Project description:Chelonus formosanus Sonan 1932 (Hymenoptera: Braconidae) is a wasp capable of parasitizing a variety of lepidopteran pests at the "egg-larval" stage which distributes throughout Taiwan, Guangdong, Zhejiang, and Hainan provinces of China. This wasp has been successfully used to control pests such as Spodoptera litura Fabricius, 1775, Spodoptera frugiperda (JE Smith, 1797), Spodoptera exigua (Hübner, 1808), and Helicoverpa armigera (Hübner, 1808). So far, there is only one genome assembled from the Chelonus genus [Chelonus insularis (Cresson, 1865)] and it is fragmented with 455 scaffolds. Here, we report a chromosome-level genome assembly of C. formosanus, which was sequenced using PacBio, Illumina, and Hi-C technologies. The long reads were 35.4 Gb (∼150× coverage) with an average length of 15.23 kb. The size of the genome assembly was 139.59 Mb. More than 99.46% of the assembled sequences were anchored to seven pseudochromosomes (138.84 Mb). The Benchmarking University Single-Copy Orthologs (BUSCO) assessment results showed 99.0% of the 1,367 genes (insect_odb10 database) were completely present. We annotated 11,242 protein-coding genes including 98.6% of BUSCO complete genes that were recovered. Nearly one-fourth of the genome assembly (22.25%) was annotated as repetitive sequences and 324 noncoding RNAs were predicted. There were 58 gene families found with significant expansion including allelopathic families (odorant receptors and ionotropic receptors), which may play a crucial role in efficiently locating a wide range of hosts. This high-quality genome assembly and annotation could provide a highly valuable resource of parasitic wasp for the biological control of Lepidoptera pest.

Project description:The molecular mechanisms that allow generalist parasitoids to exploit many, often very distinct hosts are practically unknown. The wasp Aphidius ervi, a generalist koinobiont parasitoid of aphids, was introduced from Europe into Chile in the late 1970s to control agriculturally important aphid species. A recent study showed significant differences in host preference and host acceptance (infectivity) depending on the host A. ervi were reared on. In contrast, no genetic differentiation between A. ervi populations parasitizing different aphid species and aphids of the same species reared on different host plants was found in Chile. Additionally, the same study did not find any fitness effects in A. ervi if offspring were reared on a different host as their mothers. Here, we determined the effect of aphid host species (Sitobion avenae versus Acyrthosiphon pisum reared on two different host plants alfalfa and pea) on the transcriptome of adult A. ervi females. We found a large number of differentially expressed genes (between host species: head: 2,765; body: 1,216; within the same aphid host species reared on different host plants: alfalfa versus pea: head 593; body 222). As expected, the transcriptomes from parasitoids reared on the same host species (pea aphid) but originating from different host plants (pea versus alfalfa) were more similar to each other than the transcriptomes of parasitoids reared on a different aphid host and host plant (head: 648 and 1,524 transcripts; body: 566 and 428 transcripts). We found several differentially expressed odorant binding proteins and olfactory receptor proteins in particular, when we compared parasitoids from different host species. Additionally, we found differentially expressed genes involved in neuronal growth and development as well as signaling pathways. These results point towards a significant rewiring of the transcriptome of A. ervi depending on aphid-plant complex where parasitoids develop, even if different biotypes of a certain aphid host species (A. pisum) are reared on the same host plant. This difference seems to persist even after the different wasp populations were reared on the same aphid host in the laboratory for more than 50 generations. This indicates that either the imprinting process is very persistent or there is enough genetic/allelic variation between A. ervi populations. The role of distinct molecular mechanisms is discussed in terms of the formation of host fidelity.

Project description:Six new species of the genus Distatrix Mason from Central and South America, D. loretta, D. xanadon, D. vigilis, D. pitillaensis, D. pandora Grinter, n. sp., and D. antirrheae Whitfield & Grinter, n. sp., are described from large-scale caterpillar inventory endeavors, mostly from the larvae of geometrid moths. Biological information and diagnostic features that distinguish these species from other previously described Distatrix, especially those from the Neotropical region, are provided; and the first key to New World species is presented. The new discoveries expand our knowledge of the World's widespread Distatrix fauna by about a third, suggesting that similar survey efforts in other poorly sampled regions will reveal numerous additional undescribed species.

Project description:A new species of opiine Braconidae, Opius rojam Daniels & Wharton, is described from Trinidad. The description is based in part on two individuals reared from Sepsisoma erythrocephalum infesting shoots of the grass Paspalum fasciculatum. This is the first record of members of the Opiinae attacking species in the dipteran family Richardiidae. The Opius ingenticornis species group is proposed and delineated to accommodate this and several putatively related species formerly included in Opius (Merotrachys), Opius (Pendopius), and Opius (Ilicopius). A key to the species of this group is provided. Descriptions and diagnoses are referenced to the Hymenoptera Anatomy Ontology.

