A Visual-Cue-Dependent Memory Circuit for Place Navigation.

ABSTRACT: The ability to remember and to navigate to safe places is necessary for survival. Place navigation is known to involve medial entorhinal cortex (MEC)-hippocampal connections. However, learning-dependent changes in neuronal activity in the distinct circuits remain unknown. Here, by using optic fiber photometry in freely behaving mice, we discovered the experience-dependent induction of a persistent-task-associated (PTA) activity. This PTA activity critically depends on learned visual cues and builds up selectively in the MEC layer II-dentate gyrus, but not in the MEC layer III-CA1 pathway, and its optogenetic suppression disrupts navigation to the target location. The findings suggest that the visual system, the MEC layer II, and the dentate gyrus are essential hubs of a memory circuit for visually guided navigation.

Neuronal connectome of a sensory-motor circuit for visual navigation.

Project description:Animals use spatial differences in environmental light levels for visual navigation; however, how light inputs are translated into coordinated motor outputs remains poorly understood. Here we reconstruct the neuronal connectome of a four-eye visual circuit in the larva of the annelid Platynereis using serial-section transmission electron microscopy. In this 71-neuron circuit, photoreceptors connect via three layers of interneurons to motorneurons, which innervate trunk muscles. By combining eye ablations with behavioral experiments, we show that the circuit compares light on either side of the body and stimulates body bending upon left-right light imbalance during visual phototaxis. We also identified an interneuron motif that enhances sensitivity to different light intensity contrasts. The Platynereis eye circuit has the hallmarks of a visual system, including spatial light detection and contrast modulation, illustrating how image-forming eyes may have evolved via intermediate stages contrasting only a light and a dark field during a simple visual task.

Nucleus accumbens local circuit for cue-dependent aversive learning.

Project description:Response to threatening environmental stimuli requires detection and encoding of important environmental features that dictate threat. Aversive events are highly salient, which promotes associative learning about stimuli that signal this threat. The nucleus accumbens is uniquely positioned to process this salient, aversive information and promote motivated output, through plasticity on the major projection neurons in the brain area. We describe a nucleus accumbens core local circuit whereby excitatory plasticity facilitates learning and recall of discrete aversive cues. We demonstrate that putative nucleus accumbens substance P release and long-term excitatory plasticity on dopamine 2 receptor-expressing projection neurons are required for cue-dependent fear learning. Additionally, we find that fear learning and recall is dependent on distinct projection neuron subtypes. Our work demonstrates a critical role for nucleus accumbens substance P in cue-dependent aversive learning.

Lateralized human hippocampal activity predicts navigation based on sequence or place memory.

Project description:The hippocampus is crucial for both spatial navigation and episodic memory, suggesting that it provides a common function to both. Here we adapt a spatial paradigm, developed for rodents, for use with functional MRI in humans to show that activation of the right hippocampus predicts the use of an allocentric spatial representation, and activation of the left hippocampus predicts the use of a sequential egocentric representation. Both representations can be identified in hippocampal activity before their effect on behavior at subsequent choice-points. Our results suggest that, rather than providing a single common function, the two hippocampi provide complementary representations for navigation, concerning places on the right and temporal sequences on the left, both of which likely contribute to different aspects of episodic memory.

Project description:During spatial navigation, animals use self-motion to estimate positions through path integration. However, estimation errors accumulate over time and it is unclear how they are corrected. Here we report a new cell class ('cue cell') encoding visual cues that could be used to correct errors in path integration in mouse medial entorhinal cortex (MEC). During virtual navigation, individual cue cells exhibited firing fields only near visual cues and their population response formed sequences repeated at each cue. These cells consistently responded to cues across multiple environments. On a track with cues on left and right sides, most cue cells only responded to cues on one side. During navigation in a real arena, they showed spatially stable activity and accounted for 32% of unidentified, spatially stable MEC cells. These cue cell properties demonstrate that the MEC contains a code representing spatial landmarks, which could be important for error correction during path integration.

Cue combination in goal-oriented navigation.

Project description:This study examined cue combination of self-motion and landmark cues in goal-localisation. In an immersive virtual environment, before walking a two-leg path, participants learned the locations of three goal objects (one at the path origin, that is, home) and landmarks. After walking the path without seeing landmarks or goals, participants indicated the locations of the home and non-home goals in four conditions: (1) path integration only, (2) landmarks only, (3) both path integration and the landmarks, and (4) path integration and rotated landmarks. The ratio of the length between the testing position (P) and the turning point (T) over the length between the T and the three goals (G) (i.e., PT/TG) was manipulated. The results showed the cue combination consistently for participants' heading estimates but not for goal-localisation. In Experiments 1 and 2 (using distal landmarks), the cue combination for goal estimates appeared in a small length ratio (PT/TG = 0.5) but disappeared in a large length ratio (PT/TG = 2). In Experiments 3 and 4 (using proximal landmarks), while the cue combination disappeared for the home with a medium length ratio (PT/TG = 1), it appeared for the non-home goal with a large length ratio (PT/TG = 2) and only disappeared with a very large length ratio (PT/TG = 3). These findings are explained by a model stipulating that cue combination occurs in self-localisation (e.g., heading estimates), which leads to one estimate of the goal location; proximal landmarks produce another goal location estimate; these two goal estimates are then combined, which may only occur for non-home goals.

