Denitrification performance of Pseudomonas fluorescens Z03 immobilized by graphene oxide-modified polyvinyl-alcohol and sodium alginate gel beads at low temperature.

Project description:Improving the effect of microbial denitrification under low-temperature conditions has been a popular focus of research in recent years. In this study, graphene oxide (GO)-modified polyvinyl-alcohol (PVA) and sodium alginate (SA) (GO/PVA-SA) gel beads were used as a heterotrophic nitrification-aerobic denitrification (HN-AD) bacteria (Pseudomonas fluorescens Z03) carrier to enhance nitrogen removal efficiency levels at low temperatures (6-8°C). The removal efficiency of NH4 +-N and NO3 --N and the variations in concentrations of extracellular polymeric substances (EPS) under different GO doses (0.03-0.15 g l-1) were studied. The results indicated that the addition of GO can improve the efficiency of nitrogen removal, and the highest removal efficiency level and highest carbohydrate, protein, and total EPS content levels (50.28 mg, 132.78 mg and 183.06 mg (g GO/PVA-SA gel)-1, respectively) were obtained with 0.15 g l-1 GO. The simplified Monod model accurately predicted the nitrogen removal efficiency level. These findings suggested that the application of GO serves as an effective means to enhance nitrogen removal by stimulating the activity of HN-AD bacteria.

Phosphorylated Curdlan Gel/Polyvinyl Alcohol Electrospun Nanofibres Loaded with Clove Oil with Antibacterial Activity.

Project description:Fibrous membranes based on natural polymers obtained by the electrospinning technique are a great choice for wound dressings. In order to promote an efficient wound repair, and to avoid antibiotics, antibacterial plant extracts can be incorporated. In the present work, the new electrospun nanofibre membranes based on monobasic phosphate curdlan (PCurd) and polyvinyl alcohol (PVA) were obtained for the first time. To establish the adequate mixing ratio for electrospinning, the behaviour of the PCurd and PVA mixture was studied by viscometry and rheology. In order to confer antimicrobial activity with the nanofibre membrane, clove essential oil (CEO) was incorporated into the electrospun solution. Well-defined and drop-free nanofibres with a diameter between 157 nm and 110 nm were obtained. The presence of CEO in the obtained nanofibres was confirmed by ATR-FTIR spectroscopy, by the phenolic and flavonoid contents, and by the antioxidant activity of the membranes. In physiological conditions, CEO was released from the membrane after 24 h. The in vivo antimicrobial tests showed a good inhibitory activity against E. coli and higher activity against S. aureus. Furthermore, the viability cell test showed the lack of cytotoxicity of the nanofibre membrane with and without CEO, confirming its potential use in wound treatment.

Preparation and bacteriostatic research of porous polyvinyl alcohol / biochar / nanosilver polymer gel for drinking water treatment.

Project description:Microbial contamination in drinking water has become an important threat to human health. There is thus an urgent need to develop antibacterial materials to treat drinking water. Here, porous silver-loaded biochar (C-Ag) was prepared using corn straw as the substrate and silver as the antibacterial agent. C-Ag was then uniformly distributed in polyvinyl alcohol gel beads of eluted calcium carbonate to prepare p-PVA/C-Ag antibacterial composite. The polymer composites were tested by FT-IR, XRD, SEM and TG-DSC. The results showed that C-Ag was more evenly distributed in the PVA gel spheres. Antibacterial experiments showed that p-PVA/C-Ag greatly inhibited Escherichia coli. Practical application tests revealed that p-PVA/C-Ag showed high and sustained bactericidal inhibition and reusability. Generally, p-PVA/C-Ag composite shows high potential to be applied to drinking water treatment.

Long-Term Antibacterial Effect of Electrospun Polyvinyl Alcohol/Polyacrylate Sodium Nanofiber Containing Nisin-Loaded Nanoparticles.

