experimental knockdown of Vg-like A in the fat body caused a reduction in brood care and an increase in nestmate care in young ant workers. Nestmate care is normally exhibited by older workers. We demonstrate experimentally that this task switch is at least partly based on Vg-like A-associated shifts in responsiveness from brood to worker cues. We thus reveal a novel mechanism leading to early behavioral maturation via changes in social cue responsiveness mediated by Vg-like A and associated pathways, which proximately play a role in regulating division of labor.

Task cue influences on lexical decision performance and masked semantic priming effects: The role of cue-task compatibility.

Project description:Recent research demonstrated that mere presentation of a task cue influences subsequent unconscious semantic priming by attentional sensitization of related processing pathways. The direction of this influence depended on task-set dominance. Dominant task sets with a compatible cue-task mapping were supposed to be rapidly suppressed, while weak task sets showed more sustainable activation. Building on this research, we manipulated cue-task compatibility as instance of task-set dominance in two experiments and tested how masked semantic priming was influenced by actually performing the cued task (induction-task trials) or by mere cue presentation (task cue-only trials). In induction-task trials, the results of earlier research were replicated; semantic priming was larger following a semantic induction task compared to a perceptual induction task. In task cue-only trials, priming effects were reversed compared to induction-task trials in both experiments. Priming was larger for a perceptual compared to a semantic task set in task cue-only trials, indicating suppression of task sets following mere cue presentation in preparation for the upcoming lexical decision task. This notion of an inhibition of task sets after mere cue presentation was further supported by switching-related costs and changes of task-set implementation throughout the experiment. The absence of a moderator role of cue-task compatibility for task cue effects on priming in the present study suggests that the precise time course of task-set activation and inhibition in response to task cues as a function of cue-task compatibility might depend on specific experimental settings.

Prenatal docosahexaenoic acid supplementation has long-term effects on childhood behavioral and brain responses during performance on an inhibitory task.

Project description:Introduction: Offsprings from a prenatal docosahexaenoic acid (DHA) supplementation trial, in which pregnant women were assigned to placebo or 600mg DHA/day, were followed to determine the effect of prenatal DHA supplementation on the behavior and brain function at 5.5 years (n=81 placebo, n=86 supplemented).Methods: Event-related potentials (ERP) were recorded during a visual task requiring a button press (Go) to frequent target stimuli and response inhibition to the rare stimuli (No-Go). Univariate ANOVAs were used to test differences between group and sex for behavioral measures. ERP differences were tested using a three-way mixed-design multivariate analysis of variance (MANOVA).Results: There was a significant sex × group interaction for hit rate and errors of omission; there was no difference between males and females in the placebo group, but DHA males outperformed DHA females. Males overall and the placebo group made more errors requiring response inhibition; DHA females were significantly better than placebo females and DHA males. ERP P2 amplitude was larger in the DHA group. A significant N2 amplitude condition effect was observed in females and DHA group males, but not in placebo group males.Discussion: Prenatal DHA supplementation improved inhibitory performance overall, especially for females in the DHA group, possibly accounting for their conservative behavior during Go trials. Development of brain regions responsible for visual processing may be sensitive to maternal DHA status, evidenced by greater P2 amplitude. Males may benefit more from maternal DHA supplementation, indicated by the N2 condition effect seen only in males in the DHA group.

Adaptation supports short-term memory in a visual change detection task.

Project description:The maintenance of short-term memories is critical for survival in a dynamically changing world. Previous studies suggest that this memory can be stored in the form of persistent neural activity or using a synaptic mechanism, such as with short-term plasticity. Here, we compare the predictions of these two mechanisms to neural and behavioral measurements in a visual change detection task. Mice were trained to respond to changes in a repeated sequence of natural images while neural activity was recorded using two-photon calcium imaging. We also trained two types of artificial neural networks on the same change detection task as the mice. Following fixed pre-processing using a pretrained convolutional neural network, either a recurrent neural network (RNN) or a feedforward neural network with short-term synaptic depression (STPNet) was trained to the same level of performance as the mice. While both networks are able to learn the task, the STPNet model contains units whose activity are more similar to the in vivo data and produces errors which are more similar to the mice. When images are omitted, an unexpected perturbation which was absent during training, mice often do not respond to the omission but are more likely to respond to the subsequent image. Unlike the RNN model, STPNet produces a similar pattern of behavior. These results suggest that simple neural adaptation mechanisms may serve as an important bottom-up memory signal in this task, which can be used by downstream areas in the decision-making process.

Long-term imaging reveals behavioral plasticity during C. elegans dauer exit.

