Synopsis of the cyclocephaline scarab beetles (Coleoptera, Scarabaeidae, Dynastinae).

ABSTRACT: The cyclocephaline scarabs (Scarabaeidae: Dynastinae: Cyclocephalini) are a speciose tribe of beetles that include species that are ecologically and economically important as pollinators and pests of agriculture and turf. We provide an overview and synopsis of the 14 genera of Cyclocephalini that includes information on: 1) the taxonomic and nomenclatural history of the group; 2) diagnosis and identification of immature life-stages; 3) economic importance in agroecosystems; 4) natural enemies of these beetles; 5) use as food by humans; 6) the importance of adults as pollination mutualists; 7) fossil cyclocephalines and the evolution of the group; 8) generic-level identification of adults. We provide an expanded identification key to genera of world Cyclocephalini and diagnoses for each genus. Character illustrations and generic-level distribution maps are provided along with discussions on the relationships of the tribe's genera.

Project description:The scarab beetle genus Pseudogeniates Ohaus (Scarabaeidae: Rutelinae: Rutelini) is endemic to Argentina. The genus includes three species: Pseudogeniates cordobaensis Soula, Pseudogeniates intermedius Ohaus, and Pseudogeniates richterianus Ohaus. We characterize the genus, provide a key to species, redescribe and diagnose each species, provide spatial and temporal distributions, and discuss distributions of the species in relation to eco-regions and areas of endemism in Argentina.

Mitochondrial phylogenomics reveals deep relationships of scarab beetles (Coleoptera, Scarabaeidae).

Project description:In this study, we newly sequenced the complete mitochondrial genomes (mitogenomes) of two phytophagous scarab beetles, and investigated the deep level relationships within Scarabaeidae combined with other published beetle mitogenome sequences. The complete mitogenomes of Dicronocephalus adamsi Pascoe (Cetoniinae) and Amphimallon sp. (Melolonthinae) are 15,563 bp and 17,433 bp in size, respectively. Both mitogenomes have the typical set of 37 genes (13 protein-coding genes, 22 transfer RNA genes, two ribosomal RNA genes) and an A+T-rich region, with the same gene arrangement found in the majority of beetles. The secondary structures for ribosomal RNA genes (rrnL and rrnS) were inferred by comparative analysis method. Results from phylogenetic analyses provide support for major lineages and current classification of Scarabaeidae. Amino acid data recovered Scarabaeidae as monophyletic. The Scarabaeidae was split into two clades. One clade contained the subfamilies Scarabaeinae and Aphodiinae. The other major clade contained the subfamilies Dynastinae, Rutelinae, Cetoniinae, Melolonthinae and Sericini. The monophyly of Scarabaeinae, Aphodiinae, Dynastinae, Cetoniinae and Sericini were strongly supported. The Scarabaeinae was the sister group of Aphodiinae. The Cetoniinae was sister to the Dynastinae + Rutelinae clade. The Melolonthinae was a non-monophyletic group. The removal of fast-evolving sites from nucleotide dataset using a pattern sorting method (OV-sorting) supported the family Scarabaeidae as a monophyletic group. At the tribe level, the Onthophagini was non-monophyletic with respect to Oniticellini. Ateuchini was sister to a large clade comprising the tribes Onthophagini, Oniticellini and Onitini. Eurysternini was a sister group of the Phanaeini + Ateuchini clade.

Completely engaged three-dimensional mandibular gear-like structures in the adult horned beetles: reconsideration of bark-carving behaviors (Coleoptera, Scarabaeidae, Dynastinae).

Project description:Adult horned beetles (Coleoptera, Scarabaeidae, Dynastinae) such as Trypoxylusdichotomus (Linnaeus, 1771) exhibit bark-carving behaviors to feed on tree sap, in part by using small projections of the clypeus. However, in the present experiments, adult horned beetles (T.dichotomus and Dynasteshercules (Linnaeus, 1758)) used their mandibles and not the projections of the clypeus to carve bark. Our findings show the presence of completely engaged mandibular interlocking, gear-like surface structures in molar areas that guide mandible opening and closure, and lead to completely synchronous movements of adult horned beetle mandibles. Three-dimensional shapes of these mandibular gear-like structures are complex and remained in contact after the death of a beetle. Moreover, adult horned beetles often performed bark-carving behaviors using only the mandible of one side, suggesting that the primary role of the mandibular gear-like structure is to prevent breakage of the mandible by transmitting load from one mandible to the other. Among the 22 Dynastinae and 16 other beetle species examined (not Dynastinae), the gear-like structure was found in all the Dynastinae species and in no other species.

Experimental degradation of helicoidal photonic nanostructures in scarab beetles (Coleoptera: Scarabaeidae): implications for the identification of circularly polarizing cuticle in the fossil record.

Project description:Scarab beetles (Coleoptera: Scarabaeidae) can exhibit striking colours produced by pigments and/or nanostructures. The latter include helicoidal (Bouligand) structures that can generate circularly polarized light. These have a cryptic evolutionary history in part because fossil examples are unknown. This suggests either a real biological signal, i.e. that Bouligand structures did not evolve until recently, or a taphonomic signal, i.e. that conditions during the fossilization process were not conducive to their preservation. We address this issue by experimentally degrading circularly polarizing cuticle of modern scarab beetles to test the relative roles of decay, maturation and taxonomy in controlling preservation. The results reveal that Bouligand structures have the potential to survive fossilization, but preservation is controlled by taxonomy and the diagenetic history of specimens. Further, cuticle of specific genus (Chrysina) is particularly decay-prone in alkaline conditions; this may relate to the presence of certain compounds, e.g. uric acid, in the cuticle of these taxa.