Project description:Response Surface Methodology (RSM) was used to assess the optimal conditions for a Water/Oil/Water (W/O/W) emulsion for encapsulated nisin (EN). Nano-encapsulated nisin had high encapsulation efficiencies (EE) (86.66 ± 1.59%), small particle size (320 ± 20 nm), and low polydispersity index (0.27). Biodegradable polyvinyl alcohol (PVA) and polyacrylate sodium (PAAS) were blended with EN and prepared by electrospinning. Scanning electron microscopy (SEM) revealed PVA/PAAS/EN nanofibers with good morphology, and that their EN activity and mechanical properties were enhanced. When the ultrasonication time was 15 min and 15% EN was added, the nanofibers had optimal mechanical, light transmittance, and barrier properties. Besides, the release behavior of nisin from the nanofibers fit the Korsemeyer-Peppas (KP) model, a maximum nisin release rate of 85.28 ± 2.38% was achieved over 16 days. At 4 °C, the growth of Escherichia coli and Staphylococcus aureus was inhibited for 16 days in nanofibers under different ultrasonic times. The application of the fiber in food packaging can effectively inhibit the activity of food microorganisms and prolong the shelf life of strawberries, displaying a great potential application for food preservation.

Polyvinyl Alcohol/Chitosan and Polyvinyl Alcohol/Ag@MOF Bilayer Hydrogel for Tissue Engineering Applications.

Project description:In this paper, polyvinyl alcohol/Ag-Metal-organic framework (PVA/Ag@MOF) and polyvinyl alcohol/chitosan (PVA/CS) were used as the inner and outer layers to successfully prepare a bilayer composite hydrogel for tissue engineering scaffold. The performance of bilayer hydrogels was evaluated. The outer layer (PVA/CS) has a uniform pore size distribution, good water retention, biocompatibility and cell adhesion ability. The inner layer (PVA/Ag@MOF) has good antibacterial activity and poor biocompatibility. PVA, PVA/0.1%Ag@MOF, PVA/0.5%Ag@MOF, and PVA/1.0%Ag@MOF show anti-microbial activity in ascending order. However, its use as an inner layer avoids direct contact with cells and prevents infection. The cell viability of all samples was above 90%, indicating that the bilayer hydrogel was non-toxic to A549 cells. The bilayer hydrogel scaffold combines the advantages of the inner and outer layers. In summary, this new bilayer composite is an ideal lung scaffold for tissue engineering.

Development of a polyvinyl alcohol/sodium alginate hydrogel-based scaffold incorporating bFGF-encapsulated microspheres for accelerated wound healing.

Project description:In the present study, a hybrid microsphere/hydrogel system, consisting of polyvinyl alcohol (PVA)/sodium alginate (SA) hydrogel incorporating PCL microspheres is introduced as a skin scaffold to accelerate wound healing. The hydrogel substrate was developed using the freeze-thawing method, and the proportion of the involved polymers in its structure was optimized based on the in-vitro assessments. The bFGF-encapsulated PCL microspheres were also fabricated utilizing the double-emulsion solvent evaporation technique. The achieved freeze-dried hybrid system was then characterized by in-vitro and in-vivo experiments. The results obtained from the optimization of the hydrogel showed that increasing the concentration of SA resulted in a more porous structure, and higher swelling ability, elasticity and degradation rate, but decreased the maximum strength and elongation at break. The embedding of PCL microspheres into the optimized hydrogel structure provided sustained and burst-free release kinetics of bFGF. Besides, the addition of drug-loaded microspheres led to no significant change in the degradation mechanism of the hydrogel substrate; however, it reduced its mechanical strength. Furthermore, the MTT assay represented no cytotoxic effect for the hybrid system. The in-vivo studies on a burn-wound rat model, including the evaluation of the wound closure mechanism, and histological analyses indicated that the fabricated scaffold efficiently contributed to promoting cell-induced tissue regeneration and burn-wound healing.