Project description:During their lifetime, animals must adapt their behavior to survive in changing environments. This ability requires the nervous system to adjust through dynamic expression of neurotransmitters and receptors but also through growth, spatial reorganization and connectivity while integrating external stimuli. For instance, despite having a fixed neuronal cell lineage, the nematode Caenorhabditis elegans’ nervous system remains plastic throughout its development. Here, we focus on a specific example of nervous system plasticity, the C. elegans dauer exit decision. Under unfavorable conditions, larvae will enter the non-feeding and non-reproductive dauer stage and adapt their behavior to cope with a new environment. Upon improved conditions, this stress resistant developmental stage is actively reversed to resume reproductive development. However, how different environmental stimuli regulate the exit decision mechanism and thereby drive the larva’s behavioral change is unknown. To fill this gap, we developed a new open hardware method for long-term imaging (12h) of C.elegans larvae. We identified dauer-specific behavioral motifs and characterized the behavioral trajectory of dauer exit in different environments to identify key decision points. Combining long-term behavioral imaging with transcriptomics, we find that bacterial ingestion triggers a change in neuropeptide gene expression to establish post-dauer behavior. Taken together, we show how a developing nervous system can robustly integrate environmental changes, activate a developmental switch and adapt the organism’s behavior to a new environment.

Assessment of Behavioral Disruption in Rats with Abdominal Inflammation Using Visual Cue Titration and the Five-choice Serial-reaction Time Task.

Project description:Both acute and chronic pain result in a number of behavioral symptoms in patients, including cognitive effects such as decreased attention and working memory. Intraperitoneal administration of dilute lactic acid in rodents has been used to induce abdominal inflammation and produce effects in behavioral assays of both sensory-discriminative and affective pain modalities.Intraperitoneal injection of dilute lactic acid was used to study the impact of abdominal inflammation on an operant task requiring sustained visual attention in rats (N = 7 to 15/group) that adapts dynamically to performance ability. The effects of ketoprofen and morphine on lactic acid-induced impairment were compared with those on the disruptive effects of scopolamine.Lactic acid impaired performance in a concentration-dependent manner, increasing the duration of cue presentation required to maintain optimal performance from 0.5?±?0.2?s (mean ± SD) to 17.2?±?11.4 s after the administration of 1.8% (v/v) (N = 13). The latency to emit correct responses and to retrieve the food reward were both increased by lactic acid. All effects of lactic acid injection were reversed by both ketoprofen and morphine in a dose-dependent manner. Scopolamine, however, produced dose-dependent, nonpain-related disruption in sustained attention that was not altered by either ketoprofen or morphine.These data demonstrate that abdominal inflammation induced by lactic acid produces robust disruption in a visual attention-based operant task and that this disruption is reversed by analgesics. Future studies will focus on pain-related circuitry and its impact on both limbic forebrain and frontal cortical mechanisms.

Distinct mechanism for long-term contrast adaptation.

Project description:To optimize perception, neurons in the visual system adapt to the current environment. What determines the durability of this plasticity? Longer exposures to an environment produce longer-lasting effects, which could be due to either (i) a single mechanism controlling adaptation that gains strength over time, or (ii) long-term mechanisms that become active after long-term exposure. Using recently developed technology, we tested adaptation durations an order of magnitude greater that those tested previously, and used a "deadaptation" procedure to reveal effects of a unique long-term mechanism in the longest adaptation periods. After 4 h of contrast adaptation, human observers were exposed to natural images for 15 min, which completely cancelled perceptual aftereffects of adaptation. Strikingly, during continued testing this deadaptation faded, and the original adaptation effects reappeared. This pattern strongly suggests that adaptation was maintained in a distinct long-term mechanism, whereas deadaptation affected a short-term mechanism.

Behavioral treatment of automatically reinforced SIB: 1982 - 2015.

Project description:Some individuals diagnosed with intellectual and developmental disabilities engage in automatically reinforced self-injurious behavior (SIB). For these individuals, identifying effective treatments may be difficult due to the nature of the reinforcement contingency. The purpose of this study was to review the literature on the treatment of automatically reinforced SIB to determine commonalities in procedures that produced effective and ineffective treatment outcomes, as well as historical trends in the treatment of this class of SIB. Results of this review indicated that there were many high-quality studies on this topic, but also a wide range in the quality of studies. As for effective treatments, noncontingent reinforcement (the most common treatment component) was found to be more effective when informed by a competing stimulus assessment rather than a preference assessment. Suggestions to improve the quality of the published record and areas in which additional research is needed are discussed.