Project description:Glyptapanteles Ashmead (Hymenoptera: Braconidae: Microgastrinae) is a cosmopolitan group of hyperdiverse parasitic wasps. The genus remains taxonomically challenging in India due to its highly speciose nature, morphological similarity amongst species and negligible host records. The Indian fauna is one of the most diverse and also the least studied. The present study is based on 60 populations reared from 35 host species, 100+ individual caterpillar rearings (1100 wasp specimens pinned and 2000 in alcohol) and from 12 different geographical locations of the country (11 states and one Union territory) that represent 26 provisional Glyptapanteles species within 8 species-groups. Out of 60 populations, phylogenetic analyses were performed on 38 based on mitochondrial cytochrome oxidase subunit I (COI) nucleotide sequences. Maximum likelihood and Bayesian inference methods displayed three and four major discrete Glyptapanteles clades, respectively. In clade A very few Indian species were grouped along with Neotropical and Thailand species. The other clades B and C grouped the majority of the Indian species and showed considerable host specificity in both the trees. All parasitic wasp species were gregarious in nature, except for two populations. Three different sets of data (morphology, host records, and COI) were integrated in order to generate accurate boundaries between species/species-groups. Illustrations of all parasitized caterpillars/cocoons and 42 habitus views of Glyptapanteles spp., distributional information, and GenBank accession numbers, are presented. The present study, perhaps the most comprehensive done to date in India, suggests the presence of several additional Glyptapanteles species, which were previously unrecognized.

Project description:Endoparasitoids can be killed by host encapsulation, a cellular-mediated host immunological response against parasitism that involves hemocytes aggregation. As a counteracting strategy, many parasitoids can evade this host response through self-superparasitism. The objectives of this study were: 1) to describe the parasitoid Pseudapanteles dignus (Hymenoptera: Braconidae) early immature stages (egg and larva) encapsulation by the host Tuta absoluta (Lepidoptera: Gelechiidae), and 2) to determine the occurrence of self-superparasitism and the rate of escaping to encapsulation of this parasitoid. Knowledge of host-parasitoid immunological interaction is crucial when evaluating the potential of an endoparasitoid as a biological control candidate. Parasitoid-exposed T. absoluta larvae were dissected in vivo under light stereoscope microscope at 24-h intervals, for five days after exposition to detect encapsulation. The preimaginal stages of P. dignus and numbers of healthy and encapsulated immature parasitoids per host were recorded. Samples of parasitoid eggs and larvae were processed for SEM visualization of encapsulation. Necropsies evidenced that only the early first larval instar of P. dignus (up to 96 h-old) was partially or completely encapsulated. A non-melanized capsule, formed by layers of granulocyte-type hemocytes enveloping around the parasitoid body, was recorded. Approximately 50% of the parasitized T. absoluta larvae had significantly only one P. dignus egg, meanwhile supernumerary parasitization yielded up to seven immature parasitoids per host. The proportion of single-early first larval instar of P. dignus reached ≈ 0.5 and decreased significantly as the number of parasitoid individuals per host increased. P. dignus encapsulation and its ability to overcome with the host immune defense through self-superparasitism indicate that T. absoluta is a semi-permissive host for this parasitoid.

The genus Centistidea Rohwer (Hymenoptera: Braconidae: Miracinae) is recorded for the first time from Saudi Arabia. Two species were reared from Acrocercops phaeospora Meyrick and Cosmopteryx phaeogastra (Meyrick) in India, while Centistidea tihamica was collected by Malaise trap in Saudi Arabia. Characters of these new species and their affinities with related taxa are discussed. Data on habitat, host records, and host plant species for all the parasitoid species are also provided.

Project description:The descriptive taxonomic study reported here is focused on Glyptapanteles, a species-rich genus of hymenopteran parasitoid wasps. The species were found within the framework of two independent long-term Neotropical caterpillar rearing projects: northwestern Costa Rica (Área de Conservación Guanacaste, ACG) and eastern Andes, Ecuador (centered on Yanayacu Biological Station, YBS). One hundred thirty-six new species of Glyptapanteles Ashmead are described and all of them are authored by Arias-Penna. None of them was recorded in both countries; thus, 78 are from Costa Rica and the remaining 58 from Ecuador. Before this revision, the number of Neotropical described Glyptapanteles did not reach double digits. Reasonable boundaries among species were generated by integrating three datasets: Cytochrome Oxidase I (COI) gene sequencing data, natural history (host records), and external morphological characters. Each species description is accompanied by images and known geographical distribution. Characteristics such as shape, ornamentation, and location of spun Glyptapanteles cocoons were imaged as well. Host-parasitoid associations and food plants are also here published for the first time. A total of 88 species within 84 genera in 15 Lepidoptera families was encountered as hosts in the field. With respect to food plants, these wild-caught parasitized caterpillars were reared on leaves of 147 species within 118 genera in 60 families. The majority of Glyptapanteles species appeared to be relatively specialized on one family of Lepidoptera or even on some much lower level of taxonomic refinement. Those herbivores in turn are highly food-plant specialized, and once caterpillars were collected, early instars (1-3) yielded more parasitoids than later instars. Glyptapanteles jimmilleri Arias-Penna, sp. nov. is the first egg-larval parasitoid recorded within the genus, though there may be many more since such natural history requires a more focused collection of eggs. The rate of hyperparasitoidism within the genus was approximately 4% and was represented by Mesochorus spp. (Ichneumonidae). A single case of multiparasitoidism was reported, Copidosoma floridanum Ashmead (Encyrtidae) and Glyptapanteles ilarisaaksjarvi Arias-Penna, sp. nov. both parasitoid species emerged from the caterpillar of Noctuidae: Condica cupienta (Cramer). Bodyguard behavior was observed in two Glyptapanteles species: G. howelldalyi Arias-Penna, sp. nov. and G. paulhansoni Arias-Penna, sp. nov. A dichotomous key for all the new species is provided. The numerous species described here, and an equal number already reared but not formally described, signal a far greater Glyptapanteles species richness in the Neotropics than suggested by the few described previously.
