Project description:Integration of information over the CNS is an important neural process that affects our ability to perceive and react to the environment. The visual system is required to continuously integrate information arriving from two different sources (the eyes) to create a coherent percept with high spatiotemporal precision. Although this neural integration of information is assumed to be critical for visual performance, it can be impaired under some pathological or developmental conditions. Here we took advantage of a unique developmental condition, amblyopia ("lazy eye"), which is characterized by an impaired temporal synchronization between the two eyes, to meticulously study the effect of synchronization on the integration of binocular visual information. We measured the eyes' asynchrony and compensated for it (with millisecond temporal resolution) by providing time-shifted stimuli to the eyes. We found that the re-synchronization of the ocular input elicited a significant improvement in visual functions, and binocular functions, such as binocular summation and stereopsis, were regained. This phenomenon was also evident in neurophysiological measures. Our results can shed light on other neural processing aspects and might also have translational relevance for the field of training, rehabilitation, and perceptual learning.

Project description:There is huge diversity in visual systems and color discrimination abilities, thought to stem from an animal's ecology and life history. Many primate species maintain a polymorphism in color vision, whereby most individuals are dichromats but some females are trichromats, implying that selection sometimes favors dichromatic vision. Detecting camouflaged prey is thought to be a task where dichromatic individuals could have an advantage. However, previous work either has not been able to disentangle camouflage detection from other ecological or social explanations, or did not use biologically relevant cryptic stimuli to test this hypothesis under controlled conditions. Here, we used online "citizen science" games to test how quickly humans could detect cryptic birds (incubating nightjars) and eggs (of nightjars, plovers and coursers) under trichromatic and simulated dichromatic viewing conditions. Trichromats had an overall advantage, although there were significant differences in performance between viewing conditions. When searching for consistently shaped and patterned adult nightjars, simulated dichromats were more heavily influenced by the degree of pattern difference than were trichromats, and were poorer at detecting prey with inferior pattern and luminance camouflage. When searching for clutches of eggs-which were more variable in appearance and shape than the adult nightjars-the simulated dichromats learnt to detect the clutches faster, but were less sensitive to subtle luminance differences. These results suggest there are substantial differences in the cues available under viewing conditions that simulate different receptor types, and that these interact with the scene in complex ways to affect camouflage breaking.

Project description:Amblyopia is the most common cause of monocular visual loss in children, affecting 1.3%-3.6% of children. Current treatments are effective in reducing the visual acuity deficit but many amblyopic individuals are left with residual visual acuity deficits, ocular motor abnormalities, deficient fine motor skills, and risk for recurrent amblyopia. Using a combination of psychophysical, electrophysiological, imaging, risk factor analysis, and fine motor skill assessment, the primary role of binocular dysfunction in the genesis of amblyopia and the constellation of visual and motor deficits that accompany the visual acuity deficit has been identified. These findings motivated us to evaluate a new, binocular approach to amblyopia treatment with the goals of reducing or eliminating residual and recurrent amblyopia and of improving the deficient ocular motor function and fine motor skills that accompany amblyopia.

Project description:A theory of the binocular system with asymmetric eyes (AEs) is developed in the framework of bicentric perspective projections. The AE accounts for the eyeball's global asymmetry produced by the foveal displacement from the posterior pole, the main source of the eye's optical aberrations, and the crystalline lens' tilt countering some of these aberrations. In this theory, the horopter curves, which specify retinal correspondence of binocular single vision, are conic sections resembling empirical horopters. This advances the classic model of empirical horopters as conic sections introduced in an ad hoc way by Ogle in 1932. In contrast to Ogle's theory, here, anatomically supported horopteric conics vary with the AEs' position in the visual plane of bifoveal fixations and their transformations are visualized in a computer simulation. Integrating horopteric conics with eye movements can help design algorithms for maintaining a stable perceptual world from visual information captured by a mobile robot's camera head. Further, this paper proposes a neurophysiologically meaningful definition for the eyes' primary position, a concept which has remained elusive despite its theoretical importance to oculomotor research. Finally, because the horopteric conic's shape is dependent on the AE's parameters, this theory allows for changes in retinal correspondence, which is usually considered preformed and stable.

