A role for the gibberellin pathway in biochar-mediated growth promotion.

ABSTRACT: Biochar is a carbon negative soil amendment that can promote crop growth. However, the effects of biochar on different plant species and cultivars within a species are not well understood, nor is the underlying basis of biochar-mediated plant growth promotion. This knowledge is critical for optimal use of biochar and for breeding biochar-responsive plants. Here, we investigated the genotype-specific effects of biochar on two cultivars of Solanum lycopersicum (tomato), and two wild relatives of tomato, Solanum pimpinellifolium, and Solanum pennelli, in two types of biochar. Biochar promoted shoot growth in all genotypes independent of biochar type but had genotype-dependent effects on other plant traits. Germination tests, exogenous GA4 application and mutant analysis indicated a role for GA in biochar-mediated plant growth promotion. Together, our results suggest that biochar promotes growth partially through stimulation of the GA pathway.

Rice Seedling Growth Promotion by Biochar Varies With Genotypes and Application Dosages.

Project description:While biochar use in agriculture is widely advocated, how the effect of biochar on plant growth varies with biochar forms and crop genotypes is poorly addressed. The role of dissolvable organic matter (DOM) in plant growth has been increasingly addressed for crop production with biochar. In this study, a hydroponic culture of rice seedling growth of two cultivars was treated with bulk mass (DOM-containing), water extract (DOM only), and extracted residue (DOM-free) of maize residue biochar, at a volumetric dosage of 0.01, 0.05, and 0.1%, respectively. On seedling root growth of the two cultivars, bulk biochar exerted a generally negative effect, while the biochar extract had a consistently positive effect across the application dosages. Differently, the extracted biochar showed a contrasting effect between genotypes. In another hydroponic culture with Wuyunjing 7 treated with biochar extract at sequential dosages, seedling growth was promoted by 95% at 0.01% dosage but by 26% at 0.1% dosage, explained with the great promotion of secondary roots rather than of primary roots. Such effects were likely explained by low molecular weight organic acids and nanoparticles contained in the biochar DOM. This study highlights the importance of biochar DOM and crop genotype when evaluating the effect of biochar on plants. The use of low dosage of biochar DOM could help farmers to adopt biochar technology as a solution for agricultural sustainability.

Molecular insights into biochar-mediated plant growth promotion and systemic resistance in tomato against Fusarium crown and root rot disease.

Project description:Molecular mechanisms associated with biochar-elicited suppression of soilborne plant diseases and improved plant performance are not well understood. A stem base inoculation approach was used to explore the ability of biochar to induce systemic resistance in tomato plants against crown rot caused by a soilborne pathogen, Fusarium oxysporum f. sp. radicis lycopersici. RNA-seq transcriptome profiling of tomato, and experiments with jasmonic and salycilic acid deficient tomato mutants, were performed to elucidate the in planta molecular mechanisms involved in induced resistance. Biochar (produced from greenhouse plant wastes) was found to mediate systemic resistance against Fusarium crown rot and to simultaneously improve tomato plant growth and physiological parameters by up to 63%. Transcriptomic analysis (RNA-seq) of tomato demonstrated that biochar had a priming effect on gene expression and upregulated the pathways and genes associated with plant defense and growth such as jasmonic acid, brassinosteroids, cytokinins, auxin and synthesis of flavonoid, phenylpropanoids and cell wall. In contrast, biosynthesis and signaling of the salicylic acid pathway was downregulated. Upregulation of genes and pathways involved in plant defense and plant growth may partially explain the significant disease suppression and improvement in plant performance observed in the presence of biochar.

Biochar Treatment Resulted in a Combined Effect on Soybean Growth Promotion and a Shift in Plant Growth Promoting Rhizobacteria.

