Dendritic Integration of Sensory Evidence in Perceptual Decision-Making.

ABSTRACT: Perceptual decisions require the accumulation of sensory information to a response criterion. Most accounts of how the brain performs this process of temporal integration have focused on evolving patterns of spiking activity. We report that subthreshold changes in membrane voltage can represent accumulating evidence before a choice. αβ core Kenyon cells (αβ_c KCs) in the mushroom bodies of fruit flies integrate odor-evoked synaptic inputs to action potential threshold at timescales matching the speed of olfactory discrimination. The forkhead box P transcription factor (FoxP) sets neuronal integration and behavioral decision times by controlling the abundance of the voltage-gated potassium channel Shal (K_V4) in αβ_c KC dendrites. αβ_c KCs thus tailor, through a particular constellation of biophysical properties, the generic process of synaptic integration to the demands of sequential sampling.

Integration of sensory evidence and reward expectation in mouse perceptual decision-making task with various sensory uncertainties.

Project description:In perceptual decision-making, prior knowledge of action outcomes is essential, especially when sensory inputs are insufficient for proper choices. Signal detection theory (SDT) shows that optimal choice bias depends not only on the prior but also the sensory uncertainty; however, it is unclear how animals integrate sensory inputs with various uncertainties and reward expectations to optimize choices. We developed a tone-frequency discrimination task for head-fixed mice in which we randomly presented either a long or short sound stimulus and biased the choice outcomes. The choice was less accurate and more biased toward the large-reward side in short- than in long-stimulus trials. Analysis with SDT found that mice did not use a separate, optimal choice threshold in different sound durations. Instead, mice updated one threshold for short and long stimuli with a simple reinforcement-learning rule. Our task in head-fixed mice helps understanding how the brain integrates sensory inputs and prior.

Decision Making through Integration of Sensory Evidence at Prolonged Timescales.

Project description:When multiple pieces of information bear on a decision, the best approach is to combine the evidence provided by each one. Evidence integration models formalize the computations underlying this process [1-3], explain human perceptual discrimination behavior [4-9], and correspond to neuronal responses elicited by discrimination tasks [10-14]. These findings suggest that evidence integration is key to understanding the neural basis of decision making [15-18]. But while evidence integration has most often been studied with simple tasks that limit deliberation to relatively brief periods, many natural decisions unfold over much longer durations. Neural network models imply acute limitations on the timescale of evidence integration [19-23], and it is currently unknown whether existing computational insights can generalize beyond rapid judgments. Here, we introduce a new psychophysical task and report model-based analyses of human behavior that demonstrate evidence integration at long timescales. Our task requires probabilistic inference using brief samples of visual evidence that are separated in time by long and unpredictable gaps. We show through several quantitative assays how decision making can approximate a normative integration process that extends over tens of seconds without accruing significant memory leak or noise. These results support the generalization of evidence integration models to a broader class of behaviors while posing new challenges for models of how these computations are implemented in biological networks.

Dissociating sensory from decision processes in human perceptual decision making.

Project description:A key question within systems neuroscience is how the brain translates physical stimulation into a behavioral response: perceptual decision making. To answer this question, it is important to dissociate the neural activity underlying the encoding of sensory information from the activity underlying the subsequent temporal integration into a decision variable. Here, we adopted a decoding approach to empirically assess this dissociation in human magnetoencephalography recordings. We used a functional localizer to identify the neural signature that reflects sensory-specific processes, and subsequently traced this signature while subjects were engaged in a perceptual decision making task. Our results revealed a temporal dissociation in which sensory processing was limited to an early time window and consistent with occipital areas, whereas decision-related processing became increasingly pronounced over time, and involved parietal and frontal areas. We found that the sensory processing accurately reflected the physical stimulus, irrespective of the eventual decision. Moreover, the sensory representation was stable and maintained over time when it was required for a subsequent decision, but unstable and variable over time when it was task-irrelevant. In contrast, decision-related activity displayed long-lasting sustained components. Together, our approach dissects neuro-anatomically and functionally distinct contributions to perceptual decisions.

Sensory and decision-making processes underlying perceptual adaptation.

Project description:Perceptual systems adapt to their inputs. As a result, prolonged exposure to particular stimuli alters judgments about subsequent stimuli. This phenomenon is commonly assumed to be sensory in origin. Changes in the decision-making process, however, may also be a component of adaptation. Here, we quantify sensory and decision-making contributions to adaptation in a facial expression paradigm. As expected, exposure to happy or sad expressions shifts the psychometric function toward the adaptor. More surprisingly, response times show both an overall decline and an asymmetry, with faster responses opposite the adapting category, implicating a substantial change in the decision-making process. Specifically, we infer that sensory changes from adaptation are accompanied by changes in how much sensory information is accumulated for the two choices. We speculate that adaptation influences implicit expectations about the stimuli one will encounter, causing modifications in the decision-making process as part of a normative response to a change in context.

Differentiating between integration and non-integration strategies in perceptual decision making.

Project description:Many tasks used to study decision-making encourage subjects to integrate evidence over time. Such tasks are useful to understand how the brain operates on multiple samples of information over prolonged timescales, but only if subjects actually integrate evidence to form their decisions. We explored the behavioral observations that corroborate evidence-integration in a number of task-designs. Several commonly accepted signs of integration were also predicted by non-integration strategies. Furthermore, an integration model could fit data generated by non-integration models. We identified the features of non-integration models that allowed them to mimic integration and used these insights to design a motion discrimination task that disentangled the models. In human subjects performing the task, we falsified a non-integration strategy in each and confirmed prolonged integration in all but one subject. The findings illustrate the difficulty of identifying a decision-maker's strategy and support solutions to achieve this goal.