Project description:The endogenous neurotransmitter acetylcholine (ACh) is known to affect the excitatory/inhibitory (E/I) balance of primate visual cortex, enhancing feedforward thalamocortical gain while suppressing corticocortical synapses. Recent advances in the study of the human visual system suggest that ACh is a likely component underlying interocular interactions. However, our understanding of its precise role in binocular processes is currently lacking. Here we use binocular rivalry as a probe of interocular dynamics to determine ACh's effects, via the acetylcholinesterase inhibitor (AChEI) donepezil, on the binocular visual system. A total of 23 subjects (13 male) completed two crossover experimental sessions where binocular rivalry measurements were obtained before and after taking either donepezil (5 mg) or a placebo (lactose) pill. We report that enhanced cholinergic potentiation attenuates perceptual suppression during binocular rivalry, reducing the overall rate of interocular competition while enhancing the visibility of superimposition mixed percepts. Considering recent evidence that perceptual suppression during binocular rivalry is causally modulated by the inhibitory neurotransmitter GABA, our results suggest that cholinergic activity counteracts the effect of GABA with regards to interocular dynamics and may modulate the inhibitory drive within the visual cortex.SIGNIFICANCE STATEMENT Our research demonstrates that the cholinergic system is implicated in modulating binocular interactions in the human visual cortex. Potentiating the transmission of acetylcholine (ACh) via the cholinergic drug donepezil reduces the extent to which the eyes compete for perceptual dominance when presented two separate, incongruent images.

Project description:Many species employ camouflage to disguise their true shape and avoid detection or recognition. Disruptive coloration is a form of camouflage in which high-contrast patterns obscure internal features or break up an animal's outline. In particular, edge enhancement creates illusory, or 'fake' depth edges within the animal's body. Disruptive coloration often co-occurs with background matching, and together, these strategies make it difficult for an observer to visually segment an animal from its background. However, stereoscopic vision could provide a critical advantage in the arms race between perception and camouflage: the depth information provided by binocular disparities reveals the true three-dimensional layout of a scene, and might, therefore, help an observer to overcome the effects of disruptive coloration. Human observers located snake targets embedded in leafy backgrounds. We analysed performance (response time) as a function of edge enhancement, illumination conditions and the availability of binocular depth cues. We confirm that edge enhancement contributes to effective camouflage: observers were slower to find snakes whose patterning contains 'fake' depth edges. Importantly, however, this effect disappeared when binocular depth cues were available. Illumination also affected detection: under directional illumination, where both the leaves and snake produced strong cast shadows, snake targets were localized more quickly than in scenes rendered under ambient illumination. In summary, we show that illusory depth edges, created via disruptive coloration, help to conceal targets from human observers. However, cast shadows and binocular depth information improve detection by providing information about the true three-dimensional structure of a scene. Importantly, the strong interaction between disparity and edge enhancement suggests that stereoscopic vision has a critical role in breaking camouflage, enabling the observer to overcome the disruptive effects of edge enhancement.

Project description:Establishing genotype-phenotype relationship is the key to understand the molecular mechanism of phenotypic adaptation. This initial step may be untangled by analyzing appropriate ancestral molecules, but it is a daunting task to recapitulate the evolution of non-additive (epistatic) interactions of amino acids and function of a protein separately. To adapt to the ultraviolet (UV)-free retinal environment, the short wavelength-sensitive (SWS1) visual pigment in human (human S1) switched from detecting UV to absorbing blue light during the last 90 million years. Mutagenesis experiments of the UV-sensitive pigment in the Boreoeutherian ancestor show that the blue-sensitivity was achieved by seven mutations. The experimental and quantum chemical analyses show that 4,008 of all 5,040 possible evolutionary trajectories are terminated prematurely by containing a dehydrated nonfunctional pigment. Phylogenetic analysis further suggests that human ancestors achieved the blue-sensitivity gradually and almost exclusively by epistasis. When the final stage of spectral tuning of human S1 was underway 45-30 million years ago, the middle and long wavelength-sensitive (MWS/LWS) pigments appeared and so-called trichromatic color vision was established by interprotein epistasis. The adaptive evolution of human S1 differs dramatically from orthologous pigments with a major mutational effect used in achieving blue-sensitivity in a fish and several mammalian species and in regaining UV vision in birds. These observations imply that the mechanisms of epistatic interactions must be understood by studying various orthologues in different species that have adapted to various ecological and physiological environments.