Project description:The application of biochar to soil is considered to have the potential for long-term soil carbon sequestration, as well as for improving plant growth and suppressing soil pathogens. In our study we evaluated the effect of biochar on the plant growth of soybeans, as well as on the community composition of root-associated bacteria with plant growth promoting traits. Two types of biochar, namely, maize biochar (MBC), wood biochar (WBC), and hydrochar (HTC) were used for pot experiments to monitor plant growth. Soybean plants grown in soil amended with HTC char (2%) showed the best performance and were collected for isolation and further characterization of root-associated bacteria for multiple plant growth promoting traits. Only HTC char amendment resulted in a statistically significant increase in the root and shoot dry weight of soybeans. Interestingly, rhizosphere isolates from HTC char amended soil showed higher diversity than the rhizosphere isolates from the control soil. In addition, a higher proportion of isolates from HTC char amended soil compared with control soil was found to express plant growth promoting properties and showed antagonistic activity against one or more phytopathogenic fungi. Our study provided evidence that improved plant growth by biochar incorporation into soil results from the combination of a direct effect that is dependent on the type of char and a microbiome shift in root-associated beneficial bacteria.

Plant growth improvement mediated by nitrate capture in co-composted biochar.

Project description:Soil amendment with pyrogenic carbon (biochar) is discussed as strategy to improve soil fertility to enable economic plus environmental benefits. In temperate soils, however, the use of pure biochar mostly has moderately-negative to -positive yield effects. Here we demonstrate that co-composting considerably promoted biochars' positive effects, largely by nitrate (nutrient) capture and delivery. In a full-factorial growth study with Chenopodium quinoa, biomass yield increased up to 305% in a sandy-poor soil amended with 2% (w/w) co-composted biochar (BC(comp)). Conversely, addition of 2% (w/w) untreated biochar (BC(pure)) decreased the biomass to 60% of the control. Growth-promoting (BC(comp)) as well as growth-reducing (BC(pure)) effects were more pronounced at lower nutrient-supply levels. Electro-ultra filtration and sequential biochar-particle washing revealed that co-composted biochar was nutrient-enriched, particularly with the anions nitrate and phosphate. The captured nitrate in BC(comp) was (1) only partly detectable with standard methods, (2) largely protected against leaching, (3) partly plant-available, and (4) did not stimulate N2O emissions. We hypothesize that surface ageing plus non-conventional ion-water bonding in micro- and nano-pores promoted nitrate capture in biochar particles. Amending (N-rich) bio-waste with biochar may enhance its agronomic value and reduce nutrient losses from bio-wastes and agricultural soils.

Linking the Belowground Microbial Composition, Diversity and Activity to Soilborne Disease Suppression and Growth Promotion of Tomato Amended with Biochar.

Project description:Biochar, in addition to sequestering carbon, ameliorating soil, and improving plant performance, can impact foliar and soilborne plant diseases. Nevertheless, the mechanisms associated with suppression of soilborne diseases and improved plant performances are not well understood. This study is designed to establish the relationships between biochar-induced changes in rhizosphere microbial community structure, taxonomic and functional diversity, and activity with soilborne disease suppression and enhanced plant performance in a comprehensive fashion. Biochar suppressed Fusarium crown and root-rot of tomato and simultaneously improved tomato plant growth and physiological parameters. Furthermore, biochar reduced Fusarium root colonization and survival in soil, and increased the culturable counts of several biocontrol and plant growth promoting microorganisms. Illumina sequencing analyses of 16S rRNA gene revealed substantial differences in rhizosphere bacterial taxonomical composition between biochar-amended and non-amended treatments. Moreover, biochar amendment caused a significant increase in microbial taxonomic and functional diversity, microbial activities and an overall shift in carbon-source utilization. High microbial taxonomic and functional diversity and activity in the rhizosphere has been previously associated with suppression of diseases caused by soilborne pathogens and with plant growth promotion, and may collectively explain the significant reduction of disease and improvement in plant performance observed in the presence of biochar.

The OXI1 kinase pathway mediates Piriformospora indica-induced growth promotion in Arabidopsis.

