Project description:Background:Leaching of nutrients from agricultural areas is the main cause of water pollution and eutrophication of the Baltic Sea. A variety of remedial actions to reduce nitrogen and phosphorus losses from agricultural holdings and cultivated fields have been taken in the past. However, knowledge about the risk of nutrient leaching has not yet reached many farmers operating in the water catchment area of the Baltic Sea. Methods:The nutrient balance method known as "At the farm gate" involves calculating separate balances for nitrogen (N), phosphorus (P) and potassium (K). After estimating all the components of the nutrient balance, the total balance for NPK is calculated and the data obtained is expressed as the ratio of total change (surplus) to the area of arable land on a farm. In addition, the nutrient usage efficiency on a farm is also calculated. An opinion poll was conducted in 2017 on 3.6% (n = 31) of the farms located in commune of Puck. The total area of the farms including arable and grass land ranged from 5 to 130 ha with an average of 45.82 ha. The arable land was on average 30.79 ha ranging from 4.45 to 130 ha while the grassland averaged 12.77 ha and ranged from 0 to 53 ha. Results:The average consumption of mineral fertilizer in the sample population of farms was 114.9 kg N, 9.3 kg P, and 22.9 kg K·ha-1of agricultural land (AL), respectively. N balance in the sample farms being ranged from -23.3 to 254.5 kg N·ha-1AL while nutrient use efficiency ranged from 0.40% to 231.3%. In comparison, P surplus in the sample farms was 5.0 kg P·ha-1AL with the P use efficiency of 0.4-266.5%. Discussion:Mean N fertilizer consumption in the tested farms was higher than the average usage across Poland and in the Pomeranian Voivodeship. However, mean consumption of potassium fertilizers was lower than mentioned averages. Mean P fertilizer consumption was higher than in the Pomeranian Voivodeship, but lower compared to the entire country. Generally, on the basis of designated research indicators of farm pressures on water quality, concentrations of total nitrogen and total phosphorus were obtained. CalcGosPuck (an integrated agriculture calculator) will help to raise farmers' awareness about NPK flow on farm scale and to improve nutrient management.

Project description:The ADPs were designed in response to a fall in agricultural productivity and hence a concern to sustain domestic food supplies. The study examined "Financing Agriculture in Nigeria through Agricultural Extension Services of Agricultural Development Programmes." It sought to ascertain the extent to which agricultural extension services of the agricultural development programmes have impacted the financing of agriculture in six selected local government areas in Edo South senatorial district, Nigeria using a sample of 120 respondents. Stratified random sampling was used to select the respondents. Interview schedule served as the research instrument. The research data were analyzed using t-test and Pearson correlation, which served as the inferential statistics. The research findings showed that the extension services of ADP have impacted significantly on crop development in the selected communities but have not had significant impact on employment creation and the development of infrastructural facilities. The study also revealed that there was no significant difference between the implementation of the projects in the selected communities, as revealed by the correlation test. On the basis of the research findings, the need for a complete redesign of the project to ensure that it achieves its stated goals as well as ensure proper monitoring of its implementation were suggested, among others.

Project description:Tandem mass spectrometry (MS/MS) is frequently used in the identification of peptides and proteins. Typical proteomic experiments rely on algorithms such as SEQUEST and MASCOT to compare thousands of tandem mass spectra against the theoretical fragment ion spectra of peptides in a database. The probabilities that these spectrum-to-sequence assignments are correct can be determined by statistical software such as PeptideProphet or through estimations based on reverse or decoy databases. However, many of the software applications that assign probabilities for MS/MS spectra to sequence matches were developed using training data sets from 3D ion-trap mass spectrometers. Given the variety of types of mass spectrometers that have become commercially available over the last 5 years, we sought to generate a data set of reference data covering multiple instrumentation platforms to facilitate both the refinement of existing computational approaches and the development of novel software tools. We analyzed the proteolytic peptides in a mixture of tryptic digests of 18 proteins, named the "ISB standard protein mix", using 8 different mass spectrometers. These include linear and 3D ion traps, two quadrupole time-of-flight platforms (qq-TOF), and two MALDI-TOF-TOF platforms. The resulting data set, which has been named the Standard Protein Mix Database, consists of over 1.1 million spectra in 150+ replicate runs on the mass spectrometers. The data were inspected for quality of separation and searched using SEQUEST. All data, including the native raw instrument and mzXML formats and the PeptideProphet validated peptide assignments, are available at http://regis-web.systemsbiology.net/PublicDatasets/.