Project description:Despite the complexity of the visual world, humans rarely confuse variations in illumination, for example shadows, from variations in material properties, such as paint or stain. This ability to distinguish illumination from material edges is crucial for determining the spatial layout of objects and surfaces in natural scenes. In this study, we explore the role that color (chromatic) cues play in edge classification. We conducted a psychophysical experiment that required subjects to classify edges into illumination and material, in patches taken from images of natural scenes that either contained or did not contain color information. The edge images were of various sizes and were pre-classified into illumination and material, based on inspection of the edge in the context of the whole image from which the edge was extracted. Edge classification performance was found to be superior for the color compared to grayscale images, in keeping with color acting as a cue for edge classification. We defined machine observers sensitive to simple image properties and found that they too classified the edges better with color information, although they failed to capture the effect of image size observed in the psychophysical experiment. Our findings are consistent with previous work suggesting that color information facilitates the identification of material properties, transparency, shadows and the perception of shape-from-shading.

Project description:Experience-driven neuronal plasticity allows the brain to adapt its functional connectivity to recent sensory input. Here we use binocular rivalry, an experimental paradigm in which conflicting images are presented to the individual eyes, to demonstrate plasticity in the neuronal mechanisms that convert visual information from two separated retinas into single perceptual experiences. Perception during binocular rivalry tended to initially consist of alternations between exclusive representations of monocularly defined images, but upon prolonged exposure, mixture percepts became more prevalent. The completeness of suppression, reflected in the incidence of mixture percepts, plausibly reflects the strength of inhibition that likely plays a role in binocular rivalry. Recovery of exclusivity was possible but required highly specific binocular stimulation. Documenting the prerequisites for these observed changes in perceptual exclusivity, our experiments suggest experience-driven plasticity at interocular inhibitory synapses, driven by the correlated activity (and also the lack thereof) of neurons representing the conflicting stimuli. This form of plasticity is consistent with a previously proposed but largely untested anti-Hebbian learning mechanism for inhibitory synapses in vision. Our results implicate experience-driven plasticity as one governing principle in the neuronal organization of binocular vision.

Project description:PurposeThe current understanding of binocular processing is primarily derived from static spatial visual perception: this leaves the role of temporal information unclear. In this study, we addressed this gap by testing the effect of alternating flicker on binocular information processing in adults with abnormal binocular vision. Our goal was to determine which temporal frequency optimally balanced input from both eyes.MethodsWe took measurements in four groups of human adults: 10 normal adults with the individual's nondominant eye covered by a 2% neutral density filter (aged 25.60 ± 1.43 years, experiment 1), 9 nonamblyopic anisometropes (aged 24.33 ± 1.66 years, experiment 2), 7 amblyopes (aged 26.5 ± 1.64 years, experiment 3), and 7 treated amblyopes (aged 24 ± 3.21 years, experiment 4). The balance point (BP), where participants' two eyes are equally effective, was measured using a binocular orientation combination task at four spatial frequencies (SFs; 0.5-4 c/d) and five temporal frequencies (TFs; baseline and 4, 7, 10, and 15 Hz). Its log transformation |logBP| was taken into further analysis.ResultsWe observed clear U-shaped temporal tuning of the |logBP| for the entire range of TFs (that we measured: trough occurred at 7 Hz). This pattern occurred and was significant in all four groups (P < 0.001). In addition, the effect of SFs on |logBP| was significant in normal, amblyopic, and treated amblyopic groups (all P < 0.001) and was marginally significant in the nonamblyopic anisometropic group (P = 0.086).ConclusionsAlternating flicker around 7 Hz may be the optimal temporal frequency for balancing eyes in human adults with binocular imbalance.