Carbon dioxide and blood-feeding shift visual cue tracking during navigation in <i>Aedes aegypti</i> mosquitoes.

Project description:Haematophagous mosquitoes need a blood meal to complete their reproductive cycle. To accomplish this, female mosquitoes seek vertebrate hosts, land on them and bite. As their eggs mature, they shift attention away from hosts and towards finding sites to lay eggs. We asked whether females were more tuned to visual cues when a host-related signal, carbon dioxide, was present, and further examined the effect of a blood meal, which shifts behaviour to ovipositing. Using a custom, tethered-flight arena that records wing stroke changes while displaying visual cues, we found the presence of carbon dioxide enhances visual attention towards discrete stimuli and improves contrast sensitivity for host-seeking Aedes aegypti mosquitoes. Conversely, intake of a blood meal reverses vertical bar tracking, a stimulus that non-fed females readily follow. This switch in behaviour suggests that having a blood meal modulates visual attention in mosquitoes, a phenomenon that has been described before in olfaction but not in visually driven behaviours.

Swap errors in visual working memory are fully explained by cue-feature variability.

Project description:In cue-based recall from working memory, incorrectly reporting features of an uncued item may be referred to as a "swap" error. One account of these errors ascribes them to variability in memory for the cue features leading to erroneous selection of a non-target item, especially if it is similar to the target in the cue-feature dimension. However, alternative accounts of swap errors include cue-independent misbinding, and strategic guessing when the cued item is not in memory. Here we investigated the cause of swap errors by manipulating the variability with which either cue or report features (orientations in Exp 1; motion directions in Exp 2) were encoded. We found that swap errors increased with increasing variability in memory for the cue features, and their changing frequency could be quantitatively predicted based on recall variability when the same feature was used for report. These results are inconsistent with the hypothesis that swaps are a strategic response to forgotten items, and suggest that swap errors could be wholly accounted for by confusions due to cue-dimension variability. In a third experiment we examined whether spatial configuration of memory arrays in tasks with spatial cueing has an influence on swap error frequency. We observed a specific tendency to make swap errors to non-targets located precisely opposite to the cued location, suggesting that stimulus positions are partially encoded in a non-metric format.

Behavior-dependent directional tuning in the human visual-navigation network.

Project description:The brain derives cognitive maps from sensory experience that guide memory formation and behavior. Despite extensive efforts, it still remains unclear how the underlying population activity unfolds during spatial navigation and how it relates to memory performance. To examine these processes, we combined 7T-fMRI with a kernel-based encoding model of virtual navigation to map world-centered directional tuning across the human cortex. First, we present an in-depth analysis of directional tuning in visual, retrosplenial, parahippocampal and medial temporal cortices. Second, we show that tuning strength, width and topology of this directional code during memory-guided navigation depend on successful encoding of the environment. Finally, we show that participants' locomotory state influences this tuning in sensory and mnemonic regions such as the hippocampus. We demonstrate a direct link between neural population tuning and human cognition, where high-level memory processing interacts with network-wide visuospatial coding in the service of behavior.

Medial entorhinal cortex lesions only partially disrupt hippocampal place cells and hippocampus-dependent place memory.

Project description:The entorhinal cortex provides the primary cortical projections to the hippocampus, a brain structure critical for memory. However, it remains unclear how the precise firing patterns of medial entorhinal cortex (MEC) cells influence hippocampal physiology and hippocampus-dependent behavior. We found that complete bilateral lesions of the MEC resulted in a lower proportion of active hippocampal cells. The remaining active cells had place fields, but with decreased spatial precision and decreased long-term spatial stability. In addition, MEC rats were as impaired in the water maze as hippocampus rats, while rats with combined MEC and hippocampal lesions had an even greater deficit. However, MEC rats were not impaired on other hippocampus-dependent tasks, including those in which an object location or context was remembered. Thus, the MEC is not necessary for all types of spatial coding or for all types of hippocampus-dependent memory, but it is necessary for the normal acquisition of place memory.

Sustained attention required for effective dimension-based retro-cue benefit in visual working memory.

Project description:In visual working memory (VWM) tasks, participants' performances can be improved through the use of dimension-based retro-cues, which direct internal attention to prioritize a particular dimension (e.g., color or orientation) of VWM representations even after the stimuli disappear. This phenomenon is known as the dimension-based retro-cue benefit (RCB). The present study investigates whether sustained attention is required for the dimension-based RCB by inserting interference or interruption between the retro-cue and the test array to distract attention. We tested the effects of perceptual interference or cognitive interruption on dimension-based RCB when the interference (Experiments 1 and 2 with masks) or interruption (Experiments 3 and 4 with an odd-even task) occurred concurrently with the stages for the maintenance of prioritized information (long cue-and-interference/interruption interstimulus interval, e.g., Experiments 1 and 3) or the deployment of attention (short cue-and-interference/interruption interstimulus interval, e.g., Experiments 2 and 4). Our results demonstrate that perceptual interference or cognitive interruption attenuates the dimension-based RCB. These findings suggest that sustained attention is necessary for the effective prioritization of a specific dimension of VWM representations.