Project description:Agribusiness has a great relevance in the world׳s economy. It generates a considerable impact in the gross national product of several nations. Hence, it is the major driver of many national economies. Nowadays, from each new planting to harvesting process it is mandatory and crucial to apply some kind of technology to optimize a given singular process, or even the entire cropping chain. For instance, digital image analysis joined with machine learning methods can be applied to obtain and guarantee a higher quality of the harvest, leading to not only a greater profit for producers, but also better products with lower cost to the final consumers. Thus, to provide this possibility this work describes a visual feature dataset from soybean seed images obtained from the tetrazolium test. This is a test capable to define how healthy a given seed is (e.g. how much the plant will produce, or if it is resistant to inclement weather, among others). To answer these questions we proposed this dataset which is the cornerstone to provide an effective classification of the soybean seed vigor (i.e. an extremely tiresome human visual inspection process). Besides, as one of the most prominent international commodity, the soybean production must follow rigid quality control process to be part of world trade. Hence, small mistakes in the seed vigor definition of a given seed lot can lead to huge losses.

Project description:Improved versions of inositol-1,4,5-trisphosphate (InsP3) sensors were created to follow intracellular InsP3 changes in single living cells and in cell populations. Similar to previous InsP3 sensors the new sensors are based on the ligand binding domain of the human type-I InsP3 receptor (InsP3R-LBD), but contain a mutation of either R265K or R269K to lower their InsP3 binding affinity. Tagging the InsP3R-LBD with N-terminal Cerulean and C-terminal Venus allowed measurement of InsP3 in single-cell FRET experiments. Replacing Cerulean with a Luciferase enzyme allowed experiments in multi-cell format by measuring the change in the BRET signal upon stimulation. These sensors faithfully followed the agonist-induced increase in InsP3 concentration in HEK 293T cells expressing the Gq-coupled AT1 angiotensin receptor detecting a response to agonist concentration as low as 10 pmol/L. Compared to the wild type InsP3 sensor, the mutant sensors showed an improved off-rate, enabling a more rapid and complete return of the signal to the resting value of InsP3 after termination of M3 muscarinic receptor stimulation by atropine. For parallel measurements of intracellular InsP3 and Ca2+ levels in BRET experiments, the Cameleon D3 Ca2+ sensor was modified by replacing its CFP with luciferase. In these experiments depletion of plasma membrane PtdIns(4,5)P2 resulted in the fall of InsP3 level, followed by the decrease of the Ca2+-signal evoked by the stimulation of the AT1 receptor. In contrast, when type-III PI 4-kinases were inhibited with a high concentration of wortmannin or a more specific inhibitor, A1, the decrease of the Ca2+-signal preceded the fall of InsP3 level indicating an InsP3-, independent, direct regulation of capacitative Ca2+ influx by plasma membrane inositol lipids. Taken together, our results indicate that the improved InsP3 sensor can be used to monitor both the increase and decrease of InsP3 levels in live cells suitable for high-throughput BRET applications.

Project description:AimConservation conflict takes place where food production imposes a cost on wildlife conservation and vice versa. Where does conservation impose the maximum cost on production, by opposing the intensification and expansion of farmland? Where does conservation confer the maximum benefit on wildlife, by buffering and connecting protected areas with a habitable and permeable matrix of crop and non-crop habitat? Our aim was to map the costs and benefits of conservation versus production and thus to propose a conceptual framework for systematic conservation planning in agricultural landscapes.LocationWorld-wide.MethodsTo quantify these costs and benefits, we used a geographic information system to sample the cropland of the world and map the proportion of non-crop habitat surrounding the cropland, the number of threatened vertebrates with potential to live in or move through the matrix and the yield gap of the cropland. We defined the potential for different types of conservation conflict in terms of interactions between habitat and yield (potential for expansion, intensification, both or neither). We used spatial scan statistics to find 'hotspots' of conservation conflict.ResultsAll of the 'hottest' hotspots of conservation conflict were in sub-Saharan Africa, which could have impacts on sustainable intensification in this region.Main conclusionsSystematic conservation planning could and should be used to identify hotspots of conservation conflict in agricultural landscapes, at multiple scales. The debate between 'land sharing' (extensive agriculture that is wildlife friendly) and 'land sparing' (intensive agriculture that is less wildlife friendly but also less extensive) could be resolved if sharing and sparing were used as different types of tool for resolving different types of conservation conflict (buffering and connecting protected areas by maintaining matrix quality, in different types of matrix). Therefore, both sharing and sparing should be prioritized in hotspots of conflict, in the context of countryside biogeography.