Project description:Piriformospora indica is an endophytic fungus that colonizes roots of many plant species and promotes growth and resistance to certain plant pathogens. Despite its potential use in agriculture, little is known on the molecular basis of this beneficial plant-fungal interaction. In a genetic screen for plants, which do not show a P. indica- induced growth response, we isolated an Arabidopsis mutant in the OXI1 (Oxidative Signal Inducible1) gene. OXI1 has been characterized as a protein kinase which plays a role in pathogen response and is regulated by H?O? and PDK1 (3-PHOSPHOINOSITIDE-DEPENDENT PROTEIN KINASE1). A genetic analysis showed that double mutants of the two closely related PDK1.1 and PDK1.2 genes are defective in the growth response to P. indica. While OXI1 and PDK1 gene expression is upregulated in P. indica-colonized roots, defense genes are downregulated, indicating that the fungus suppresses plant defense reactions. PDK1 is activated by phosphatidic acid (PA) and P. indica triggers PA synthesis in Arabidopsis plants. Under beneficial co-cultivation conditions, H?O? formation is even reduced by the fungus. Importantly, phospholipase D (PLD)?1 or PLD? mutants, which are impaired in PA synthesis do not show growth promotion in response to fungal infection. These data establish that the P. indica-stimulated growth response is mediated by a pathway consisting of the PLD-PDK1-OXI1 cascade.

Microbial-Mediated Plant Growth Promotion and Pest Suppression Varies Under Climate Change.

Project description:Climate change is altering the dynamics of crop pests and diseases resulting in reduced crop yields. Using beneficial soil bacterial to increase crop health is a quickly developing area in sustainable agriculture, but it is unknown if climate change or interactions with other species could alter their effect. The plant growth-promoting rhizobacterium Acidovorax radicis N35 is known to increase barley (Hordeum vulgare) plant growth under laboratory conditions, and we tested the stability of the plant-bacterial interactions when exposed to elevated carbon dioxide (CO2) and ozone (O3) levels while infesting the aboveground leaves with cereal aphids (Sitobion avenae) and the soil with beneficial earthworms. Acidovorax radicis N35 increased plant growth and reduced insect growth - with greatest effect in a high-stress elevated O3 environment, but reduced effects under elevated CO2. Earthworms promoted both plant and insect growth, but inoculation with A. radicis N35 alleviated some of the earthworm-mediated increase in pest abundance, particularly in the ambient environment. The consistency of these beneficial effects highlights the potential of exploiting local species interactions for predicting and mitigating climate change effects in managed systems. We conclude that microbial bioprotectants have high potential for benefiting agriculture via plant-growth promotion and pest suppression.

Does gibberellin biosynthesis play a critical role in the growth of Lolium perenne? Evidence from a transcriptional analysis of gibberellin and carbohydrate metabolic genes after defoliation.

Project description:Global meat and milk production depends to a large extent on grazed pastures, with Lolium perenne being the major forage grass in temperate regions. Defoliation and subsequent regrowth of leaf blades is a major and essential event with respect to L. perenne growth and productivity. Following defoliation, carbohydrates (mainly fructans and sucrose) have to be mobilized from heterotrophic tissues to provide energy and carbon for regrowth of photosynthetic tissues. This mobilization of reserve carbohydrates requires a substantial change in the expression of genes coding for enzymes involved in carbohydrate metabolism. Here we tested the hypothesis that gibberellins (GA) are at the core of the processes regulating the expression of these genes. Thus, we examined the transcript profiles of genes involved in carbohydrate and GA metabolic pathways across a time course regrowth experiment. Our results show that following defoliation, the immediate reduction of carbohydrate concentrations in growing tissues is associated with a concomitant increase in the expression of genes encoding carbohydrate mobilizing invertases, and was also associated with a strong decrease in the expression of fructan synthesizing fructosyltransferase genes. We also show that the decrease in fructan levels is preceded by increased expression of the GA activating gene GA 3-oxidase and decreased expression of the GA inactivating gene GA 2 -oxidase in sheaths. GA 3-oxidase expression was negatively, while GA 2 -oxidase positively linked to sucrose concentrations. This study provides indicative evidence that gibberellins might play a role in L. perenne regrowth following defoliation and we hypothesize that there is a link between gibberellin regulation and sugar metabolism in L. perenne.
