Project description:Engaging executive function often requires overriding a prepotent response in favor of a conflicting but adaptive one. Language may play a key role in this ability by supporting integrated representations of conflicting rules. We tested whether experience with contrastive language that could support such representations benefits executive function in 3-year-old children. Children who received brief experience with language highlighting contrast between objects, attributes, and actions showed greater executive function on two of three 'conflict' executive function tasks than children who received experience with contrasting stimuli only and children who read storybooks with the experimenter, controlling for baseline executive function. Experience with contrasting stimuli did not benefit executive function relative to reading books with the experimenter, indicating experience with contrastive language, rather than experience with contrast generally, was key. Experience with contrastive language also boosted spontaneous attention to contrast, consistent with improvements in representing contrast. These findings indicate a role for language in executive function that is consistent with the Cognitive Complexity and Control theory's key claim that coordinating conflicting rules is critical to overcoming perseveration, and suggest new ideas for testing theories of executive function.

Project description:The present study examined whether processing words with affective connotations in a listener's native language may be modulated by accented speech. To address this question, we used the Event Related Potential (ERP) technique and recorded the cerebral activity of Spanish native listeners, who performed a semantic categorization task, while listening to positive, negative and neutral words produced in standard Spanish or in four foreign accents. The behavioral results yielded longer latencies for emotional than for neutral words in both native and foreign-accented speech, with no difference between positive and negative words. The electrophysiological results replicated previous findings from the emotional language literature, with the amplitude of the Late Positive Complex (LPC), associated with emotional language processing, being larger (more positive) for emotional than for neutral words at posterior scalp sites. Interestingly, foreign-accented speech was found to interfere with the processing of positive valence and go along with a negativity bias, possibly suggesting heightened attention to negative words. The manipulation employed in the present study provides an interesting perspective on the effects of accented speech on processing affective-laden information. It shows that higher order semantic processes that involve emotion-related aspects are sensitive to a speaker's accent.

Project description:The large majority of humankind is more or less fluent in 2 or even more languages. This raises the fundamental question how the language network in the brain is organized such that the correct target language is selected at a particular occasion. Here we present behavioral and functional magnetic resonance imaging data showing that bilingual processing leads to language conflict in the bilingual brain even when the bilinguals' task only required target language knowledge. This finding demonstrates that the bilingual brain cannot avoid language conflict, because words from the target and nontarget languages become automatically activated during reading. Importantly, stimulus-based language conflict was found in brain regions in the LIPC associated with phonological and semantic processing, whereas response-based language conflict was only found in the pre-supplementary motor area/anterior cingulate cortex when language conflict leads to response conflicts.

Project description:Knowing where north is provides a navigator with invaluable information for learning and recalling a space, particularly in places with limited navigational cues, like complex indoor environments. Although north is effectively used by orienteers, pilots, and military personnel, very little is known about whether nonexpert populations can or will use north to create an accurate representation of an indoor space. In the current study, we taught people 2 nonoverlapping routes through a complex indoor environment, with which they were not familiar-a university hospital with few windows and several turns. Along 1 route, they wore a vibrotactile compass on their arm, which vibrated continuously indicating the direction of north. Along the other route, they were only told where north was at the start of the route. At the beginning, the end, and back at the beginning of each route, participants pointed to well-known landmarks in the surrounding city and campus (external landmarks), and newly learned landmarks in the hospital (internal landmarks). We found improved performance with the compass only for external landmarks, driven by people's use of the availability of north to orient these judgments. No such improved orientation occurred for the internal landmarks. These findings reveal the utility of vibrotactile compasses for learning new indoor spaces. We speculate that such cues help users map new spaces onto familiar spaces or to familiar reference frames. (PsycINFO Database Record

Project description:When plant cells are under environmental stress, several chemically distinct reactive oxygen species (ROS) are generated simultaneously in various intracellular compartments and these can cause oxidative damage or act as signals. The conditional flu mutant of Arabidopsis, which generates singlet oxygen in plastids during a dark-to-light transition, has allowed the biological activity of singlet oxygen to be determined, and the criteria to distinguish between cytotoxicity and signalling of this particular ROS to be defined. The genetic basis of singlet-oxygen-mediated signalling has been revealed by the mutation of two nuclear genes encoding the plastid proteins EXECUTER (EX)1 and EX2, which are sufficient to abrogate singlet-oxygen-dependent stress responses. Conversely, responses due to higher cytotoxic levels of singlet oxygen are not suppressed in the ex1/ex2 background. Whether singlet oxygen levels lower than those that trigger genetically controlled cell death activate acclimation is now under investigation.