Decoupling sensory from decisional choice biases in perceptual decision making.

Project description:The contribution of sensory and decisional processes to perceptual decision making is still unclear, even in simple perceptual tasks. When decision makers need to select an action from a set of balanced alternatives, any tendency to choose one alternative more often-choice bias-is consistent with a bias in the sensory evidence, but also with a preference to select that alternative independently of the sensory evidence. To decouple sensory from decisional biases, here we asked humans to perform a simple perceptual discrimination task with two symmetric alternatives under two different task instructions. The instructions varied the response mapping between perception and the category of the alternatives. We found that from 32 participants, 30 exhibited sensory biases and 15 decisional biases. The decisional biases were consistent with a criterion change in a simple signal detection theory model. Perceptual decision making, thus, even in simple scenarios, is affected by sensory and decisional choice biases.

Mesoscale cortical dynamics reflect the interaction of sensory evidence and temporal expectation during perceptual decision-making.

Project description:How sensory evidence is transformed across multiple brain regions to influence behavior remains poorly understood. We trained mice in a visual change detection task designed to separate the covert antecedents of choices from activity associated with their execution. Wide-field calcium imaging across the dorsal cortex revealed fundamentally different dynamics of activity underlying these processes. Although signals related to execution of choice were widespread, fluctuations in sensory evidence in the absence of overt motor responses triggered a confined activity cascade, beginning with transient modulation of visual cortex and followed by sustained recruitment of the secondary and primary motor cortex. Activation of the motor cortex by sensory evidence was modulated by animals' expectation of when the stimulus was likely to change. These results reveal distinct activation timescales of specific cortical areas by sensory evidence during decision-making and show that recruitment of the motor cortex depends on the interaction of sensory evidence and temporal expectation.

The Dilution Effect and Information Integration in Perceptual Decision Making.

Project description:In cognitive science there is a seeming paradox: On the one hand, studies of human judgment and decision making have repeatedly shown that people systematically violate optimal behavior when integrating information from multiple sources. On the other hand, optimal models, often Bayesian, have been successful at accounting for information integration in fields such as categorization, memory, and perception. This apparent conflict could be due, in part, to different materials and designs that lead to differences in the nature of processing. Stimuli that require controlled integration of information, such as the quantitative or linguistic information (commonly found in judgment studies), may lead to suboptimal performance. In contrast, perceptual stimuli may lend themselves to automatic processing, resulting in integration that is closer to optimal. We tested this hypothesis with an experiment in which participants categorized faces based on resemblance to a family patriarch. The amount of evidence contained in the top and bottom halves of each test face was independently manipulated. These data allow us to investigate a canonical example of sub-optimal information integration from the judgment and decision making literature, the dilution effect. Splitting the top and bottom halves of a face, a manipulation meant to encourage controlled integration of information, produced farther from optimal behavior and larger dilution effects. The Multi-component Information Accumulation model, a hybrid optimal/averaging model of information integration, successfully accounts for key accuracy, response time, and dilution effects.

Limitation of switching sensory information flow in flexible perceptual decision making.

Project description:Humans can flexibly change rules to categorize sensory stimuli, but their performance degrades immediately after a task switch. This switch cost is believed to reflect a limitation in cognitive control, although the bottlenecks remain controversial. Here, we show that humans exhibit a brief reduction in the efficiency of using sensory inputs to form a decision after a rule change. Participants classified face stimuli based on one of two rules, switching every few trials. Psychophysical reverse correlation and computational modeling reveal a reduction in sensory weighting, which recovers within a few hundred milliseconds after stimulus presentation. This reduction depends on the sensory features being switched, suggesting a constraint in routing the sensory information flow. We propose that decision-making circuits cannot fully adjust their sensory readout based on a context cue alone, but require the presence of an actual stimulus to tune it, leading to a limitation in flexible perceptual decision making.

Dynamic integration of reward and stimulus information in perceptual decision-making.

Project description:In perceptual decision-making, ideal decision-makers should bias their choices toward alternatives associated with larger rewards, and the extent of the bias should decrease as stimulus sensitivity increases. When responses must be made at different times after stimulus onset, stimulus sensitivity grows with time from zero to a final asymptotic level. Are decision makers able to produce responses that are more biased if they are made soon after stimulus onset, but less biased if they are made after more evidence has been accumulated? If so, how close to optimal can they come in doing this, and how might their performance be achieved mechanistically? We report an experiment in which the payoff for each alternative is indicated before stimulus onset. Processing time is controlled by a "go" cue occurring at different times post stimulus onset, requiring a response within msec. Reward bias does start high when processing time is short and decreases as sensitivity increases, leveling off at a non-zero value. However, the degree of bias is sub-optimal for shorter processing times. We present a mechanistic account of participants' performance within the framework of the leaky competing accumulator model [1], in which accumulators for each alternative accumulate noisy information subject to leakage and mutual inhibition. The leveling off of accuracy is attributed to mutual inhibition between the accumulators, allowing the accumulator that gathers the most evidence early in a trial to suppress the alternative. Three ways reward might affect decision making in this framework are considered. One of the three, in which reward affects the starting point of the evidence accumulation process, is consistent with the qualitative pattern of the observed reward bias effect, while the other two are not. Incorporating this assumption into the leaky competing accumulator model, we are able to provide close quantitative fits to individual participant data.
