Project description:BACKGROUND:Episodic memory (EM) declines with age and the rate of decline is variable across individuals. A single nucleotide polymorphism (rs17070145) in the WWC1 gene that encodes the KIBRA protein critical for long-term potentiation and memory consolidation has previously been associated with EM performance, as well as differences in hippocampal engagement during EM tasks using functional magnetic resonance imaging (fMRI). In the current study, we explore the effect of this polymorphism on EM-related activity and cognitive performance across the adult life span using fMRI. METHODS:Two hundred thirty-two healthy, Caucasian subjects (18-89 years) completed a battery of cognitive tests, as well as an EM task during an fMRI scan. RESULTS:WWC1 T carriers had significantly better delayed recall performance than CC individuals (p = .006). The relationship between increasing age and recall scores (immediate and delayed) was also significantly different between WWC1 genotype groups (p = .01). In addition to the age-related decline in hippocampal formation (HF) activation (p < .05; false discovery ratesmall volume correction-HF-region of interest), we observed an age by WWC1 genotype interaction on HF activation during encoding and retrieval. The CC group showed a significant negative association between HF activity and increasing age, while no such association was observed in the T carrier group (left HF p = .04; r-z correlation difference during encoding and retrieval; right HF p = .0008; r-z correlation difference during retrieval). CONCLUSIONS:Our results show a dynamic relationship between rs17070145 polymorphism and increasing age on neuronal activity in the hippocampal region.

Project description:The present research explores the effects of contextualized material on age-related working memory performance. Two experiments examining younger and older adults are reported. ANOVA results of the first experiment showed age effects in both a standard operation span and a contextual task of parallel structure (air travel task). The second experiment also revealed a significant age effect in a standard operation span task. However, there was no age difference in a contextual task providing additional visual context (rail travel task), mainly due to older adults being unaffected by task type manipulation and younger adults performing worse in the contextual than in the standard task. The present research suggests that contextual task material may not necessarily lead to improved working memory performance in older adults. Several methodological and conceptual conclusions for future research are discussed.

Project description:Executive function (EF) refers to a set of cognitive functions that support goal-directed behaviors. Recent findings have suggested that the frontoparietal network (FPN) subserves neural processes that are related to EF. However, the FPN structural and functional network properties that mediate age-related differences in EF components remain unclear. To this end, we used three experimental tasks to test the component processes of EF based on Miyake and Friedman's model: one common EF component process (incorporating inhibition, shifting, and updating) and two specific EF component processes (shifting and updating). We recruited 126 healthy participants (65 females; 20 to 78 years old) who underwent both structural and functional MRI scanning. We tested a mediation path model of three structural and functional properties of the FPN (i.e., gray matter volume, white matter fractional anisotropy, and intra/internetwork functional connectivity) as mediators of age-related differences in the three EF components. The results indicated that age-related common EF component differences are mediated by regional gray matter volume changes in both hemispheres of the frontal lobe, which suggests that structural changes in the frontal lobe may have an indirect influence on age-related general elements of EF. These findings suggest that the FPN mediates age-related differences in specific components of EF.

Project description:The ability to remember the details of our own experiences declines gradually as we get old. The reason for this decay has been attributed to several factors besides age, such as education, nutrient intake and health status. However, the influence of these factors has mainly been examined individually and rarely together. Here we identify those factors that jointly act as mediators of episodic memory decay across the adult life span. We examined source memory in a lifespan sample of 1557 healthy adults. A total of 70 physical, biological and lifestyle variables were measured and introduced into a structural equation model as potential mediators that intervene between age and source memory. Only 14 mediator variables reliably mediated source memory decay; notably, eight of these variables have an effect on the cardiovascular system. The model unequivocally highlights that the mediators that may impair cardiovascular functioning also impact brain resources involved in episodic memory. We identified the factors that are relevant to episodic memory decline when they interact together as occurs in real life.