Termitotroxvenus sp. n. (Coleoptera, Scarabaeidae), a new blind, flightless termitophilous scarab from Cambodia.

Project description:Termitotroxvenus sp. n. is described from Cambodia and represents the second discovery of Termitotrox Reichensperger, 1915 from the Indo-Chinese subregion of the Indomalayan region. Most of the type series was collected from refuse dumps in fungus garden cells of Macrotermescf.gilvus (Hagen, 1858). Macrotermes Holmgren, 1910 was previously an unknown host of Termitotrox species. The new species is easily distinguished from all known congeners by having wing-shaped trichomes on the elytra and the larger body size, at 2.5 mm in length. We also describe the mouthparts to complement the description of the genus Termitotrox.

Description of Onthophagus humboldti and Uroxys bonplandi, two new scarab beetles (Coleoptera, Scarabaeidae, Scarabaeinae) from Costa Rica, with notes on tropical mountain brachyptery and endemicity.

Project description:Two new endemic species of scarab beetles are described from Costa Rica, Onthophagus humboldti sp. nov. and Uroxys bonplandi sp. nov. Onthophagus humboldti sp. nov. is also the tenth brachypterous Onthophagus species to be described worldwide, representing also a case of extreme brachyptery in Onthophagini. Illustrations for both new species, as well as marking differences with closely related species are included. Maps showing the distribution of the new species, as well as the distribution of brachypterous and endemic scarab-beetle species for Costa Rica are presented and discussed. The Cordillera de Talamanca represents an area where Scarabaeinae (four genera) show very high known levels of brachypterism in Mesoamerica. A reconstruction of the montane environment in the Cordillera de Talamanca during the Last Glacial Maximum (~24 ka) is analyzed, in order to try to understand a possible historical biogeography model that might promote high levels of brachypterism in scarab-beetles. The present study supports previous proposals that brachyptery is correlated with stable environments associated with deeply incised valleys. Tropical mountain ranges are also identified as having more endemics than lowland rain forests, contradicting accepted wisdom. Lastly, a mitochondrial DNA analysis supports the existence of the Onthophagus dicranius and the O. clypeatus species-groups as two well-defined and closely related branches.

Bait attractiveness changes community metrics in dung beetles (Coleoptera: Scarabaeidae: Scarabaeinae).

Project description:Species relative abundance (SRA) is an essential attribute of biotic communities, which can provide an accurate description of community structure. However, the sampling method used may have a direct influence on SRA quantification, since the use of attractants (e.g., baits, light, and pheromones) can introduce additional sources of variation in trap performance. We tested how sampling aided by baits affect community data and therefore alter derived metrics. We tested our hypothesis on dung beetles using data from flight interception traps (FITs) as a baseline to evaluate baited pitfall trap performance. Our objective was to assess the effect of bait attractiveness on estimates of SRA and assemblage metrics when sampled by pitfall traps baited with human feces.Dung beetles were sampled at three terra firme primary forest sites in the Brazilian Amazon. To achieve our objective, we (i) identified species with variable levels of attraction to pitfall baited with human feces; (ii) assessed differences in SRA; and (iii) assessed the effect of bait on the most commonly used diversity metrics derived from relative abundance (Shannon and Simpson indices). We identified species less and highly attracted to the baits used, because most attracted species showed greater relative abundances within baited pitfall traps samples compared with our baseline. Assemblages sampled by baited pitfall traps tend to show lower diversity and higher dominance than those sampled by unbaited FITs. Our findings suggest that for ecological questions focused on species relative abundance, baited pitfall traps may lead to inaccurate conclusions regarding assemblage structure. Although tested on dung beetles, we suggest that the same effect could be observed for other insect taxa that are also sampled with baited traps. We highlight a need for further studies on other groups to elucidate any potential effects of using baits.

Potential cooperations between odorant-binding proteins of the scarab beetle Holotrichia oblita Faldermann (Coleoptera: Scarabaeidae).

Project description:It was previously thought that the odorant binding proteins (OBPs) in the sensillum lymph might serve as carriers, which could carry lipophilic odorant molecules to olfactory receptors. In this study, two novel OBP genes of the scarab beetle Holotrichia oblita were screened using an antennal cDNA library. The full cDNA of HoblOBP3 and HoblOBP4 was cloned using reverse transcription PCR and rapid amplification of the cDNA ends. Homology modeling of both OBPs was performed using SWISS-MODEL on-line tools. Next, the two OBPs were expressed in Escherichia coli and purified using Ni ion affinity chromatography. The ligand-binding properties of HoblOBP3 and HoblOBP4 in 42 ligands respectively were measured using the fluorescence probe N-phenyl-naphthylamine (1-NPN). The results obtained from competitive binding assays demonstrated that HoblOBP4 showed a broader range of binding affinities to the test compounds, while HoblOBP3 displays more specific binding affinity. Furthermore, other OBPs and CSPs were expressed in Escherichia coli and purified using Ni ion affinity chromatography. Binding curves were measured for binary mixtures of OBPs and CSPs using 1-NPN, and the Scatchard plots exhibited "J"-like nonlinear correlation trends in some samples. In addition, competitive binding assays of the HoblOBP1 and HoblOBP2 mixtures and of the HoblOBP2 and HoblOBP4 mixtures with representative compounds unexpectedly demonstrated good affinity, which revealed extreme differences that were only obtained using the individual proteins. In the immunocytochemical analysis, colocalization of HoblOBP1 and HoblOBP2, and of HoblOBP2 and HoblOBP4, was detected in the sensilla basiconica and sensilla placodea, respectively. All of these results suggested that HoblOBP1 and HoblOBP2, as well as HoblOBP2 and HoblOBP4, may serve as heterodimers in the sensillum lymph.