Project description:Telework has been promoted for decades as one of the traffic demand management policies to alleviate congestion during peak periods and reduce work-related trips, along with other benefits. However, less clear is the role played by life stages (i.e., gender, marital status and parenthood) on telework behavior. This study investigated to which extent telework frequency associated with life stages, and how these associations could be explained based on the work-life conflict perspective. Representative data were obtained from German Microcensus 2010 (N = 188,081 participants). The outcome variable was measured as ordered telework participation levels (i.e., never, infrequently and frequently). After testing for multicollinearity, a zero-inflated ordered probit regression model was applied to assess the associations between telework and family-life stages, while adjusting for individual, household, job-related and environmental characteristics. Results suggest that life stages associate with telework behavior in a complex way. Three patterns have been distinguished. Specifically, irrespective of gender and marital status, parents are less likely to telework compared to those without children. Regarding individuals without children, single individuals are more likely to telework than married ones, and males more likely than females. In contrast, for individuals with children, the partnered parents are more likely to telework than single parents, and females more likely than males. Our findings suggest that as the most important feature in family-life stages, children play a vital role in telework behavior. It not only increases both work-to-family conflict and family-to-work conflict, but also triggers housework re-division within couples and aggravates gender differences. Policies that support formal childcare resources could relieve the family-to-work conflict and encourage people to work at home.

Project description:The "costly signaling" hypothesis proposes that animal signals are kept honest by appropriate signal costs. We show that to the contrary, signal cost is unnecessary for honest signaling even when interests conflict. We illustrate this principle by constructing examples of cost-free signaling equilibria for the two paradigmatic signaling games of Grafen (1990) and Godfray (1991). Our findings may explain why some animal signals use cost to ensure honesty whereas others do not and suggest that empirical tests of the signaling hypothesis should focus not on equilibrium cost but, rather, on the cost of deviation from equilibrium. We use these results to apply costly signaling theory to the low-cost signals that make up human language. Recent game theoretic models have shown that several key features of language could plausibly arise and be maintained by natural selection when individuals have coincident interests. In real societies, however, individuals do not have fully coincident interests. We show that coincident interests are not a prerequisite for linguistic communication, and find that many of the results derived previously can be expected also under more realistic models of society.

Project description:Genomic DNA in the eukaryotic cell nucleus is present in the form of chromatin. Histones are the principal protein component of chromatin, and their post-translational modifications play important roles in regulating the structure and function of chromatin and thereby in determining cell development and disease. An understanding of how histone modifications translate into downstream cellular events is important from both developmental and therapeutic perspectives. However, biochemical studies of histone modifications require access to quantities of homogenously modified histones that cannot be easily isolated from natural sources or generated by enzymatic methods. In the past decade, chemical synthesis has proven to be a powerful tool in translating the language of histone modifications by providing access to uniformly modified histones and by the development of stable analogues of thermodynamically labile modifications. This Review highlights the various synthetic and semisynthetic strategies that have enabled biochemical and biophysical characterization of site-specifically modified histones.

Project description:Ubiquitin (Ub) is a small 76 amino acid long protein that is highly conserved in all eukaryotes studied to date. In humans, more than 600 ligases are involved in the reversible modification of specific lysine side-chain amines in substrate proteins by conjugation with the C-terminal carboxylate of Ub. Initially monoubiquitylated proteins can undergo repetitive ubiquitylation starting from one of seven lysine residues or the α-amine in the first Ub to generate a variety of polyUb chains with different topologies and functions. The most well known role for protein ubiquitylation is in targeting substrates for proteolytic destruction by 26S proteasomes. However, a growing body of evidence indicates that both mono- and polyubiquitylation play proteasome-independent roles in modulating the structure, function, and localization of protein substrates. Understanding the complexity of Ub-mediated functions in our cells is a major challenge for modern biology. In addition to well-established in vivo genetic methods, biochemical and biophysical investigations of ubiquitylated proteins in vitro can shed light on the direct mechanistic roles for Ub in different contexts. Such studies have traditionally been limited by the ability to obtain sufficient quantities of homogenously ubiquitylated proteins with precisely defined linkages. This review focuses on recent advances in both synthetic and recombinant protein-based methods that have yielded access to homogenously site-specifically ubiquitylated proteins. Mechanistic studies of the roles for protein ubiquitylation and of the enzymes involved in protein deubiquitylation that are enabled by these chemical tools are highlighted.

Project description:All natural languages have formal devices for communicating about number, be they lexical (e.g., two, many) or grammatical (e.g., plural markings on nouns and/or verbs). Here we ask whether linguistic devices for number arise in communication systems that have not been handed down from generation to generation. We examined deaf individuals who had not been exposed to a usable model of conventional language (signed or spoken), but had nevertheless developed their own gestures, called homesigns, to communicate. Study 1 examined four adult homesigners and a hearing communication partner for each homesigner. The adult homesigners produced two main types of number gestures: gestures that enumerated sets (cardinal number marking), and gestures that signaled one vs. more than one (non-cardinal number marking). Both types of gestures resembled, in form and function, number signs in established sign languages and, as such, were fully integrated into each homesigner's gesture system and, in this sense, linguistic. The number gestures produced by the homesigners' hearing communication partners displayed some, but not all, of the homesigners' linguistic patterns. To better understand the origins of the patterns displayed by the adult homesigners, Study 2 examined a child homesigner and his hearing mother, and found that the child's number gestures displayed all of the properties found in the adult homesigners' gestures, but his mother's gestures did not. The findings suggest that number gestures and their linguistic use can appear relatively early in homesign development, and that hearing communication partners are not likely to be the source of homesigners' linguistic expressions of non-cardinal number. Linguistic devices for number thus appear to be so fundamental to language that they can arise in the absence of conventional linguistic input.