Project description:Typical cognitive aging may be defined as age-associated changes in cognitive performance in individuals who remain free of dementia. Ideally, the full adult age spectrum should be included to assess brain imaging findings associated with typical aging.To compare age, sex, and APOE ?4 effects on memory, brain structure (adjusted hippocampal volume [HVa]), and amyloid positron emission tomography (PET) in cognitively normal individuals aged 30 to 95 years old.Cross-sectional observational study (March 2006 to October 2014) at an academic medical center. We studied 1246 cognitively normal individuals, including 1209 participants aged 50 to 95 years old enrolled in a population-based study of cognitive aging and 37 self-selected volunteers aged 30 to 49 years old.Memory, HVa, and amyloid PET.Overall, memory worsened from age 30 years through the 90s. The HVa worsened gradually from age 30 years to the mid-60s and more steeply beyond that age. The median amyloid PET was low until age 70 years and increased thereafter. Memory was worse in men than in women overall (P <?.001) and more specifically beyond age 40 years. The HVa was lower in men than in women overall (P <?.001) and more specifically beyond age 60 years. There was no sex difference in amyloid PET at any age. Within each sex, memory performance and HVa were not different by APOE ?4 status at any age. From age 70 years onward, APOE ?4 carriers had significantly greater median amyloid PET than noncarriers. However, the ages at which 10% of the population were amyloid PET positive were 57 years for APOE ?4 carriers and 64 years for noncarriers.Male sex is associated with worse memory and HVa among cognitively normal individuals, while APOE ?4 is not. In contrast, APOE ?4 is associated with greater amyloid PET (from age 70 years onward), while sex is not. Worsening memory and HVa occur at earlier ages than abnormal amyloid PET. Therefore, neuropathological processes other than ?-amyloidosis must underlie declines in brain structure and memory function in middle age. Our findings are consistent with a model of late-onset Alzheimer disease in which ?-amyloidosis arises in later life on a background of preexisting structural and cognitive decline that is associated with aging and not with ?-amyloid deposits.

Project description:It is well established that brain structures and cognitive functions change across the life span. A long-standing hypothesis called "age differentiation" additionally posits that the relations between cognitive functions also change with age. To date, however, evidence for age-related differentiation is mixed, and no study has examined differentiation of the relationship between brain and cognition. Here we use multigroup structural equation models (SEMs) and SEM trees to study differences within and between brain and cognition across the adult life span (18-88 years) in a large (N > 646, closely matched across sexes), population-derived sample of healthy human adults from the Cambridge Centre for Ageing and Neuroscience (www.cam-can.org). After factor analyses of gray matter volume (from T1- and T2-weighted MRI) and white matter organization (fractional anisotropy from diffusion-weighted MRI), we found evidence for the differentiation of gray and white matter, such that the covariance between brain factors decreased with age. However, we found no evidence for age differentiation among fluid intelligence, language, and memory, suggesting a relatively stable covariance pattern among cognitive factors. Finally, we observed a specific pattern of age differentiation between brain and cognitive factors, such that a white matter factor, which loaded most strongly on the hippocampal cingulum, became less correlated with memory performance in later life. These patterns are compatible with the reorganization of cognitive functions in the face of neural decline, and/or with the emergence of specific subpopulations in old age.SIGNIFICANCE STATEMENT The theory of age differentiation posits age-related changes in the relationships among cognitive domains, either weakening (differentiation) or strengthening (dedifferentiation), but evidence for this hypothesis is mixed. Using age-varying covariance models in a large cross-sectional adult life span sample, we found age-related reductions in the covariance among both brain measures (neural differentiation), but no covariance change among cognitive factors of fluid intelligence, language, and memory. We also observed evidence of uncoupling (differentiation) between a white matter factor and cognitive factors in older age, most strongly for memory. Together, our findings support age-related differentiation as a complex, multifaceted pattern that differs for brain and cognition, and discuss several mechanisms that might explain the changing relationship between brain and cognition.