Project description:ObjectiveTo evaluate the need for survey mode adjustments to hospital care evaluations by discharged inpatients and develop the appropriate adjustments.Data sourceA total of 7,555 respondents from a 2006 national random sample of 45 hospitals who completed the CAHPS Hospital (HCAHPS [Hospital Consumer Assessments of Healthcare Providers and Systems]) Survey.Study design/data collection/extraction methodsWe estimated mode effects in linear models that predicted each HCAHPS outcome from hospital-fixed effects and patient-mix adjustors.Principal findingsPatients randomized to the telephone and active interactive voice response (IVR) modes provided more positive evaluations than patients randomized to mail and mixed (mail with telephone follow-up) modes, with some effects equivalent to more than 30 percentile points in hospital rankings. Mode effects are consistent across hospitals and are generally larger than total patient-mix effects. Patient-mix adjustment accounts for any nonresponse bias that could have been addressed through weighting.ConclusionsValid comparisons of hospital performance require that reported hospital scores be adjusted for survey mode and patient mix.

Project description:Women's empowerment is a process that includes increases in intrinsic agency (power within); instrumental agency (power to); and collective agency (power with). We used baseline data from two studies-Targeting and Realigning Agriculture for Improved Nutrition (TRAIN) in Bangladesh and Building Resilience in Burkina Faso (BRB)-to assess the measurement properties of survey questions operationalizing selected dimensions of intrinsic, instrumental, and collective agency in the project-level Women's Empowerment in Agricultural Index (pro-WEAI). We applied unidimensional item-response models to question (item) sets to assess their measurement properties, and when possible, their cross-context measurement equivalence-a requirement of measures designed for cross-group comparisons. For intrinsic agency in the right to bodily integrity, measured with five attitudinal questions about intimate partner violence (IPV) against women, model assumptions of unidimensionality and local independence were met. Four items showed good model fit and measurement equivalence across TRAIN and BRB. For item sets designed to capture autonomy in income, intrinsic agency in livelihoods activities, and instrumental agency in: livelihoods activities, the sale or use of outputs, the use of income, and borrowing from financial services, model assumptions were not met, model fit was poor, and items generally were weakly related to the latent (unobserved) agency construct. For intrinsic and instrumental agency in livelihoods activities and for instrumental agency in the sale or use of outputs and in the use of income, items sets had similar precision along the latent-agency continuum, suggesting that similar item sets could be dropped without a loss of precision. IRT models for collective agency were not estimable because of low reported presence and membership in community groups. This analysis demonstrates the use of IRT methods to assess the measurement properties of item sets in pro-WEAI, and empowerment scales generally. Findings suggest that a shorter version of pro-WEAI can be developed that will improve its measurement properties. We recommend revisions to the pro-WEAI questionnaire and call for new measures of women's collective agency.

Project description:Sub-Saharan Africa (SSA) faces climate change and food insecurity challenges, which require action to create resilient farming systems. Conservation agriculture (CA) is widely promoted across SSA but the impacts on key soil physical properties and functions such as soil structure and hydraulic properties that govern water storage and transmission are not well understood. The aim of this study was to assess the impacts of long term (10-12 years) maize-based CA on soil hydraulic conductivity, water retention and pore size distribution. Root zone (0-30 cm depth) soil total porosity, pore size distribution, saturated hydraulic conductivity (Ksat) and plant available water capacity (PAWC) of conventional maize monocrop farming systems (CP) are compared with those of adjacent CA trials with either sole maize or maize intercrop/rotation with cowpea (Vigna unguiculata L.), pigeon pea (Cajanus cajan L.) or velvet bean (Mucuna pruriens L) in trial locations across central and southern Malawi. Results show that maize-based CA systems result in significant changes to soil hydraulic properties that correlate with improved soil structure. Results demonstrate increases of 5-15 % in total porosity, 0.06-0.22 cm/min in Ksat, 3-7 % in fine pores for water storage and 3-6 % in PAWC. Maize monocrop CA had similar effect on the hydraulic properties as the maize-legume associations. The values of Ksat for CA systems were within optimum levels (0.03-0.3 cm/min) whereas PAWC was below optimum (<20 %). There was no significant build-up in soil organic matter (OM) in the CA systems. The results lead to a recommendation that crop residue management should be more pro-actively pursued in CA guidance from agricultural extension staff to increase soil OM levels, increase yields and enhance climate resilience of sub-Saharan African farming systems.

Project description:Previous studies on the adoption and impacts of improved crop varieties have relied on self-reported adoption status of the surveyed households. However, in the presence of weak variety maintenance and poorly functioning seed certification system, measurement errors in self-reported adoption status can be considerable. This paper investigates how such measurement errors can lead to biased welfare estimates. Using DNA-fingerprinting based varietal identification as a benchmark, we find that misclassification in self-reported adoption status is considerable, with significant false negative and positive response rates. We empirically show that such measurement errors lead to welfare estimates that are biased towards zero and substantially understate the poverty reduction effects of adoption. While the empirical evidence suggests attenuation bias, our theoretical exposition and simulations demonstrate that upward bias and sign reversal effects are also possible. The results point to the need for improved monitoring of the diffusion process of improved varieties through innovative adoption data collection approaches to generate robust evidence for prioritizing and justifying investments in agricultural research and extension.
