Paleocene Ipomoea (Convolvulaceae) from India with implications for an East Gondwana origin of Convolvulaceae.

ABSTRACT: The morning glory family, Convolvulaceae, is globally important in medicine and food crops. The family has worldwide distribution in a variety of habitats; however, its fossil record is very poorly documented. The current fossil record suggests an origin in North America, which is in contrast to molecular data that indicate an East Gondwana origin. We report Ipomoea leaves from the late Paleocene (Thanetian; 58.7-55.8 million years ago) of India, which was a part of East Gondwana during this time. This is the earliest fossil record for both the family Convolvulaceae and the order Solanales. This suggests that the sister families Convolvulaceae and Solanaceae diverged before the Eocene in Gondwana-derived continents. The evidence presented here supports the conclusion from molecular phylogenetic analysis of an East Gondwana origin of Convolvulaceae.

Project description:A monograph of the 425 New World species of Ipomoea is presented. All 425 species are described and information is provided on their ecology and distribution, with citations from all countries from which they are reported. Notes are provided on salient characteristics and taxonomic issues related to individual species. A full synonymy is provided and 272 names are lectotypified. An extensive introduction discusses the delimitation and history of Ipomoea arguing that a broad generic concept is the only rational solution in the light of recent phylogenetic advances. Although no formal infrageneric classification is proposed, attention is drawn to the major clades of the genus and several morphologically well-defined clades are discussed including those traditionally treated under the names Arborescens, Batatas, Pharbitis, Calonyction and Quamoclit, sometimes as distinct genera, subgenera, sections or series. Identification keys are provided on a regional basis including multi-entry keys for the main continental blocks. Six species are described as new, Ipomoea nivea J.R.I. Project description:BackgroundIpomoea aquatica (Convolvulaceae), an aquatic edible plant, is traditionally used against heavy metal toxicity in India. The current study intended to explore the protective role of edible (aqueous) extract of I. aquatica (AEIA) against experimentally induced Pb-intoxication.MethodsThe cytoprotective role of AEIA was measured on mouse hepatocytes by cell viability assay followed by Hoechst staining and flow cytometric assay. The effect on ROS production, lipid peroxidation, protein carbonylation, intracellular redox status were measured after incubating the hepatocytes with Pb-acetate (6.8 ?M) along with AEIA (400 ?g/ml). The effects on the expressions of apoptotic signal proteins were estimated by western blotting. The protective role of AEIA was measured by in vivo assay in mice. Haematological, serum biochemical, tissue redox status, Pb bioaccumulation and histological parameters were evaluated to estimate the protective role of AEIA (100 mg/kg) against Pb-acetate (5 mg/kg) intoxication.ResultsPb-acetate treated hepatocytes showed a gradual reduction of cell viability dose-dependently with an IC50 value of 6.8 ?M. Pb-acetate treated hepatocytes exhibited significantly enhanced levels (p < 0.01) of ROS production, lipid peroxidation, protein carbonylation with concomitant depletion (p < 0.01) of antioxidant enzymes and GSH. However, AEIA treatment could significantly restore the aforementioned parameters in murine hepatocytes near to normalcy. Besides, AEIA significantly reversed (p < 0.05-0.01) the alterations of transcription levels of apoptotic proteins viz. Bcl 2, Bad, Cyt C, Apaf-1, cleaved caspases [caspase 3, caspase 8 and caspase 9], Fas and Bid. In in vivo bioassay, Pb-acetate treatment caused significantly high intracellular Pb burden and oxidative pressure in the kidney, liver, heart, brain and testes in mice. In addition, the haematological and serum biochemical factors were changed significantly in Pb-acetate-treated animals. AEIA treatment restored significantly the evaluated-parameters to the near-normal position.ConclusionThe extract may offer the protective effect via counteracting with Pb mediated oxidative stress and/or promoting the elimination of Pb by chelating. The presence of substantial quantities of flavonoids, phenolics and saponins would be responsible for the overall protective effect.

Project description:The latex of Ipomoea (Convolvulaceae) is a source of a special kind of acylsugars called resin glycosides, which are highly appreciated because of their biological activities (i.e. laxative, antimicrobial, cytotoxic etc.). Most research has been conducted in perennials with tuberous roots, where resin glycosides are stored. However, their content and variation are unknown in annual vines that lack this type of root, such as in the case of Ipomoea parasitica. This species contains research/biological and human value through its fast growth, survival in harsh environments, and employment in humans for mental/cognitive improvements. These qualities make I. parasitica an ideal system to profile resin glycosides and their variations in response to edaphoclimate. Topsoil samples (0-30 cm depth) and latex from petioles of I. parasitica were collected in two localities of central Mexico. The latex was analyzed through UHPLC-ESI-QTOF, and soil physico-chemical characteristics, the rainfall, minimum, average, and maximum temperatures were recorded. We also measured canopy (%), rockiness (%), and plant cover (%). A Principal Component Analysis was conducted to find associations between edaphoclimate and the resin glycosides. Forty-four resin glycosides were found in the latex of I. parasitica. Ten correlated significantly with three components (47.07%) and contained tetrasaccharide, pentasaccharide, and dimers of tetrasaccharide units. Five resin glycosides were considered constitutive because they were in all the plants. However, exclusive molecules to each locality were also present, which we hypothesize is in response to significant microhabitat conditions found in this study (temperature, clay content, pH, and potassium). Our results showed the presence of resin glycosides in I. parasitica latex and are the basis for experimentally testing the effect of the conditions above on these molecules. However, ecological, molecular, and biochemical factors should be considered in experiments designed to produce these complex molecules.