Project description:Working memory (WM) is the ability to maintain and manipulate task-relevant information in the absence of sensory input. While its improvement through training is of great interest, the degree to which WM training transfers to untrained WM tasks (near transfer) and other untrained cognitive skills (far transfer) remains debated and the mechanism(s) underlying transfer are unclear. Here we hypothesized that a critical feature of dual n-back training is its reliance on maintaining relational information in WM. In Experiment 1, using an individual differences approach, we found evidence that performance on an n-back task was predicted by performance on a measure of relational WM (i.e., WM for vertical spatial relationships independent of absolute spatial locations); whereas the same was not true for a complex span WM task. In Experiment 2, we tested the idea that reliance on relational WM is critical to produce transfer from n-back but not complex span task training. Participants completed adaptive training on either a dual n-back task, a symmetry span task, or on a non-WM active control task. We found evidence of near transfer for the dual n-back group; however, far transfer to a measure of fluid intelligence did not emerge. Recording EEG during a separate WM transfer task, we examined group-specific, training-related changes in alpha power, which are proposed to be sensitive to WM demands and top-down modulation of WM. Results indicated that the dual n-back group showed significantly greater frontal alpha power after training compared to before training, more so than both other groups. However, we found no evidence of improvement on measures of relational WM for the dual n-back group, suggesting that near transfer may not be dependent on relational WM. These results suggest that dual n-back and complex span task training may differ in their effectiveness to elicit near transfer as well as in the underlying neural changes they facilitate.

Project description:Although research has identified age-related changes in neural recruitment during emotional memory encoding, it is unclear whether these differences extend to retrieval. In this study, participants engaged in a recognition task during a functional magnetic resonance imaging scan. They viewed neutral titles and indicated whether each title had been presented with an image during the study phase. Neural activity and connectivity during retrieval of titles associated with positive and negative images were compared with age (treated as a continuous variable) included as a regressor of interest. Aging was associated with increased prefrontal activation for retrieval of positive and negative memories, but this pattern was more widespread for negative memories. Aging also was associated with greater negative connectivity between a left hippocampal seed region and multiple regions of prefrontal cortex, but this effect of age occurred during negative retrieval only. These findings demonstrate that age-related changes in prefrontal recruitment and connectivity during retrieval depend on memory valence. The use of a life span approach also emphasized both continuities and discontinuities in recruitment and connectivity across the adult life span, highlighting the insights to be gained from using a full life span sample.

Project description:The aim of the study was to identify nutrients that have the ability to impact brain functioning and, as a consequence, influence episodic memory. In particular, we examined recollection, the ability to recall details of previous experiences, which is the episodic memory process most affected as age advances. A sample of 1,550 healthy participants between 21 and 80 years old participated in the study. Nutritional intake was examined through a food frequency questionnaire and software developed to determine the daily consumption of 64 nutrients based on food intake during the last year. Recollection was measured through a computerized source memory paradigm. First, we identified which nutrients influence recollection across the entire adult life span. Then, moderator analyses were conducted by dividing the sample into young (21-40 years old), middle-aged (41-60 years old) and older (61-80 years old) adults to establish in which life stage nutrients influence episodic memory. Across the adult life span, recollection accuracy was shown to benefit from the intake of sodium, heme, vitamin E, niacin, vitamin B6, cholesterol, alcohol, fat, protein, and palmitic, stearic, palmitoleic, oleic, gadoleic, alpha-linoleic and linoleic acid. The effects of energy, maltose, lactose, calcium and several saturated fatty acids on recollection were modulated by age; in older adults, the consumption of these nutrients negatively influenced episodic memory performance, and in middle-aged adults, only lactose had negative effects. Several brain mechanisms that support episodic memory were influenced by specific nutrients, demonstrating the ability of food to enhance or deteriorate episodic memory.

Project description:BackgroundA number of large-scale population studies have provided valuable information about physical performance in aged individuals; however, there is little information about trajectories of function and associations with age across the adult life span. We developed a mobility-focused physical performance screener designed to be appropriate for the adult life span.MethodsThe physical performance battery includes measures of mobility, strength, endurance, and balance. Physical activity (PA) was assessed with accelerometry. We examined age-related trends in physical performance and PA, and the relationship between physical performance and PA across the age range (30-90+), by decade, in 775 participants enrolled in the study 2012-2014.ResultsPhysical performance was worse with increasing age decade. Although men performed better than women across all ages, the decrement by age group was similar between genders. Worsening physical performance was observed as early as the fifth decade for chair stands and balance and in the sixth decade for gait speed and aerobic endurance. The number and strength of significant associations between physical performance and PA increased with greater age: the greatest number of significant associations was seen in the 60-79 age groups, with fewer reported in the 30-59 and 80-90+ age groups. More PA was associated with better physical function.ConclusionThese results emphasize the importance of a life span approach to studies of function and aging. This work points to the need for a physical performance screener that spans across adulthood as a clinical tool for identifying functional decline.
